Change of Fibrinolytic and Antioxidative Activities of Codonopsis lanceolata According to Various Storage Conditions, and Heat or Salt Treatments

  • We investigated the changes of fibrinolytic and antioxidative activities of wild or cultivated Codonopsis lanceolata, which were packed in woven polypropylene (WP) film or low density polyethylene (LDPE) film and stored for 0, 15, and 30 days at refrigerated $(2\~4^{\circ}C)$ or room $(l8\~20^{\circ}C)$ temperature (So we have 16 samples, and wrote them as the abbreviated words such as W-WP-RE-15, W-WP-RO-15, W-LDPE-RE-15, W-LDPE-RO-15, W-WP-RE-30, W-WP-RO-30, W-LDPE-RE-30, W-LDPE-RO-30, C-WP-RE-15, C-WP-RO-15, C-LDPE-RE-15, C-LDPE-RO-15, C-WP-RE-30, C-WP-RO-30, C-LDPE-RE-30, C-LDPE-RO-30). Fibrinolytic activity of fresh Codonopsis lanceolata cultivated in the mountain or field were 0.8 unit (plasmin unit/ml) or 7.3 units, respectively. In descending order, the activities of wild Codonopsis lanceolata stored for 15 days were as followed; LDPE-RE (0.70 unit), WP-RO (0.52 unit), WP-RE (0.45 unit), and LDPE-RO (0.30 unit). After 30 days, fibrinolytic activities of them decreased to 0.47 unit (LDPE-RE), 0.28 unit (WP-RO), 0.21 unit (WP-RE), and 0.30 unit (LDPE-RO). Considering from the point of fibrinolytic activity, the optimal storage condition of wild Codonopsis lanceolata was packing with LDPE film and storing at $4^{\circ}C$. The change of fibrinolytic activities of Codonopsis lanceolata cultivated in the field revealed the similar trend as wild samples, but the fibrinolytic activities of 30 days-stored samples were maintained better than the wild Codonopsis lanceolata. Fibinolytic activity of wild Codonopsis lancealata was increased by heating for 5 min at $100^{\circ}C$ and decreased by addition of NaCl. Antioxidative activities of Codonopsis lanceolata were also compared from the electron donating activity. Fresh Codonopsis lanceolata had about $70\%$ of electron donating activity. Independent of cultivation area, electron donating activity dropped to $19\~74\%$ (wild Codonopsis lanceolata) and $27\~59\%$ (cultivated Codonopsis lanceolata) during 15 days storage. But after 30 days storage, we obtained the unexpected results, which meant that the activities were higher activities than 15 days-stored samples or even though the fresh samples. In general, Codonopsis lanceolata could maintain antioxidative activities most strongly with LDPE film and chilled condition.

The properties of algal degradation and gas emission by thermophilic oxic process (고온호기발효장치를 이용한 조류 분해 및 가스 발생특성)

  • The purpose of this study is to establish effective conditions for controlling $CH_4$, $N_2O$ emission from organic Waste / wastewater treatment processes. Continuous and batch experiments were conducted to treat the micro algae from polluted and eutrophicated lakes through the thermophilic oxic process. The microalgae used were mainly Microcystis sp.(collected from eutrophic lake) and Chlorella sp. (cultured in laboratory) Wasted cooking oil was added by aid-heating source. Physico-chemical components of sludges and microalgae were analyzed. In batch experiments, air supply was changed from 50ml/min to 150ml/min. The temperature. water content and drained water were affected by the air flow rate at initial stage. However, there was almost no influence of air flow rate on them in middle and last stages. At air flow rate of 100ml/min, the degradation rate of organic material was higher than that at other air flow rates. $CO_2$ concentration in exhaust was proportional to the strength of aeration, especially at initial stage when degradation was active. $CH_4$ with low concentration was detected only at starting stage when air diffusion was not enough. $N_2O$ production was not affected by variation of air supply. In continuous experiments no matter what the dewatering methods (with PAC and without PAC) and media (wood chip and reed chip) were changed, $N_2O$ was almost not affected by variation of injected air. Result showed that the reed chips using for lake purification could be used as media for thermophilic oxic process in lake and marshes area. $CO_2$ concentration was not so much affected by the change of dewatering methods and media types. $CH_4$ was not detected in the experimental period. So it can be shown that the thermophilic oxic process had been well operated in wide handling conditions regardless of media and dewatering methods.

Development of Thermo-Cosmetics Using Photothermal Effect of Gold Nanoparticles (금 나노입자의 광열효과를 이용한 온열화장품 개발)

  • Many applications of nanoparticles have been developed since 1970s. Surface plasmon resonance (SPR) effect can be generated at the surface of nanoparticles by illumination. SPR is the resonant oscillation of conduction electrons at the surface material stimulated by incident light. The collisions between excited electrons and metal atoms can cause the production of thermal energy (photothermal effect). Here, we presented the development of thermo-cosmetics using photothermal effect of gold nanoparticles. Gold nanoparticles (GNPs) were chosen for it's low toxicity. We also and investigated the cell biocompatibility and heating effectiveness for photothermal effect of GNPs. Synthesized GNPs were verified by UV-vis spectrophotometer, where GNP has a characteristic absorbance spectrum. Concentration of GNP was measured by atomic absorption analyzer. The cytotoxicity was confirmed by MTT assay and double staining assay. Photothermal effect of GNP was demonstrated by the thermal increasing properties depending on GNP concentration, which was taken by an IR-thermal camera with a xenon lamp as the light source. If the thermal effect of GNP is applied for thermo-cosmetics, it can supply heat to skin by converting solar energy into thermal energy. Thus, cosmetics containing GNPs can provide benefits to people in the cold region or winter season for maintaining skin temperature, which lead to a positive effect on skin health.

