Transformation-Associated Recombination (TAR) cloning technique allows selective isolation of chromosomal regions and genes from complex genomes. The procedure requires knowledge of relatively small genomic sequences that reside adjacent to the chromosomal region of interest. This technique involves homologous recombination during yeast spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences. In this study, we examined the effect of non-homologous spacing sequence in target hooks on homologous recombination using a plasmid model system. The efficiency of homologous recombination between the modified his3-TRP1-his3 fragments and HlS3 gene on plasmid were analyzed by the characterization of $Ura^+$ transformants. The numbers of $Ura^+$ transformant showed same level when seven different modified his3-TRP1-his3 fragments were used. But the percentage of positive recombinants. $Trp^+His^-$, dramatically decreased when used the modified his3-TRP1-his3 fragments contained incorrect spacing of nonhomologous region. As a result, we suggest that incorrect spacing inhibits the homologous recombination between target hook and substrate DNA. Therefore, we should consider the correct spacing in target hook when the target hook are used for cloning of orthologue gene.

The entire nucleotide sequence of the HIS3 gene encoding imidazoleglycerol phosphate dehydratase (IGPD) in yeast Candida milleri CBS 8195 was determined. Sequence analysis revealed an open-reading frame of C. milleri HIS3 that spans 678 bp, encodes 226 amino acids, and shares 80.5% amino acid identity to Torulaspora delbrueckii IGPD, followed by that to Saccharomyces cerevisiae (79.6%). The cloned HIS3 gene complemented a his3 mutation in S. cerevisiae, suggesting that it encodes a functional IGPD in C. milleri CBS 8195. A new auxotrophic marker is now available for acid-tolerant yeast C. milleri.

The HIS5 gene of Saccharomyces cerevisiae host was encoded histidinol phosphate aminotransferase(E.C.: 2.6. 1.9). The HIS5 gene of Saccharomyces cerevisiae was cloned on plasmid pSH 530. This gene mighted be transcripted from a promoter of yeast gene both in E. coli and yeast hosts. We have determined the nucleotide sequence of the yeast HIS5 gene and its 5' and 3' flanking sequences. There are no large differences between the relative levels of HIS5 mRNA molecules with different 5' termini in represent and derepressed cell. In the DNA sequence upstream from the 5' termini of HIS5 mRNA we have found live closely related copies of a 9 base pair sequence. The sequence is also repeated in the 5' noncoding regions of HIS1, HIS3, HIS4, HIS5 and TRP5. Closely related sequence are not found flanking repeat sequence plays a role in the regulation of amino acid biosynthetic genes subject to the general amino acid control.

The purpose of this study is to define concept and characteristics of "Unphenomenon", the design concept conceived by Shiro Kuramata who is a world renowned Japanese interior designer and to explore how his ideas were materialized in his works. This study 1) examines his experience during his childhood reflected in his essay The scenery of the Unphenomenon' and training in Kuwazawa Design Institute that made Shiro Kuramata a true interior designer, and also considers how his design philosophy had been built up and realized under the unique social situation at the time. 2) defines the concept and characteristics of "Unphenomenon" as substance of his design philosophy by surveying remarks in his books and comments made by people around him in press interviews. 3) concludes his own design method and uniqueness by analyzing his works based on them. As a result, the study has founded that 1) transparency as recognitive realization of existence and non-existence: non-existence of physical property, connotation, ambiguity 2) expression of uncommonness by correcting and reinterpreting common prejudices: flotation, distortion of scale and shape 3) realization of insubstantiality by removing and replacing material and shape: replacement into light, removal of the structure, multiple layers of meaning.

- I aimed to isolate trans-acting factors involved in the basal expression level of eukaryotic genes. One of the yeast histidine biosynthetic gene, HIS5 was taken as a model for this study. HIS5 gene has a substantial basal level in amino acid rich medium and is derepressed if starved for any single amino acid. The derepression is mediated by cis-acting DNA sequences 5'-TGACTC-3' found in 5' non-transcribed region of the gene and trans-acting factors including GCN4 as positive factor and its negative factor GCDI 7, and GCNZ as a negative factor of GCD17. I first investigated the role of these trans-acting factors in HIS5 basal expression level by using HIS5-pH05 fusion in which expression of pH05 gene encoding inorganic phosphate-repressible acid phosphatase (APase) is regulated by HIS5 promoter. Strain with gcn2 or gcn4 mutation showed 3 to 4 fold lower APase activity than wild type. The level of APase activity was similar in gcn2 and gcn4 mutants. Trans-acting factors involved in basal level were identified by isolating 14 mutants showing increased expression of HISSPH05 fusion from gcn4 background. All the mutants carry a single nuclear recessive mutation and fall into four complementation groups, designated as bell (basal expression level), be12, be23 and be14.

