A Genome Wide Association Study on Age at First Calving Using High Density Single Nucleotide Polymorphism Chips in Hanwoo (Bos taurus coreanae)

  • Age at first calving is an important trait for achieving earlier reproductive performance. To detect quantitative trait loci (QTL) for reproductive traits, a genome wide association study was conducted on the 96 Hanwoo cows that were born between 2008 and 2010 from 13 sires in a local farm (Juk-Am Hanwoo farm, Suncheon, Korea) and genotyped with the Illumina 50K bovine single nucleotide polymorphism (SNP) chips. Phenotypes were regressed on additive and dominance effects for each SNP using a simple linear regression model after the effects of birth-year-month and polygenes were considered. A forward regression procedure was applied to determine the best set of SNPs for age at first calving. A total of 15 QTL were detected at the comparison-wise 0.001 level. Two QTL with strong statistical evidence were found at 128.9 Mb and 111.1 Mb on bovine chromosomes (BTA) 2 and 7, respectively, each of which accounted for 22% of the phenotypic variance. Also, five significant SNPs were detected on BTAs 10, 16, 20, 26, and 29. Multiple QTL were found on BTAs 1, 2, 7, and 14. The significant QTLs may be applied via marker assisted selection to increase rate of genetic gain for the trait, after validation tests in other Hanwoo cow populations.

  • 본 시험은 한우연구소 시험우사와 표고가 700 m 지역에 위치한 개량목초지 (오차드그라스, 티머시, 톨페스큐 우점)에서 2014년 5월~10월동안 수행되었는데, 방목 처리구별 방목지를 선정하고 3구역으로 나눈 후, 방목지 초지식생을 고려하여 윤환방목을 4회 (1회당 38.5일) 실시하였다. 국립축산과학원 한우연구소에서 암소 육성우 및 성우 27두(시험구별 9두)를 공시하여 2014년 5~10월동안 수행하였다. 방목대상지는 오차드그라스, 티머시, 톨페스큐가 우점한 해발표고 700 m의 개량목초지를 이용하였다. 시험구 배치는 대조구, 처리1구, 처리2구로 구분하였으며, 대조구가 우사관리구 (건초급여와 농후사료급여), 처리1구는 방목사육과 농후사료 급여구, 처리2구는 방목사육과 농후사료 급여와 미량광물질제 급여구로 처리하였다. 본 연구에 사용된 미량광물질제는 마그네슘, 셀레늄, 반추위보호콜린이 포함된 시제품제제 (30g/두/일)를 급여하였다. 사료급여기준은 2012 한국 사양표준 (한우)에 따라 성장단계별 표준 목표체중 도달 또는 체중별 영양소요구량 충족을 위해 영양조절 급여를 수행하였다. 처리구별 방목지를 선정하고 각 세부적으로 3구역으로 나눠 5~10월 중 윤환방목을 실시했다. 측정항목은 생초수량, 체중, 사료섭취량, 혈중 대사물질, 경제성 분석, 수태율을 실시하였다. 윤환방목지의 생초수량 및 건물수량은 처리1구와 처리2구의 생초 총 생산량은 30,473.9 kg/ha, 30744.3 kg/ha였으며 방목지간의 차이를 보이지 않았다(P>0.05). 한우 14개월령 목표체중 240 kg에 모든 처리구가 도달했으며 생후 15개월령 체중은 대조구, 처리1구, 처리2구가 각각 259.9, 257.7 및 262.1 kg였다. 일당증체량은 대조구, 처리1구, 처리2구가 각각 평균 0.52, 0.51, 0.54 kg으로 시험구간에 차이가 없었다. 혈중 대사물질 중 글루코스의 함량이 처리1구(방목사육)에서 높았다. 수태율은 대조구, 처리1구, 처리2구가 88.9%, 100%, 100%였다. 시험구 간에 경제성 분석을 실시한 결과 두당 비용절감이 대조구에 비해 시험1구와 시험2구가 각각 143,790, 125,970원으로 나타났다.

