타액반 피검물에서 개인식별을 위한 DNA의 유전자형 검사 (Typing of DNA Extracted from Cigarette Butts for Individual Identification)

  • Cigarette butts from 5 smokers were gathered and then, placed in room temperature for 1, 3, 5, 7, 15 days. The possible use of the cigarette butts for individual identification was evaluated in sex determination, amplification of D1S80 locus, polymorphisms of HLA-DQA1 gene from the extracted DNA. 1. DNA extraction was possible in cigarette butts weree left in room temperature for 15days, so it can be applicatable to individual identification by polymerase chain reaction(PCR). 2. Amplification of X-Y homologous amelogenin gene by PCR made it possible to identify the sex in saliva stains (cigarette butts). 3. Amplification of D1S80 locus can be acquired from adding the boving serum albumin and hot start PCR procedures from forensic samples such as saliva stains (cigarette butts), so the AMP-FLPs examining is possible. 4. Genotype could be determined simply and rapidly using Amplitype$TM$ HLA-DQ$\alpha$ forensic kit in examining the HLA-DQA1 gene. From the investigation, DNA extraction, sex determination, amplification of D1S80 locus, polymorphisms of HLA-DQA1 gene was successfully done even though the cigarette butts were left for 15 days at room temperature. Therefore cigarette butts are highly reliable and applicatable as molecular biologic samples for individual identification.

Replicated Association Study between Tuberculosis and CLCN6, DOK7, HLA-DRA in Korean

  • Tuberculosis is a global public health problem and manifests itself as a difference in the genetic susceptibility of the host, along with the properties of Mycobacterium tuberculosis (MTB). The single nucleotide polymorphisms (SNPs) and candidate genes proposed in the Genome-wide association study (GWAS) on tuberculosis in a recently published Chinese population were reported. In this study, we investigated whether the genetic polymorphism of candidate genes related to tuberculosis is reproduced when targeting Koreans. The CLCN6 (rs12404124, rs198391, rs535107), DOK7 (rs1203104, rs1203103) and HLA-DRA (rs1051336) gene polymorphisms showed statistically significant results. In addition, it was also found whether it acts as an expression quantitative trait loci (eQTL) that can influence gene expression. This study confirmed that the genetic polymorphism of the three genes (CLCN6, DOK7, HLA-DRA) affects the development of tuberculosis and will help to understand the genetic specificity of tuberculosis and the interaction between pathogens and hosts.

다중경로 음파전달이 HLA의 근거리 방위탐지에 미치는 영향 (Effects of the Multipath Propagation on the Source Bearing Detection of HLA at near range)

  • 다중경로의 영향을 분석하기 위하여 다중경로가 방위로 나타나는 과정을 설명하고, 동해와 남해의 대표적인 수직 음속구조를 이용하여 계산한 다중경로로부터 방위를 추정하였다. 또한 HLA의 빔형성 시뮬레이션을 실시하여 빔출력에 나타나는 잘못된 방위를 관찰하고 분석하였다. 수온 구조가 강한 음(-)의 기울기를 갖는 동해의 경우에 근거리에서 탐지된 방위가 실제 수평방위와 다르게 나타나고, 음원의 수가 잘못 탐지되는 구간이 발생할 가능성도 있는 것으로 보인다.

  • 단백질 항원에 존재하는 에피토프는 에피토프를 기반으로 한 백신 개발의 표적이 되고 있다. 인간의 주조 직적합 복합체 (MHC-1)에 결합하는 펩타이드를 확인할 수 있는 여러 서버들이 보고되고 있으나 인간의 MHC-I 분자의 수가 매우 많고 각 서버 검색 방법의 표준화 부재 등의 문제로 인해 펩타이드 예측에 적절한 서버를 선정하는 것이 쉽지 않다. 본 논문에서는 MHC-I 결합 펩타이드를 예측하는 서버 30 종을 비교하였으며, 다발성 골수종에 적용하기 위해 survivin 단백질로부터 사람의 HLA-A2 제한 펩타이드를 예측하였다. 본 연구의 결과는 MHC-I 결합 예측의 표준화된 방법을 제시하고 펩타이드 에피토프를 예측하는데 도움을 줄 것이다.