This paper surveys the meaning of the pen name ′Yi Sang(李箱)′ in the light of the arbitariness of language and the relationship among names, observing in three ways how the images of ′Sang′ (Box) appear differently in his works. In Particular, we look into (1) the relationship between a box and its inner boxes, (2) the divisional relationship in the box, and (3) the relationship between the box and an outer box. In Yi Sang′s work, image (1) gives shape to the relationship between the writer himself and his ancestors who are responsible for his present existence. Image (2) gives shape to two internally divided Yi Sangs, and is symbolized as a mirror that reflects two selves. Image (3) gives shape to a relationship between himself in the closed world and other people and/or a bigger outside world. All these symbolic relationships are fully represented in his work "Nalgae" (The Wing)". What is interesting is that Yi Sang has already employed these box images in his earlier poetry "Geonchuk-muhan-yukmyeon-gaucho" (Infinite Architectural Hexagon). furthermore, by visualizing his name as a sqaure($\square$), he means to suggest the intentional image of his pen name as a ′box′.

This paper examines the crisis of the protagonist's Jewish identity in Philip Roth's Portnoy's Complaint. Jewish values are centered on the philosophy of Judaism and Jewish history. Judaism is based on an ethical monotheism which is Bible-centered. It is characterized by its covenant with God, its humanism, and its emphasis on moral action. It provides essential reasons for man's existence and stresses human confidence and sufficiency. Jewish values can be found in words such as "good," "humanity," "dignity," "responsibility," and "sense of community." These positive Jewish values pervade Philip Roth's fiction paradoxically. Throughout especially Portnoy's Complaint, the protagonist fails to embrace Jewish values in contrast to Bellow or Malamud's heroes and repeat the same mistakes eliciting fits of laughter from readers. The protagonist suffers from his strongly-felt ethical and altruistic impulses perpetually warring with his extreme sexual longings against which he struggles. His desperation grows as he finds himself unable to channel his dissatisfaction and change his situations. His dominating mother and his confusion over Jewishness and Americanism are the main obstacles to his establishment of self-identity. He attempts to build up his gender identity and Jewish identity through his ego-centric sexual relationship with shikses(female gentiles). His inability to embody Jewish values leads to the failure to fulfill his identity. Roth paradoxically shows that the protagonist's realization of Jewishness is essential to the cure for his fragmented self.

This study was conducted to determine phytotoxicity of domestic rice varieties and characteristic of sensitive gene involved in herbicide reaction of 4-HPPD (4-hydroxy phenylpyruvate dioxygenase) inhibiting herbicides. The five rice varieties were grown for 2 to 3 leaf stage on seedling trays and then transplanted into plastic pot: 4-Japonica type (Sangjubyeo, Sambaekbyeo, Sanduljinmi and Kumyoung) varieties and 1-Indica type variety (IR8). We trialled standard (14 g a.i. $10a^{-1}$) and double fold (28 g a.i. $10a^{-1}$) dose of benzobicyclon treatment at 10 days after transplanting in order to investigate phytoxicity. The Japonica-type Sangjubyeo showed no rice injury but Indica-type IR8 show 4-5 (standard) and 5-6 (double fold) phytoxicity levels. In spite of Japonica-type, Sanduljinmi and kumyoung showed 3-4 and 4-5 levels. Target resistant gene, Hydroxyphenylpyruvate dioxygenase inhibitor sensitive gene No.1 (HIS1) have been isolated from five domestic rice cultivars (Sangjubyeo, Sambaekbyeo, Sanduljinmi, Kumyoung and IR8). Results of the sequence through PCR, all five tested rice cultivars had HIS1 gene regardless to rice cultivars. And the difference between rice varieties from sequence of HIS1 were identified some variation in genes.

This study aims to examine the life of Chugang Nam Hyo-On (1454~1492) from his Chinese poem and to figure out how he overcame and healed his agony and wound. Nam Hyo-On was one of the Saengyuksin who submitted Sureungbokuiso and spent his whole life with alcohol and wandering. Thus, he is known to be a hermit writer or stranger. He had to suffer poverty during his life thanks to his only one choice. He was weak by nature but also feeble mentally as he always worried about death. In his 30s, he regarded himself as an aged man and always worried about death due to his frequent illness. Sometimes, he used to dream of being forever young. He composed self-elegy poem that is about his death, which is the ever first self-elegy poem in our history. He sent it in his letter to his master Kim Jong Jik (1431-1492) in 1489. His agony and motivation of writing such poem can be conjectured from his master's reply and his self-elegy poem. Nam Hyo-On tried to overcome and heal his real discontent and wound from his literary works. And it seems appropriate to regard him that he attempted to get compensation of his limitation and discontent from literature. We sometimes heal our wound by writings (or literature) and by reflecting through such writings. Nam Hyo-On reminded of his beloved person through his poet writing and healed his fear of wound of death.

The subject of 'Emotional Design' is appeared recently as one of the important issues in design field. 'Emotion', which has been underestimated since Kant and should be reevaluated from now on, is one of the basic two elements with reason which constitutes human nature. The purpose of this study is to research emotional design method appeared in the works of Steven Holl who is considered one of the influencing architects because of his unique methodology in developing the emotional design. The results of this study are as follows; 1) Emotional aspects were implied in his concepts by the way of emphasis on phenomenology in dealing with question of perception. 2) Emotional aspects were interwoven in his form and space as related to his concept. 3) Emotional aspects were shown in his way of developing his ideas with water coloring, in deriving his concept from other genre(especially from literature) and in the articulation of details. As a result, his unique way of implying emotional aspects which is coming from in-depth research of human nature as well as his own instinct were all interwoven in concept, form, and space together as a total way of thinking.
