The aim of this study was to determine whether Hounsfield units (HUs), volume, and various void parameters can predict stone fragility in extracorporeal shock wave lithotripsy (ESWL). HU, volume, porosity, number of voids/stone volume, and void distribution of 30 struvite stones were estimated using helical computed tomography (CT) and micro-CT. The number of shock waves necessary for full fragmentation was accepted as a measure of the stone fragility in ESWL. The correlations between the number of shock waves and the HU, volume, porosity, and number of voids/stone volume were examined. The number of shock waves of the two groups according to the void distribution was also compared. Stone volume correlated with the number of shock waves. Shell-patterned struvite stones were significantly less susceptible to fragmentation in ESWL than non-shell-patterned struvite stones. Stone volume and void distribution may be predictors of the outcome of ESWL treatment.
Shiga toxins (Stxs) are major virulence factors from the enterohemorrhagic Escherichia coli (EHEC), a subset of Stx-producing Escherichia coli. Stxs are multi-functional, ribosome-inactivating proteins that underpin the development of hemolytic uremic syndrome (HUS) and central nervous system (CNS) damage. Currently, therapeutic options for the treatment of diseases caused by Stxs are limited and unsatisfactory. Furthermore, the pathophysiological mechanisms underpinning toxin-induced inflammation remain unclear. Numerous works have demonstrated that the various host ribotoxic stress-induced targets including p38 mitogen-activated protein kinase, its downstream substrate Mitogen-activated protein kinase-activated protein kinase 2, and apoptotic signaling via ER-stress sensors are activated in many different susceptible cell types following the regular retrograde transportation of the Stxs, eventually leading to disturbing intercellular communication. Therapeutic options targeting host cellular pathways induced by Stxs may represent a promising strategy for intervention in Stx-mediated acute renal dysfunction, retinal damage, and CNS damage. This review aims at fostering an in-depth understanding of EHEC Stxs-mediated pathogenesis through the toxin-host interactions.

Previously, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein, here named RCMF [${\underline{R}}hus$ verniciflua Stokes (RVS) ${\underline{c}}hloroform-{\underline{m}}ethanol\;{\underline{f}}raction$], was prepared from a crude acetone extract of RVS which is traditionally used as a food additive and as an herbal medicine. In the present study, we investigated the effects of $TNF-{\alpha}$ on RCMF-induced growth inhibition and apoptosis induction using human osteosarcoma (HOS) cells. The results from tritium uptake and MTT assays showed that $TNF-{\alpha}$ treatment itself (10 ng/ml) did not induce any cytotoxicity, but it actively accelerated RCMF-mediated cytotoxicity of HOS cells. RCMF-induced cytotoxicity and its facilitation by $TNF-{\alpha}$ was verified to be apoptotic, based on the increased DNA fragmentation and low fluorescence intensity in nuclei after propidium iodide (PI) staining of HOS cells. This speculation was further demonstrated by monitoring the Annexin V/PI double staining which could discriminate the difference between apoptotic and necrotic deaths. Collectively, our findings indicate that $TNF-{\alpha}$ accelerates RCMF-induced cytotoxicity in HOS cells.
For a long time Rhus verniciflua Stokes (RVS) has traditionally been used as a herbal plant. It is known to contain various biological activities. Previously, a crude ethanol extract from RVS was reported to have antioxidant effects, and antiproliferative activities, on human cancer cell lines. In this report, we prepared a highly purified ethanol extract from RVS, which did not contain the urushiol derivatives, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), to investigate the mechanisms of the scavenging activity of hydroxyl radicals using mouse thymocytes. The results from the deoxyribose, DNA nicking, and glucose/glucose oxidase enzyme assays showed that REEE-1 contained a strong scavenging activity of oxygen free radicals, especially of hydroxyl radicals. However, interestingly, REEE-1 also showed cytotoxicity against the thymocytes, although the effect was variable, depending on the concentrations and times of treatment. The REEE-1-mediated cytotoxicity against thymocytes, which has been used as one of the well-characterized models for apoptosis studies, was verified to be apoptotic. This was proven by the following: the appearance of DNA laddering, increases in DNA fragmentation, low fluorescence intensity in the nuclei after propidium iodide staining, and positive Annexin V staining of the cells. These results suggested that REEE-1 had both antioxidative activity and cytotoxicity against the thymocytes, although the effect of the cytotoxicity was variable, depending on the dose and time of the treatment.
Shiga toxin (Stx)-producing Escherichia coli (STEC) strains can cause broad spectrum of human disease, including diarrhea, hemorrhagic colitis, and the life-threatening hemolytic uremic colitis (HUS). We examined 868 samples was taken from bovine feces and carcass from January to December 2008 in Seoul. Twenty two (9.5%) shiga toxin -producing Escherichia coli were isolated from the 230 of bovine feces, and two (0.31%) were isolated from the 638 of carcasses. Serotype of E. coli isolates were O157 (10, 41.6%), O26 (10, 41.6%), O111 (1, 4.2%) and UT (3, 12.6%). In PCR, the isolates displayed three different stx gene combination (stx1 [2, 8.4%]), stx2 [3, 12.6%] and stx1 and stx2 [19,87.5%]). The eaeA and hlyA gene were found in 11 (45.8%) of the 24 strains. Saa gene was present only one strains (4.2%). Toxin typing using reverse passive latex agglutination test showed the same result in VT 1. But it showed different result in VT 2. In antimicrobial susceptibility test, all isolates were sensitive to amikacin, amoxicillin/clavulanic acid, ciprofloxacin and colistin. Eighteen strains (75.0%) of 24 isolates showed the multi-resistant patterns with over 3 drugs. PFGE was performed after the genomic DNA of twenty four isolates was digested with Xba I. the 24 isolates showed 7 (A~G) PFGE type.