  • BamHI restriction pattern and genomic library of Herpes simplex virus type 2 (HSV-2) strain G were constructed, and locations of the glycoproteins gB and gD, and tk genes on the fragments were detected by Southern blot analysis. HSV-2 genomic DNAs were cleaved into twenty-seven fragments by BamHI enzyme in the range of 0.72 to 15.08 (total 150.44 kb), which were cloned into the BamHI site of pBluescript SK(+) to construct genome library of the HSV-2. The library was named by the order of the fragment size from smallest one to largest one. HSV-2 glycoprotein gD gene was located in pHLA2-21 and pHLA2-22 recombinant plasmids, gB gene in pHLA2-24 plasmid, and tk gene in pHLA2-11 clone by Southern blot analysis.

SVM-based Drone Sound Recognition using the Combination of HLA and WPT Techniques in Practical Noisy Environment

  • In recent years, the development of drone technologies has promoted the widespread commercial application of drones. However, the ability of drone to carry explosives and other destructive materials may bring serious threats to public safety. In order to reduce these threats from illegal drones, acoustic feature extraction and classification technologies are introduced for drone sound identification. In this paper, we introduce the acoustic feature vector extraction method of harmonic line association (HLA), and subband power feature extraction based on wavelet packet transform (WPT). We propose a feature vector extraction method based on combined HLA and WPT to extract more sophisticated characteristics of sound. Moreover, to identify drone sounds, support vector machine (SVM) classification with the optimized parameter by genetic algorithm (GA) is employed based on the extracted feature vector. Four drones' sounds and other kinds of sounds existing in outdoor environment are used to evaluate the performance of the proposed method. The experimental results show that with the proposed method, identification probability can achieve up to 100 % in trials, and robustness against noise is also significantly improved.

시스템 엔지니어링을 통한 HLA 기반 선박 안전성 평가 시뮬레이션 개발에 관한 연구 (Research into the Development of HLA-based Ship Safety Assessment Simulation with Systems Engineering)

  • Many accidents occur on the oceans. Accidents involving ships can cause enormous damage. When an accident occurs, the cost of environmental restoration can bankrupt even a top-ranked international company. The potential damages resulting from ship accidents have resulted in many international agreements such as MARPOL, SOLAS, ISM Code, etc. The viability of damaged ships has become more and more important. A damaged ship's viability and cargo safety can be improved by a safety assessment design procedure. We consider th£ shipbuilding industry's distributed characteristics to define a safety assessment system with software engineering-based requirements analysis. We used a distributed network-based simulation method (HLA - High level architecture), because of ease of reuse and expansion of existing components to other situations, and because it is a military standard. HLA is the standard for distributed network-based simulation in many countries including the United States, Canada, Israel, and Korea. The paper describes research to develop a prototype of a network-based safety assessment simulation system by software engineering based analysis.

Genetic factors associated with development of cerebral malaria and fibrotic schistosomiasis

