Determination of Net Energy and Protein Requirements for Growth in Hanwoo Steers by Comparative Slaughter Experiment (비교도체법에 의한 한우 거세우의 증체에너지 및 단백질 요구량)

  • Data from a comparative slaughter experiment with two hundreds of Korean native (Hanwoo) beef steers were utilized to determine net energy and protein requirements for growth (NEg and NPg). Eight randomly selected steers were used in the initial slaughter group of 6 month of age and the remaining steers allocated to treatments within two groups. The restricted groups were fed 1.2 to 1.5% concentrate per kg body weight (BW) in the growing period (6~12 month of age), 1.7 to 1.8% concentrate per kg BW in the early fattening period (13~18 month of age), and concentrate feed ad libitum in the late fattening period (19~30 month of age). Ad libitum groups were fed concentrate ad libitum through the whole period. Rice straw was offered ad libitum for 24 month of two feeding systems. The steers were slaughtered when animals reached every 2 month from 8 to 30 month of age. For all body composition determinations, whole empty body components were weighed, taken each proportional subsample was ground for chemical analysis. Equations developed with the pooled data to predicted NEg and NPg were similar to the equations of Japanese Feeding Standard for Beef Cattle (2000). First equations were developed to predicted NEg; NEg = 0.05332×LBW0.75×DG for restricted treatment and NEg = 0.04912×LBW0.75×DG for ad libitum treatment. Second equations were developed to predicted NPg; NPg = DG × (224.7-0.251×LBW) for restricted treatment and NPg=DG×(210.1-0.214×LBW) for ad libitum treatment.

Effect of Seed Mixture on the Forage Yield and Botanical Composition in the Hanwoo Grazing Pasture (한우 방목초지에서 목초 혼파조합이 식생구성과 수량에 미치는 영향)

  • This study was conducted to select the optimum seed mixture for beef cattle (Hanwoo) grazing in middle mountain area at 450m from sea level during 2 years from October 1, 1999. Seed mixtures were composed of four different treatment according to main species such as orchardgrass (T1), perennial ryegrass (T2), timothy (T3) and Italian ryegrass (T4). Grass composition rate of different seed mixtures was decreased mostly at first year of grazing, like 44.9% of orchardgrass in T1 plot, 27.6% of perennial ryegrass in T2 plot, 40.9% of timothy in T3 plot and 40.7% of Italian ryegrass in T4 plot, respectively. Main species in seed mixture were decreased in grazing pasture, but kentucky bluegrass and tall fescue were increased in all plots. Italian ryegrass in T4 plot was well grown in early grown stage and inhibited the growth of other grass species, and 44.2% of Italian ryegrass was maintained in second year in the grazing plot. Average dry matter yields of first you showed 6,673 kg/ha and second year was 10,177 kg/ha. Average TDN yields of different seed mixtures were 6,399 kg/ha in T1, 7,769 kg/ha in T2, 4,383 kg/ha in T3 and 7,146 kg/ha in T4 plot. Average herbage dry matter intakes by different seed mixtures during 2 years were 5,937 kg/ha in T1, 6,375 kg/ha in T2, 4,925 kg/ha in T3 and 6,456 kg/ha in T4. This result indicated that perennial ryegrass would recommended in main species of Hanwoo grazing pasture.

Effect of Protein Supplementation, O2 Concentration and Co-Culture on the Development of Embryos Produced by Nuclear Transfer Using Cultured Cumulus Cells in Hanwoo (Korean Cattle)

