Three Pseudomonas strains (Bk8, Bk9, Bk10) selected from soil for their ability to degrade herbicide diuron were tested for their heavy metal resistance. The growth of these catabolic strains on a minimal medium with various concentrations of $Cd^{2+},\;Zn^{2+},\;Ni^{2+}$, and $Hg^{2+}$ revealed a minimal effect on the carbon source for the inhibitory effect of the metals. One of these strains, namely, Bk8, exhibited a high resistance to the heavy metals as compared to the two other strains. This strain harbors plasmid pBk8 (110 kb) and contains at least fur determinants encoding heavy metal resistance. Nickel and zinc resistance are encoded by genes located on the chromosome, while cadmium and mercury resistance are on plasmid pBk8. Accordingly, the characteristics of strain Bk8 suggest that it would be useful in the bioremediation of aromatic compounds in the presence of toxic heavy metals as co-contaminants.
A new strain of Bacillus capable of growing upto pH 11 was isolated from a local pond polluted with detergents. This strain elicited unimpaired growth in media supplemented with heavy metals such as As, Cu, Cd, Hg, Ni and Zn. A MIC value of 200, 3, 1.60, 2.25, 7.50 and 3.70 mM was noticed for As, Cu, Cd, Hg, Ni and Zn respectively. Analysis of total DNA revealed the presence of a plasmid of 26 kb. This plasmid was lost by acriflavine treatment to the cultures. Such cured strains were found sensitive to heavy metals. Our findings suggest that incidence of heavy metal resistance is widely distributed and resistant strains could be isolated from heavy metal unpolluted sites.
Effects of rare earth metals addition and aging treatment on corrosion resistance and mechanical properties of super duplex stainless steels were investigated using optical/SEM/TEM metallographic examination, an X-ray diffraction test, a potentiodynamic anodic polarization test and a tensile test. The performance of the experimental alloy with 0.32% REM addition was compared with commercial super duplex stainless steel such as SAF 2507 when they were exposed to solution annealing heat treatment and aging treatment. The corrosion resistance in CF environments and mechanical properties of the experimental alloy were found superior to those of the commercial duplex stainless steel. The REM with larger atomic radii than those of Cr, Mo and W may fill vacancies inside the matrix and around the grain boundaries, retarding formation of harmful intermetallic σ and χ phases. In addition, fine REM oxides/oxy-sulfides (1-3㎛) seemed to enhance the retardation effects. With REM additions, strength and ductility increased due to the phase and grain refinement caused by fine REM oxides and oxy-sulfides.
Recently a fuel oil of the diesel engine of the marine ship is being changed with heavy oil of low quality as the oil price is higher more and more. Therefore the wear and corrosion in all parts of the engine such as cylinder liner, piston crown, spindle and seat ring of exhaust valves are predominantly increased. In particular the degree of wear and corrosion of piston crown is more seriously compared to the other parts of the engine due to operating in severe environment such as the high temperature of exhaust gas and repeating impact. Thus the repair weldment of the piston crown is a unique method to prolong the its life in a economical point of view. In this case, filler metals having a high corrosion and wear resistance such as stellite 6, Inconel 625 and Inconel 718 are mainly being used for repair welding. However it has been often happened that piston crown on the ship,s job site is being actually inevitably welded with mild filler metals. Therefore in this study, filler metals such as E4301, E4313 and E4316 were welded at SS401 steel as the base metal, and corrosion property of their weld metals in the case of post weld heat treatment or not was investigated with some electrochemical methods such as measurement of corrosion potential, cathodic and anodic polarization curves, cyclic voltammogram and polarization resistance etc. in 0.1% $H_2SO_4$ solution. Corrosion resistance of the weld metal of E4301 was better than the other weld metals in the case of no heat treatment, however, its resistance was considerably decreased with post weld heat treatment(annealing:$625^{\circ}C$, 2 hr) compared to other weld metals. The weld metals of E4313 and E4316 showed a relatively good corrosion resistance by post weld heat treatment.
Metal ions contamination may inhibit microorganisms involved in the biodegradation of organic compounds and affect biodegradation rates. Therefore, it is likely that bioremediation of xenobiotics-contaminated soils and waste will require inoculation with efficient biodegrading microbial communities, with capabilities of being resistant to heavy metals as well. Two different transconjugants (Pseudomonas sp. KMl2TC and P. aeruginosa TC) were constructed by conjugation experiments. Results on MIC, induction and growth inhibition strongly indicated that arsenic-resistant plasmid, pKM20, could be mobilized, and the newly acquired phenotype of pKM20 was not only expressed but also well regulated, resulting in newly acquired resistances to $As^{5+},\;As^{3+},\;and\;Sb^{3+} in\;addition\;to\;Cd^{2+},\;Zn^{2+},\;and\;Hg^{2+}$. The phenol- degradation efficiencies of Pseudomonas sp. KMl2TC were maintained significantly even at high heavy metal concentrations at which these efficiencies of P. aeruginosa TC were completely impaired. The results in this study on the effects of heavy metals on phenol degradation, especially after conjugation, are the first ever reported. All the results described in this study encourage to establish a goal of making "designer biocatalysts" which could degrade certain xenobiotics in the area contaminated with multiple heavy metals.