National Genetic Evaluation (System) of Hanwoo (Korean Native Cattle)

  • Hanwoo (Also known as Korean native cattle; Bos taurus coreanae) have been used for transportation and farming for a long time in South Korea. It has been about 30 yrs since Hanwoo improvement began in earnest as beef cattle for meat yield. The purpose of this study was to determine the trend of improvement as well as to estimate genetic parameters of the traits being used for seedstock selection based on the data collected from the past. Hanwoo proven bulls in South Korea are currently selected through performance and progeny tests. National Hanwoo genetic evaluations are implemented with yearling weight (YW), carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS). Yearling weights and MS are used for selecting young bulls, and EMA, BF, and MS are used for selecting proven bulls. One individual per testing room was used for performance tests, and five individuals per room for progeny tests. Individuals tested were not allowed to graze pasture, but there was enough space for them to move around in the testing room. Feeds including roughages and minerals were fed ad libitum, and concentrates were provided at the rate of about 1.8% of individual weight. Overall means of the traits were $352.8{\pm}38.56$ kg, $335.09{\pm}44.61$ kg, $77.85{\pm}8.838\;cm^2$, $8.6{\pm}3.7$ mm and $3.293{\pm}1.648$ for YW, CW, EMA, BF and MS. Heritabilities estimated in this study were 0.30, 0.30, 0.42, 0.50 and 0.63 in YW, CW, EMA, BF and MS, respectively, which are similar to results from previous research. Yearling weight was 315.54 kg in 1998, and had increased to 355.06 kg in 2011, resulting in about 40 kg of improvement over 13 yrs. YW and CW have improved remarkably over the past 15 yrs. Breeding values between 1996 and 2000 decreased or did not change much, but have moved in a desirable direction since 2001. These improvements correspond with the substantial increase in use of animal models since the late 1990s in Korea. Hanwoo testing programs have practically contributed to the improvement in aspects of quality and quantity. In sum, the current selection system is good enough to accommodate circumstances where fewer sires are used on many more cows. Although progeny tests take longer and cost more, they seem to be appropriate under the circumstances of the domestic market with its higher requirement for better meat quality. Consequently, accumulative data collection, genetic evaluation model development, revision of selection indices, as well as cooperation among farms, associations, National Agricultural Cooperative Federation, universities, research institutes, and government agencies must be applied to the Hanwoo selection program. All these efforts will assist the domestic market to secure a competitive position against imported beef under Free Trade Agreement trade system and will provide farmers with higher profits as well as the public with a higher quality of beef.

수단그라스 사일리지 급여가 한우 경산우의 번식 성적에 미치는 영향 (Effects of Feeding of Sudangrass Silage on the Reproductive Characteristics of Hanwoo Cows)

  • This study was carried out between November 2008 and October 2009 in the experimental farm of Chunnam National University to investigate the effect of feeding of Sudangrass silage on the reproductive performance of Hanwoo cows. Two diets, rice straw or Sudangrass silage separately from concentrate were fed 19 Hanwoo cows. In control group (=CON), cows (n=9) were fed 5 kg (/head) rice straw and 3 kg (/head) commercial diet. In Sudangrass silage group (=SGS), cows (n=10) were fed 12.5 kg (/head) Sudangrass silage and 1 kg (/head) commercial diet. 1. Days to post-partum insemination were $78.8{\pm}25.3$ days for CON and $84.7{\pm}24.6$ days for SGS group and days to post-partum conception in CON or SGS were $90.1{\pm}24.1$ and $87.7{\pm}26.8$ days, respectively. 2. Post-partum conception rates for first service in CON or SGS was 66.7% and 90.0%, respectively, and caving interval was $375.1{\pm}46.7$ days for CON and $370.2{\pm}34.4$ days for SGS group. 3. Body Condition Scores of Hanwoo cows at artificial insemination was 5.36 for CON group and 4.93 for SGS group.

Effect of γ-aminobutyric acid producing bacteria on in vitro rumen fermentation, growth performance, and meat quality of Hanwoo steers

