In vitro endonucleolytic cleavage of synthesized cucumber mosaic virus RNA by hammerhead ribozyme (인공적으로 합성한 오이모자이크 바이러스 RNA의 헤머헤드 ribozyme에 의한 시험관내에서의 절단)

  • Oligonucleotides for a conserved region of the coat protein gene of cucumber mosaic virus (CMV) and a hammerhead structure ribozyme against CMV RNA were synthesized using a DNA synthesizer. Both strands of oligonucleotides were annealed and restricted with BamHI/SacI, then cloned into a plasmid pBS SK (+). The cloned CMV substrate and ribozyme were sequenced to verify correct constructions. In vitro transcriptions were carried out by using T7 RNA polymerase with BssHII or SspI digests of $1\;{\mu}g$ of substrate and ribozyme clones. The size of substrate RNA was 176 nucleotides (nt) containing 50 nt of CMV RNA sequence, 6 nt of XbaI restriction site and 120 nt of vector-derived sequence in the case of BssHII digest. The size of ribozyme RNA was 164 nt containing 40 nt of ribozyme RNA sequence and same sequences of substrate. Substrate RNA was efficiently cleaved into two fragments (96 nt and 80 nt) by ribozyme RNA. This endonucleolytic cleavage occurred more efficiently at $55^{\circ}C$ than $37^{\circ}C$. SspI digest-derived substrate RNA (2234 nt) was also cleaved into two fragments by the same ribozyme. SspI digest-derived ribozyme RNA (2222 nt) cleaved the above substrate to two fragments. In vitro-tested ribozyme construct is being cloned into a plant transformation vector to develop virus-resistant plants.

Transition Flow Analysis According to the Change of Reynolds Number for Supersonic Launch Vehicle Fairing Expansion Area (초음속 발사체 선두 팽창부의 레이놀즈수 변화에 따른 천이 유동 해석)

  • RANS computational analysis was performed on the head of the launch vehicle including the hammerhead nose pairing in the supersonic regime. The two-dimensional axisymmetric analysis was performed by using laminar, fully turbulent and transition models and compared with the experimental data. It was observed that different flow phenomena occurred depending on the Reynolds number. Under the high Reynolds number condition, the boundary layer becomes turbulent, which is not separated from the surface of the launch vehicle. With the low Reynolds number condition, laminar separation bubble was produced due to the separation and reattachment of the boundary layer on the expansion-compression edge of the hammerhead type nose fairing. The three-dimensional computations with the angle of attack showed a fully detached vortical structure due to the laminar separation bubble. It is proved that the turbulent transition should be considered to predict the separation bubble with the Reynolds number.

  • Biochemical amounts of RNA molecules can be synthesized in vitro, which is functionally equivalent or similar to those transcripts normally existing at extremely low levels in vivo. In this study we described a method for efficient preparation of pure T7 RNA polymerase from Escherichia coli strain BL21/pAR1219. The procedure, which used ammonium sulfate fractionation and preparative column chromatography on sephadex SP, was shown to be simple, rapid, and cost effective in comparison with other methods reported previously, Using the purified T7 RNA polymerase we were able to synthesize very long RNA transcript of 1.54 kb length, which is not feasible by conventional chemical synthesis. RNA molecule that was also synthesized by the purified T7 RNA polymerase, such as hammerhead ribozyme, retained its biochemical activity by cleaving the target RNA successfully in vitro. Thus, the procedure shown in this study can be useful to synthesize any length of RNA molecules in vitro in a simple and cost effective way for a variety of purposes.

  • Ribozymes are catalytic RNAs that can cleave RNAs at specific sites, thus they have been employed to degrade a target mRNA in vivo. Development of allosterically controllable ribozymes is of great current interest, but it remained difficult to furnish such functions to ribozymes in cultured cells or in animals. Recently, we designed allosterically controllable ribozymes termed maxizymes, which have sensor arms that recognize target mRNA sequences and, in the presence of such target sequences only, they form a cavity that can capture catalytically indispensable $Mg^{2+}$ ions, cleaving the target. The maxizyme was applied to therapy for chronic myelogenous leukemia (CML). It cleaved specifically the chimeric BCR-ABL mRNA, which caused CML, without damaging the normal ABL or BCR mRNA in mammalian cells and also in mice, providing the first successful example for allosteric control of the activity of artificial ribozymes in vivo.

