Study on Ovum-pick up for Improvement of Embryo Transfer Efficiency in Hanwoo Cows (한우의 수정란이식 효율성 향상을 위한 생체난포란 채취에 관한 연구)

  • Commercial applications of OPU/IVP were to produce embryos and calves from high genetic cows. The aim of this present study was to compare the number of recovered oocytes and cultured In vitro produced embryos from Ovum Pick-up (OPU). OPU derived embryo production was carried out of oocytes by ultrasonographic guided follicular aspiration and then produced in vitro produced blastocysts by IVP culture system. In result, the rate of recovered oocytes was obtained 612 (57.2%) and 451(73.7) G1+G2 grade oocytes. No difference of recovered rate (51.1~62.1%) was seen in six donor. The rate of cleavage and blastocyst development were obtained 320 (70.9%) and 78 (24.4%) that was $3.3{\pm}0.4$ cleaved embryo and $0.9{\pm}0.2$ blastocysts per session. Cleavage rate of OPU oocytes in No. 6 donor was 90.6%, significantly (P<0.05) higher than that in the other donors, However, blastocysts was similar (25.8~30.0%). In conclusion, limited numbers of OPU oocytes had competent development when cultured in SOF culture medium.

Estimation of Genetic Parameters for Ultrasound and Carcass Traits in Hanwoo (한우의 초음파 측정 형질과 도체 형질의 유전모수 추정)

  • This study was conducted to estimate genetic parameters for ultrasound and carcass traits in Hanwoo. Heritabilities and genetic and phenotypic correlations were estimated for carcass and ultrasound measurements collected from Hanwoo cows (n=312) born at Hanwoo experiment station. Traits evaluated were eye muscle area (EMA), backfat thickness (BF), marbling score (MS) from carcass, and ultrasound eye muscle area (UEMA), ultrasound backfat (UBF), and ultrasound marbling score (UMS). Parameters were estimated using multi-trait animal models byderivative-free restricted maximum likelihood procedures. Estimated heritabilities for UBF, UEMA and UMS were 0.43, 0.23 and 0.32, while heritabilities for BF, EMA and MS were 0.33, 0.13 and 0.33 in fattened cows, respectively. Genetic correlations between ultrasound and carcass measurements were estimated to -0.19, -0.61, and -0.36 for UBF: UEMA, UBF: UMS, and UEMA: UMS in fattened cows, respectively. Phenotypic correlations between ultrasound and carcass measurements were 0.03, 0.13 and 0.26 for UBF: UEMA, UBF: UMS, and UEMA: UMS in fattened cows, respectively. As for the steer, genetic correlations between ultrasound and carcass measurements were 0.36, -0.80 and 0.27 for UBF: UEMA, UBF: UMS, and UEMA: UMS in steers, respectively. Phenotypic correlations between ultrasound and carcass measurements were 0.13, 0.07 and 0.41 for UBF: UEMA, UBF: UMS, and UEMA: UMS in steers, respectively. In conclusion, this finding would be very useful to implement into Hanwoo breeding program.

Fatty acid compositions, free radical scavenging activities, and antioxidative enzyme activities of high-preference and low-preference beef cuts of Hanwoo (Bos taurus coreanae) cows

  • Objective: This study compared fatty acid compositions and antioxidant activities of high-preference cuts (loin, tenderloin, and rib) and low-preference cuts (brisket, topside, and shank) of Hanwoo (Bos taurus coreanae) cows to obtain potentially useful information for promoting the consumption of various low-preference cuts. Methods: Individual 500 g samples of fresh beef were collected from each of the six cuts from 10 Hanwoo cows (quality grade 1) and immediately freeze-dried. The dried samples were evaluated for fatty acid composition, free radical scavenging activities (hydroxyl, alkyl, and 2, 2'-diphenyl-1-picrylhydrazyl [DPPH] radical), and antioxidative enzyme activities (glutathione peroxidase [GPx], glutathione-S-transferase [GST], and superoxide dismutase [SOD]). Results: The percentages of total polyunsaturated fatty acids were significantly higher in low-preference cuts than in high-preference cuts (p<0.05). Hydroxyl, alkyl, and DPPH radical scavenging activities were significantly higher in low-preference cuts than in high-preference cuts (p<0.05). In addition, the activities of antioxidant enzymes, such as GPx, GST, and SOD, were significantly higher in low-preference cuts compared with high-preference cuts (p<0.05). Conclusion: These results may influence consumers to include more low-preference cuts in their selections based on the nutritional facts, which could help to balance the beef market in South Korea.

