Antioxidant Effect of Garlic Supplement against Exercise-Induced Oxidative Stress in Rats (운동으로 유발된 산화 스트레스와 마늘의 항산화 작용)

  • This study was to investigate lipid peroxidation, antioxidant enzyme activity and DNA damage after exercise, and the protective effect of garlic against exercise-induced oxidative stress. Male Sprague-Dawley rats(4 weeks old) were randomly divided into three groups of 6 rats each; control group(Con) without garlic and exercise, Ex group with exercise alone, and Ex-G group with 2% garlic and exercise. For 4 weeks, rats were given diets containing 15% corn oil and 1% cholesterol with or without garlic. The swimming was selected as a model for exercise performance. Rats swam for 40 min a day, for 5 days a week. Group Ex and Ex-G showed significant lowering in body weight gain and fat accumulation compared to control. No significant changes were observed in levels of plasma cholesterol and triglyceride among three groups, demonstrating that exercise and garlic had no effects on changes of blood lipid. This finding of blood lipid seems to be due to higher plant sterol content in corn oil. The DNA tail moment of lymphocytes showed greater tendency in Ex and Ex-G than in control, but garlic supplements failed to suppress DNA damages. Compared to control, Ex had higher plasma TBARS which was lowered to the control's level in Ex-G with 2% garlic supplementation(p<0.05). Ex-G led to a higher hepatic superoxide dismutase(SOD) activity than control and Ex(p<0.05). Activity of hepatic catalase was also increased in Ex-G, while in Ex it was significantly low(p<0.05). It seemed that TBARS levels were related to the activities of SOD and catalase, and that garlic contributed to increasing the enzyme activities and led to decrease of TBARS. These results demonstrate that lipid peroxidation and DNA damage occur as a consequences of oxidative stress after exercise, and that antioxidant defense against oxidative stress could be enhanced by garlic supplementation through the induction of antioxidant enzymes. However, further investigations should be done on the garlic effect on DNA damage.

Effects of Red-Yeast-Rice Supplementation on Bone Mineral Density and Bone Mineral Content in Overiectomized Rats (홍국의 첨가가 난소절제 쥐에서 골밀도 및 골함량에 미치는 영향)

  • This study was performed to evaluate the effect of red-yeast-rice on bone metabolism in overiectomized (OVX) rats. Forty female Sprague-Dawley rats (body weight 210 $\pm$ 5 g, 9 weeks old age) were divided into two groups. One group were OVX, and the other group received sham operation (SHAM), and received either control diet (20% casein) or a red-yeast-rice power supplemented diet (0.1%) for 9 weeks. And then each rat group was further divided into control diet (casein 20%) and red-yeast-rice powder supplemented (0.1%) diet group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur on 5, 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, IGF-l and calcitonin were analyzed using radioimmunoassay kits. Urinary Ca and P excretion were not significantly different among the groups. Within the OVX group, the red-yeast-rice group had a lower crosslinks value than the casein group. Therefore the red-yeast-rice supplemented groups had a lower bone resorption ratio than the casein group in the ovariectomized rats. And, the red-yeast-rice group had significantly higher IGF-l hormone than casein group in ovariectomized rats. The red-yeast-rice group had higher spine bone mineral content than those of control group within the OVX groups. This study was an important first step in establishing that the observed beneficial effects of red-yeast-rice on bone, and this study also established the need for a study on the long-term effect of this supplement in a human.

Effects of Pinitol Supplementation and Strength Training on Anaerobic Performance and Status of Energy Substrates in Healthy Young Men

  • To assess the effect of pinitol supplementation and strength training for two weeks on the anaerobic capacity during and after exercise, and improvement of glucose metabolism during the recovery period of muscular fatigue with repeated acute bouts of cycling exercise, a total of 24 healthy young men were recruited and randomly and equally divided into three groups; pinitol supplementation group (PSG), placebo group (PLG), and control group (CON). Using a randomized double-blinded design, subjects in PSG were provided pinitol supplement, consumed orally 1.2 g/day, and participated in the resistance exercise program and cycling exercise for two weeks. Subjects in PLG underwent the same protocol as those in PSG but consumed the same amount of placebo. No supplementation and exercise program was given to CON. Before and after the intervention, all subjects were tested for their anaerobic capacities evaluated by Wingate test twice separated by 30 min. During the test, peak anaerobic power (PP), mean anaerobic power, total work, and fatigue index were evaluated During resting and recovery, blood samples were drawn and plasma pinitol, myo-inositol, chiro-inositol, insulin, free fatty acid, glucose, and lactate levels were analyzed After two weeks, PP and relative PP of the second biking were improved from the first biking in PSG only (p<0.05). No changes were found in all other variables of Wingate test in all groups. No statistical differences between groups and pre- and post-intervention were observed in concentrations of pinitol, myo-inositol, and chiro-inositol, but pinitol concentration was higher during recovery compared to the baseline in all groups and testings (p<0.05). Lactate level during recovery was higher than the resting level, but no other blood parameters were significantly changed. In conclusion, two weeks of pinitol supplementation in conjunction with short duration of anaerobic training in healthy young men did not induce any obvious benefits in terms of anaerobic capacity and energy metabolism Individual and/or population susceptibility may be one factor responsible for adopting pinitol supplementation.

