Morphological Development and Reproductive Behavior of Hedgehog Seahorse Hippocampus spinosissimus (Teleostei: Syngnathidae) (Hedgehog Seahorse (Hippocampus spinosissimus)의 산출습성 및 형태발달)

  • This study described the reproductive behavior and the morphological development of the larvae and juvenile of the hedgehog seahorse (Hippocampus spinosissimus). Larvae released from male seahorse were reared for 30 days in the laboratory. At the birth, the small flap-like mouth and anus of the yolk-sac larvae were open, and the yolk was completely absorbed within 1 day. The size of newborn larvae ranged 7.01-13.19mm (mean $9.26\pm1.9mm$, n=9) in TL (total length). The larvae had 15-16 pectoral fin rays and 19 dorsal fin rays, but 4 anal fin rays were not completely developed. The larval size ranged 13.38-14.38 mm (mean $13.96\pm0.3mm$, n=6) in TL at 1 day after release, and they had 17 pectoral fin rays, 20 dorsal fin rays and 4 anal fin rays when the larvae reached the juvenile stage. Juvenile size ranged 14.98-16.42mm (mean $15.84\pm0.5mm$, n=5) in TL at 3 days after release, and 1, 4, 6, 8 trunk spines extended quickly. Terminal part of the coronet became a four-forked wedge shape concurrently. Juvenile became 23.25mm (n=l) in TL after 15 days and all 11 trunk rings was connected completely. After 20 days, the juveniles were 24.66-28.65mm (mean $27.26\pm1.6mm$, n=5) in TL, and many melanophores appeared in integument. In the eye lenses, 5-6 lusters were formed. After 30 days, the juveniles were 26.78-28.26 mm (mean $27.36\pm0mm$, n=4)in TL and 8-9 lusters appeared on the eye lenses. Coronet terminal part developed to a five-forked crown-shape completely.

An immunohistochemical study of the endocrine cells in the stomach of the Korean hedgehog(Erinaceus korean us) (고슴도치 위점막의 내분비세포에 관한 면역조직화학적 연구)

  • The gastric endocrine cells of the Korean hedgehog, Erinaceus korean us were studied immunohistochemically. Seven kinds of endocrine cells-, gastrin-, somatostatin-, 5-HT-, glucagon-, BPP-, motiIin-and GIP-immunoreactive cells- were identified in this study. The chracteristic findings of the regional distribution and relative frequency of them were examined. Gastrin-immunoreactive cells were very numerously detected only in the pyloric region. Somatostatin-immunoreactive cells were more numerous in the pyloric region than in the cardiac and fundic regions. 5-HT-immunoreactive cells were more numerous in the cardiac and pyloric regions than in the tundic one. Glucagon-immunoreactive cells were found few or rarely in the fundic and pyloric regions. BPP-imunoreactive cells were numerously distributed in the pyloric region, moderately in the fundic region and few in the cardiac region. Motilin-immunoreactive cells were found rarely or few in the fundic and pyloric regions. GIP-immnuoreactive Cells were detected onIy in the pyloric region.

Molecular Mechanism of Runx2-Dependent Bone Development

  • Runx2 is an essential transcription factor for skeletal development. It is expressed in multipotent mesenchymal cells, osteoblast-lineage cells, and chondrocytes. Runx2 plays a major role in chondrocyte maturation, and Runx3 is partly involved. Runx2 regulates chondrocyte proliferation by directly regulating Ihh expression. It also determines whether chondrocytes become those that form transient cartilage or permanent cartilage, and functions in the pathogenesis of osteoarthritis. Runx2 is essential for osteoblast differentiation and is required for the proliferation of osteoprogenitors. Ihh is required for Runx2 expression in osteoprogenitors, and hedgehog signaling and Runx2 induce the differentiation of osteoprogenitors to preosteoblasts in endochondral bone. Runx2 induces Sp7 expression, and Runx2, Sp7, and canonical Wnt signaling are required for the differentiation of preosteoblasts to immature osteoblasts. It also induces the proliferation of osteoprogenitors by directly regulating the expression of Fgfr2 and Fgfr3. Furthermore, Runx2 induces the proliferation of mesenchymal cells and their commitment into osteoblast-lineage cells through the induction of hedgehog (Gli1, Ptch1, Ihh), Fgf (Fgfr2, Fgfr3), Wnt (Tcf7, Wnt10b), and Pthlh (Pth1r) signaling pathway gene expression in calvaria, and more than a half-dosage of Runx2 is required for their expression. This is a major cause of cleidocranial dysplasia, which is caused by heterozygous mutation of RUNX2. Cbfb, which is a co-transcription factor that forms a heterodimer with Runx2, enhances DNA binding of Runx2 and stabilizes Runx2 protein by inhibiting its ubiquitination. Thus, Runx2/Cbfb regulates the proliferation and differentiation of chondrocytes and osteoblast-lineage cells by activating multiple signaling pathways and via their reciprocal regulation.

