Vanillic Acid Stimulates Anagen Signaling via the PI3K/Akt/β-Catenin Pathway in Dermal Papilla Cells

  • The hair cycle (anagen, catagen, and telogen) is regulated by the interaction between mesenchymal cells and epithelial cells in the hair follicles. The proliferation of dermal papilla cells (DPCs), mesenchymal-derived fibroblasts, has emerged as a target for the regulation of the hair cycle. Here, we show that vanillic acid, a phenolic acid from wheat bran, promotes the proliferation of DPCs via a PI3K/Akt/Wnt/β-catenin dependent mechanism. Vanillic acid promoted the proliferation of DPCs, accompanied by increased levels of cell-cycle proteins cyclin D1, CDK6, and Cdc2 p34. Vanillic acid also increased the levels of phospho(ser473)-Akt, phospho(ser780)-pRB, and phospho(thr37/46)-4EBP1 in a time-dependent manner. Wortmannin, an inhibitor of the PI3K/Akt pathway, attenuated the vanillic acid-mediated proliferation of DPCs. Vanillic acid-induced progression of the cell-cycle was also suppressed by wortmannin. Moreover, vanillic acid increased the levels of Wnt/β-catenin proteins, such as phospho(ser9)-glycogen synthase kinase-3β, phospho(ser552)-β-catenin, and phospho(ser675)-β-catenin. We found that vanillic acid increased the levels of cyclin D1 and Cox-2, which are target genes of β-catenin, and these changes were inhibited by wortmannin. To investigate whether vanillic acid affects the downregulation of β-catenin by dihydrotestosterone (DHT), implicated in the development of androgenetic alopecia, DPCs were stimulated with DHT in the presence and absence of vanillic acid for 24 h. Western blotting and confocal microscopy analyses showed that the decreased level of β-catenin after the incubation with DHT was reversed by vanillic acid. These results suggest that vanillic acid could stimulate anagen and alleviate hair loss by activating the PI3K/Akt and Wnt/β-catenin pathways in DPCs.

Effect of Black Rice (Oryza sativa L. indica) Ethanolic Extract on Tyrosinase Activity and Antioxidant Activity Related to Melanin Production (흑미의 주정 추출물이 melanin 생성과 관련된 tyrosinase 및 항산화 활성에 미치는 영향)

  • Hair graying is processed by loss of melanin production caused by the decrease of activity and number of melanocyte and the accumulation of hydrogen peroxide ($H_2O_2$) in the hair follicle with increase of age. The purpose of this study was to investigate the effect the Black oryzasativa ethanolic extract (BLEE) on the melanin production. In this study BLEE at $8{\mu}g/ml$ or more showed a significant 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and reduction power. BLEE at $16{\mu}g/ml$ or more showed promoted tyrosinase activity and melanin production. In addition BLEE scavenged intracellular $H_2O_2$ in 2',7'-dichlorodihydrofluorescein (DCF) fluorescence assay in B16F1 cells. However, Western blot analyses displayed that BLEE decreased the expression level of catalase, but no effect on the expression level of tyrosinase, tyrosinase associated protein-1 (TRP-1), tyrosinase associated protein-2 (TRP-2) and microphthalmia-associated transcription factor (MITF) transcription factor involved in melanogenesis. Thus, the promotive effect of BLEE on melanin production is attributed to the increase of tyrosinase activity and the reduction of intracellular $H_2O_2$ level. In conclusion, BLEE played a key role in in promoting melanin production, which suggests that the BLEE could be applied as a potential functional material in the development of hair care cosmetics related to the promotion of melanin production for the growth of black hair.

A case of distichiasis treatment using electroepilation in a dog (개에서 전기제모술을 이용한 첩모중생의 치료 1례)

  • Distichiasis is one of the diseases commonly encountered in companion animals, and these abnormal eyelashes cause corneal ulcers, continuous eye irritation, eye pain, glare, epiphora, foreign body sensation and can cause corneal opacity and vision loss in severe cases. In this study, an eyelash epilation needle for animals was developed and applied to a real case, and the results were observed. In a case of corneal ulcer caused by distichiasis of a 2-year-old Shih Tzu, a high-frequency surgical instrument for animals was converted into an electric epilation needle to attempt a procedure to destroy the eyelash hair follicles on the upper eyelid. A epilation needle was developed to have a diameter of 0.1 mm and a length of 4 mm at the end of the handle of DOCTANZ 400, an electrosurgical instrument for animals only. In the procedure, 2~3 mm of an epilation needle was inserted into the hair follicle, and 1 watt of electric power was applied to the hair follicle for about 5 sec. to carry out electrolysis until white bubbles were generated around the meibomian glands thereby destroying the hair follicle. As a result, no eyelashes grew any longer in the treated area indicating that the treatment was successful. It is hoped that the method developed in this study will be applied so that it will be widely used as a treatment method for distichiasis in companion animals that can be frequently seen hereafter.

