Isolation, Identification, and Probiotic Properties of Lactobacillus reuteri HY701 from Human Feces

  • Strain HY701 was isolated from human feces for probiotic use by selecting highly resistant isolates to artificial gastric acid and bile acid. Strain HY701 was identified as Lactobacillus reuteri using 16S rDNA sequencing, and tentatively named L. reuteri HY701. The resistance of L. reuteri HY701 to artificial gastric acid (PH 2.5) was high with a survival rate of over 90%. L. reuteri HY701 also showed high tolerance to artificial bile acid after incubation in artificial gastric acid. Using the API ZYM test kit, the carcinogenic enzymes (${\beta}$-glucuronidase and (${\beta}$-glucosidase were not detected with L. reuteri HY70l, while the beneficial enzyme (${\beta}$-galactosidase was weakly detected. L. reuteri HY701 was sensitive to $100\;{\mu}g/mL$ nisin, $20\;{\mu}g/mL$ roxithromycin, $15\;{\mu}g/mL$ erythromycin, but resistant to $20\;{\mu}g/mL$ streptomycin, $10\;{\mu}g/mL$ tetracycline, $20\;{\mu}g/mL$ ciprofloxacin, $20\;{\mu}g/mL$ nystatin, $20\;{\mu}g/mL$ gentamycin, $10\;{\mu}g/mL$ doxycycline, $10\;{\mu}g/mL$ chloramphenicol, and $20\;{\mu}g/mL$ ampicillin. L. reuteri HY701 was shown to possess bactericidal activity as it inhibited the growth of Listeria monocytogenes ATCC 19111 and Escherichia coli JM109 completely within 24 hr of incubation. These results indicate that L. reuteri HY701 could be used as a probiotic strain.

향사양위탕(香砂養胃湯) 전탕액이 노화주의 비장, 췌장, 위장 세포의 항산화능에 미치는 영향 (Anti-oxidant Effect of Hyangsayangyi-tang Decoction in Stomach, Spleen and Pancreas Cell of SD Rats)

  • 1. Objectives The purpose of this study was to observe the anti-oxidant effects of Hyangsayangyi-tang(HY) in SD rats. 2. Methods This experiment was used the tissue of stomach, spleen and pancreas cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was HY-treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of stomach, spleen and pancreas cells. 3. Results and Conclusions The activity of SOD were significantly increased in spleen cell of 52, 68 w-HY group, pancreas cell of 68 w-HY group and in stomach cell of 52, 68 w-HY group compared with those of the normal and the control groups. The level of GSH were significantly increased in spleen cell of 52, 68w-HY group and in pancreas cell of 68w-HY group compared with those of the normal and the control groups. The level of MDA were significantly decreased in pancreas cell of 68 w-HY group compared with those of the normal and the control groups. The level of NO were significantly decreased in spleen cell of 68 w-HY group, pancreas cell of 52, 68w-HGD groups compared with those of the control groups. According to results, HY showed anti-oxidant effect. Investigation into the clinical use of the HY is suggested for future research.

Lactobacillus plantarum HY7712 Ameliorates Cyclophosphamide-Induced Immunosuppression in Mice

  • Lactic acid bacteria (LAB) in fermented foods have attracted considerable attention recently as treatment options for immune diseases, the incidence of which has been increasing worldwide. The ability of 500 strains of LAB, isolated from kimchi, to induce TNF-${\alpha}$ production in peritoneal macrophages was investigated. Lactobacillus plantarum HY7712 most strongly induced TNF-${\alpha}$ production as well as NF-${\kappa}B$ activation. However, HY7712 inhibited NF-${\kappa}B$ activation in LPS-stimulated peritoneal macrophages. When HY7712 was orally treated in cyclophosphamide (CP)-immunosuppressed mice for 5 or 15 days, it reversed the body and spleen weights, blood RBC and WBC levels, and splenocyte and bone marrow cells that were reduced by CP. Orally administered HY7712 increased concanavalin A-induced T cell proliferation to 84.5% of the normal group on day 15, although treatment with CP alone markedly reduced it to 53.7% of the normal group. Furthermore, orally administered HY7712 significantly induced the expressions of IL-2 and IFN-${\gamma}$ in ConA-induced splenic cytotoxic T cells of CP-treated mice. Orally administered HY7712 restored the CP-impaired phagocytosis of macrophages in mice. Orally administered HY7712 also restored the cytotoxicity of NK and cytotoxic T cells derived from spleen and bone marrow against YAC-1 in CP-immunosuppressed mice. Based on these findings, orally administered HY7712 may accelerate the recovery of cyclophosphamide-caused immunosuppression, without evident side effects, by immunopotentiating NK and Tc cells, and may provide a mechanistic basis for using HY7712 as an alternative means in lessening chemotherapyinduced immunosuppression in cancer patients.