Effects of Legume Supplementation on the Glucose and Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats (두류의 첨가가 당뇨병 쥐의 내당능과 지질대사 및 지질과산화에 미치는 영향)

  • The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. Long-Term Evaluation of the Adequacy of Korean Diet to Meet the Protein-Energy Requirement of Young Korean Male Adult (한국인(韓國人) 단백질(蛋白質)-열량(熱量) 소요량(所要量)에 대(對)한 연구(硏究) (장기간급식(長期間給食)에 의(依)한 한국식이(韓國食餌)의 적정성(適正性)에 대(對)한 평가(評價)))

  • To evaluate long-term metabolic response of free-living Korean subjects consuming local sources adequately or slightly below habitual intake. Subjects, six male medical students aged 23-25 years were free-living, but consumed diets in the metabolic unit. They were healthy as determined from medical laborarory tests. They belong to the Korean middle-class. Temperature of outdoor during study were ranged between -2 to $26^{\circ}C$, whereas the indoor were $18-22^{\circ}C$. Animal protein ranged between 25-30% of total calorie. The three meals and a vitamin supplement to meet recommendation were given daily. Three 50day periods following the design were used to test the protein-energy adequacy of the diet. Urinary nitrogen excretion was measured daily. Fecal samples were pooled for the last 5 days of each week. Nitrogen balance was computed using 5mg N/Kg skin and miscellaneous losses. Body weight was measured daily. The study was covered on 126 metabolic observations weekly. Body weight changes were relatively stable with the energy intake of 37-44Kca1/kg, and N-balances were also relatively stable, but it was influenced by physical and or mental stress somehow. It could be assumed that the mean protein and energy requirement were $190{\pm}10mg$ N/Kg with $41{\pm}2$ Kcal/kg, and the mean digestibility of protein was 81% on these subjects with the experimental conditions. The protein requirement, however, could be cut down somewhat with some limited increaing of energy intake under parameters of N-balance and maintaining body weight. It was also observed a tendency of the requirement were getting smaller with the increase of body size.

Purpose: This study was intended to consider through a bibliographical comparison between the system of Kabo-Bon ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ and that of Shinehook-Pan ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$. 2. Method: This study was based on the ${\ulcorner}$Hamsansachon Dongyi Soose Bowon old Kabo-Bon${\lrcorner}$, which was compared with a bibliographical data from Shinchook-Pan ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ and the collection of manuscripts left unpublished in Lee Je-ma's lifetime. 3. Result & Conclusion: Exterior-Interior Pathological system was found in the system of Kabo-Bon ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$, and it might be referred to that of Jang Joong-kyung's ${\ulcorner}$Shanghan Ron${\lrcorner}$. There were no 'the Discourse on the Origin of oriental Medicine' and 'the Discourse on the General Health Maintenance' in the Kabo-Bon ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$. And it was supposed to be many changes of contents on 'the Discourse on Nature and Order', 'the Discourse on the Four Principles', 'the Discourse on the Establishment and Supplement' and 'the Discourse on Viscera and Bowels' in the Shinchook-Pan ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ compared with those in the Kabo-Bon ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$.

Effect of Momordica charantia on Glucagon Secretion in High-fat diet(HFD)/Streptozotocin(STZ)-induced Diabetic Rat (고지방식이(HFD)/stereptozotocin(STZ) 유도 당뇨모델에서 여주가 글루카곤 분비에 미치는 영향)

  • In present study, we investigated the antidiabetic effect of Momordica charantia(as well known "bitter melon"). This study was conducted to determine antidiabetic mechanism of Bitter Melon Extract (BME). We measured blood glucose, insulin, glucagon level in a Sprague-Dawley rat model of high-fat diet/streptozotocin(HFD/STZ)-induced diabetes. Five experimental groups were used: normal, HFD/STZ, BME 62.5 mg/kg HFD/STZ, BME 125 mg/kg HFD/STZ and BME 250 mg/kg HFD/STZ. BME was orally administered to the rats every other day for 9 weeks. Results showed that fasting blood glucose levels were significantly lower in the BME 125 mg/kg(150.17 ± 20.22 mg/dL) and 250 mg/kg(124.17 ± 22.17 mg/dL) groups than in the vehicle group(188.83 ± 26.63 mg/dL)(p<0.05). In addition, glucagon levels were lower in the three BME treatment groups than in the vehicle group(p<0.05). Oral glucose tolerance tests revealed that the BME 250 mg/kg group had significantly(p<0.05) reduced 120-minute blood glucose levels and areas under the curve. Our results suggest that BME induces antidiabetic effects via the reduction of glucagon and blood glucose levels.