Anti-bacterial Effect of Psidium guajava and Geranium thunbergii Extracts on Listeria sp. Isolated from Fishery Products (수산가공품에서 분리한 Listeria sp.에 대한 구아바(Psidium guajava)잎과 현초(Geranium thunbergii) 추출물의 항균활성)

  • Listeria sp. is one of the pathogenic bacteria causes the infection listeriosis, through mainly raw food such as fishery food, dairy food and vegetables. Listeria sp. is a Gram-positive, non-spore-forming, motile, and facultative anaerobic bacterium. Because of the tolerance of Listeria sp. to low temperature and high salt concentration, it is very difficult to prevent them contaminated in the food, which do not require heating, especially, such as raw fishery products. So prevention and removal of bacterial contamination at the food manufacturing stage is the best method. In this study, therefore, several natural products including Psidium guajava and Geranium thunbergii were screened to investigate the antibacterial activity against Listeria sp., with expectation of fewer side effects and fewer resistance problems. Significant effects of two extracts were confirmed by well diffusion assay, MIC assay, and growth inhibition assay. P. guajava and G. thunbergii showed MIC values at 64-256 ㎍/mL meaning strong antibacterial activities against 6 kind of Listeria sp. tested. And the growth of Listeria sp. in the liquid media was actually inhibited by the addition of these two extracts.

Deterioration and Microclimate in the Shelter for the Gaetaesajiseokbulibsang (Standing Triad Buddha Statues in Gaetaesaji Temple Site), Nonsan, Korea (논산 개태사지석불입상의 손상도와 보호각 내부의 미기후 환경)

  • The Gaetaesajiseokbulibsang (Treasure No. 219) consists of light gray and coarse to medium-grained granodiorite with feldspar phenocrysts in part. Magnetic susceptibility of the rock material was measured as 12.06(${\times}10^{-3}$ SI unit), being different from the granite($0.19{\times}10^{-3}$ SI unit) in the Mt. Cheonho. This indicates the raw material has been supplied from the outside. As a result of deterioration assessment, exfoliation of the Right Buddha, cement and dust of the Main Buddha were estimated as 35.2%, 21.1% and 25.0%. The ultrasonic velocity was measured as 2850.2m/s(Main Buddha), 2648.4m/s(Left Buddha) and 2644.5m/s(Right Buddha). The compressive strength calculated from the velocity showed low in the Right Buddha among three and the all pedestal parts which corresponds to the result of deterioration assessment. The indoor mean temperature and relative humidity of the shelter were $13.7^{\circ}C$ and 79.0%. It is evaluated that the indoor microclimate was stable and the shelter functioned to reduce climatic fluctuation of the outdoor. However, water condensation was occurred on the surface of the pedestal part during spring and summer, and freezing in winter season might also be done. These factors were probable to be a main cause of the surface exfoliation of the Triad Buddha Statues. Therefore, dehumidification and heating system in the shelter should be applied to prevent further deterioration.

Changes to the Anti-oxidative Activity and Amino Acid Content of Arctium lappa Tea by Heat Treatment (열처리 조건에 따른 우엉차의 항산화 활성과 아미노산 함량의 변화)

  • In this study, the effect on the free amino acid content and anti-oxidative activity of Arctium lappa samples by heat treatment were compared to obtain data on the conditions required for properly processing the root as a raw material for tea. The results of DPPH and ABTS radical scavenging activity assays showed that 180℃-4 min and 200℃-3 min treatments retained relatively high activity. All heat treatment conditions showed increased polyphenol content compared to the initial sample value of 42.72 mg/l, and the 180℃-4 min and 200-3 min treatments were higher than the others at 60.09 mg/l and 62.74 mg/l, respectively. In the free amino acid analysis of the A. lappa root tea, the tendency of the initial content was confirmed to decrease as temperature and time increased. Changes in phenylalanine, histidine, leucine, and isoleucine were found to be high, while aspartic acid and proline changes were relatively low. Overall, amino acid content decreased significantly under heating conditions above 180℃-3 min and 200℃-2 min. From these results, heat treatment of A. lappa root at 180℃-4 min was deemed optimal based on anti-oxidative activity and free amino acid content.