  • The effect of protein supplementation, $O_2$ concentration and co-culture on the development of embryos produced by nuclear transfer using cultured cumulus cell was investigated. Recipient oocytes and cumulus cells were obtained from the ovaries of the slaughtered Hanwoo cows. Donor cumulus cells were cultured in Dulbecco's modified Eagle medium containing 10% fetal bovine serum at 5% $CO_2$ in air at $38.5^{\circ}C$. The 1 to 6 passages of cumulus cells were isolated and used as donor cells. The in vitro matured oocytes were enucleated and then the isolated donor cells were introduced. One $15{\mu}s$ pulse of 180 volts was applied to induce the fusion between karyoplast and cytoplast. The fused embryos were activated with $10{\mu}M$ calcium ionophore for 5 min and 2 mM 6-dimethylaminopurine for 3 h. To examine the effect of protein supplementation, nuclear transfer (NT) embryos were cultured in one of the following 4 treatments : 1) CR1aa + 3 mg/ml BSA for 7 days ; 2) CR1aa + 10% FBS for 7 days ; 3) CR1aa + 1.5 mg/ml BSA + 5% FBS for 7 days ; and 4) CR1aa + 3 mg/ml BSA for first 3 days and then CR1aa + 1.5 mg/ml BSA + 5% FBS for 4 days. Culture took place at 5% $CO_2$, 5% $O_2$ and 90% $N_2$ at $38.5^{\circ}C$. Although there were no significant differences in cleavage rate among different protein supplements, the rates of blastocyst formation were significantly different. When NT embryos were cultured in the medium supplemented with only BSA, they could develop to only morula not to blastocyst. However, when FBS was supplemented, NT embryos developed to blastocyst stage. In order to investigate the effect of $O_2$ concentration and co-culture, NT embryos were cultured in CR1aa + 1.5 mg/ml BSA + 5% FBS with or without cumulus cell co-culture at an atmosphere of 5% $CO_2$ in air (20% $O_2$) or 5% $CO_2$, 5% $O_2$, 90% $N_2$ (5% $O_2$) at $38.5^{\circ}C$ for 7 days. The percentage of blastocyst development was significantly higher when the NT embryos were cultured at an atmosphere of 5% $O_2$ than that of 20% $O_2$ (p<0.05). However, there was no significant difference between with and without cumulus cell co-culture at an atmosphere of 5% $O_2$ or 20% $O_2$. Fifty embryos were transferred to 25 recipients and 5 recipients were pregnant at 100 days. From 5 pregnant cows, only one cow was delivered of female twin. In conclusion, the embryos reconstructed by enucleation of metaphase II oocytes and introduction of the cycling and quiescent cumulus donor cells in Hanwoo had developmental potential to term after embryo transfer to recipient cows.

The Effects of Fig Fermented Product Supplementation on Animal Performance, Serum Profile and Meat Quality in Hanwoo Bulls (무화과 발효물의 급여가 한우의 생산성, 혈액성상 및 육질에 미치는 영향)

  • We investigated the effect of fig fermented product(FFP) supplementation on growth performance, serum profile, carcass performance, meat performance and meat quality in 10 bulls of Korean cattle. Concentrates diet was supplemented with substrate fermented from fig fruit and leaves at 10% of the diet. The feed intake of FFP were slightly higher than the control, but the final weight showed no sifnificant difference between the two. Daily weight gain and feed intake were increased in FFP. The serum profile had no significant difference in the treatment. In carcass performance, the meat quantity grade of the treatment had no significant difference, but in meat quality grade the marbling score of FFP was significantly(P<0.05) increased therefore it showed a positive effect on meat quality grade. Also there was no significant(P<0.05) difference of meat cut performance in the treatment. Due to the proximate characteristics of longissimuss muscles the crude fat content of the FFP was significantly(P<0.05) increased. There was no significant difference(P<0.05) in physical characteristics ; pH level, meat color and heat loss of the treatment, but the shear force value and the cholesterol content of FFP significantly(P<0.05) decreased. Crude fat was increased(P<0.05) and cooking loss, shear force and cholesterol concentration were decreased. In fatty acids composition of the FFP, the linoleic acid from the longissimus increased significantly(P<0.05). In subcutaneous fat of longissimus of the FFP, C16:0(palmitic acid) significantly(P<0.05) decreased, but C18:1 significantly (P<0.05) increased. Therefore in FFP, the concentration of saturated fatty acid significantly decreased (P<0.05), but on the other hand the concentration of unsaturated fatty acids significantly (P<0.05) increased. In sensory evaluation of the FFP, the evaluation of odor increased slightly in a positive manner, also the appearance and the taste increased significantly(P<0.05). In conclusion when annexing additional fig fermented product to Hanwoo bulls, the carcass grade improves and the livestock production increases. Also the shear force, lower cholesterol, improved appearance and taste will open the doors to high quality meat production.

A Comparison of Discriminating Powers Between 14 Microsatellite markers and 60 SNP Markers Applicable to the Cattle Identification Test (소 동일성 검사에 적용 가능한 14 Microsatellite marker와 60 Single Nucleotide Polymorphism marker 간의 판별 효율성 비교)

  • When 14 microsatellite (MS) markers were applied in the identifying test for 480 Hanwoo, the discriminating power was estimated as $3.43{\times}10^{-27}$ based on the assumption of a random mating group (PI). This rate is 1,000 times higher than that of 60 single nucleotide polymorphism (SNP) markers. On the other hand, the power of the 60 SNP markers was estimated as $4.69{\times}10^{-20}$ and $8.02{\times}10^{-12}$ on the assumption of a half-sib mating group ($PI_{half-sibs}$) and a full-sib mating group ($PI_{sibs}$), respectively. These powers were 10 times and 10,000 times higher than those of the 14 MS markers. The results indicated that the total number of alleles (MS vs SNP = 146 vs 120) acted as a key factor for the discriminating power in a random mating population, and the total number of markers (MS vs SNP = 14 vs 60) was a dominant influence on the power in half-sib and full-sib populations. In the Hanwoo population, in which it was assumed that the entire population is the enormous half-sib group formed by the absolute genetic contribution of a few nuclear bulls, there will be only a 10 times difference in the discriminating power between the 14 MS markers and the 60 SNP makers. However, the probability of not excluding a candidate parent pair from the parentage of an arbitrary offspring, given that only the genotype of the offspring ($PNE_{pp}$) was 1,000 times higher as shown by the 14 MS markers than that by the 60 SNP markers. The strong points of SNP makers are the stability of the variation (low mutation rate) and automation of high-throughput genotyping. In order to apply these merits for the practical and constant Hanwoo identity test, research and development are required to set a cost-effective platform and produce a homemade apparatus for SNP genotyping.