Physiological Responses and Lactation to Cutaneous Evaporative Heat Loss in Bos indicus, Bos taurus, and Their Crossbreds

  • Cutaneous evaporative heat loss in Bos indicus and Bos taurus has been well documented. Nonetheless, how crossbreds with different fractional genetic proportions respond to such circumstances is of interest. A study to examine the physiological responses to cutaneous evaporative heat loss, also lactation period and milk yield, were conducted in Sahiwal (Bos indicus, n = 10, $444{\pm}64.8kg$, $9{\pm}2.9years$), Holstein Friesian (Bos taurus, HF100% (n = 10, $488{\pm}97.9kg$, $6{\pm}2.8years$)) and the following crossbreds: HF50% (n = 10, $355{\pm}40.7kg$, $2{\pm}0years$) and HF87.5% (n = 10, $489{\pm}76.8kg$, $7{\pm}1.8years$). They were allocated so as to determine the physiological responses of sweating rate (SR), respiration rate (RR), rectal temperature (RT), and skin temperature (ST) with and without hair from 06:00 h am to 15:00 h pm. And milk yield during 180 days were collected at days from 30 to 180. The ambient temperature-humidity-index (THI) increased from less than 80 in the early morning to more than 90 in the late afternoon. The interaction of THI and breed were highly affected on SR, RR, RT, and ST (p<0.01). The SR was highest in Sahiwal ($595g/m^2/h$) compared to HF100% ($227g/m^2/h$), and their crossbreds both HF50% ($335g/m^2/h$) and HF87.5% ($299g/m^2/h$). On the other hand, RR was higher in HF87.5% (54 bpm) and both HF100% (48 bpm) and HF50% (42 bpm) than Sahiwal (25 bpm) (p<0.01). The RT showed no significant differences as a result of breed (p>0.05) but did change over time. The ST with and without hair were similar, and was higher in HF100% ($37.4^{\circ}C$; $38.0^{\circ}C$) and their crossbred HF50% ($35.5^{\circ}C$; $35.5^{\circ}C$) and HF87.5% ($37.1^{\circ}C$; $37.9^{\circ}C$) than Sahiwal ($34.8^{\circ}C$; $34.8^{\circ}C$) (p<0.01). Moreover, the early lactation were higher at HF100% (25 kg) and 87.5% (25 kg) than HF50% (23 kg) which were higher than Sahiwal (18 kg) while the peak period of lactation was higher at HF100% (35 kg) than crossbreds both HF87.5% and HF50% (32 kg) which was higher than Sahiwal (26 kg) (p<0.05). In conclusion, sweating and respiration were the main vehicle for dissipating excess body heat for Sahiwal, HF and crossbreds, respectively. The THI at 76 to 80 were the critical points where the physiological responses to elevated temperature displayed change.

The first Korean case of Waardenburg-Shah syndrome with novel endothelin receptor type B mutations

  • Waardenburg syndrome (WS) is a rare genetic disorder, including clinical features of pigmentary abnormalities of irides, skin, hair and sensorineural hearing loss and facial dysmorphism. Among the four types, WS type IV (Waardenburg-Shah syndrome) additionally represents Hirschsprung's disease. Mutations in the SOX10, END3, or EDNRB genes are known to cause WS type IV. Here, we report a 6 year-old girl who was diagnosed as WS type IV by typical clinical manifestations, including skin hypopigmentation, heterochromia of both irides, unilateral sensorineural hearing loss, mild developmental delay and Hirschsprung's disease. The diagnosis was confirmed by molecular genetic analysis of EDNRB. Two novel EDNRB mutations were identified, and each mutation was segregated from each of her parents. During the follow-up period, the patient underwent a surgery for spleen torsion and was medically managed due to recurrent enterocolitis. Also, she suffered from impaired immunity including Hirschsprung's associated enterocolitis.