Effect of Timed Artificial Insemination Protocols on the Pregnancy Rate Per Insemination and Pregnancy Loss in Dairy Cows and Korean Native Cattle under Heat Stress

  • We aimed to determine the effect of timed artificial insemination (TAI) protocols on the pregnancy rate per insemination and pregnancy loss compared with AI performed at detected estrus in dairy cows and Korean Hanwoo cattle under heat stress. In dairy cattle, 1,250 sets of data that underwent AI during heat stress (temperature-humidity index ≥ 72) were categorized according to their TAI protocols or as controls: 1) PGF-36 h-estradiol benzoate (EB)-36 h-TAI (PG-EB group, n = 113); 2) GnRH-7 days-PGF-56 h-GnRH-16 h-TAI (Ovsynch group, n = 455); or 3) GnRH-6 days-Ovsynch (G6G group, n = 136). The remaining cows underwent AI at detected estrus (AIDE group, n = 546). The probability of pregnancy per AI 45 days after AI was higher (P < 0.01) in the PG-EB (odds ratio [OR]: 1.68), Ovsynch (OR: 1.48), and G6G (OR: 1.79) groups than in the AIDE group. However, the prevalence of pregnancy loss between 30 and 45 days after AI did not differ among the groups. In Hanwoo cattle, 617 sets of data inseminated artificially under heat stress were categorized into AIDE (n = 281), PG-EB (n = 194), and combined Ovsynch or G6G (n = 142) groups. The probability of pregnancy per AI 45 days after AI and the prevalence of pregnancy loss between 30 and 45 days after AI did not differ among the groups. Thus, implementation of a TAI protocol (PG-EB, Ovsynch, or G6G) in dairy cows under heat stress improves the pregnancy rate per AI versus AIDE, whereas there is no beneficial effect of TAI on the pregnancy rate of Hanwoo cattle under heat stress.

Relationship between Estrous Expression, Body Condition Score (BCS) and Transferable Embryos Hanwoo Donors (한우 공란우의 발정발현율, 신체 충실 지수(BCS)와 이식가능 수정란과의 관계)

  • The objective of this study was to investigate the relationship between estrous expression, body condition score (BCS), blood urea nitrogen (BUN) and number of transferable embryos for the purpose of improving reproductive performance in blood of Hanwoo donors. Sixty, at random stages of the estrous cycle, received a CIDR. Four days later, the animals were superovulated with a total of 28AU FSH (Antorin, 2AU=1 ml) administered twice daily in constant doses over 4 days. On the 3th administration of FSH, CIDR was withdrawn and 25 mg PGF2 ${\alpha}$ was administered. Cows were artificially inseminated twice after estrous detection at 12 hr intervals. The cows received 100 ${\mu}g$ GnRH at the time of 1nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. The estrous inducement rate and estrous expression rate were significantly lower for cows with BCS below 2.25 than for cows with BCS above 2.25. There was 50.0% of rate of mounting in cows with BCS below 2.25 whereas the rate of mounting was markedly increased in cows with BCS above 2.25 (94.1% and 89.5% for BCS 2.25~2.75 and BCS above 2.75 cows, respectively). Cows with BCS <2.25, 2.25~2.75 and ${\geq}$2.75 had number of transferable embryos of $4.5{\pm}0.7$, $5.9{\pm}1.8$ and $5.6{\pm}2.3$ respectively.

Determination of in situ degradation parameters and feeding level of pineapple (Ananas comosus L.) cannery by-product to Hanwoo steers