Estimation of Genetic Parameters of Body Weight by Growth Periods for Hanwoo Using Animal Model (개체모형에 의한 한우의 성장단계별 체중의 유전모수 추정)

  • The objectives of this study were to investigate the genetic characteristics of body weight by growth periods for Hanwoo. A total of 1,736 records were used for body weight. The data for body weights were collected from 1990 to 2000 in Daekwanryong branch, National Livestock Research Institute(NLRI). Estimates of (co)variance components were obtained by derivative-free Restricted Maximum Likelihood (DF-REML). The results are summarized as follows; The means for the weights were 25.60, 79.31, 98.91, 145.40, 283.26, 392.32, 545.65kg at birth, 3, 4, 6, 12, 18, 24month postpartum, respectively. The effects of calving year-season were significant for the milk yield of cow. Heritability estimates of direct genetic effects for birth weight were 0.54(all), 0.52(female), 0.36(male) in modelⅠ, 0.45(all), 0.41(female), 0.24(male) in modelⅡ, and heritabilities estimates of direct genetic effects for 4 month(weaning) weight were 0.47(all), 0.33(female), 0.28(male) in modelⅠ, 0.38(all), 0.21(female), 0.21(male) in modelⅡ. Heritability estimates for male and female data differed from those for combined data. The estimates became smaller for the body weights at 12 month or later(0.13~0.05). The heritabilities of average daily gain were smaller than those for body weights, but showed that the similar pattern to body weights.

Imputation Accuracy from 770K SNP Chips to Next Generation Sequencing Data in a Hanwoo (Korean Native Cattle) Population using Minimac3 and Beagle (Minimac3와 Beagle 프로그램을 이용한 한우 770K chip 데이터에서 차세대 염기서열분석 데이터로의 결측치 대치의 정확도 분석)

  • Whole genome analysis have been made possible with the development of DNA sequencing technologies and discovery of many single nucleotide polymorphisms (SNPs). Large number of SNP can be analyzed with SNP chips, since SNPs of human as well as livestock genomes are available. Among the various missing nucleotide imputation programs, Minimac3 software is suggested to be highly accurate, with a simplified workflow and relatively fast. In the present study, we used Minimac3 program to perform genomic missing value substitution 1,226 animals 770K SNP chip and imputing missing SNPs with next generation sequencing data from 311 animals. The accuracy on each chromosome was about 94~96%, and individual sample accuracy was about 92~98%. After imputation of the genotypes, SNPs with R Square ($R^2$) values for three conditions were 0.4, 0.6, and 0.8 and the percentage of SNPs were 91%, 84%, and 70% respectively. The differences in the Minor Allele Frequency gave $R^2$ values corresponding to seven intervals (0, 0.025), (0.025, 0.05), (0.05, 0.1), (0.1, 0.2), (0.2, 0.3). (0.3, 0.4) and (0.4, 0.5) of 64~88%. The total analysis time was about 12 hr. In future SNP chip studies, as the size and complexity of the genomic datasets increase, we expect that genomic imputation using Minimac3 can improve the reliability of chip data for Hanwoo discrimination.

Warner-Bratzler Shear Force, Sarcomere Length, Total Collagen Contents and Sensory Characteristics of Hanwoo Beef (Korean Native Cattle) Quality Grade (한우육의 육질등급에 따른 부위별 전단력, 근절길이, 콜라겐의 함량 및 관능 특성)