Introduction of tmie Gene Can Recover the Hearing Impairment and Abnormal Behavior in the Circling Mouse

  • The spontaneous mutant circling mouse (cir/cir) shows a circling behavior and hearing loss. We produced transgenic mice overexpressing transmembrane inner ear (tmie) gene, the causative gene, for the phenotypic rescue of the circling mouse. Through the continuous breeding with circling mice, the cir/cir homozygous mice carrying the transgene (cir/cir-tg) were produced. The rescued cir/cir-tg mice were able to swim in the water with proper orientation and did not show any circling behavior like wild type mice. Western blot and immunohistochemical analysis exhibited that the transgenic tmie was expressed in the inner ear. Inner and outer hair cells were recovered in the cochlea and spiral ganglion neurons were also recovered in the rescued mice. Auditory brainstem response (ABR) test demonstrated that the cir/cir-tg mice are able to respond to sound. This study demonstrates that tmie transgene can recover the hearing impairment and abnormal behavior in the circling mouse.

Experimental Study on the Effect of L-Glutamate to Acute Myocardial Toxicity by Doxorubicin (Doxorubicin 에 의한 급성 심근독성과 L-Glutamate 의 효과에 관한 실험적 연구)

  • The effect of Glutamate to myocardial toxicity induced by doxorubicin was studied with 20 male rats. 20 rats divided into 4 subgroups, 1st group was taken for normal control group without any treatment, 2nd group was injected with only doxorubicin, 3rd group was injected with L-glutamate and doxorubicin, and 4thd group was injected with only L-glutamate [all injections were done intraperitoneally]. Observations were made to each group on their gross findings, body weight, and electrocardiography, complete blood count and serum level of creatine phosphokinase. The results were as follows; l. In 1st group, we found no changes. 2. In 2nd group, there were many changes which were loss of body weight, dehydration, loss of body hair, diarrhea and death, in addition, elevation of CPK-MB isoenzyme and changes in EKG due to myocardial damage, leukopenia, thrombocytopenia were also found. 3. In 3rd group, there were more toxic effects containing 2 death cases, compared to 2nd group. 4. In 4th group, we found no specific changes except weight gain. These results suggest that L-glutamate which is intermediate of Krebs cycle may worsen the doxorubicin-induced myocardial toxicity.

  • Objective : The purpose of this study is to report the effect of Pharmacopuncture Therapy and Needle-embedding Therapy on Androgenetic Alopecia in woman. Methods : The patient was treated by acupucture, pharmacopuncture, needle-embedding. The improvement of the patient was judged by phtograpies and VAS(Visual Analogue Scale) score. Results & Conclusions : Her hair loss and physical condition were improved remarkably, and an author consider that continuous clinical study will be needed in other needle-embedding therapy and korean medical treatment.

Suggestion of experimental model on the innate kidney essence insufficiency syndrome for the development of humanitas traditional medicine

  • According to the traditional Korean medicine (TKM), kidney has been recognized as the roof of innate endowment because it plays an important role in the birth, growth, sexual reproduction, and aging. Kidney essence insufficiency syndrome (KEIS) is caused when kidney is impaired. KEIS is characterized by retarded development, decreased reproduction, tinnitus, loosening of teeth, and loss of hair and forgetfulness. In traditional Chinese medicine, KEIS mouse model was established by use of threatinjuring the kidney combined with over-fatigue. However, the TKM theory-based KEIS experiment model has not been described correctly. In the present study, we suggest a new KEIS experiment model including following cases; 1) weakness of father essence and mother blood, 2) life nurturing during pregnancy, 3) full-term gestation period.

Inhibitory Effects of Corni Fructus Extract on Angiogenesis and Adipogenesis

  • Natural products in Chonnam, Korea were screened via anti-angiogenesis experiments, and 1 candidate product was identified, Corni fructus, which exerted dose-dependent inhibitory effects against angiogenesis, adipogenesis, and cell adhesion. C. fructus extract (CFE) exhibits an angiogenesis inhibitory effect superior to that of the EGCG from green tea leaves. The expression level of angiogenesis and adipogenesis-related signal molecules in the western blotting was reduced by increasing the amount of added CFE. Moreover, a diet supplemented with CFE was deemed more effective in inducing weight loss in LB mice than a representative synthetic diet drug, orlistat, which incidently caused the side effect of denuding the mice of their hair. These results indicate that C. fructus may prove to be a useful anti-adipogenic compound, and these in vitro results may be reflected later under in vivo conditions.
