탈모 유효 약침액 한약 소재에 대한 동물모델을 이용한 국내 연구보고 고찰 (Effect of Herbal Medicines Pharmacopuncture on Hair Growth, a Review of Animal Study Reports Published in Koreae)

  • Objectives : The aim of this study was to review the effect of herbal medicine on hair growth in animal models reported in Korean domestic journals. Methods : Korean core databases were searched with terms alopecia and hair, and animal study reports on hair growth with herbal medicine were reviewed. Animal model, intervention, and hair growth measure data were extracted. Results : A total of 28 reports were reviewed. C57BL/6 mice with depilation was the most frequent animal models used. Polygoni multiflori radix, Cnidii rhizoma and Biotae Folium were frequently used herbal medicine. Positive effects were reported. Conclusions : Herbal medicines may be promising resource for pharmacopuncture materials for positive effects on hair growth.

모발생장기 유도 C3H 생쥐에 있어서 미녹시딜과 생약추출 혼합 조성물의 모발 재성장 유도 효능 (Evaluation of the Herbal Extract Mixture for the Effects of Hair-Regrowth Compared to 3% Minoxidil; Elongation of Anagen Period on C3H Mice)

  • The hair cycle consists of three phases, growth (anagen), involution (catagen) and quiescence (telogen) phases. In order to evaluate hair re-growth effect of herbal extracts mixture containing the 70% ethanol extracts of Polygoni Multiflori Radix, Mori Cortex Radicis, Gingko Biloba Folium and Pine bud, we have examined the induction of the anagen phase and/or elongation of the anagen period using C3H mice. Morphological examination was done by Hattori' and Ogawa's method. Enzyme activities of ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GT) and alkaline phosphatase (ALP) was detected by Bessey-Lovry-Brock's method. Enzyme activity as a biochemical marker of hair cycle was investigated in the third hair cycle period of C3H mice after depilation. 3% Minoxidil treated group and herbal extract mixture treated group were shown 3 days earlier initiation of anagen than control group. In cycling mouse skin, ${\gamma}$-GT activity is pronounced during anagen and greatly diminished during telogen. Herbal extract mixture has shown promising hair re-growth effect on hair follicular cycles of C3H mice.

청호의 육모활성연구 (A Study on Hair-growth Activity of Artemisia apiacea Hance)

  • To investigate the effect of Artemisia apiacea Hance on the hair growth of black mouse (C57BL/6), this study was carried out. We obtained chloroform fraction, ethyl acetate fraction, butanol fraction from ethanol extract. When these fractions were administered to the black mouse by the method of skin paste, chloroform extract increased remarkably the hair growth of black mouse. thus Chloroform fraction was devided into 5 fractions from 20:1 to 1:1 with chloroform : methanol. the detail fractions of chloroform fraction were administered to the black mouse. We obtained the result that fraction I, II, III, IV are superior to minoxidil of positive control. Most of all fraction II have an dramatical effect on hair growth of black mouse.

삼황사심탕의 항산화능 및 C57BL/6 마우스 모델에서의 발모 촉진효과 (Antioxidant Ability and Hair Growth Effect of Samhwang-Sasimtang in C57BL/6 Model)

  • The purpose of this study is to evaluate the antioxidant activity and hair growth effects of a natural herbal ethanol extract, Samhwang-Sasimtang(SS). In case of antioxidant ability of SS, the content of phenolic compounds was 28.44mg/g. The extract showed strong electron donating ability and free radical scavenging activity in a concentration-dependent manner. SOD-like activity also rose through increasing the concentrations of SS. In order to estimate the hair growth effects, the extract was applied to the back of seven-week-old C57BL/6 male mice ($150{\mu}{\ell}$ a day, five days a week, for four weeks) in four groups (C, control, saline; PC, positive control, 3% minoxidil; E1, experimental 1, 1% SS; E2, experimental 2, 2% SS). Ten mice were assigned to each group and five mice in each group were sacrificed at weeks 2 and 4, respectively. There was no a significant difference in body weight change among experimental groups. In macroscopic observation of hair growth at week 4, the scores of hair growth on the backs of mice were 80, 60, 40 and 20% in the PC, E2, E1 and C groups. In terms of histological observation, the ratio and thickness of hair follicles, the enzyme activities of ALP and ${\gamma}$-GT, immunohistological examination of IGF-1 and VEGF which are the hair growth factors, in each group were significantly high in this order: PC, E2, E1 and C group, at week 4. Meanwhile, hair growth inhibition factors, TGF-${\beta}1$ and Caspase-3, were reduced in PC, E1 and E2 groups compared with C group. These results indicate that SS extract may be effective in promoting hair growth, and suggest that it can be used practically as a superior natural agent for hair growth promotion.

