  • Oxygen is necessary to sustain life, yet cellular oxygen metabolism creates destructive elements called free radicals. Free radicals are chemically unbalanced and carrying free electrons that can damage molecules, potentially damaging the cell itself. For this reason, many antioxidant products, including supplements and functional foods, are being developed. In particular, natural products are rich sources of pharmacologically active compounds. The purpose of this study was to investigate the antioxidant effects of target biomaterials in Korean traditional spices such as diallyl sulfide (DAS), capsaicin (CAP), and gingerol (GGR), and to investigate the response of the antioxidant defense system to oxidative stress by hydrogen peroxide ($H_2O_2$) compared to sulforaphane (SFN) in HepG2 cells. After the analysis of the cell viability using Cell Counting kit-8 (CCK-8) assay, we determined that the optimum levels were $200{\mu}M$ DAS, $25{\mu}M$ CAP, $50{\mu}M$ GGR, and $12.5{\mu}M$ SFN. Antioxidant enzymes were measured and protein expression was detected by Western blotting. All treatments showed a significant decrease in antioxidant enzyme activity such as superoxide dismutase, catalse, and glutathione peroxidase in HepG2 cells. Additionally, DAS, CAP, GGR and SFN increased the antioxidant system-related transcription factor Nrf2 which was found to be regulated by the activation of MAPK-JNK in this study. In conclusion, these results indicate the protective effects of DAS CAP, GGR, and SFN against $H_2O_2$-induced oxidative stress.

  • There is growing evidence that soy isoflavones play beneficial role in the prevention of cardiovascular disease. However, current data are not sufficient to determine the effective dose of isoflavone on improving plasma lipid profiles and total antioxidant capacity in Korean menopausal women. In the present study, 24 perimenopausal and 23 postmenopausal women were divided into 3 groups and were given soy isoflavone supplements of 100, 150, 200mg/day for 12weeks. Blood samples were taken before and after 6, 12weeks of soy isoflavone supplementation for analysis of total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride and total antioxidant status(TAS). In the perimenopausal women, serum total cholesterol level decreased significantly in all three groups after 12weeks of isoflavone supplementation(p< 0.05). However, serum LDL cholesterol level significantly lowered only in 150 and 200mg groups, and serum triglyceride level decreased significantly in 200mg group(p<0.05). In the postmenopausal women, the 150mg isoflavone supplementation group demonstrated significant effects on lowering serum total cholesterol, LDL cholesterol levels and raising HDL cholesterol level after 12weeks of isoflavone supplementation(p<0.05). Two hundred mg isoflavone supplementation group had a significantly higher TAS than other groups in both perimenopausal and postmenopausal women. (Korean J Nutrition 34(3) : 322∼329, 2001)

  • Lactobacillus (LAB) have been reported to exert both harmful and beneficial effects on human and animal health. Recently, it has been reported that dysbiosis and bacterial translocation contribute to liver fibrosis. However, the role of Gram-positive LAB in the situation of chronic liver diseases has not been yet elucidated. Liver injury was induced by bile duct ligation (BDL) in LAB or control-administered mice. Liver fibrosis was enhanced in LAB-administered mice compared with control-treated mice as demonstrated by quantification of Sirius-red positive area, hydroxyproline contents and fibrosis-related genes ($Col1{\alpha}1$, Acta2, Timp1, Tgfb1). Moreover, LAB-administered mice were more susceptible to BDL-induced liver injury as shown by increased ALT and AST level of LAB group compared with control group at 5 days post BDL. Consistent with serum level, inflammatory cytokines ($TNF-{\alpha}$, IL-6 and $IL-1{\beta}$) were also significantly increased in LAB-treated mice. Of note, LAB-treated liver showed increased lipoteichoic acid (LTA) expression compared with control-treated liver, indicating that LAB-derived LTA may translocate from intestine to liver via portal vein. Indeed, responsible receptor or inflammatory factor (PAFR and iNOS) for LTA were upregulated in LAB-administered group. The present findings demonstrate that administration of LAB increases LTA translocation to liver and induces profibrogenic inflammatory milieu, leading to aggravation of liver fibrosis. The current study provides new cautious information of LAB for liver fibrosis patients to prevent the detrimental effect of LAB supplements.