  • This study was conducted to investigate the Warner-Bratzler shear force (WBS), sarcomere length, total collagen contents and consumer tests of different cuts of Hanwoo beef based on quality grade. Five cuts [Cheggt (striploin), Dngsim (loin), Moksim (chuck roll), Udoon (top round), Yanggi (brisket)] were obtained from Hanwoo beef (bulls and steers, 24-30 months old) and grouped by their quality grades ($1^{++}$, $1^+$, 1, 2 and 3). The WBS values of chuck cut in $1^{++}$, $1^+$ and 1 quality grade groups were 5.47 (kg/0.5 $inch^2$), 5.46 (kg/0.5 $inch^2$) and 6.54 (kg/0.5 $inch^2$), respectively, which were significantly lower than those of chuck in quality grade 3, which was 8.41 (kg/0.5 $inch^2$) (p<0.05). The sarcomere length of the quality grade groups were significantly longer for brisket (3.72 ${\mu}m$) of $1^{++}$ quality grade than for that of 3 quality grade brisket (p<0.05). Brisket of $1^{++}$ and 2 quality grade had significantly greater sarcomere lengths than strip loin and top round in the same quality grade (p<0.05). The total collagen contents of strip loins in the $1^{++}$, $1^+$, and 1 quality grade groups (1.17, 1.18 and 1.14 g/100 g, respectively) were significantly lower than those of the 3 quality grade group (1.63 g/100 g) (p<0.05). The overall sensory scores of tenderness and juiciness for the five cuts of $1^{++}$ quality grade were significantly higher than those of the other quality grade (p<0.05). Among the five cuts, loin had the highest tenderness and juiciness scores (p<0.05), whereas top round had the lowest tenderness and juiciness scores (p<0.05). Overall, there were significant differences in the WBS and sensory properties among quality grades and cuts (p<0.05). However, the sarcomere length and total collagen contents of chuck roll, top round and loin did not differ among quality grades.

The Effect of Alcohol Fermented Feedstuff Made of Byproducts on Fermentation Characteristics and Dry Matter Disappearance in the Rumen (비지박 및 맥주박을 이용한 알코올 발효사료의 반추위내 발효특성 및 건물 분해율에 미치는 영향)

  • A study was conducted to determine the effect of alcohol-fermented feedstuff formulated with byproducts on the fermentation characteristics and dry matter disappearance in the rumen. Dietary treatments were either a soybean curd-based alcohol-fermented feedstuff(AFS) and brewery grain-based alcohol-fermented feedstuff(AFB). The AFS and AFB are composed of 50% commercial beef cattle feed, 50% soybean curd dreg, 5% molasses and 0.5% yeast, and 25% commercial beef cattle feed, 25% brewery grain, 25% soybean curd dreg, 25% corn grit, 5% molasses and 0.5% yeast, respectively. The ruminally cannualted Korean cattle were utilized to investigate the change of ammonia, pH alcohol, volatile fatty acids, and DM digestibility at 0, 2, 4, 8 and 12 hr after feeding. The rumen ammonia concentrations were significantly lower in AFS and AFB with incubation time, especially at 6 hr incubation(AFS, 0.7 mg/dl; AFB, 1.5 mg/dl; control 2.5 mg/dl). Lower rumen pH was observed in AFS and AFB during the early stage of incubation, but no significant difference was found at late stage of incubation. The total VFA concentrations were not affected by diet treatments at 2 hr incubation time, but the concentration significantly decreased after that. The dry matter disappearance was significantly lower in AFS and AFB during the early stage of incubation. However, the dry matter disappearance of AFS and AFB was similar to that of control during the late stage of incubation. It is concluded that the industrial byproducts such as soybean curd dreg and brewery grain were effective materials to make an alcohol fermented feedstuffs and resulted in better fermentation characteristics in the rumen when both were applied to Hanwoo.

Methods for Genetic Parameter Estimations of Carcass Weight, Longissimus Muscle Area and Marbling Score in Korean Cattle (한우의 도체중, 배장근단면적 및 근내지방도의 유전모수 추정방법)

  • This study is to investigate the amount of biased estimates for heritability and genetic correlation according to data structure on marbling scores in Korean cattle. Breeding population with 5 generations were simulated by way of selection for carcass weight, Longissimus muscle area and latent values of marbling scores and random mating. Latent variables of marbling scores were categorized into five by the thresholds of 0, I, 2, and 3 SD(DSI) or seven by the thresholds of -2, -1, 0,1I, 2, and 3 SD(DS2). Variance components and genetic pararneters(Heritabilities and Genetic correlations) were estimated by restricted maximum likelihood on multivariate linear mixed animal models and by Gibbs sampling algorithms on multivariate threshold mixed animal models in DS1 and DS2. Simulation was performed for 10 replicates and averages and empirical standard deviation were calculated. Using REML, heritabilitis of marbling score were under-estimated as 0.315 and 0.462 on DS1 and DS2, respectively, with comparison of the pararneter(0.500). Otherwise, using Gibbs sampling in the multivariate threshold animal models, these estimates did not significantly differ to the parameter. Residual correlations of marbling score to other traits were reduced with comparing the parameters when using REML algorithm with assuming linear and normal distribution. This would be due to loss of information and therefore, reduced variation on marbling score. As concluding, genetic variation of marbling would be well defined if liability concepts were adopted on marbling score and implemented threshold mixed model on genetic parameter estimation in Korean cattle.