지모(知母) 에탄올 추출물이 쥐의 모발 성장에 미치는 영향 (The effect of Anemarrhena asphodeloides ethanolic extracts on hair growth effect in mice)

  • 여포 상피세포에 우수한 모발성장 촉진효과를 보이는 minoxidil의 단점을 보완하고자 대체의학제재로서의 모발 성장 촉진제로 지모를 연구하였다. 지모과(知母科)의 뿌리식물인 지모(知母)는 해열제, 항 당뇨병, 항우울제, 항염제 등으로 이용되고 있는 한국의 전통적인 약용식물이다. 지모 에탄올 추출물을 마우스의 등에 도포하여 관찰하였는데 모든 그룹의 마우스에서 체중과 음식 섭취량은 유의한 차이를 보이지 않았다. 지모 에탄올 추출물은 혈청 조성의 변화 없이 왕성한 모발성장 촉진효과를 보임으로써 향후 모발성장 촉진제로서 유용한 가치가 있으리라 사료된다.

Hair Growth-Promoting Effects of Lavender Oil in C57BL/6 Mice

  • The purpose of this study was to determine the hair growth effects of lavender oil (LO) in female C57BL/6 mice. The experimental animals were divided into a normal group (N: saline), a vehicle control group (VC: jojoba oil), a positive control group (PC: 3% minoxidil), experimental group 1 (E1: 3% LO), and experimental group 2 (E2: 5% LO). Test compound solutions were topically applied to the backs of the mice ($100{\mu}L$ per application), once per day, 5 times a week, for 4 weeks. The changes in hair follicle number, dermal thickness, and hair follicle depth were observed in skin tissues stained with hematoxylin and eosin, and the number of mast cells was measured in the dermal and hypodermal layers stained with toluidine blue. PC, E1, and E2 groups showed a significantly increased number of hair follicles, deepened hair follicle depth, and thickened dermal layer, along with a significantly decreased number of mast cells compared to the N group. These results indicated that LO has a marked hair growth-promoting effect, as observed morphologically and histologically. There was no significant difference in the weight of the thymus among the groups. However, both absolute and relative weights of the spleen were significantly higher in the PC group than in the N, VC, E1, or E2 group at week 4. Thus, LO could be practically applied as a hair growth-promoting agent.

3-Deoxysappanchalcone Promotes Proliferation of Human Hair Follicle Dermal Papilla Cells and Hair Growth in C57BL/6 Mice by Modulating WNT/β-Catenin and STAT Signaling

  • 3-Deoxysappanchalcone (3-DSC) has been reported to possess anti-allergic, antiviral, anti-inflammatory and antioxidant activities. In the present study, we investigated the effects of 3-DSC on the proliferation of human hair follicle dermal papilla cells (HDPCs) and mouse hair growth in vivo. A real-time cell analyzer system, luciferase assay, Western blot and real-time polymerase chain reaction (PCR) were employed to measure the biochemical changes occurring in HDPCs in response to 3-DSC treatment. The effect of 3-DSC on hair growth in C57BL/6 mice was also examined. 3-DSC promoted the proliferation of HDPCs, similar to Tofacitinib, an inhibitor of janus-activated kinase (JAK). 3-DSC promoted phosphorylation of ${\beta}$-catenin and transcriptional activation of the T-cell factor. In addition, 3-DSC potentiated interleukin-6 (IL-6)-induced phosphorylation and subsequent transactivation of signal transducer and activator of transcription-3 (STAT3), thereby increasing the expression of cyclin-dependent kinase-4 (Cdk4), fibroblast growth factor (FGF) and vascular endothelial growth factor (VEGF). On the contrary, 3-DSC attenuated STAT6 mRNA expression and IL4-induced STAT6 phosphorylation in HDPCs. Finally, we observed that topical application of 3-DSC promoted the anagen phase of hair growth in C57BL/6 mice. 3-DSC stimulates hair growth possibly by inducing proliferation of follicular dermal papilla cells via modulation of $WNT/{\beta}$-catenin and STAT signaling.