Camphor Inhibits Adipocyte Differentiation via Its Impact on SMO-dependent Regulation of Hedgehog Signaling (Camphor의 Hedgehog 신호 SMO 조절을 통한 지방구세포 분화 억제효과)

  • In this study, we examined inhibition of adipocyte differentiation associated with the administration of camphor, a substance identified in extracts of the flowering plant Chrysanthemum indicum L. (CI). No camphor-mediated cytotoxicity was observed over a period of 1-10 days in studies targeting cells of the 3T3-L1 adipocyte-like line. Experiments that featured siRNA-mediated suppression of the transmembrane proteins Patched (PTCH) and Smoothened (SMO) resulted in inhibition and activation of differentiation, respectively. Interestingly, inhibition of PTCH typically activates SMO protein targeting and serves to activate hedgehog (HH)-mediated signaling. The results of our study suggest that activation of HH-mediated signaling can inhibit adipocyte differentiation. Furthermore, expression of glioma-associated oncogene homologue 1 (Gli1) was detected by flow cytometry in 62.7±1.5% of cells in response to administration of Lactobacillus rhamnosus (KCTC 3237) and in 60.4±2.2% of cells in response to camphor; these levels are higher than those detected in undifferentiated controls (24.9±3.1%). No change in the state of fermented camphor was identified by gas chromatography-mass spectrometry (GC-MS), but a 15.41% quantitative increase was confirmed in KCTC 3237. Overall, we conclude that administration of camphor resulted in overexpression of SMO and modulated the differential expression of Gli1. Animal studies focused on the impact of camphor as an agent to counteract obesity might be considered in the future. Indeed, camphor and similar physiologically active compounds from fermented CI might be developed as new and effective treatments for obesity.

Recent advances of basal cell carcinoma, squamous cell carcinoma, and carcinoma in situ (기저세포암, 편평세포암 및 피부내암의 최신지견)

  • Incidence of basal and squamous cell carcinoma is increasing among Korean. With increasing this incidence we need to attend the management and prevention of these cutaneous malignancies. Basal cell carcinoma (BCC) is the most common skin malignancy. Sun exposure is the most important environmental cause of BCC. Daily sun protection especially avoiding sun exposure between 10 a.m. and 4 p.m. provides most effective prevention against chronic ultraviolet-induced skin damage. Hedgehog pathway inhibitors are used locally advanced and metastatic BCC lesions in Korea. Until recently, there are no reports whether it developed resistance of Hedgehog pathway inhibitors with advanced BCC in Korean patients. Among malignant skin tumors, 19.1% were squamous cell carcinoma (SCC). The incidence of SCC was rapidly increased over the past years. However, the incidence rate of SCC in Japan has not been changed from the period 1976-1980 to 1986-1990. Sentinel lymph node biopsy for SCC did not provide diagnostic value. Bowen's disease and Erythroplasia of Queyrat are considered as carcinoma in situ of the skin. Erythroplasia of Queyrat have worse prognosis with a higher rate of malignant degeneration. Dermoscopy may helpful tool in assisting the noninvasive diagnosis of carcinoma in situ of the skin.

Hedgehog Oral Viral Papilloma (고슴도치 구강유두종)

  • Here we describe the histological lesion of a viral fibro-papilloma in a hedgehog. After surgical removal from maxilla, the solitary swollen mass was round to oval, yellowish with rough surface and measuring $6{\times}3{\times}2mm$ approximately. The tumor mass was submitted to the laboratory of pathology, college of veterinary medicine, Kyungpook National University for pathological diagnosis. Pathological examination of the tumor was established, the tumor was described grossly and sample was trimmed, sectioned and routinely prepared for histopathological evaluation. The tumor mass was diagnosed as viral fibro-papilloma, as the histological picture showed characteristic features of warts caused by papillomaviruses. The tumor characterized by thickening of the stratum spinosum (acanthosis) and basale, koilocytosis, intra-nuclear inclusion bodies in keratinocytes and fibrosis of submucosa. Further, viral inclusion bodies were demonstrated by Machiavello stain giving red color to the nuclei. No lymphocytes that responsible for regression of the wart could be detected, suggesting the poor possibility of spontaneous regression of the tumor. Papillomatosis is a disease of young animals, but in our case the infected Hedgehog was 5 years old, that maybe due to an impaired immune system, which is also shown by absence of lymphocytes. To the best knowledge of the author, this case presents the first report of viral fibropapillomatosis in Hedgehog.