HyRAM+를 이용한 수소 누출 시뮬레이터 구현 (Inplementation of a Hydrogen Leakage Simulator with HyRAM+)

  • 수소는 청정, 무탄소, 고에너지등 다양한 특징을 가진 재생에너지원으로 국제적으로 '미래에너지'로 인정받고 있다. 수소에너지 산업의 급속한 발전과 더불어 수소 수요를 충족시키기 위하여 더 많은 수소 인프라가 필요한 실정이다. 그러나 수소 인프라 사고가 빈번한 발생함으로 인해, 수소산업 발전에 걸림돌이 되고 있다. 미국 Sandia National Laboratories에서 개발한 HyRAM+는 수소충전소를 포함한 다양한 저장 응용 분야에 대한 수소 안전 평가와 관련된 데이터와 방법을 통합하는 소프트웨어 툴킷이다. HyRAM+의 물리 모드는 수소충전소 컴포넌트에 따라 수소누출 결과를 모사하여 가스 플룸 분산, 제트 프레임 온도와 궤적 그리고 복사 열속 등을 그래프로 나타낸다. 본 논문에서는 강원도 삼척시에 있는 수소충전소를 대상으로하여 HyRAM+ 소프트웨어를 이용하여 수소 누출 데이터를 추출하였다. HyRAM+에서 추출한 데이터를 이용하여 수소 누출(Leakage) 발생 시뮬레이터를 개발하였다. 데이터베이스와 그라파나(Grafana)를 이용하여 시뮬레이터에서 발생한 데이터를 보여주는 대쉬보드(Dashboard)로 구현하였다.

SGML에 기반한 하이퍼미디어를 위한 HyTime 엔진 설계 (HyTime Engine Design for Hypermedia Systems based on SGML)

  • HyTime(Hypermedia/Time-Based Structuring Language)는 SGML 문서에 하이퍼미디어를 포함하는 기능으로 확장하고, 하이퍼미디어의 논리 구조를 기술하기 위한 언어로 1992년 4월에 국제표준으로 채택되었다. 기존의 문서들은 각기 다른 플렛폼마다 그들만의 문서포맷이 정해져 있는 이유로 상이한 플랫폼에서도 문서를 서로 교환할 수 있도록 하기 위한 SGML 표준을 채택하게 되었다. SGML 환경하에서 많은 문서들은 하이퍼링크와 멀티미디어 정보를 포함하는 문서로 발전하게 된다. 하이퍼미디어문서의 처리를 위한 표준으로 HyTime이 정의되고 많은 어플리케이션들이 이를 적용하고 있으며 하이퍼문서로의 확장을 위한 HyTime 엔진이 요구된다. 이에 본 논문에서는 하이퍼미디어 문서 시스템을 구현하기 위한 필수 요소인 HyTime 엔진의 설계와 HyTime 엔진의 처리 절차에 관해 연구하였다.

Bifidobacterium longum HY8001의 섭취가 사람의 장내세균층 및 장내세균 효소에 미치는 영향 (Effect of Bifidobacterium longum HY8001 Administration on Human Fecal Bacterial Enzymes and Microflora)

  • The effects of Bifidobacterium longum HY8001 supplement intake on the fecal microflora and fecal bacterial enzyme activity were studied in ten healthy human volunteers, before, during and after intake (respectively for 3 weeks). During intake of B. longum HY8001 supplement, fecal, $\beta$-glucuronidase and nitroreductase activities significantly decreased 44.6%(p<0.005) and 32.3%(p<0.01), respectively. Although numbers of major bacterial groups of fecal microflora were not affected by B. longum HY8001 intake for 3 weeks, the number of Bifidobacterium was significantly increased (p<0.05). This result indicates that intake of B. longum HY8001 might be potentially beneficial for the prevention and inhibition of colon cancer and improvement of human intestinal microflora composition.

다국어를 지원하는 XML 문서 검색 시스템: HyREX (HyREX: Universal XML Retrieval Engine for XML)

  • HyREX는 연구용 프로토타입 XML 하이퍼미디어 문서 검색시스템으로 다국어를 지원하고 있다. HyREX는 검색을 위한 효율적인 접근 경로들을 처리하는 물리적 계층 HyPath와 질의어를 처리하는 논리적 계층 XIRQL 그리고 사용자 인터페이스인 HyGate 계층으로 이루어져 있다. 이 연구에서는 영어와 독일어 등의 검색을 지원하는 기존의 HyREX 시스템을 한글 XML 문서 검색시스템으로 확장하기 위해 먼저 한글 데이터타입을 위한 클래스를 구현하였다. 앞으로 한글 XML 문서 검색에서 정확율과 재현율을 향상하기 위해 각 문서의 인덱스에 대해 $tf{\cdot}idf$ 공식을 이용하여 가중치를 부여하고 이를 개발하고자 한다.