  • Objective: The present study aimed to evaluate the effects of γ-aminobutyric acid (GABA)-producing bacteria (GPB) on in vitro rumen fermentation and on the growth performance and meat quality of Hanwoo steers. Methods: The effects of GPB (Lactobacillus brevis YM 3-30)-produced and commercially available GABA were investigated using in vitro rumen fermentation. Using soybean meal as a substrate, either GPB-produced or commercially available GABA were added to the in vitro rumen fermentation bottles, as follows: control, no additive; T1, 2 g/L GPB; T2, 5 g/L GPB; T3, 2 g/L autoclaved GPB; T4, 5 g/L autoclaved GPB; T5, 2 g/L GABA; and T6, 5 g/L GABA. In addition, 27 Hanwoo steers (602.06±10.13 kg) were subjected to a 129-day feeding trial, during which they were fed daily with a commercially available total mixed ration that was supplemented with different amounts of GPB-produced GABA (control, no additive; T1, 2 g/L GPB; T2, 5 g/L GPB). The degree of marbling was assessed using the nine-point beef marbling standard while endotoxin was analyzed using a Chromo-Limulus amebocyte lysate test. Results: In regard to in vitro rumen fermentation, the addition of GPB-produced GABA failed to significantly affect pH or total gas production but did increase the ammonia nitrogen (NH3-N) concentration (p<0.05) and reduce total biogenic amines (p<0.05). Animals fed the GPB-produced GABA diet exhibited significantly lower levels of blood endotoxins than control animals and yielded comparable average daily gain, feed conversion ratio, and beef marbling scores. Conclusion: The addition of GPB improved in vitro fermentation by reducing biogenic amine production and by increasing both antioxidant activity and NH3-N production. Moreover, it also reduced the blood endotoxin levels of Hanwoo steers.

Two diets, rice straw or whole crop barley silage separately from concentrate were fed 11 Hanwoo heifers and 26 Hanwoo cows. In control group (=CON), heifers (n=6) and cows (n=13) were fed 7 kg (/head) rice straw and 4 kg (/head) commercial diet. In whole crop barley silage group (=WBG), heifers (n=5) and cows (n=13) were fed 8 kg (/head) whole crop barley silage and 1 kg (/head) commercial diet. 1. Conception rates for first service in CON or WBG heifers were 66.7% (4/6) and 60.0% (3/5), respectively, and the services per conception cows were $1.5{\pm}0.2$ for CON and $1.4{\pm}0.2$ for WBG group. 2. Days to post-partum insemination were $106.6{\pm}26.3$ days for CON and $85.6{\pm}12.6$ days for WBG group, and days to post-partum conception in CON or WBG were $128.4{\pm}27.1$ and $96.8{\pm}16.8$ days, respectively. 3. Post-partum conception rates for first service in CON or WBG were 76.9% (10/13) and 84.6% (11/13), respectively, and caving interval was $418{\pm}50.7$ days for CON and $392.8{\pm}20.7$ days for WBG group.

Effect of supplementation of acetaminophen on the intake and growth of Hanwoo steers in summer

  • The aim of this study was to investigate the effects of dietary supplementation of acetaminophen on feed intake and growth performance of Hanwoo steers in the summer. Eight steers participated in a twelve-week experiment. The steers were randomly assigned into one of two treatments: Control and acetaminophen supplementation. Commercial acetaminophen was top-dressed to a concentrate mix at a rate of 3 g/kg based on dry matter. Individual feed intake was measured daily during the entire period of the experiment, and the intake of forage and concentrate diet were measured separately. The body weight of the steers were measured every 4 weeks. During the experimental period, ambient temperatures were recorded using an automatic temperature and humidity recorder. Acetaminophen had no effect (p > 0.05) on body weight, average daily gain, dry matter intake (DMI) and feed conversion ratio. Numerically, total DMI and concentrate DMI in the acetaminophen treated group were 5.7% and 7.6% lower than those of the control group, respectively, and the daily gain was 9.7% higher. As a result, the feed conversion rate of the acetaminophen treated group improved by 17% compared to the control; however, it was not statistically significant (p = 0.237). The results of this study suggest that acetaminophen supplementation may reduce the feed conversion ratio of Hanwoo steers under high temperature stress. However, further studies are needed to confirm the optimal level of acetaminophen supplementation which has a significant effect on the intake and weight gain of Hanwoo steers in the summer.

Effect of Feeding a Mixed Microbial Culture Fortified with Trace Minerals on the Performance and Carcass Characteristics of Late-fattening Hanwoo Steers: A Field Study