  • Collaborative studies have identified some genetic factors contributing to the development of severe forms of malaria and schistosomiasis. In Thailand, the $TNF-{\alpha}{\;}5'-flanking$ region shows biallelic polymorphic sites at nucleotides -238, -308, -857, -863, and -1031, and seven alleles have been identified in patients from Myanmar. We found that the TNF promoter (TNFP)-D allele was significantly associated with cerebral malaria in populations from Karen (P < 0.0001. OR = 124.86) and ethnic Burma (P < 0.0001, OR = 34.50) . In China, we have identified two major genes related to the severity of liver fibrosis, one an HLA class II gene, and the other the IL-13 gene. The frequency of the HLA- DRB5*0101 allele and that of the IL-13 promoter A/A (IL- l3P- A/A) genotype were elevated in fibrotic patients, although the two genes are located on different chromosomes, chromosomes 6p and 5q, respectively Subjects with both genotypes had odds ratios (OR = 24.5) much higher than the sum of the ratios for each individual genotype (OR = 5.1,95% Confidence Interval 1.3-24.7 for HLA-DRB5*0101, OR = 3.1 95% CI 1.5 - 6.5 for IL-l3P-A/A). That the effects of the two susceptibility markers are synergistic rather than additive, strongly suggests that the pathogenic Th2 response directly influences the prognosis of post-schistosomal liver fibrosis.

  • 현대는 시뮬레이션을 통하여 많은 모의 훈련을 대체하고 있다. 시뮬레이터는 고유의 훈련 임무를 수행하기 위하여 기획, 제작 되어진다. 하지만 현대와 같은 종합적인 입체훈련 체계는 고유의 훈련 임무 이외에 개개의 시뮬레이터의 임무가 가상의 종합 시뮬레이션 환경에 참가한 통합 훈련 체계의 필요성을 야기하게 되었다. 또한 각각의 시뮬레이터의 기능은 보다 세분화 되었으며 높은 성능을 요구하게 되었다. 현재 KA-32 헬기 시뮬레이터 영상 프로그램을 제작하고 있는 한국소프트스페이스(주)는 독립적인 시뮬레이터들에 HLA(High Level Architecture)를 적용하여 하나의 종합 훈련 시스템을 구축하였다. 고유의 임무를 지닌 시뮬레이테는 각각의 임무를 가상의 종합 훈련 환경에 참가하여 서로간의 훈련 효과를 극대화 시켜화 시켜준다. HLA 기반의 종합 훈련 시스템의 RTI(Run-Time Infrastructure)는 Pitch 사의 pRTI를 사용하였으며, 가상의 종합 시뮬레이션 환경의 핵심이 되는FOM(Federation Object Model)은 호환성을 및 모든 객체의 표현을 위하여 RPR FOM(Real-time Reference FOM)을 사용하였다. 본 종합 훈련 시스템에서는 각각의 시뮬레이션간에 최고의 성능을 내기 위하여, 기능 세부화 하여 구성하였다. Terrion 사의 FLAMES는 종합 훈련 시스템의 시뮬레이션 진행/설정 및 분석을 담당하며, Mantis는 훈련 상황을 사실감 있는 3 차원 영상으로 참가자에게 전달하여 준다.

Role of natural killer cells for immunotherapy in chronic myeloid leukemia (Review)

  • The majority of natural killer (NK) cells serve an important role in eliminating malignant cells. The cytotoxic effects of NK cells were first identified against leukemia cells, and it is now hypothesized that they may have a critical role in leukemia therapy. The cellular functions of NK cells are mediated by their cell surface receptors, which recognize ligands on cancer cells. The role of NK cells is specifically regulated by the activating or inhibitory killer cell immunoglobulin-like receptors (KIRs) on their surface, which bind to the human leukocyte antigen (HLA) class I ligands present on the target cells. The association between KIR and HLA is derived from the diversity of KIR/HLA gene profiles present in different individuals, and this determines the cytotoxic effect of NK cells on cancer cells. Chronic myeloid leukemia (CML) is a hematological leukemia characterized by the hyper-proliferation of myeloid cells, with the majority of patients with CML presenting with abnormal immune cells. Tyrosine kinase inhibitors are the present standard therapy for CML, but are associated with numerous adverse side effects. Various studies have proposed CML therapy by immunotherapeutic approaches targeting the immune cells. This review summarizes the contents of NK cells and the association between KIR/HLA and leukemia, especially CML. This is followed by a discussion on the development of NK cell immunotherapy in hematological malignancies and research into strategies to enhance NK cell function for CML treatment.