Hair growth promoting effects of emodin in telogenic C57BL/6 mice

  • Emodin is an anthraquinone derivative from the roots of Rheum officinale Baill that possesses a variety of biological activities, including inhibition of $5{\alpha}$-reductase and prostaglandin D2. In this study, we investigated whether emodin promotes hair growth. After emodin was topically applied to the shaved dorsal skin of telogenic C57BL/6 N mice, the hair growth rate and morphological analysis were evaluated in dorsal skin for 15 days. After 13 days of treatment, minoxidil or emodin (0.01% or 0.1%)-treated groups showed remarkable regrowth of hairs relative to the vehicle control group. Scoring of the hair growth and rate of hair growth area for 15 days revealed that groups treated with minoxidil and 0.1% emodin were significantly higher than the vehicle control group. Histological examination revealed the emodin and minoxidil groups markedly recovered the number and morphology of hair follicles, including the subcutis depth, relative to the vehicle group. These results suggest that emodin has an excellent promoting effect in hair growth similar to that of minoxidil and might be useful for treatment of baldness or alopecia.

Immunohistochemical Study on the Hair Growth Promoting Effect of Yonnyuniksoogobon-dan

  • Objectives : This study evaluates the hair growth promoting effect of Yonnyuniksoogobon-dan on shaved C57BL6 mice. Methods : Yonnyuniksoogobon-dan was administered orally (Group I) and both orally and by skin application (Group II) once a day for 12 days. The experimental groups were compared to Control, which was orally administered physiological saline solution. Hair regrowth was photographically and histologically determined during the experimental period. The levels of hair growth cycle related factors (EGF, TGF-${\beta}$1) and vascular factors (VEGF, iNOS) were also determined by immunohistochemistry. On gross observation of hair growth, both Group I and Group II shaved C57BL6 mice showed accelerated hair regrowth. Results : The hair regrowth index of Group I increased significantly from 7 days (P<0.05) to 12 days (P<0.01) after shaving and that of Group II was significantly higher at 12 days (P<0.05). On histological observation, both Group I and Group II demonstrated histological improvement and increases in the number and diameter of the hair follicles. EGF expression on the root sheath of hair follicles was up-regulated in both Group I and II. TGF-${\beta}$1 expression on the root sheath of hair follicles was unchanged in both Group I and II. VEGF expression in the tissues surrounding hair follicles was up-regulated in both groups. iNOS expression in the tissues surrounding hair follicles was down-regulated in both groups. Conclusions : These results suggest that Yonnyuniksoogobon-dan promotes hair growth and this effect is related to up-regulation of EGF and VEFG expression and down-regulation of TGF-${\beta}$1 and iNOS expression on hair roots.

봉독약침요법(蜂毒藥鍼療法)에 의한 발모관련 인자들의 발현에 대한 연구 (Experimental Study on the Expression of Hair Growth Related Factors after Bee Venom Acupuncture Therapy)

  • The present study was undertaken to investigate the effect of bee venom acupuncture therapy on the hair follicle growth by macroscopic, microscopic and immunohistochemical observation of skin 10 and 17 days after treatment. The results were as follows : Macroscopic hair follicle growth of 0.2 ml S.B.V. acupuncture treated group was more prominent than those of 0.1 ml S.B.V. acupuncture treated group and control group. Microscopic observation indicated that the hair follicle growth of control group reached anagen phase IV of hair growing cycle, and that of 0.1 ml and 0.2 ml S.B.V. acupuncture treated groups reached anagen phase VI and catagen, respectively. Immunohistochemical observations of the expression of various cytokines, enzymes and receptors in association with hair follicle cycle after local treatment of S.B.V. acupuncture therapy are as follows: Expression of fibroblast growth factor was more intense in epidermis and outer root sheath in 0.2 ml S.B.V. acupuncture treated group than that of 0.1 ml S.B.V. acupuncture treated group and control group. Expression of epidermal growth factor was more intense in bulge and outer root sheath in 0.2 ml S.B.V. acupuncture treated group than that of 0.1 ml S.B.V. acupuncture treated group and control group. Expression of c-kit receptor was more intense in epidermis, bulge and outer root sheath in 0.2 ml S.B.V. acupuncture treated group than that of control group. Expression of vascular endothelial growth factor was more intense in epidermis, bulge and outer root sheath in 0.2 ml S.B.V. acupuncture treated group than that of control group. Expression of protein kinase C-${\alpha}$ was more intense in epidermis, bulge and outer root sheath in 0.2 ml S.B.V. acupuncture treated group than control group. It is concluded that bee venom acupuncture therapy promoted the expression of various cytokines, enzymes and receptors related to the hair growth cycle for hair growth. This findings indicates that bee venom acupuncture therapy is applicable to the treatment of hair loss.