A study on in-flight acoustic load reduction in launch vehicle fairing by FE-SEA hybrid method (FE-SEA 하이브리드 기법을 이용한 비행 중 발사체 페어링 내부 음향하중 저감에 관한 연구)

  • Launch vehicles are subject to airborne acoustic loads during atmospheric flight and these effects become pronounced especially in transonic region. As the vibration due to the acoustic loads can cause malfunction of payloads, it is essential to predict and reduce the acoustic loads. In this study, a complete process has been developed for predicting airborne vibro-acoustic environment inside the payload pairing and subsequent noise reduction procedure employing acoustic blankets and Helmholtz resonators. Acoustic loads were predicted by Reynolds-Averaged Navier-Stokes (RANS) analysis and a semi-empirical model for pressure fluctuation inside turbulent boundary layer. Coupled vibro-acoustic analysis was performed using VA One SEA's Finite Element Statistical Energy Analysis (FE-SEA) hybrid module and ANSYS APDL. The process has been applied to a hammerhead launch vehicle to evaluate the effect of acoustic load reduction and accordingly to verify the effectiveness of the process. The presently developed process enables to obtain quick analysis result with reasonable accuracy and thus is expected to be useful in the initial design phase of a launch vehicle.

Expression of in vitro-tested ribozyme against cucumber mosaic virus RNA in tobacco plant (시험관내에서 합성한 오이모자이크 바이러스 RNA단편을 성공적으로 절단한 ribozyme의 식물체내의 발현)

  • In vitro-tested ribozyme against synthesized cucumber mosaic virus (CMV) RNA (Agric. Chem. & Biotech. 37:56-63(1994)) was expressed in tobacco plant to develop virus resistant plants. The ribozyme sequence was linked to cauliflower mosaic virus 35S promoter and nopaline synthase(nos) terminator and this chimeric 35S-ribozyme-nos gene was sequenced. The sequenced chimeric gene was transferred to Agrobacterium tumefaciens LBA4404 using tri-parental mating system. The E. coli HB101 containing chimeric gene was incubated with E. coli HB101(pRK2073) as a helper and Agrobacterium tumefaciens LBA4404. Then Agrobacterium cells containing the ribozyme construct was cocultivated with tobacco leaf pieces. Ten different plants were regenerated from kanamycin containing MS medium. The presence of the ribozyme construct in the transgenic tobacco plants was confirmed by polymerase chain reaction (PCR). Seven different transgenic plants in ten different kanamycin resistant plants showed the expected size (570 base pairs) of 35S-ribozyme-nos gene fragment. Total RNAs were isolated from four different transgenic plants and separated on a 1% agarose gel containing formamide. Northern hybridization with 35S-ribozyme-nos gene fragment as a probe indicated that ribozyme transcripts may be degraded tv nuclease. Therefore, nuclease-resistant ribozymes are needed for the development of virus-resistant transgenic plants using ribozymes.

Monitoring Methylmercury in Abyssal Fish (심해성 어류 중 메틸수은 모니터링)

  • The aim of this study was to determine the methylmercury (MeHg) levels in abyssal fish species. The MeHg in the fishes was extracted with hydrochloric acid and toluene and then purified using an L-cysteine solution. The extract was analyzed with a gas chromatography-electron capture detector (GC-${\mu}ECD$) with a thermon Hg-capillary column. The detection limit and the recovery of the method were 0.002 and 84.2-98.5% (mean, 93.4%), respectively. The MeHg content in 492 abyssal fishes ranged from 0.037 to 2.009 mg/kg. The levels of MeHg [range, mg/kg (mean)] were significantly dependent on fish species and presented as the following; 0.157-2.009 (0.546) in Scalloped hammerhead shark, 0.211-0.878 (0.501) in Blue shark, 0.121-0.993 (0.482) in Spiny dogfish, 0.243-0.658 (0.397) in Salmon shark, 0.074-1.958 (0.353) in Blacktip shark, 0.038-0.807 (0.302) in Southern hake, 0.099-0.511 (0.300) in Scorpion fish, and 0.037-0.133 (0.067) in Ling. The monitoring results showed that the estimated weekly intake of MeHg from sharks, Southern hake, and Ling were lower than the provisional tolerable weekly intake recommended by the Joint FAO/WHO expert committee on food additives.