Development of Natural and Ecological Wastewater Treatment System for Decentralized Regions and Rural Communities (분산지역 및 농촌마을 하수처리를 위한 자연정화 고도처리 공법 개발)

  • The feasibility of the Natural and Ecological Wastewater treatment System (NEWS) was examined for rural wastewater treatment in Korea. The intermittent trickling biofilter with high hydrophilic filter media was used for pretreatment for suspended solids and organic pollutants. The subsequent constructed wetland with porous granule materials was used for promoting nutrient removal. The results show that the removal efficiencies of the system were high with respect to the water quality parameters except COD. Even if the effluent from the biofilter did not meet the guidelines for wastewater treatment plant effluent in Korea in terms of $BOD_5$ and TN, the final effluent of the system meets the guidelines us to good performance of the constructed wetland. The regression analysis between pollutant loading rate and removal rate indicated that the system could have stable removal for SS, $BOD_5$, TN, and TP in the given influent ranges. The analysis in the winter period indicated that the wetland covered with transparent polycarbonate glass had the statble performance during the winter period dus to increase of temperature inside the wetland without any heating system. With the stable performance, effective poilutant removal, low maintenance, and cost-effectiveness, the NEWS could be considered as an alternative treatment system for decentralized regions and rural communities in Korea.

Thermally Stimulated Current Analysis of (Ba, Sr)TiO$_3$ Capacitor ((Ba, Sr)TiO$_3$ 커패시터의 Thermally Stimulated Current분석)

  • It has been known that the leakage current in the low field region consists of the dielectric relaxation current and intrinsic leakage current, which cause the charge loss in dynamic random access memory (DRAM) storage capacitor using (Ba,Sr)TiO$_{3}$ (BST) thin film. Especially, the dielectric relaxation current should be seriously considered since its magnitude is much larger than that of the intrinsic leakage current in giga-bit DRAM operation voltage (~IY). In this study, thermally stimulated current (TSC) measurement was at first applied to investigate the activation energy of traps and relative evaluation of the density of traps according to process change. And, through comparing TSC to early methods of I-V or I-t measurement and analyzing, we identify the origin of the dielectric relaxation current and investigate the reliability of TSC measurement. First, the polarization condition such as electric field, time, temperature and heating rate was investigated for reliable TSC measurement. From the TSC measurement, the energy level of traps in the BST thin film has been investigated and evaluated to be 0.20($\pm$0.01) eV and 0.45($\pm$0.02) eV. Based on the TSC measurement results before and after rapid thermal annealing (RTA) process, oxygen vacancy is concluded to be the origin of the traps. TSC characteristics with thermal annealing in the MIM BST capacitor have shown the same trends with the current-voltage (I-V) and current-time (I-t) characteristics. This means that the TSC measurement is one of the effective methods to characterize the traps in the BST thin film.

Removal of Cyanogenic Compounds in Apricot Kernel during Heating Process (가열조리방법을 통한 행인 내 시안화합물의 저감화)

  • Apricot Kernel, consumed as herbal medicine, contains amygdalin which generate HCN upon hydrolysis. Dyspnea was reported by ingesting large amount of apricot kernel, and neurological disorders such as tropic ataxic neuropathy or konzo were known as chronic toxicity of amygdalin. Other cyanogen containing plants, including flaxseed and almond, are consumed as food around the world. Moreover, some of them are promoted as functional food, leading to higher consumption, and posing health risk by cyanogenic components. The objective of this study was to find a method for the reduction of the cyanogenic compound, using apricot kernel as a model food. The most effective reduction was obtained by boiling the slices of the kernel for one hour in pH 1 HCl solution, showing 90% removal. However, the common process known to reduce the cyanogen contents, i.e., long incubation at the low temperature, did not show significant change in cyan concentration. Our data contribute to the safety of the plants containing cyanogenic compounds if they were to be developed as foodstuff.

Physicochemical Properties and Cu Sorption of the Biochar Derived from Woody Biomass (목질계 바이오매스에서 생산된 바이오차의 물리화학적 특성 및 Cu 흡착제거 특성)

  • In this study, the adsorption of $Cu^{2+}$ from aqueous solution by the biochar derived from woody biomass at different pyrolysis temperatures has been investigated. The woody biomass wastes used in this study were branch of willow ($Salix$ $koreensis$ $Andersson$) and bark of chestnut ($Castanea$ $crenata$ $var.$ $dulcis$). Three biochar samples prepared by heating each biomass at temperature of $300^{\circ}C$, $500^{\circ}C$, and $700^{\circ}C$were tested for the adsorption capacity of Cu. Also the physicochemical properties of the developed biochars were studied using different characterization techniques such as FT-IR, SEM, BET surface area, and cation exchange capacity (CEC). The adsorption of Cu could be well described by Langmuir model for both willow and chestnut biochars with $R^2{\geq}0.98$. The maximum adsorption capacities of the biochar produced at $700^{\circ}C$ from the Langmuir equation were found to be 12.5 mg $g^{-1}$ and 16.9 mg $g^{-1}$ for willow and chestnut, respectively. Chestnut biochar was found to interact more effectively with the active sites available for Cu, resulting higher removal of Cu(II) than wiloow biochar. Ion exchange and surface complexation found to be the main mechanisms involved in the adsorption process. This study demonstrated the feasibility of the biochars derived from woody biomass to be as a low-cost potential adsorbent for heavy metals as Cu(II) removal in aquatic system.
