Conformation and Linkage Studies of Specific Oligosaccharides Related to H1N1, H5N1, and Human Flu for Developing the Second Tamiflu

  • The interaction between viral HA (hemagglutinin) and oligosaccharide of the host plays an important role in the infection and transmission of avian and human flu viruses. Until now, this interaction has been classified by sialyl(${\alpha}2-3$) or sialyl(${\alpha}2-6$) linkage specificity of oligosaccharide moieties for avian or human virus, respectively. In the case of H5N1 and newly mutated flu viruses, classification based on the linkage type does not correlate with human infection and human-to-human transmission of these viruses. It is newly suggested that flu infection and transmission to humans require high affinity binding to the extended conformation with long length sialyl(${\alpha}2-6$)galactose containing oligosaccharides. On the other hand, the avian flu virus requires folded conformation with sialyl(${\alpha}2-3$) or short length sialyl(${\alpha}2-6$) containing trisaccharides. This suggests a potential future direction for the development of new species-specific antiviral drugs to prevent and treat pandemic flu.

The Effect of Cimetidine, Ranitidine and Famotidine on the Immune Response in ICR Mice (마우스에 있어서 Cimetidine, Ranitidine 및 Famotidine이 면역반응에 미치는 영향)

  • Experiments were performed on mice to investigate the influences of cimetidine, ranitidine and famotidine on the immune response. Immune response were evaluated by antibody, Arthus reaction (Arthus), delayed type hypersensitivity (DTH), rosette forming cell (RFC), phagocyte activity and whit( blood cell (WBC) in mice, sensitized and challenged with sheep red blood cells (SRBC). The weight of liver, spleen and thymus were measured. Following results obtained in this experiment. 1) The administration of cimetidine as compared to normal group significantly decreased Arthus, Hemagglutinin titer (HA), RFC, DTH, WBC and phagocyte activity, but increased the activity of serum albumin. 2) The administration of ranitidine as compared to normal group decreased RFC and HA. 3) The administration of Famotidine as compared to normal group decreased DTH and RFC, and significantly decreased HA, Arthus and serum protein. 4) The administration of ranitidine and famotidine decreased more humoral immune response than cellular immune response, but the administration of cimetidine significantly decreased humoral and cellular immune response, WBC and phagocyte activity.

  • An attempt was carried out to purify the cell components of Bordetella bronchiseptica by the mild procedures. From the supernatant of sonic treated colls, the K-agglutinogen, hemagglutinogen and skin necrotizing factor were separated by a successive use of DEAE-cellulose column chromatography and ammonium sulfate precipitation. The specific activity of purified K-agglutinogen was demonstrated by a gel-diffusion test and was also found to be free from other biologically active substances of B. bronchiseptica: namely hemagglutinin, skin-necrotizing factor and O-antigen.

  • The object of thes research is to lucidate the clinical effects of Gami-Cheongshimyeonjayeum on the anti-allergic and gimmune response to rats and mice. The obtaind results of Gami-Cheongshimyeonjayeum administration are as follows : 1. The cytotoxic index increased slightly at over concentration $1{\times}10^{-2}/ml$, but it is thought that the cytotoxic effect show no significant value under the concentration $1{\times}10^{-3}/ml$. 2. Gami-Cheongshimyeonjayeum water extract seems to suppress the leakage volume of Evans blue to decrease the vascular permeability. 3. Gami-Cheongshimyeonjayeum show the inhibitory effect of footpad edema. 4. Protein leakage is inhibited by the administration of Gami-Cheongshimyeonjayeum. 5. The delayed type hypersensitivity to SRBC is suppressed at the 24hours after the administration of Gami-Cheongshimyeonjayeum on the experimental group 1, 2. 6. Hemolysin and hemagglutinin titer, and RFC increase to bulid up the immune function.

