Assessment of Genetic Diversity of Hedera spp. Using RAPD Marker Technique (RAPD Marker를 이용한 Hedera속 식물의 다양성 조사)

  • Eleven accessions of Hedera helix, three accessions of Hedera rhombea, one accession of Fatshedera lizei, and one accession of Fatsia japonica were collected and their genetic diversity was measured by using 10 RAPD primers. Approximately ninety seven percentage of polymorphism was detected, because broad germplasm, three genus, was used. Total 97 bands were scored and a dendrogram was constructed by using an UPGMA method. Accessions belonging to Hedera helix tightly clustered in one group: eight accessions showed extremely narrow genetic differences and the other three accessions also showed significant similarity. Despite of their genetic similarity, they showed morphological variations. The morphological variation with a narrow genetic differences indicated that the ivy cultivars have been indeed developed from a mutation breeding program. Genetically most unrelated Fatsia japonica showed a genetic distance of 0.63 on the average between other species. An accession from Fatshedera lizei developed by crossing between two genus, Hedera helix and Fatsia japonica, was allocated together with accessions from Hedera rhombea.

Fluorometric Measurement of Acid Phosphatase Activity in the Angiosperm Parasite (Cuscuta cephalanthi) and its Host (Hedera helix) (기생피자식물(Cuscuta cephalanthi)과 그 숙주(Hedera helix)에서 acid phosphatase활동의 형광분석법에 의한 측정)

  • Acid phosphatase activities were analyzed in $\mu\textrm{g}$ tissue samples from an angiosperm parasite (Cuscuta cephalanthi) and its host plant (Hedera helix) by a fluorometric microtechnique. The apex and the coiling portion of the parasite axis exhibited greater enzyme activies than other portions of the hypha. Acid phosphatase activity in the haustorium was 2-3 times that in the hyphal axis. The vascular bundles of the normal host exhibited the greatest enzyme activity. The acid phosphatase activity in the host infected by the parasite decreased to the activity level of the haustorium.

Evaluation of Salt Tolerance of Three Foliage Plant as affected by Salinity Concentration in Indoor Ornamental Hydroculture (실내 관상용 물재배에서 염분농도에 따른 3가지 관엽식물의 내염성 평가)

  • This study investigated the growth characteristics of Euonymus japonicus, Hedera helix, and Peperomia puteolata treated with different calcium chloride (CaCl2) concentrations to evaluate salt tolerance limits in hydroculture cultivation. Six concentrations of CaCl2 (0, 1, 2, 5, 10, and 15 g·L-1 referred to as Cont., C1, C2, C5, C10, and C15) were applied to solution - grown plant species. The survival rate, growth index, plant height, plant width, leaf width, leaf length, number of leaves, and relative chlorophyll contents were measured at monthly intervals. Euonymus japonicus, Hedera helix, and Peperomia puteolata survived up to C2, C5, and C10 at each CaCl2 concentration. The Euonymus japonicus was higher in the C1 treatment than in the Cont. for most growth characteristics. Hedera helix had the highest leaf width, leaf length, and number of leaves in the Cont., a significant difference was observed compared with the C1 treatment. The chlorophyll content did not differ significantly between the C5 and Cont. treatments. The leaf width and length of Peperomia puteolata were greater in the C2 and C1 treatments than in the Cont., whereas the number of leaves and chlorophyll content were the highest in C5. Dry weight analysis revealed that Euonymus japonicus, Hedera helix, and Peperomia puteolata were the lowest in the Cont. treatments. Euonymus japonicus was 74% in C15, and Hedera helix, and Peperomia puteolata were analyzed at approximately 37%- 50% and 9%-14%, respectively, regardless of the concentration in the CaCl2 treatment groups. In indoor hydroponic cultivation, the salt tolerance limit concentrations of Euonymus japonicus, Hedera helix, and Peperomia puteolata are 2, 5, and 10 g·L-1, respectively, indicating that hydroculture management techniques should be applied at higher concentrations.

