Evaluation of concurrent immunizations with equine influenza virus and strangles vaccines

  • Despite regular vaccinations, equine influenza virus (EIV) and Streptococcus equi subsp. equi (strangles) are the cause of highly contagious respiratory infections in horses. Many recent studies have reported that the concurrent administration of two vaccines could simplify horse management and minimize veterinary expenses. However, there is little information available regarding the efficacy of concurrent vaccinations against EIV and strangles. In this study, we evaluated EIV-specific antibody responses following the single EIV vaccination with the recombinant viral-vectored EIV vaccine or concurrent vaccination with the EIV and inactivated strangles vaccines. Blood samples were collected at 1-, 2-, 4-, and 8 weeks post-immunization (wpi) from each group. EIV-specific antibodies were evaluated by enzyme-linked immunosorbent assay (ELISA) and hemagglutination inhibition (HAI) assay. Both single and concurrent vaccination showed similar levels of EIV-specific serum immunoglobulin g (IgG) at 1 and 2 wpi. However, at 4 to 8 wpi, the EIV-only vaccination group showed significantly higher serum IgG levels than those from the concurrently vaccinated group. The HAI titers showed similar trends as the ELISA data, except at 8 wpi when both groups presented HAI titers with no significant differences. These data demonstrate that the concurrent vaccination against EIV and strangles could compromise the humoral immune response to equine influenza between vaccination intervals, which suggests the use of the consecutive vaccination protocol for EIV and strangles rather than concurrent vaccination.

Effects of Methionine Diets on the Immunotoxicity of Ethanol in ICR Mice (생쥐에 있어서 에탄올의 면역독성에 미치는 메치오닌 식이의 영향)

  • Experiments were performed on mice to investigate the effect of methionine diets (MET) on the immunotoxicity of ethanol. ICR female mice were divided into 5 groups, Met (Basal (B)+0.19% methionine(M), B+1.71% M and B+5.13%W) and ethanol(4%) were administered ad libitum for 21 days. The mice were evaluated for changes in immune status as measured by antibody titer, Arthus reaction, delayed type hypersensitivity (DTH), rosette forming cell(RFC) and plaque forming cell (PFC) to sheep red blood cells (S-RBC). To investigate the change of the non-specific immune response, the number of leukocytes in peripheral blood and phagocyte activity were measured. The results were summarized as follows: (1) The weight ratios of spleen and thymus to body weight were significantly increased by the B+0.19% M, B+0.57% M and B+1.71% M groups in comparison with control group(B), but B+5.13% M group was significantly decreased. (2) Humoral immune responses were significantly increased by the B+0.19% M and B+0.57% M groups in comparison with control group, but B+5.13% M group was significantly decreased. (3) Cellular immune responses were significantly decreased by the B+1.71% M and B+5.13% M groups in comparison with control group. (4) Phagocyte activities were significantly increased by the B+0.19% M, B+0.57% M and B+1.71% M groups in comparison with control groups, but B+5.13% M group was significantly decreased. (5) The number of circulating leukocyte was significantly increased in the B+0.19% M and B+0.57% M groups in comparison with control group, but B+5.13% M group was significantly decreased.

Clinical Evaluation of Mycoplasma pneumoniae Antibody Titer and Laboratory Tests in Acute Mycoplasma pneumoniae Pneumonia (급성기 Mycoplasma 폐렴의 항체가와 검사소견에 관한 고찰)

