Saccharomyces cerevisiae deletion mutant의 세라마이드 생합성 (Biosynthesis of ceramide by deletion mutant of Saccharomyces cerevisiae)

  • Saccharomyces cerevisiae의 deletion mutant를 이용하여 ydc1, ypc1, scs7, sur1, csg2, ipt1, Icb4, Icb5, dpll의 deletion이 세라마이드의 생산에 미치는 영향을 고찰하였다. 세라마이드는 ELSD가 연결된 HPLC를 통하여 분석하였으며 ${\triangle}$ydc1 mutant의 세라마이드 생산량이 6 mg ceramide/g cell로 최대량을 나타내었으며 ${\triangle}$sur1 mutant, ${\triangle}$lcb5 mutant, ${\triangle}$dpll mutant의 경우 control로 사용한 BY4742와 비슷한 세라마이드 생산량을 나타내었고, 그 외 ${\triangle}$ypc1 mutant, ${\triangle}$scs7 mutant, ${\triangle}$csg2 mutant, ${\triangle}$ipt1 mutant, ${\triangle}$lcb4 mutant는 BY4742보다 낮은 세라마이드 생산량을 나타내었다.

효소적 Acyl migration을 이용한 비대칭형 재구성지질(1,2-disaturated-3-unsaturated glycerol)의 합성 및 분석 (Enzymatic synthesis of asymmetric structured lipids containing 1,2-disaturated-3-unsaturated glycerol using acyl migration)

  • The enzymatic interesterification was performed to produce structured lipids (SLs) with palm mid fraction (PMF) and stearic ethyl ester (STEE) for 1, 3, 6, 9, 12 and 15 hr at $80^{\circ}C$. The reaction was catalyzed by Lipozyme TLIM (immobilized lipase from Thermomyces lanuginosus, amount of 20% by weight of total substrates) in a shaking water bath set at 180 rpm. The optimum condition for synthesis of asymmetric SLs were: substrate molar ratio 1:0.5 (PMF:STEE, by weight), reaction time 6 hr, enzyme 20% (wt%, water activity=0.085) of total substrate and reaction temperature $80^{\circ}C$. After reaction at optimized condition, triacylglycerols (symmetrical and asymmetrical TAGs) from reactants were isolated. POP/PPO (1,3-palmitoyl-2-oleoyl glycerol or 1,2-palmitoyl-3-oleoyl glycerol), POS/PSO (palmitoyl-oleoyl-stearoyl glycerol or palmitoyl-stearoyl-oleoyl glycerol), SOS/SSO (1,3-stearoyl-2-oleoyl glycerol or 1,2-stearoyl-3-oleoyl glycerol) were obtained by solvent fractionation. Finally, refined SLs contained stearic acid of 16.91%. Solid fat index and thermogram of the refined SLs were obtained using differential scanning calorimetry. The degree of asymmetric triacylglycerol in the refined SLs was analyzed by Ag-HPLC equipped with evaporated light scattering detector (ELSD). The refined SLs consisted of symmetric TAG of 41.15 area% and asymmetric TAG of 58.85 area%.

  • For the purpose of the quality control of Poria cocos, three major compounds were isolated and identified as pachymic acid (1), $3{\beta}-hydroxylanosta-7,9(11),24-trien-21-oic$ acid (2), and dehydroeburicoic acid (3). The optimal extraction conditions for the quantification of pachymic acid its analogues were the 3 hours of reflux with 15g of P. cocos in 100ml 95% ethanol. HPLC conditions were as follows: Column; ZORBAX Eclipse XDB C18 $(4.6{\times}250\;mm,\;Agillent)$, mobile phase; 1% HOAc in 70% $MeOH{\rightarrow}1%$ HOAc in 100% MeOH for 25 min, then 1% HOAc in 100% MeOH for 15 min, detector, ELSD, flow rate; 1ml/min. The mean contents of 1,2, and 3 in Poria cocos cultivated at 18 different site were $0.65{\pm}0.19,\;0.88{\pm}0.72,\;and\;0.84{\pm}0.54\;mg/g$, respectively, and values might be the guide line for the quality control of P. cocos.