Antiviral Activity of Hederasaponin B from Hedera helix against Enterovirus 71 Subgenotypes C3 and C4a

  • Enterovirus 71 (EV71) is the predominant cause of hand, foot and mouth disease (HFMD). The antiviral activity of hederasaponin B from Hedera helix against EV71 subgenotypes C3 and C4a was evaluated in vero cells. In the current study, the antiviral activity of hederasaponin B against EV71 C3 and C4a was determined by cytopathic effect (CPE) reduction method and western blot assay. Our results demonstrated that hederasaponin B and 30% ethanol extract of Hedera helix containing hederasaponin B showed significant antiviral activity against EV71 subgenotypes C3 and C4a by reducing the formation of a visible CPE. Hederasaponin B also inhibited the viral VP2 protein expression, suggesting the inhibition of viral capsid protein synthesis.These results suggest that hederasaponin B and Hedera helix extract containing hederasaponin B can be novel drug candidates with broad-spectrum antiviral activity against various subgenotypes of EV71.

Effects of Different Light Conditions on Wintering in Evergreen Ground Covers (차광처리가 상록지피식물의 내한성에 미치는 영향)

  • Six evergreen ground-cover species including Ajuga reptans, Carex morrowii 'Variegata', Dianthus 'Night Star', Hedera helix 'Ivalace', Pachysandra terminalis, and Sedum rupestre, were tested for cold acclimation with three different light treatments (0, 30, and 70% of shading, respectively) to select potential candidates for over-wintering in central Korea. Among those species, Ajuga reptans, Carex morrowii 'Variegata', and Sedum rupestre performed well, which had more than 50% of survival rate in all light conditions. However, Dianthus 'Night Star' had less than 40% under any light treatment and the other two species, Hedera helix 'Ivalace' and Pachysandra terminalis, survived more than 40% only with 70% of shading condition. These results imply that those candidate species having less cold hardness in central Korea could have the potential to could over-winter as long as a proper light condition is provided.

  • 화학적 살충제는 빠르고 정확하게 해충을 방제하는 효과를 나타내지만, 인간과 다른 유기체에 대한 살충제 잔류의 위험성, 화학적 살충에 대한 저항성을 가진 해충의 출현, 생태계의 교란 등 다양한 부작용이 있다. 이 때문에 생물적 방제를 통한 해충 방제는 효과적인 수단이 될 수 있다. 특히 채소 작물 등에 주로 발생하는 응애는 생산량 저하 및 품질 저하 등과 같은 다양한 문제를 일으킬 수 있다. 특히 진딧물이나 점박이응애는 화학적 살충제에 대해 높은 저항성이 발달되어 문제가 되고 있다. 본 연구에서는 아이비(Hedera helix), 란타나(Lantana camara), 예루살렘체리(Solanum pseudocapsicum)를 통한 천연 식물 추출물의 응애에 대한 생물적 방제 효과를 확인하였다. 각 추출물은 0.5%, 0.05% 주정 및 메탄올로 추출하였으며, 응애 방제 효과(%)는 아이비>란타나>예루살렘체리로 나타났다. 각 추출물은 GC/MS 분석을 통해 생리활성 후보 물질을 확인하였다. 이를 통해 농업 이익 증대, 생산량 유지와 더불어 환경 및 인간과 다른 유기체에 피해를 주지 않으며 효과적으로 응애를 방제 할 수 있는 천연 생물적 방제제로의 가치를 확인하였다.

  • Experiments were conducted to investigate propagation, shade tolerance, cold resistance and growth rate of evergreen ground cover plants ; Vinca minor K , Ajuga reptans L., Ophiopogon japonicus ker. f nanus hort , and Hedera helix.. The results were as follows : 1.It is convenient to use the growing case for rice seed to grow and transplant Vinca minor seed-ling. The most proper density of transplanting Vinea was 180 plants per 1m$^2$. At the end of growing season, the coverage came up to 90% The rooting ability of Hedera helix was best to cut from April to May (temperature 15~23˚C). The seedling length of Parthenocissus quinguefohlia were irregular and ranged from 5 cm of 200cm. 2. Ajuga reptans L., Ophiopogon j. and Vinca minor L. grew better under 50% light intensity than full sunlight. Particularly, under full sunlight and aestival high temperature, there were a few withering plants in Ajuga reptans L. , and V Vilica minor L. 3. Of Vinca minor K., Ajuga reptans L. , Ophiopogon japonicus ker. f. nanus hort. and Hedera helix. acclimatized in open field, where temperature was from -7~8˚C. There were few plants had damage in low temperature incubator to -16˚C after field acclimatization. 4.Tendrils of Parthenocissus quinguefolia strongly clung to the wire netting stone fence but intruded into the gap and cranny of the precast concrete fence and so clung to, so that it needed to install the bending net on the precast concrete fence.