  • Purpose: The Cow's Milk-related Symptom Score (CoMiSSTM), which considers crying, regurgitation, stools, skin and respiratory symptoms, was developed as an awareness tool for evaluating cow's milk-related symptoms. The scoring ranges from 0 to 33. A score ≥12 was proposed as being likely cow's milk-related and suggestive of allergy to cow's milk. This study aimed to determine the age-related CoMiSSTM values in presumed healthy infants in Poland. Methods: This was a cross-sectional study conducted in well-child clinics in two locations. Parents of the presumed healthy infants aged ≤6 months were approached during a routine checkup/vaccination visit. The exclusion criteria were as follows: presence of acute or chronic diseases, preterm delivery, treatment with therapeutic formula, and use of any food supplements (except vitamins) or medications. Results: Data from 226 infants were obtained (median age [Q1-Q3], 4 months [3-4]). The overall median (Q1-Q3) and mean (standard deviation) CoMiSSTM values were 4 (2-7) and 4.7 (3.5), respectively. The 95th percentile was 11. Scores on some, albeit not all, components of the CoMiSSTM significantly differed between age groups (crying, stools) or feeding type groups (stools and skin symptoms). Eleven children (4.9%) scored ≥12. Conclusion: This study adds to earlier age-related CoMiSSTM data by providing CoMiSSTM values in presumed healthy infants in Poland.

  • This study was performed to determine the health and nutritional risk factors associated with hypertension in Koreans over the age of 50 in a high-income class (more than twice as much family income as the 2005 Korean minimum cost of living, 668,540 Won). A total of 505 subjects aged over 50 from the 2005 Korean National Health and Nutrition Examination Survey (KNHANES) were divided into two groups: A hypertension group (HG) (N=151, Systolic Blood Pressure ${\geq}$140 mmHg or Diastolic Blood Pressure ${\geq}$90 mmHg) and normal group (NG) (N=354). Subjects who took hypertension medicines or underwent diet therapy were excluded. In HG, mean daily alcohol intake and the amount of alcohol consumption per one occasion were significantly higher than in NG, respectively. A greater number of hypertension subjects answered that they drank alcohol to reduce stress as compared to normal subjects. HG also took fewer dietary supplements than NG. Mean body mass index (BMI), waist circumference, fasting blood sugar level, and 2 hour postprandial blood sugar following a glucose load were significantly higher in HG than in NG, respectively. Also, iron, thiamin, and niacin intakes and the consumption frequency of seaweeds were significantly lower in HG than in NG, respectively. Finally, obesity (BMI ${\geq}$25 $kg/m^2$), abdominal obesity (waist circumference ${\geq}$90 cm for males, ${\geq}$ 80 cm for females), high blood sugar level 2 hours after an oral glucose load (${\geq}$140~200 mg/dl), and hypertriglyceridemia (serum TG ${\geq}$200 mg/dl) were related to a significantly higher risk of hypertension in the subjects (odds ratio: 1.884~3.040). In conclusion, dietary factors such as higher alcohol consumption; lower intakes of iron, thiamin, and niacin; lower consumption frequency of seaweeds; and metabolic syndrome were associated with hypertension in the study subjects.

  • The juvenile red variant of the sea cucumber Stichopus japonicus is distributed worldwide. It is a valuable food source in Korea, China, and Japan. Major issues in farming the juvenile red variant sea cucumber include growth rates and disease resistance. In recent years, studies have focused on feed additives to enhance immune system and health. In this study, we used a common diet (CD), a nutritional diet (ND; mixed nutritional supplements), and an immunity diet (ID; mixed probiotics, spirulina, and levan) and compared the growth rates and immunity of juvenile red variant sea cucumbers fed the experimental diets for 12 weeks. The growth and survival rates in the ID group were significantly increased (P>0.05). This suggests that the feed additives positively influenced immunity and growth in the ID group. However, the immune activity was exhibit a stabilizing effect, and further investigation of immune effects is required.

  • Platycodon grandiflorum root (Platycodi radix) saponins, platycosides, have been used as health supplements and food items for the treatment of respiratory disorders and pulmonary diseases. Deglycosylated saponins have been known to exert stronger biological effects than their glycosylated forms. In the present study, glycosylated platycosides in Platycodi radix extract were biotransformed into deglycosylated 3-O-β-ᴅ-glucopyranosyl platycosides, including 3-O-β-ᴅ-glucopyranosyl platycodigenin, 3-O-β-ᴅ-glucopyranosyl polygalacic acid, and 3-O-β-ᴅ-glucopyranosyl platyconic acid, by pectinase from Aspergillus aculeatus. This is the first report on the quantitative enzymatic production of 3-O-β-ᴅ-glucopyranosyl platycosides. The chemical structures of 3-O-β-ᴅ-glucopyranosyl platycosides were identified with LC/MS. Moreover, the biotransformation pathways of the three types of platycosides in Platycodi radix into 3-O-β-ᴅ-glucopyranosyl platycosides were established.