초임계 $CO_2$와 에탄올 혼합물에 의한 대두인지질의 선택적 추출 (Selective Extraction of Phospholipids from Soybeans with Mixture of Supercritical Carbondioxide and Ethanol)

  • Supercritical carbondioxide is very effective in removing oils from a variety of seed matrices, devoid of any appreciable amount of phospholipid content. However, the limited solubility of phosphalipids in supercritical carbondioxide leaves behind a potentially valuable by-product in spent seed matrix. Any phospholipid extraction process from the spent matrix must maintain the structure and the quality of phospholipids and must be compatible with the end use of the seed protein meal an animal feed or for human consumption. An initial supercritical carbondioxide extraction of soybean flakes was performed at 32 MPa and $80^{\circ}C$ to extract the oils, leaving the phospholipids in the deflatted soybean flakes, A second step was performed on the defatted soybean flake using $X_{eth}$=0.10, Varying the pressure from 175 MPa to 70.6 MPa and temperature from $60^{\circ}C$ to $80^{\circ}C$. For all supercritical carbon dioxide/ethanol mixture extractions, a fraction rich in phospholipids was obtained. The fractions extracted from defatted soybean flakes were dried and them redissolved in chloroform before HPLC-ELSD analysis. Quantitative and qualitative analysis of phospholipids on soybean seeds, defatted soybean flake, and different extracted phospholipid fractions was carried out, to ascertain the effect of extraction pressure and temperature.

프랜차이즈 치킨과 대형마트 내 판매 치킨의 당과 나트륨 함량 조사 (Sugar and Sodium Content of Franchise Chickens and Market Chickens)

  • 서울시내에 위치한 프랜차이즈 매장과 대형마트 내 판매되고 있는 4종(후라이드, 양념, 간장, 치즈가루 치킨)의 치킨제품 123건에 대하여 당과 나트륨 검사를 실시하였다. 전체 치킨제품의 평균 당 함량은 100 g 당 4.5 g이었으며, 양념치킨(8.7 g), 치즈가루 치킨(4.3 g), 간장치킨(3.6 g), 후라이드 치킨(0.6 g) 순으로 나타났다. 평균 나트륨 함량은 전체 치킨제품의 경우 100 g 당 552.4 mg이었으며, 치즈가루 치킨(627.0 mg), 간장치킨(536.3 mg), 양념치킨(511.2 mg), 후라이드 치킨(448.0 mg) 순으로 나타났다. 프랜차이즈와 대형마트 내 판매 치킨제품을 비교하였을 때, 평균 당 함량의 경우 100 g당 대형마트 내 판매 후라이드 치킨(0.8 g)과 양념치킨(8.9 g)에서 모두 프랜차이즈의 후라이드 치킨(0.5 g)과 양념치킨(8.6 g)보다 높았으며, 프랜차이즈와 대형마트 내 판매 제품에 따른 유의적으로 차이를 보였다(P<0.05). 평균 나트륨 함량의 경우 100 g당 프랜차이즈 양념치킨(516.0 mg)과 대형마트의 후라이드 치킨(471.5 mg)이 높은 값을 보였으며, 유의적인 차이는 보이지 않았다. 포장량 전체를 섭취했을 경우, 평균 당 함량의 섭취비율은 후라이드 치킨은 모두 WHO에서 권고하는 1일 당류 섭취권고량인 50 g 이하였고, 간장치킨(47.6%)과 치즈가루 치킨(55.4%)은 절반 수준이었으나, 양념치킨은 프랜차이즈(140.4%)와 대형마트(148.8%) 제품 모두 권고량을 넘어섰다. 나트륨은 WHO의 1일 나트륨 섭취권고량인 2,000 mg 대비 섭취비율이 후라이드 치킨은 프랜차이즈(140.1%)와 대형마트(170.3%) 제품과 간장치킨(167.9%)에서 높게 나타났다. 특히, 양념치킨은 프랜차이즈(203.7%)와 대형마트(208.4%) 제품 모두와 치즈가루 치킨(202.4%)에서 섭취권고량의 2배를 넘어선 것으로 나타났다.

코르티코스테론 및 멜라토닌 수치 조절을 통한 동애등에 물 추출물의 항스트레스 및 수면 개선 효과 (Anti-stress and Sleep-enhancing Effects of Ptecticus tenebrifer Water Extract Through the Regulation of Corticosterone and Melatonin Levels)