A Growth Responses of Indoor Ground Cover Plants according to a Light Source of Aritificial Light (인공광의 광원에 따른 실내 지피식물의 생육반응)

  • This study was carried out to obtain fundamental information of growth response of ground cover plants under artificial light quality at indoor. Aglaonema 'Silver Queen', Hedera helix L., Hoya carnosa 'Tricolor' and Saintpaulia ionantha 'Frances' were examined under a 400lux light intensity consisted of Mercury lamp, True-lite lamp, incandescent lamp, dark-room and sunlight indoor condition. A data analysis were performed by GLM, Duncan's multiple range test and mean score with SAS program. Results of experiments are as follows; 1. A plant growth status was better showed under the True-lite lamp than sunlight. 2. A Saintpaulia ionantha flower color was responded in the first place, the deep pinkish red color of Saintpaulia ionantha flower was obtained under Mercury lamp and "True-lite lamp", "sunlight", and incandescent lamp were follow. Flower numbers of Saintpaulia ionantha after 60 days tended to decrease under every artificial light quality. 3. Leaf length and leaf width were increased under True-lite lamp, but most of plants was not significantly affected by artificial light quality. 4. A stem length of Hedera helix was increased the highest rank under sunlight also, one of artificial light, the highest increase rank was showed under incandescent lamp. 5. Chlorophyll content was highly increased under Mercury lamp, but was responded poor under incandescent lamp.

An Analysis Method on Injury Symptoms Utilizing Infrared Thermal Imaging under the Freezing Stress of Hedera helix L. (헤데라 헬릭스 식물의 적외선 열영상에 의한 저온 및 한풍피해에 관한 연구)

  • The experiments, which analyze the injury symptoms and diagnose growth conditions utilizing IRVT and analyzing each parts of H. helix L., had been held under a low temperature. Greenhouse and outdoor growing Genus hedera had been prepared and compared with each Genus hedera's peak and bottom leaves' surface temperature under the experimental categories $-6^{\circ}C$ and $-12^{\circ}C$. As results, analyzing the surface thermal property of peak part leaves' of outdoor growing Genus hedera, at experimental categories $-6^{\circ}C$, $-12^{\circ}C$ were ranged from $-2^{\circ}C{\sim}-7^{\circ}C$ and $-2^{\circ}C{\sim}-15^{\circ}C$. On the other hand, the surface thermal property of bottom part leaves at experimental categories $-6^{\circ}C$, $-12^{\circ}C$ were ranged $-2^{\circ}C{\sim}-11^{\circ}C$ and $-1^{\circ}C{\sim}-12^{\circ}C$. It appears that the thermal properties of leaves' surface on $-6^{\circ}C$ peaks and $-12^{\circ}C$ bottoms were more broadband than bottoms and peaks. It means that the peaks were more sensitive than bottoms, as like $-2^{\circ}C{\sim}-15^{\circ}C$, $-1{\sim}-12^{\circ}C$. Moreover, as similar results had seen to leaves surface temperature added to cold wind conditions. How the cold wind damaged the outdoor growing Genus hedera, analyzed the surface thermal property by IRVT data under $0^{\circ}C$, $-2^{\circ}C$, $-4^{\circ}C$ condition, it resulted to $-6.2^{\circ}C$, $-6.8^{\circ}C$, $-7.5^{\circ}C$. It appeared more $3.5{\sim}6.2^{\circ}C$ low temperature than experimental setting point. In addition, each parts thurmal property of peaks and bottoms was not similar, it referred to each parts' sensitivities of low temperature were different on the peak and bottom leaves surface temperature.