  • This study was conducted to determine the effects of feeding a trace minerals-fortified microbial culture (TMC) on the performance and carcass characteristics of late-fattening Hanwoo steers. A mixture of microbes (0.6% [v/w] of Enterobacter sp., Bacillus sp., Lactobacillus sp., and Saccharomyces sp.) was cultured with 99% feedstuff for ensiling and 0.4% trace minerals (zinc, selenium, copper, and cobalt). Sixteen late-fattening steers (mean age, 21.8 months) were allocated to two diets: a control diet (concentrate mix and rice straw) and a treated diet (control diet+3.3% TMC). At a mean age of 31.1 months, all the steers were slaughtered. The addition of TMC to the diet did not affect the average daily weight gain of the late fattening steers, compared with that of control steers. Moreover, consuming the TMC-supplemented diet did not affect cold carcass weight, yield traits such as back fat thickness, longissimus muscle area, yield index or yield grade, or quality traits such as meat color, fat color, texture, maturity, marbling score, or quality grade. However, consumption of a TMC-supplemented diet increased the concentrations of zinc, selenium, and sulfur (p<0.05) in the longissimus muscle. With respect to amino acids, animals consuming TMC showed increased (p<0.05) concentrations of lysine, leucine, and valine among essential amino acids and a decreased (p<0.05) concentration of proline among non-essential amino acids. In conclusion, the consumption of a TMC-supplemented diet during the late-fattening period elevated the concentrations of certain trace minerals and essential amino acids in the longissimus muscle, without any deleterious effects on performance and other carcass characteristics of Hanwoo steers.

High-level dietary crude protein decreased backfat thickness and increased carcass yield score in finishing Hanwoo beef cattle (Bos taurus coreanae)

  • Recently, a high level of dietary crude protein (CP) has become of interest as a possible practice to improve the carcass quality of beef cattle, and its level has been increasing in the field. However, there is little scientific evidence that supports this. This study was conducted to test whether a high dietary CP level would improve growth performance, body metabolism, and carcass traits in Hanwoo beef cattle. A total of 32 Hanwoo finishing beef cattle (18 multiparous cows, six heifers, and eight steers) participated in a 12-weeks feeding trial. Two kinds of total mixed rations were prepared to contain two different CP; 156 g/kg for the control (CON) and 173 g/kg of CP for the treatment (HCP), while maintaining a similar level of metabolizable energy. The experiment was ended when more than half of the steers reached the target body weight (730 kg). Blood was collected at the end of the experiment. After harvesting, the carcass trait was evaluated at the slaughterhouse according to Korean standards. The carcass yield score and grade were also calculated based on revised criteria. Overall, dry matter intake, average daily gain, blood metabolites concentration, and the carcass traits, except for backfat thickness and the yield score, did not differ between the treatments. The HCP had lower backfat thickness than those of CON. There was no difference in the carcass yield grade, but the yield score was higher in the HCP treatment. According to the newly revised carcass grading criteria, both yield score and grade were higher in HCP than in CON. Increasing CP supply decreased the carcass's backfat thickness without altering growth performance and body metabolism, resulting in improved yield score and grade. Therefore, feeding a high CP diet may be beneficial in the farm income, although it may also increase feed cost and nitrogen excretion to the environment.

Effects of feeding system on growth performance, plasma biochemical components and hormones, and carcass characteristics in Hanwoo steers

  • Objective: This study was conducted to compare growth performance, blood components and carcass traits by two feeding systems (concentrate with roughage separately [CON] vs total mixed ration [TMR]) in Hanwoo steers, and to learn the relationship between blood components during fattening or finishing phases and carcass traits in Hanwoo steers. Methods: Sixty steers aged 8 months were allotted to two feeding systems and fed similar amounts of average dry matter and total digestible nutrient throughout whole experimental period according to each feeding program. Steers were weighed monthly, taken blood at the end of growing, fattening and finishing periods, and slaughtered at 30 month of age. Results: Growing performance was higher (p<0.05) in the CON group compared to the TMR group during fattening and finishing periods. The CON group was lower (p<0.05) in blood aspartic acid transaminase, blood urea nitrogen and retinol levels during growing period, but higher in triglyceride and cholesterol levels during fattening and finishing periods compared to the TMR group. The CON group was greater (p<0.05) in rib-eye area, and lighter (p<0.05) red in meat color compared to the TMR group. In the correlation coefficients between blood components of steers and carcass traits, retinol had a negative (p<0.05) correlation with marbling score and rib-eye area. Leptin had a positive (p<0.05) correlation with back fat thickness. Blood cholesterol and triglyceride were positively (p<0.05) correlated with carcass weight and rib-eye area. Conclusion: Growth performance, carcass ribeye area and meat color showed a more desirable result in the CON compared to the TMR in Hanwoo steers. Assessing the accumulated data of carcass traits with blood components including hormones-particularly retinol, cholesterol, triglyceride, and leptin-during the fattening or finishing phases, it may be possible to find a biomarker for determining beef quality in living animals.