  • A lectin from the edible mushroom, Lentinus edodes, was purified through physiological saline extraction, ammonium sulfate fractionation and column chromatographies. On polyacrylamide gel electrophoresis, 0.05M fraction from hydroxyapatite column exhibited adjacent four sharp bands. The partially purified lectin agglutinated the erythrocytes of rabbit, mouse and rat, but not agglutinated human erythrocytes. The lectin's mitogenic effects were tested by its application to human and murine splenic lymphocytes. The results showed that the 0.05M fraction from hydroxyapatite was mitogenic, and the optimal dose of Lentinus edodes lectin was slightly lower than Con A by the culture with murine splenic and human peripheral lymphocytes. Meanwhile, its ability to agglutinate transformed cells was tested by its administration to continuous cell lines L1210 and HeLa cells. The leetin was found to be an agglutinin of tumor cell lines tested by L1210 and HeLa cells.

  • In order to investigate the effect of Mori Folium on immunologic control function, the various immune responses were studied. The results were obtained as follows : 1. The transformation of lymphocyte was increased significantly in all concentration of Mori Folium treated group as compared with negative and positive control. 2. In FACS analysis, the population of helper T cell and T cell were increased significantly in Mori Folium treated group as compared with control. 3. IL-12(p35) and INF-${\gamma}$ were upregulated in Mori Folium treated group 4. NO and carbon clearance were significantly increased in Mori Folium treated group as compared with control. 5. The hemagglutinin titer and hemolysin titer were increased but insignificantly in Mori Folium treated group as compared with control.

  • In order to investigate the effects of shipjuntaepotang (少陰人 十全大補湯) on immune response, the author performed this experimental study. Delayed type hypersensitivity (DTH) and rosette forming cells (RFC) for cell-mediated immune response, hemagglutinin (HA) titers, hemolysin (HL) titers, and carbon clearance for phagocytic function of MPS (mononuclear phagocyte system) were measured in ICR mice. The results were summarized as follows. 1. DTH in the experimental group was increased, as compared with the control group, with statistical significance. 2. RFC in the experimental group was increased, as compared with the control group, with statistical significance. 3. HA-titers were increased in the experimental group, as compared with the control group, with statistical significance. 4. HL-titers were increased in the experimental group, as compared with the control group, with statistical significance. 5. Carbon clearance was increased in the experimental group, as compared with the control group, with statistical significance. Through invivo experimental study in ICR mice, these findings suggest that shipjuntaepo-tang enhance both cellmediated and humoral immune responce.

In this experiments, effect of PP on sheep red bood cells (sRBC)-induced Arthus reaction (AR) were studied in ICR male mice. SRBC were challenged by i.p. injection two weeks after sensitization of Lp. injection of sRBC. Five days after the challenge of antigen, paw edema induced 3 hours after the last challenge by AR.   • Effects of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on sheep red blood cells (sRBC)-induced arthus reaction (AR) were studied in ICR male mice and determined the plaque forming cells (PFC) numbers and hemagglutinin (HA) titer. Two weeks after sensitization of i.p. injection of sRBC (4x10$^{8}$ cells), ICR male mice were challenged by i.p. injection of sRBC (2$\times{10}^8$ cells). Five days after the challenge of antigen, paw edema induced three hours after the last challenge by Arthus reaction. (omitted)

  • Prime editors (PEs), which are CRISPR-Cas9 nickase (H840A)-reverse transcriptase fusion proteins programmed with prime editing guide RNAs (pegRNAs), can not only edit bases but also install transversions, insertions, or deletions without both donor DNA and double-strand breaks at the target DNA. As the demand for in-locus tagging is increasing, to reflect gene expression dynamics influenced by endogenous genomic contexts, we demonstrated that PEs can be used to introduce the hemagglutinin (HA) epitope tag to a target gene locus, enabling molecular and biochemical studies using in-locus tagged plants. To promote genome-wide in-locus tagging, we also implemented a publicly available database that designs pegRNAs for in-locus tagging of all the Arabidopsis genes.