  • 동애등에는 파리목(Diptera) 동애등에과(Stratiomyidae)에 속한다. 최근에 곤충 산업은 식품, 동물사료 및 환경개선 이점으로 주목받고 있다. 가바 및 멜라토닌은 수면 및 우울증 조절에 관여하며, 가바는 억제성 신경전달물질이며, 유산균에 의해 MSG가 가바로 생체전환되어 합성된다. 본 연구에서는 MSG를 동애등에에 급여하여 가바 생성이 증가된 동애등에 추출물을 사용하였다. 스트레스 및 불면증에 대한 작용은 코르티코스테론에 의해 유도된 ER 스트레스 및 만성 구속 스트레스(CRS)에 노출된 마우스 모델과 마우스에 펜토바르비탈(45 mg/kg)-유도 수면개선에 미치는 메커니즘에 대해 조사하여 확인하였다. 본 연구에서 MSG를 급여하지 않은 동애등에 추출물과 MSG를 급여한 동애등에 추출물을 HPLC-ELSD기기분석을 통해 GABA 피크를 확인한 결과, MSG를 급여한 동애등에 추출물에서만 GABA 피크가 나타남을 확인하였다. 또한, MSG를 급여한 동애등에 물 추출물 및 동애등에 70% 에탄올 추출물이 CORT에 의해 유도된 인산화된 ERK1/2 및 CREB의 하향 조절에 대해 보호함으로써 신경세포 보호 효과가 있을 확인하였다. 동애등에 물 추출물(300 mg/kg)은 CRS-노출된 마우스에서 혈청 코르티코스테론 수치를 유의적으로 감소시켰다. 더 나아가, 동애등에 물 추출물(100 및 300 mg/kg)는 혈청 멜라토닌 증가에 관여하여 펜토바르비탈(45 mg/kg)로 유도된 수면 행동에서 입면시간 감소 및 총 수면 시간을 증가시켰다. 결론적으로, 동애등에 물 추출물은 혈청 코르티코스테론 및 멜라토닌 수치를 조절함으로써 항스트레스 및 수면 향상 효과가 있는 것으로 사료된다.

Comparison of Chemical Constituents in Mung bean (Vigna radiata L.) Flour between Cultivation Regions and Seeding Dates

  • Legumes are one of the largest families of crop plants and are widely consumed and produced for their nutritional and commercial benefits. Mung bean (Vigna radiata L.) is a legume crop that contains various functional compounds ; moreover, it has strong antioxidant properties and is becoming an increasingly important food crop. However, most previous studies on mung beans have focused on their primary metabolites. In this study, we investigated the composition and contents of phenolic compounds, fatty acids, soyasapogenol and tocopherol in mung beans cultivated in different regions and cultivated at different seeding dates. Material analysis was conducted using the following methods: LC-MS/MS, GC-FID and HPLC-ELSD. In total, 57 different samples were analyzed. Thirteen phenolic compounds were detected in mung beans. Of these, vitexin and isovitexin were the most abundant compounds, accounting for approximately 99% of phenolic compounds. The difference in phenol compounds according to the seeding dates of mung bean was not statistically significant. The total fatty acid content in beans was the highest in Pyeongchang. Significant differences in total fatty acid content were found according to the cultivation regions. Crops grown in Sohyeon and Dahyeon showed the highest soyasapogenol B content in the Suwon region, and these were the lowest in Jeonju. The total tocopherol content of beans cultivated in Dahyeon and Sohyeon was the lowest and highest in Pyeongchang. Soyasapogenol B and total tocopherol content were not significantly different according to seeding dates. This study was conducted to obtain basic data for the cultivation of mung beans with a high content of various functional materials in terms of regional specialization and optimal seeding time.

국내산 백삼과 태극삼의 크기 및 연근별 인삼사포닌 함량 (Ginsenoside Contents of Korean White Ginseng and Taegeuk Ginseng with Various Sizes and Cultivation Years)

  • 고려인삼의 수출확대를 위해서 백삼 및 태극삼의 사포닌(saponin) 함량에 대한 중국 고려인삼 수입의약품 둥록기준 설정의 기초 자료를 얻고자 조사하였다. 백삼 50구의 크기에 따른 초특대편, 특대편, 대편, 중편 및 소편의 ginsenoside-Rg1, -Re 및 -Rb1의 평균 함량은 각각 664.7, 796.9, 674.7, 839.0 및 646.6 mg%이었으며, Rg1/Re의 비율은 각각 1.0, 1.2, 0.8, 1.0 및 1.0의 분포였다. 태극삼 13구의 ginsenoside-Rg1, -Re및 -Rb1의 평균 함량은 755.1 mg%, Rg1/Re의 비율은 1.28이었다. 그리고 백삼 50구의 Rg1 평균값은 $232.7{\pm}110.2 mg%$, Re평균값은 $235.3{\pm}101.5 mg%$, Rb1 평균값은 $280.1{\pm}121.3 mg%$으로 이들의 합은 $748.2{\pm}299.4 mg%$이었으며, Rg1/Re의 비율은 1.02이었다. 또한 태극삼 13구의 사포닌 성분의 분석결과, Rg1 평균값은 $262.1{\pm}127.2 mg%$, Re 평균값은 $213.1{\pm}55.7 mg%$, Rb1 평균값은 $279.9{\pm}92.1 mg%$으로 이들의 합은 $755.1{\pm}233.6 mg%$이었다. 백삼과 태극삼의 사포닌 조성 및 함량은 중국수입의약품 등록기준인 ginsenoside-Rg1, -Re 및 -Rb1 값의 합이 0.4% 이상이라는 기준규격에 적합하였고, HPLC-ELSD로 분석시 인삼의 분석방법별 기준인 ginsenoside -Rg1과 -Re의 함량비($Rg1/Re{\Leq}3.87$)에 부합되었다.