  • Purpose : For evaluation of acute Mycoplasma pneumoniae(M. pneumoniae) pneumonia in children, we have studied the Mycoplasma indirect particle agglutination test, cold hemagglutinin test, ESR, CRP, and total white blood cell counts and it's differential count retrospectively. Methods : The total numbers of patients whom compatible with diagnostic criteria of acute M. pneumoniae peumonia were 56 cases from Jan. to Dec. 1997. The diagnostic criteria were 1) onset of fever(${\geq}38.0^{\circ}C$) and coughing were within 7 days, 2) rhonchi and/or role was audible on chest, 3) pneumonic infiltration on chest X-ray, and 4) M. pneumoniae indirect particle agglutination test titer was higher than 1:640, or initial titer was less than 1:640 but increased more than 4 folds after week. We classified the enrolled patients according to initial antibody titer, such as soup A(${\leq}1:640$) and group B(${\geq}1:320$). We compared group A and B by demographic findings, clinical symptoms and signs, and laboratory findings. Results : 1) The male and female sex ratio was 1:1.4, and average onset age was $5.8{\pm}2.96$ years. 2) The average body temperature on admission was $38.5{\pm}0.1^{\circ}C$ and productive coughing was noticed in 52 cases(93%). 3) The average total white cell counts were $10,470{\pm}877.0/mm^3$ in group B patients, which was significantly higher compared to $7,761{\pm}508.5/mm^3$ in group A(p<0.014). 4) The average value of ESR and CRP were within normal range in both group. 5) The most common site of pneumonic infiltration was right lower lobe of lung in both groups. 6) There were no correlation between antibody titer and cold hemagglutinine titer in patients and cold hemagglutination titer were less than 1:64 in 25 cases(45%). Conclusion : The clinical manifestations of pneumonia, findings of chest x-ray, and indirect particle agglutination test were useful on diagnosis of M. pneumoniae pnumonia onset within 7 days, but cold hemagglutinin test was a little diagnostic meaning.

Identification and molecular characterization of a rabbit hemorrhagic disease virus variant (KV0801) isolated in Korea

  • Rabbit hemorrhagic disease (RHD) is caused by RHD virus (RHDV) and is one of the most fatal diseases of rabbits. Acute death of rabbits occurred in a farm located in the Gyeonggi province of South Korea. The virus was isolated and confirmed as RHDV based on reverse transcription polymerase chain reaction and hemagglutination assay (HA), and the isolate was designated as KV0801. The nucleotide sequence of the complete VP60 gene of KV0801 was determined and the corresponding amino acid sequence was deduced. Molecular analysis showed that the KV0801 isolate can be classified as a pandemic antigenic variant strain, RHDVa. The VP60 nucleotide sequence and deduced amino acid homology between KV0801 and other Korean isolate, RHF89, which was isolated in 1988, were 92.1 and 94.3%, respectively. The pathogenicity of the KV0801 isolate at an HA titer ranging from 16,384 to 0.16 HA units was evaluated in five-month-old SFP rabbits. The rabbits inoculated with KV0801 isolate containing more than 1.63 HA units died within six days of inoculation. These results suggest that a highly pathogenic RHDVa is circulating in the rabbit populations of Korea.

Isolation of Getah virus from racehorse in Korea (국내(國內) 경주마(競走馬)에서의 Getah 바이러스 분리(分離) 동정(同定))

  • Getah virus is known as a causative agent of recognized febrile illness of horses characterized by fever, rash and edema. A serological survey indicated that hemagglutination inhibition antibody against Getah virus was detected in 34% of 464 racehorses from Korean Horse Affairs Association and 57% of 262 ponies from Cheju island, respectively. Several field strains of Getah virus isolated were from the racehorse that have been shown fever and febrile signs in 1989. The field isolates produced cytopathic effect in Vero, MA-104, BHK-21 cell cultures. Especially, they multiplied to the highest titer($10^6TCID_{50}/0.1ml$) in Vero cell cultures. When day-old mice were inoculated with field isolates by the intracerebral route, they showed a typical paralysis sign and died within seven days after inoculation. The guinea pig exhibited skin rash and edema, and died with neural signs after inoculation with the field isolates. In the cross neutralization test and indirect immunofuorescent assay, the field isolates were proved to be closely related to the Sakai strain of Getah virus antigenically.

Serum Parvovirus Antibody Titers among Healthy Adult Dogs in Korea (국내 성견의 개 파보바이러스에 대한 항체가 조사)

  • This study was performed to monitor the level of serum canine parvovirus(CPV) antibody titers in adult dogs throughout the Korea from January 2003 to April 2004. A total of clinically healthy 885 dogs between 1 year and 17 years old were included in this study. Serum antibody titers against CPV were measured by means of hemagglutination inhibition(HI) titers at the time dogs were brought to the hospital for revaccination. Most of dogs had been primarily vaccinated or previously revaccinated. Dogs were grouped by age, breed, sex, and primary vaccination and revaccination to determine whether these factors were associated with antibody titers. Serum CPV titers ${\geq}80$ were considered protective. Protective antibodies against CPV were present in 95.0% of the population. Breed, age, and primary vaccination and revaccination were not significantly associated with serum CPV antibody titers. But sex was significantly associated with CPV antibody titers. The results of this study have shown that there is a need to reconsider the annual revaccination strategy against CPV infection.