The role of soil microorganisms, specifically rhizobacteria, in the development of rhizoremediation techniques is important to speed up the process and to increase the rate of mobilization or absorption of heavy metals to the plant. In this study, Methylobacterium sp. SY-NiR1 was isolated from the rhizosphere soils of plants in oil and heavy metal-contaminated soil. Based on its pink pigmented colony, rod-shape cells, and belonging in $\alpha-Proteobacteria$, Methylobacterium sp. SY-NiR1 is considered a pink-pigmented facultative methylotroph. SY-NiR1 had the ability to produce indole acetic acid which is one of phytohormones. This bacterium showed resistance against multiple heavy metals such as Cd, Cr, Cu, Pb, Ni, Zn, and the order of its resistance based on $EC_{50}$ was Zn > Ni > Cu > Pb > Cd > Cr. Therefore, Methylobacterium sp. SY-NiR1 can stimulate seed germination and plant growth in soil contaminated with heavy metals.
Different polluted water samples were collected from a wastewater treatment plant, agricultural drainage canals, the River Nile, and irrigation canals. The samples were examined for the enumeration of Pseudomonas aeruginosa in the Sohag area, Egypt over a period of one year. A total of 240 isolates were collected and tested for their resistance to 12 common antibiotics and 6 heavy metals. The isolates were found to be less resistant to norfloxacin(1.7%), ofloxacin(4.6%), amikacin(9.6%), tobramycin (10.4), carbenicillin (15.4), and gentamycin (41.3%), yet more sensitive to rifampicin (75%), kanamycin (89.6%), ampicillin (90.8%), chloramphenicol (91.7%), streptomycin (92.9%), and tetracyclin(96.3%). In contrast, 7.1%, 12.9%, 25.4%, and 53.7% of the isolates were resistant to lead, cadmium, mercury, and zinc, respectively. None of the isolates had developed a resistance to silver or molybdenum. The high frequency of metal-antibiotic double resistance existed between lead and amikacin (56.5%), cadmium and ofloxacin (72.7%), zinc and norfloxacin (100%), and mercury and carbenicillin (94.6%). The high occurrence of antibiotic-resistant bacteria in natural water could be related to the widespread use of antibiotics, with possible public health hazard.
Chemical resistance of the cement mortar containing the Thermally Activated Diaomite(TAD) and H.M.(Heavy Metals) has been studied. The H.M.. extracted from EAF(Electrica1 Arc Furnace) Dust. were saturated with diatomite. The diatomite was then thermally activated at $750{\circ}C$ for 30minutes and powdeled. The powder was mixed with a portland cement on a weight basis from 0%. 2.5%. 5.0%. 10%. 20%. The optimum mixture. after those mixtures were subjected to compressive strength(7 and 28days) and leaching bchaviour of H.M.. was tested for its experiment on Wet/Dry cycles and chemical resistance(e.q. imrncrsion in 5%(Conc.) of H2S04, CaC12 and hlgSO4. It was shown that the cement, mortar containing 10% of' P.D. gave a rise to the remarkable increase in compressive strength. The compressive strength was generally decrease beyond the addition of 10% of P.D. The maximum $496kgf/cm^2$ of 28days compressive strength was acheiveti when 10% of P.D. was added to the cement mortar.
Recently, the fuel using in the diesel engines of marine ships has been changed to a low quality of heavy oil because of the steady increase in the price of oil. Therefore, the wear and corrosion in all parts of the engine such as the cylinder liner, piston crown, and spindle and seat ring of exhaust valves has correspondingly increased. The repair welding of a piston crown is a unique method for prolonging its lifetime from an economic point of view. In this case, filler metals with a high corrosion and wear resistance are mainly being used for repair welding. However, often at a job site on a ship, a piston crown is actually welded with mild filler metals. Therefore, in this study, mild filler metals such as CSF350H, E8000B2, and 435 were welded to SS401 steel as the base metal, and the corrosion properties of the weld metals with and without post weld heat treatment were investigated using some electrochemical methods in a 0.1% $H_2SO_4$ solution. The weld metal welded with CSF350H filler metal exhibited the best corrosion resistance among these filler metals, irrespective of the heat treatment. However, the weld metal zones of the E8000B2 and 435 filler metals exhibited better and worse corrosion resistance with the heat treatment, respectively. As a result, it is suggested that in the case of repair welding with CSF350H and 435 filler metals, no heat treatment is advisable, while heat treatment is desirable if E8000B2filler metal is used with repair welding.
This study was conducted to investigate the tolerance of some resistant fungal strains from soils contaminated with heavy metals. Various fungal strains were isolated from soil samples collected from studied sites which heavy metals and other pollutants have been emitted in effluents for several years. Fungi isolated belong to different genera; however, Penicillium spp. showed the most frequent species. The microbial number was remarkably higher in the control soil than contaminated soil samples collected from mining areas. $Pb^{2+}$ and $Zn^{2+}$ had the highest concentration in the polluted soils ranging from 89 - 3,521 ppm and 98 - 4,383 ppm, respectively. The minimum inhibition concentrations (MICs) of $Pb^{+2}$ and $Zn^{+2}$ showed the highest values against the fungal strains. $Ni^{+2}$ and $Co^{+2}$ were the lowest contaminants in the polluted soils with the concentration of 5 to 12.1 ppm and 1.8 to 4.8 ppm, respectively. The tested resistant strains showed the strongest inhibition for $Ni^{+2}$ and $Co^{+2}$ up to 200-400 ppm. Cadmium was the most highly toxic heavy metal for most of strains, however, 1 mM of $Cr^{3+}$, $Cu^{2+}$ and $Pb^{2+}$ accelerated the growth of Penicillium verrucosum KNU3. $Cu^{+2}$ and $Zn^{+2}$ at concentration of 1 mM did not affect the growth rate P. funiculosum KNU4. Tolerance of fungal species to heavy metals appears to be strain and origin dependent.