광주지역 학교주변 길거리 음식 중 당, 나트륨, 인공감미료 함량 실태조사 (A Survey on Total Sugar, Sodium and Artificial Sweetener Contents of Light Meals from the School Zone in Gwangju)

  • This survey was conducted to monitor the total sugar, sodium, and artificial sweetener contents of light meals from the school zone in Gwangju, from November, 2008 to April, 2009. A total of 100 samples were tested. HPLC/ELSD was used for the determination of total sugar contents and AAS for sodium contents. Total sugar amounts were the sum of both mono-and disaccharide according to nutritional information standard defined by the Korea Food and Drug Administration. The results were as follows by the form of total sugar content(sodium content): $5.7\;{\pm}\;1.9%$($4.6\;{\pm}\;1.6\;mg/g$) for Tteokbokki, $7.6\;{\pm}\;5.1%$($5.4\;{\pm}\;2.7\;mg/g$) for Chicken(skewed), $15.4\;{\pm}\;3.2%$($3.0\;{\pm}\;0.9\;mg/g$) for Bun(fish-shaped), $0.6\;{\pm}\;0.3%$($3.7\;{\pm}\;1.0\;mg/g$) for Sundae, $0.9\;{\pm}\;0.3%$($7.4\;{\pm}\;1.4\;mg/g$) for Oden(with broth), $20.5\;{\pm}\;6.2%$($2.9\;{\pm}\;0.6\;mg/g$) for Waffle, $6.8\;{\pm}\;2.2%$($4.7\;{\pm}\;1.0\;mg/g$) for Hotdog, $14.2\;{\pm}\;2.8%$($3.1\;{\pm}\;1.6\;mg/g$) for Hotteok, $6.6\;{\pm}\;2.1%$($3.9\;{\pm}\;0.6\;mg/g$) for Toast, $10.1\;{\pm}\;3.7%$($2.3\;{\pm}\;0.7\;mg/g$) for Fry(sweet potato), $1.6\;{\pm}\;0.7%$($4.0\;{\pm}\;0.7\;mg/g$) for Fry(etc), and $9.3\;{\pm}\;2.4%$($4.0\;{\pm}\;0.6\;mg/g$) for Doughnut. In addition, sodium saccharin, one of the artificial sweeteners, was tested. Among 60 samples from February to April, 2009, 11 samples had sodium saccharin(6.7~101.0 mg/kg).

Comparison of Soyasapogenol A, B Concentrations in Soybean Seeds and Sprouts

  • Soybean seeds contain many biologically active secondary metabolites, such as proteins, saponins, isoflavones, phytic acids, trypsin inhibitors and phytosterols. Among them, saponins in soybeans have attracted considerable interest because of their health benefits. Soyasaponin A and B are the most abundant types of saponins found in soybeans along with soyasapogenol (aglycone), which is a precursor of soyasaponin. The main purpose of this experiment was to determine the concentration of soyasapogenol in soybean seeds and sprouts as a function of seed size, usage, seed coat color and seed cotyledon color. The 79 Korean soybean varieties were cultivated at Yesan of Chungnam in 2006 for the analysis of soyasapogenol using HPLC with Evaporative Light Scattering Detection (ELSD). The total average concentration of soyasapogenol was $1313.52{\mu}g\;g^{-1}$ in soybean seeds and $1377.22{\mu}g\;g^{-1}$ in soybean sprouts. Soybean sprouts were about 5% higher than soybean seeds in average total soyasapogenol concentration. In the process of sprouting, the average soyasapogenol A content decreased by approximately 1.6%, but soyasapogenol B and total soyasapogenol increased by 8.31% and 4.88%, based on the content of soybean seeds. When classified according to the size of seeds, the total soyasapogenol concentration of soybean seeds were not significantly different (p<0.05) On average, small soybean seeds were increased by as much as $103.14{\mu}g\;g^{-1}$ in sprouting process. As a function of the use of the seeds, The total soyasapogenol in soybean seeds were significantly different (p<0.05). While, the soybean sprouts were not significant different (p<0.05). Altogether, sprout soybean seeds show the greatest change in content during the germination process. When seeds with different coat colors were compared, the total soyasapogenol concentration of soybean with yellow seed coats ($1357.30\mu g\;g^{1}$) was slightly higher than that of soybean with black ($1260.30{\mu}g\;g^{-1}$) or brown ($1263.62{\mu}g\;g^{-1}$) seed coats. For the color of the cotyledon, the total soyasapogenol concentration was significantly increased in green cotyledon during the germination and seedling process. The results of this study suggest the functional characteristics of soybeans through quantitative analysis of soyasapogenol. In addition, the concentration of soyasapogenol exhibited a change during the germination process, which was evaluated by the nutritional value of the soybean sprouts.