Establishment of a Pancreatic Cancer Stem Cell Model Using the SW1990 Human Pancreatic Cancer Cell Line in Nude Mice

  • Aim: To establish a pancreatic cancer stem cell model using human pancreatic cancer cells in nude mice to provide a platform for pancreatic cancer stem cell research. Materials and Methods: To establish pancreatic cancer xenografts using human pancreatic cancer cell line SW1990, nude mice were randomly divided into control and gemcitabine groups. When the tumor grew to a volume of $125mm^3$, they treated with gemcitabine at a dose of 50mg/kg by intraperitoneal injection of 0.2ml in the gemcitabine group, while the mice in control group were treated with the same volume of normal saline. Gemcitabine was given 2 times a week for 3 times. When the model was established, the proliferation of pancreatic cancer stem cells was observed by clone formation assay, and the protein and/or mRNA expression of pancreatic stem cell surface markers including CD24, CD44, CD133, ALDH, transcription factors containing Oct-4, Sox-2, Nanog and Gli, the key nuclear transcription factor in Sonic Hedgehog signaling pathway was detected by Western blot and/or RT-PCR to verify the reliability of this model. Results: This model is feasible and safe. During the establishment, no mice died and the weight of nude mice maintained above 16.5g. The clone forming ability in gemcitabine group was stronger than that of the control group (p<0.01). In gemcitabine group, the protein expression of pancreatic cancer stem cell surface markers including CD44, and ALDH was up-regulated, the protein and mRNA expression of nuclear transcription factor including Oct-4, Sox-2 and Nanog was also significantly increased (P<0.01). In addition, the protein expression of key nuclear transcription factor in Sonic Hedgehog signaling pathway, Gli-1, was significantly enhanced (p<0.01). Conclusions: The pancreatic cancer stem cell model was successfully established using human pancreatic cancer cell line SW1990 in nude mice. Gemcitabine could enrich pancreatic cancer stem cells, simultaneously accompanied by the activation of Sonic Hedgehog signaling pathway.

An electron microscopic study on endocrine cells in the cardiac and fundic regions of the Korean hedgehog(Erinaceus koreanus) (한국산 고슴도치 분문부와 위저부의 내분비세포에 관한 전자현미경적 연구)

  • Endocrine cells in the cardiac and fundic regions of the Korean hedgehog were studied ultrastructurally. Five types of endocrine cells classified as EC, ECL, $D_1$. G and A-like cells were identified in these regions. EC cells contained pleomorphic granules, 170~500nm in diameter, with high electron density and highly dense bodies in a dense matrix. ECL cells were characterized by the presence of round or oval granules, 220~450nm in diameter, with high electron density. Some granules of ECL cells showed a small amounts of contents or empty. $D_1$ cells contained round and relatively small granules, 140~400nm in diameter, with low to moderate electron density. G cells were characterized by the presence of round or oval granules, 200~400nm in diameter, with moderate electron density. Some granules of these cells showed a narrow halo between the limiting membrane and the granular matrix. A-like cells contained round granules, 170~260nm in diameter, With high electron density. The granules of these cells showed homogeneous matrix surrounded by the tight membrane.

Cytologic Aspects of Oral Squamous Cell Carcinoma in a Captive African Hedgehog (Altelerix albiventris) (고슴도치의 구강 편평상피암종의 세포학적 고찰 1증례)

  • A 3-year-old intact female hedgehog (Atelerix albiventris) was presented for evaluation of mandibular swelling. Fine needle aspiration of swollen mandibular was performed and smears were stained with Romanowsky type stain for cytological evaluation. Smears were highly cellular with predominance of variably shaped keratinized or non keratinized squamous cells with low numbers of spindloid to abnormally elongated cells. Cytologic impression was squamous cell carcinoma. The mass was surgically removed for histological examination. Microscopically tortuous and anastomosing delicate to broad pegs and nests of neoplastic squamous epithelial cells were supported by a moderate collagenous and spindloid fibroblast stroma. Tumor cells had moderate anisocytosis and mild anisokaryosis and range from moderately to well keratinized, with areas of intratumoral acantholysis accompanied by mixed stromal lymphoplasmacytic, neutrophilic inflammation. The mitotic index is 2-3 per high-power field. Tumor cells were expanding the subcutis subjacent to the layer of skeletal muscle and incorporating the osseous tissue fragments. The final diagnosis was squamous cell carcinoma. The patient survived three months after surgery without any further medical treatments.