  • Objective: The objectives of this study were to determine the in situ degradation parameters and appropriate feeding level of pineapple cannery by-products (PCB) based on the growth performance and blood parameters of growing Hanwoo (Bos taurus coreanae) steers fed various levels of PCB. Methods: Two ruminally cannulated Holstein cows were used for in situ disappearance rate measurements. Nylon bags (5×10 cm, 45 ㎛ pore size) filled with 5 g of PCB in triplicate were inserted into the ventral sac of the two cannulated Holsteins cows and incubated for 0, 2, 4, 8, 16, 24, and 48 h. A total of 16 castrated growing Hanwoo steers (12.5±0.5 months old, 302.9±25.7 kg of initial body weight [BW]) were used for the experiment. Animals were stratified by initial BW and then randomly assigned to one of four experimental diets (0%, 1.5%, 3.0%, or 6.0% of PCB, on the dry matter [DM] basis) fed for 91-d, including 30-d of adaptation. Results: Soluble fraction a of DM and crude protein (CP) was 61.9% and 86.0%, fraction b of DM and CP was 32.7% and 11.2%, and indigestible fraction c of DM and CP was 5.4% and 2.8%. The 6.0% PCB feeding group showed lower productivity compared to animals in the other treatments. Increasing the dietary level of PCB did not alter DM intake, but it was numerically lowest in the 6.0% feeding group. The gain to feed ratio was linearly decreased by increasing of PCB. The quadratic broken-line test estimated that 2.5% (DM basis) was the maximum feeding level of PCB in growing Hanwoo steers (y = 0.103 - 0.001×[1.245-x]2, R2 = 0.18). Conclusion: Diets containing up to 2.5% PCB can be fed to growing Hanwoo steers without adverse effects on growth performance.

Effects of Maternal Genetic Potential and Parity with Pre- and Postpartum on Body Weights, Body Condition Score and Blood Metabolites in Hanwoo Cows (한우 암소의 유전능력과 산차에 따른 분만 전?후 체중, Body Condition Score 및 혈중 대사물질의 변화)

  • This study was conducted to investigate effects of maternal genetic potential and parity with pre- and postpartum periods on body weights, body condition score (BCS) and blood metabolites in relation to physiological stress and nutritional metabolism in Hanwoo cows. Also, this study was designed to develop effective husbandry technique for Hanwoo cows concerning of pre- and postpartum periods, and to get basic data for it. Forty five cows were allocated into two groups, 24 cows with high maternal genetic potentials and 21 cows with low maternal genetic potentials. The average parity of experimental cows with high and low maternal genetic potentials were 2.83±1.63 and 3.00±1.77, respectively. The growth performances such as body weights, average daily gain (ADG) and BCS were not different between two groups regardless of maternal genetic potential. However, pre- and postpartum periods had effects on the growth performances (p<0.05). Parity had no effects on ADG and BCS (p>0.05), but effect on body weight of cows (p<0.05). The metabolites of physiological stress such as neutrophil, hematocrit and cortisol, and nutritional metabolites such as albumin, blood urea nitrogen (BUN), triglyceride, and non-esterified fatty acid (NEFA) concentrations in blood of cows were affected by pre- and postpartum periods in a large scale, while those were partially affected by maternal genetic potential. However, among the metabolites in blood, only neutrophil and triglylceride concentrations were affected by different parity of cows. Therefore, the present study suggests that nutritional intake and digestion are affected by physiological stress due to the parturition, and it should need to consider different husbandry technique based on the maternal genetic potential, and pre- and postpartum periods of cows.

Effects of High Protein Diet on Meat Productivity and Quality in Multiparous Hanwoo Cull Cows (고단백질 사료의 급여가 다산 한우 암소의 육생산량과 육질에 미치는 영향)

  • Multiparous Hanwoo cull cows have been generally regarded to have poor meat quality compared to young and primiparous Hanwoo in Korea, and there have been few studies attempting to understand various feeding programs for the multiparous Hanwoo cull cow. In this study, the effects of a feeding program consisting of two different diets for multiparous Hanwoo cull cows on meat production and quality were tested in comparison to a commercially used diet. Diets for treatment consisted of two levels of crude protein contents, 14.28% and 12.70% for early fattening and finishing, respectively. For a control, commercially used fattening feed (12.39% crude protein) was used. Feeding trials were performed at three different farms. In farm A, 29 herds of multiparous Hanwoo cull cows were used for the treatment group and 3 herds for the control. In farms B and C, the number of animal herds for treatment and control were 8 vs. 3 and 11 vs. 4 herds, respectively. Experiment diets were fed for an average of 211 days and in treatment, early fattening diet was fed for 4 months and then finishing feed was fed until slaughter. Average daily gains, thickness of back fat, area of Longissimus dorsi, carcass weight, index of carcass weight, intramuscular fat, meat color and texture were analyzed after slaughter. Random effect model [8] was employed in effect analysis. Positive effects of treatment were found in terms of average daily gain, back fat thickness, Longissimus dorsi area, carcass weight, and intramuscular fat. Carcass yield index, meat color and texture showed a negative effect. In this study, significant results were not found in all factors analyzed because the variance between experiment farms was large, however the 90% confidence interval of summary effects of ADG, back fat thickness, Longissimus dorsi area and carcass weight were significant and that of carcass yield index, intramuscular fat, meat color and texture were less so. Conclusively, a high protein diet fed early during the fattening period in multiparous Hanwoo cull cows could have positive effects on meat production.

