Traditionally, dyes extracted from onion shells have been used as natural pigments but dyeing methods with reproducibility have not been developed. In this study, the dyes were analyzed by high performance thin layer chromatography(HPTLC) to obtain the ratio of pigments in water and ethyl acetate extract. Furthermore, the present study was also carried out to obtain the information characteristic of wavelength which proportionally decrease for continuous dyeing reaction. Scanning of water extract showed 4 peaks by the mobile phase of benzene/ethyl acetate/acetic acid(40/10/5) and the peak 1 exhibited yellow color with the maximal absorption spectra of 306nm and 309nm while ethyl acetate extract showed 9 peaks in the same mobile phase. The water extract after ethyl acetate extraction was adjusted to 0.5 of O.D.(optical density) at 550nm by adding of water and compared wavelength of the pigments from 200 to 600nm to find the proportional decrease of wavelength. As the result, it showed that wavelength of 306nm could be the standard of dyeing monitoring points.

  • Ganglioside GM3 and sialidase activities in human fetal liver have been investigated. Gangliosides were extracted from fetal livers by the Folch-Suzuki method and analyzed by high-performance thin layer chromatography (HPTLC). GM3 increased, but lactosylceramide (LacCer) decreased predominantly over the developmental stages. Sialidase in human fetal liver was mainly localized in the lysosomal fraction and its activity was high in the earlier stages of development. The optimum pH for this enzyme was 4.3~4.4. Sialidase was more active with the ganglioside mixture than with GM3, sialyllactose or fetuin. Fetal liver sialidase was still active (20% activity) in the presence of 25% methanol. These results suggested that the changes of the ganglioside GM3 and sialidase activity may be involved in the regulation of cell growth in human fetal liver during development.

  • The entire plant of Leptadenia reticulata (Asclepidaceae) is extensively used as lactogen, traditionally, in veterinary practice. The plants of Dregea volubilis and Pentatropis microphylla (Asclepidaceae) are now used as its substitute and sometimes replace the original drug as traditional lactogen. The lactogenic potential of these drugs was studied in rats using, pup weight, weight of mother, parenchyma percentage, secretary rating, estimation of total protein content and glycogen content of mammary glands tissues as assessment parameters. HPTLC profiles of bioactive extracts were also generated to serve the authentification needs. The results of present studies show that P.microphylla forms a better substitute over D. volubilis.

  • The distribution of phenolic components of Korean ginseng (Panax ginseng KG) and American ginseng (Panax quinquefolium, AG) were compared by thin-layer chromatography (TLC). Silica gel TLC gave 3~4 spots, while $NH_2$ HPTLC 5~6 spots, which were colored by both $FeCl_3$/$K_3Fe(CN)_6$ and Folin-Ciocalteu. The distribution of phenolic components was quite different between KG and AG. Especially, a polyphenol (m.w. 578), which had been isolated from KG by the author, was not found in AG. This result suggests that the polyphenol could be used as an index compound for the differentiation of KG from AG.

  • Chlorophytum borivilianum Baker (Antharicaceae) commonly referred as 'Safed Musli' has been widely used in the Indian traditional systems of medicine to treat various diseases like rheumatism apart from having immunomodulating property and is used as general tonic. It is also known as 'Ayurvedic viagra' for its aphordisiac properties. C. borivilianum was screened for the first time to determine its antioxidant activity, isolation of the sapogenins and standardization of the isolated sapogenin fraction using HPTLC. Potent antioxidant activity of ethanolic extract was found by their ability to scavenge DPPH (84.51%), hydroxyl radical (48.95 %), ferryl bi-pyridyl complex (84.53%) along with the inhibition of lipid peroxidation (67.17%) at $100\;{\mu}g/ml$ concentration. The ethanolic extract also exhibited significant inhibition of superoxide anion radical generated by photochemiluminescence. Thus, the potent antioxidant activity validates the innumerable therapeutic claims of the plant in the traditional system especially its use as a Rasayana drug.