Enterohemorrhagic Escherichia coli (EHEC) O157:H7 is well-characterized as an important food-borne pathogen worldwide and causes human diseases such as diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome (HUS) by producing shiga-like toxin (Stx). It has been reported that a number of dairy foods, including cheese, can act as the source of EHEC O157:H7 infections. In addition to the toxicity of Stx, recently it has been indicated that EHEC O157:H7 possesses virulence factors related to attachment, quorum sensing, and biofilms. Moreover, these novel virulence factors might become critical points to be considered in the future production of food derived from animals. Here, we review the evidences that support these insights on new virulence factors and discuss the potential mechanisms mediating the pathogenesis of EHEC O157:H7 in the dairy food industry.
Rhus verniciflua Stokes (RVS) has been used as a traditional herbal medicine. Several earlier studies indicated that an ethanol extract of RVS has both anti-oxidant and anti-tumor properties, although the mechanism for the activity remains to be elucidated. In this report, we prepared a highly purified ethanol extract from RVS, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), and investigated the mechanism involved in its growth-inhibitory effect on the human B and T lymphoma cell lines, BJAB and Jurkat, respectively. Results from tritium uptake proliferation assays showed that the proliferative capacities of both BJAB and Jurkat cells were strongly suppressed in the presence of REEE-1. This was further confirmed through trypan blue exclusion experiments that revealed a dose-dependent decrease in viable cell numbers after REEE-1 treatment. REEE-1-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. In particular, REEE-1 exerted its anti-oxidant activity through the inhibition of hydroxyl radical-mediated degradation by iron ion chelation rather than direct scavenging of hydroxyl radicals. Furthermore, REEE-1 was revealed to be a potential scavenger of superoxide anions. Collectively, our findings suggest that REEE-1 is a natural anti-oxidant that could be used as a cancer chemo-preventive and therapeutic agent.
CGCM3.1 SRES B1 시나리오의 2D 변수들을 입력값으로 인공신경망 모형을 이용한 스케일 상세화기법으로 강부식(2009)은 소양강댐 유역의 월 누적강수 경향분석을 실시하였다. 원시 GCM 시나리오를 스케일 상세화 시키기 위한 기법의 하나로 인공신경망 모형을 사용할 수 있는데, 이 경우 GCM에서 모의되는 강수플럭스, 해면기압, 지표면 근처에서의 일 평균온도, 지표면 근처에서의 일평균온도, 지표면으로부터 발생하는 잠열플럭스 등과 같은 22개의 변수를 잠재적인 예측인자로 사용하여 신경망을 구성하게 된다. 입력변수세트의 구성은 인공신경망의 계산 효율을 좌우하는 중요한 요소라 할 수 있다. 본 연구에서는 변수의 물리적 특성을 고려하여 순차적인 변수선택을 통한 신경망 입력변수 세트를 구성하고 입력세트 간의 학습성과 비교를 통하여, 최적 입력변수 선정 및 신경망의 학습효과를 높일 수 있는 방법에 대해 연구하였다. 물리적 상관성이 높다고 판단되는 GCM_Prec, huss, ps를 입력변수로 하여 순차적인 케이스를 학습해본 결과 huss와 ps를 입력변수로 하는 케이스에 대해서 적은 오차와 높은 상관성을 보였다, 또한, 신경망의 학습 효과를 높이기 위해 홍수기와 비홍수기로 구분하여 학습한 결과 홍수기와 비홍수기로 구분하여 신경망을 구성하였을 경우가 향상된 모의값을 나타내었다. 기후변화모의자료는 CCCma(Canadian Center for Climate Modeling and Analysis)에서 제공되는 CGCM3.1/T63 20C3M 시나리오를 사용하였으며, 관측값으로는 AWS에서 제공된 일 누적강수를 사용하였다. 인공신경망의 학습기간은 1997년부터 2000년이며, 검증기간은 2001년부터 2004년으로 구성하였다.
지식정보사회에서 발전된 네트워크 인프라를 바탕으로 전산망의 구조는 보안성을 확보한 다양한 형태의 망 구성을 구축하여 운영하고 있다. 국가·공공기관 전산망의 경우 각 기관별 특성과 연계 기관까지 고려한 기술적, 관리적 보안환경 구축이 필요하며, 이를 위해 기관별 특성에 따른 사이버 위협을 분류 및 위협 지도를 바탕으로 기술적·관리적 취약점 및 사이버 위협을 분석 등을 통한 사이버 훈련을 위한 기본 연구의 중요성이 대두되고 있다. 따라서 본 연구에서는 인터넷망과 국가정보통신망의 이원적 인프라 망을 기반으로 구축되어진 국가·공공기관 전산망의 내·외부 사이버 위협에 따른 유형별 분석을 사례 기반의 시나리오를 통해, 실질적인 사이버 위협 요소를 도출 및 분석하여 사이버보안 훈련 요소 도출용 사이버 위협 MAP을 제시하고자 한다.