Production of Calves by Transfer of Vitro Produced Hanwoo Embryos (한우 체외수정란이식에 의한 산자 생산)

  • The objective of this study was to improve the efficiency of bovine embryo transfer by transferring of Hanwoo embryos into Hanwoo or Holstein recipients. The cryopreserved or fresh in vitro produced(IVP) embryos were transferred into uterine horn contralaterally or ipsilaterally to the corpus luteum. The recipients were inseminated by artificially on the next day of estrus. The pregnancy was diagnosed by rectal palpation at 60∼90 days after transfer of the embryos. The pregnancy rate by transfer of one or two embryos was 78%(7/9) and 74%(31/42), respectively. The pregnancy rates according to the grade of corpus lutea of recipients was 75% (20/27) and 82.0%(18/22) at the grade of A and B, respectively. Ten(67.0%) of 15 Holstein recipients transferred with IVP Hanwoo embryos and 5(42.0%) of 12 Holstein recipients transferred with frozen IVP Hanwoo embryos were pregnant. The single and twin calving ratio in Hanwoos was 77.0%(10/13) and 23.0%(3.13) in the recipients transferred with IVP embryos and 64.0%(7/10) and 27.0%(3/10) in the recipients transferred with frozen IVP embryos, respectively. Twenty-four pregnant cows following transfer of IVP embryos, 21(88.0%) calved the normal calves, and 2(8.3%) aborted. When the frozen IVP embryos were transferred, 16 pregnant cows calved 14(88.0%) normal calves and 2(13.0%) aborted. In conclusion, when one or two IVP bovine embryos were transferred into recipients, the A and B grade of corpus luteum resulted in high pregnancy rates. For the production of twin calves, transfer of the IVP or frozen IVP embryos could be suitable.

Production of Twin Calves Following Transfer of Hanwoo Embryos Produced In Vitro (한우 체외수정란을 이용한 쌍자생산에 관한 연구)

  • This study was carried out to improve a technique of embryo transfer for twin calves production in Hanwoo cattle. Blastocysts for the donor of embryo transfer were classified into three criteria by accessment of morphology; early blastocyst, blastocyst and expanded blastocyst. Tow embryos were introduced transcervically into utrerine horn either of Hanwoo or Holstein by ipsilaterally or contralaterally to the corpus luteum. Thiry-six out of 57 recipients cows were inseminated by artificially on the next day of estrus, and followed by transfer of embryos into contralaterally. The pregnance rates of recipients following transfer of bovine embryos of day 7, 8 and 9 was 43.5, 18.2 and 8.3%, respectively. These results appeared that these was a significant (P<0.05) difference between on day-7 embryos and day-9 embryos, but not between on day-8 and day-9 embryos. Although there was not significant(P<0.05) difference in the pregnancy rates between the blastocysts(11/25, 44%) and expanded blastocysts(2/19, 10.5%) and between the blastocysts and early blastocysts(2/13, 15.4%), the embryos at blastocyst stage are more suitable than others for obtaining higher rate of pregnancy. There was no significant difference on pregnancy of the embryos transferred prior to presence(6/21, 29%) or absence (9/36, 25%) of artificial insemination. On pregnancy of Holstein, 2(15.4%) out of 13 recipients were pregnant in heifer. Similar Pregnancy rates were obtained between 1∼2 parities and 3∼4 parities by 30% (6/20) and 27.3%(3/11), respectively. Taken together, there was not significant difference in pregnancy rate due to small number of recipients used for this experiment. Both of Hanwoo and Holstein introduced the embryos by contralsterally to the corpus luteum were slightly higher pregnancy rate compare to by ipsilaterally (12/41, 29.3% vs, 3/16, 18.8%). The ratio of production of twin and single calves in Holstein was 20% (9/45) and 2.2% (1/45), respectively. However, in Hanwoo cows both of production of twin and single were similar as 8%. This result suggests that Holstein as recipients was superior to Hanwoo cows for production of twin calves. Out of all 15 pregnant, 12(80%) were produced a total of 22 normal calves in which the others composed of abnormal, as judging as 2(13.3%) for abortion and 1(6.6%) for stillbirth during the pregnant period.