Development of Fruit Harvest Basket and Assessment of its Effectiveness

  • Objective: The purpose of this study is to develop products that prevent muscular skeletal disease of farm workers, which would bring improvement for an efficient harvesting work. Background: A pretest for product development and a survey research was carried out to inspect the problems of current harvesting work procedure. The product was developed and assessed based on ideas to supplement and improve these weak points. Method: A total of twenty men were recruited to evaluate the effects of harvest baskets on the upper-limb muscle activity, working hours. For the usability evaluation, electromyogram and working hours of previous working method (disuse of harvest basket, one-hand carriage of the basket) and new working method with the usage of newly-developed harvest basket was examined. The whole anterior deltoid, biceps brachii and erector spinae on both sides, which make a total of six muscle parts, were used for this experiment. The results were analyzed using ANOVA with muscle activity (%MVC) of each three forms of work and required work time from SPSS 18.0. Results: According to the test result, all muscles did not demonstrate any statistically significant difference, with an exception of the backbone erector (right). The muscle activity of backbone erector (right) in the work method that uses the harvest basket developed in this study was detected to be approximately 23% less than that of the previous working method. Moreover, compared to the previous working method, the required work time decreased to a statistically significant degree. Conclusion: As such, the decrease in the amount of waist muscle usage at harvesting work would enable the prevention of muscular skeletal disease and stabilization of lumbar spine. Application: Which in turn would increase the effectiveness and reduce personnel expense (labor costs) while enhancing productivity with a decreased working hours.

Enhancing Effect of Pleurotus ostreatus Extracts on Mouse Spleen and Cytokine Cells Activation (느타리버섯 물 추출물 투여에 의한 마우스 비장세포 및 사이토카인 활성효과)

  • Pleurotus ostreatus have been used as a traditional remedy and food source. Its anti-inflammatory, anti-oxidation effects have been previously reported. The production of cytokines (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$) secreted by LPS- and non LPS-stimulated macrophages, were detected by ELISA assay using a cytokine kit. Mouse splenocytes proliferation increased with Pleurotus ostreatus water extracts supplement at 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after a 48hr pre-treatment with the mitogen (ConA or LPS). The cytokine production (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$), measured by a cytokine ELISA kit, increased on water extracts supplementation. This in vitro study suggested that supplementation with Pleurotus ostreatus water extracts may enhance the immune function by regulating the splenocyte proliferation and enhancing cytokine production.

Effects of Red-Yeast-Rice Supplementation on Blood Lipids and Relations among Cholesterol, Bone Markers, and Hormones in Overiectomized Rats (난소 절제 쥐에서 홍국의 첨가가 혈중 지질에 미치는 영향과 혈중 콜레스테롤과 골대사 지표 및 호르몬과의 관련성)

  • This study was performed to evaluate the effect of red-yeast-rice on blood lipids and also to investigated the relation of blood lipids, bone related hormones, and bone mineral density in overiectomized rats. Forty female rats (body weight $210\pm5$g) were divided into two groups. The rats in the first group were ovariectomized which the others received sham operation (SHAM). And then each rat group was further divided into control diet $(casein\;20\%)$ and red-yeast-rice powder supplemented $(0.1\%)$ diet group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus in spine and femur. Bone formation and bone resorption were measured by serum osteocalcin and urinary deoxypyridinoline (DPD) crosslinks value. Serum growth hormone, IGF-1 and calcitonin were analyzed using radioimmunoassay kits. The red-yeast-rice group had significantly lower the blood triglyceride concentration and had lower the blood total cholesterol concentration than casein group did in ovariectomized rats. And the red-yeast-rice group than casein group in ovariectomized rats. The red-yeast-rice group had significantly lower the atherogenic index than control group within the ovariectomized groups. In the experimental rats, serum total cholesterol concentration was negatively correlated with femur BMC and crosslink value. Crosslink value was negatively correlated with spine BMC and IGF-1. Osteocalcin concentration was negatively correlated with crosslink value. Growth hormone was negatively correlated with crosslink value and positively correlated with spine BMC. The findings of this study suggest that red-yeast-rices are beneficial for blood lipid in ovariectomized rats. And when considering a population-based public health approach to lowering cholesterol and bone loss, and preventing coronary heart disease and osteoporosis, the red-yeast-rice dietary supplement could provide a new approach for the maintenance of healthier cholesterol and bone.