Sero-prevalence against a H3 subtype isolate of swine influenza virus (돼지인플루엔자바이러스 A형 H3 국내 분리주에 대한 혈청학적 역학조사)

  • A Total of 703 swine sera from 55 swine farrns (Mar. 1998 through Feb. 2001) were nation-wide collected for the presence of the antibody to influenza A virus H3 subtype isolate. The presence of antibody was tested by hernagglutination inhibition with chicken red blood cells and seropositiveness was determined by HI titer ${\geq}1$: 40. Sero-prevalence was evaluated based on year, season, region and age, respectively. In consequence, there were seme differences by year, season and region, respectively. High susceptibility was routinely observed in 60 and 90 day-old piglets. Therefore, it seems that the sero-prevalence to swine influenza virus H3 subtype isolate is useful for the prevention and control of swine influenza in Korea.

  • The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.

Antigenicity of CFC-101(Pseudomonas vaccine) in Guinea Pigs and Mice (기니픽과 마우스에서 CFC-101(녹농균 백신)의 항원성시험)

  • As a part of the safety evaluation of Pseudomonas vaccine(CFC-101), antigenicity tests were carried out in guinea pigs and mice. In active systemic anaphylaxis(ASA) test, guinea pigs showed no sign or only moderate sign(1/5) when sensitized and challenged with up to 200 $\mu\textrm{g}$/kg. In homologous passive cutaneous anaphylaxis(PCA) test using guinea pigs, inoculation of CFC-101 alone did not produce CFC-101-specific antibody. When inoculated with 200 $\mu\textrm{g}$/kg plus adjuvant, challenge of 200 $\mu\textrm{g}$/kg produced PCA titer of 32(5/5) but challenge of 20 $\mu\textrm{g}$/kg did not produce CFC-101-specific antibody. In heterologous PCA test using mice, CFC-101-specific antibody was not detected when sensitized with CFC-101 alone. Some animals(3/12) showed positive PCA response when inoculated with 200 $\mu\textrm{g}$/kg plus alum. In passive hemagglutination (PHA) test, although no antibody was detected at 20 $\mu\textrm{g}$/kg, inoculation of 200 $\mu\textrm{g}$/kg alone or with alum produced positive response in all animals. This result has already been predicted because CFC-101 is a vaccine developed for the purpose of immunization. From the above results, it can be concluded that there is no adverse antigenic potential up to 10 times clinical dose of 200 $\mu\textrm{g}$/kg.

Sample size of pooled sera for detection of chicken infectious bronchitis virus infection (닭 전염성 기관지염을 검출하기 위한 합병혈청의 표본크기)

  • The sample sizes required to detect at least one chicken infectious bronchitis virus(IBV) infection at flock-level were determined using pooled samples for 48 submissions with different samples in each. A total of serum samples of 9,980 layers from Kangwon, Chungpook and Chungnam province were collected and tested hemagglutination inhibition(HI) antibody titers against IBV both individually and with pooling size of 10. Of the 48 submissions, 72.9% were required less than 5 pools to detect at least one infected pool at 95% confidence level, and the corresponding rate was 77.1% at 90% confidence level. Overall, the number of pools was decreased as the percent of positive pools increased. At two different cut-of HI titer${\geq}9\;and{\geq}10$ for individual samples the seroprevalence was 50.1% and 33.4%, respectively while 59.9% were seropositive for pooled samples at HI $titer{\geq}8$. The correlation coefficients between pooled and individual samples at each submission were 0.592(p<0.001) for HI $titer{\geq}9$ and 0.561(p<0.001) for ${\geq}10$, with common correlation coefficient of 0.576. This study indicated that pooled testing for the detection of IBV infection may be an alternative strategy when only the pooled results are of interest and the prevalence has not known exactly.