Lactobacillus brevis HY7401 섭취가 쥐의 혈중 알코올 수준에 미치는 영향 (Effect of Lactobacillus brevis HY7401 Intake on the Serum Ethanol Concentration in Rats)

  • Lactobacillus 균주들의 에탄올과 acetaldehyde의 대사 가능성을 시험관내 및 생체내에서 시험하였다. ADH와 ALDH 활성은 L. brevis 균주들이 다른 Lactobacillus 균주들과 비교하여 높게 나타났으며, 특히 L. brevis HY7401은 가장 높은 ADH와 ALDH 활성을 나타냈다. L. brevis HY7401$(10^9\;cfu/mL)$을 2주간 하루에 1 mL씩 쥐에게 급여한 후 시험 14일째에 에탄올(4g/kg BW)을 경구투여하고 혈중 에탄올 양을 측정한 결과, L. brevis HY7401 급여에 의해 혈중 에탄올 수준이 대조구에 비해 유의적으로 감소하는 것으로 나타났다. 에탄올 투여 6시간후의 혈중 에탄올 양이 각각 $1,195{\pm}92.9$(대조군) 그리고 $454{\pm}150mg/L$(HY7401군)으로 나타났다. 또한 L. brevis HY7401 급여한 군에서 소장내 에탄올 농도가 대조구와 비교하여 약 1/2수준이었으며, 초산 농도는 2배정도 높은 것으로 확인되었다. 따라서 이 연구결과들은 사람으로부터 분리된 L. brevis HY7401이 높은 ADH와 ALDH 활성을 갖고 있어 에탄올과 acetaldehyde를 대사한다는 것을 의미하고 있다. 또한 L. brevis HY7401을 쥐에게 급여함으로써 혈중 에탄올 수준을 낮출 수 있는 것으로 확인되었다.

100ksi급 고장력강의 모재 및 용접부 특성 (A characteristics of base metal and weldment of 100ksi class high strength steel)

  • A study was performed to investigate the properties of base metal and weldment for two HSLA steels and one HY-100 steel. Tensile, yield strength and elongation of HSLA-A steel were superior to those of HY-100 steel and yield ratios in HSLA-A and HSLA-B steels were higher than HY-100 steel owing to the precipitation of $\varepsilon$-Cu phase. The impact energy of HSLA-A steel was greater at all aging temperatures than that of HY-100 steel. HSLA-A and HY-100 steels had low impact transition temperature of about -l$25^{\circ}C$ and high upper shelf energy, The peak hardness of weldment in HSLA-A, HSLA-B and HY-100 steels were Hv 299, Hv 275 and Hv 441, respectively. The hardenability of HY-100 steel was largest due to the higher amount of carbon. The y-groove test showed that HSLA steels had superior resistance to cold cracking. Toughness of weld joint at the F. L. and F. L. +1mm in HSLA-A was almost the same as HY-100, but those at F. L.+3mm and F. L.+5mm was greater in HSLA-A steel.

Induction of Apoptosis in the HepG2 Cells by HY53, a Novel Natural Compound Isolated from Bauhinia forficata

  • In the search for a novel cytotoxic substance from medicinal plants, HY53 ($C_{17}H_{32}O_2N_2$; molecular weight 296) was isolated from the leaves of Pata de Vaca (Bauhinia forficata). The growth of the HepG2 cells was inhibited in a dose-dependent manner when treated with 0.07 to 0.40 mM HY53 for 24 h (IC$_{50}$: 0.13 mM). Furthermore, nuclear DAPI staining revealed the typical nuclear features of apoptosis in the HepG2 cells exposed to 0.27 mM HY53, whereas a flow cytometric analysis of the HepG2 cells using propidium iodide showed that the apoptotic cell population increased gradually from 8% at 0 mM to 23% at 0.14 mM and 45% at 0.40 mM after being exposed to each concentration of HY53 for 24 h. Moreover, a TUNEL assay also exhibited the apoptotic induction of the HepG2 cells treated with HY53. To obtain further information on the HY53-induced apoptosis, the expression level of certain apoptosis-associated proteins was examined using a Western blot analysis. Treatment of the HepG2 cells with HY53 resulted in the activation of caspase-3, and subsequent proteolytic cleavage of poly(ADP-ribose) polymerase (PARP). Consequently, the results confirmed that the apoptosis in the HepG2 cells was induced by HY53 and the involvement of caspase-3-mediated PARP cleavage in the apoptotic process.
