히스톤 탈아세틸화 효소 억제제 trichostatin A가 C2C12 myoblast 세포 분화와 세포주기 조절인자의 발현에 미치는 영향 (Effects of Histone Deacetylase Inhibitor, Trichostatin A, on the Differentiation of C2C12 Myoblasts and the Expression of Cell Cycle Regulators)

  • 본 연구는 분화 전단계인 C2C12 myoblast세포에 중요한 후천적 기작의 하나인 DNA 히스톤 단백질의 아세틸화를 조절하였을 때 일어나는 변화를 살펴본 결과, 히스톤 탈아세틸화 효소를 trichostatin A로서 억제시키자 C2C12 myoblast 세포가 smooth muscle로 분화하였다. 이는 immunofluorescentstaining을 통해 smooth muscle ${\alpha}-actin$의 발현 증가를 trishostatin A로 처리한 세포에서 관찰하였으며, DAPI 염색을 통해 대조군 세포와 비교하여 세포의 증식이 많이 억제됨을 관찰하였다. 또한 real-time PCR 결과는 smooth muscle ${\alpha}$-actin과 transgelin mRNA의 발현이 trichostatin A 처리군 세포에서 현저히 증가함을 보여주었다. 이러한 결과를 바탕으로 히스톤 단백질의 탈아세틸화 억제는 C2C12 myoblast 세포의 분화에 매우 중요한 역할을 하며, 또한 C2C12 myoblast 세포를 골격근인 다핵의 myotube로 분화시키지 않고, smooth muscle로 분화시킴을 알 수 있었다. 이것은 분명히 HDAC억제제 인 trichostatin A가 DNA 히스톤 단백질의 HDAC 효소에 의한 탈아세틸화를 강력히 억제하고, 이러한 HDAC효소의 억제는 세포주기에 있어서 증식과 분화를 조절하는 유전자들의 발현을 조절하였음을 시사한다. 이를 검증하기 위해 세포주기 조절인자인 p21과 cyclin Dl mRNA의 발현을 조사한 결과 세포를 증식단계로 진행하는데 있어서 필수적인 cdk 억제제인 p21 mRNA의 발현이 trichostatin A로 처리한 세포에서 현저히 증가함을 보였으며, 세포 증식을 유도하는 cyclin Dl mRNA의 발현은 trichostatin A를 처 리 한 후 24시간 후 유의하게 감소함을 보였는데 이는 trichostatin A가 세포증식을 억제하는 초기단계에서 cyclin Dl 유전자의 발현을 조절함을 보여준다. 향후 연구에서는 또 하나의 중요한 후천적 기작인 DNA 메틸화와 히스톤 아세틸화가 유전자 발현을 조절하는데 있어서 상호작용에 대한 연구가 필요할 것으로 생각된다.

섬유소의 이용과 butyrate의 최근 연구 (Use of Cellulose and Recent Research into Butyrate)

  • 지구상에는 약 5,400여 종의 포유동물이 있고 그 중 약 1,000여 종은 풀을 뜯어 먹고 사는 초식동물이다. 초식동물 중에서 약 250여 종이 반추동물로 알려져 있다. 반추동물인 소와 양은 반추위에서 주로 발효가 일어나지만 비반추동물인 돼지와 사람은 맹장과 결장, 직장에서 주로 발효가 일어난다. 반추위 미생물의 종류와 우점도 Bacteroidetes 51%, Firmicutes 43% 존재하며, 사람의 대장미생물의 우점도Firmicutes 65%, Bacteroidetes 25%로 존재한다. 풀의 세포벽 구성성분은 미생물에 의해 분해, 발효에 의해 SCFA (short chain fatty acid)를 생성하게 되고 acetate, propionate, butyrate 생성비율은 60:25:15이다. 장내 primary butyrate transporter인 MCT1(monocarboxylatetransports-1)에 의해서 흡수된 butyrate는 SCFA receptor GPR43과 GPR41을 활성화시킨다. Butyrate는 강력한 anti-tumorigenic 기능을 가지고 있다. Butyrate는 다양한 cancer cell에 효과를 나타내며 세포내의 기능 조절에 기여하고, 암세포사멸을 유도하는 특성이 있다. Butyrate는 caspase의 활성화, HDAC (histone deacetylase) 활성을 억제하여apoptosis를 유도하고, p53 발현증가로 cell cycle arrest와 apoptosis를 유도한다. SCFA의 항 염증작용으로는 장 상피세포에서 IL-8 발현 감소, NO합성과 NF-${\kappa}B$ (nuclear factor ${\kappa}B$)의 활성을 억제하여 염증으로 인한 암 발생을 억제한다. Butyrate는 장 점막의 생리적 기능을 유지하는데 중요한 역할을 하며 IBD (inflammatory bowel disease) 치료법으로 이용되고 있다.

Effects of Macrolide and Corticosteroid in Neutrophilic Asthma Mouse Model

  • Background: Asthma is a disease of chronic airway inflammation with heterogeneous features. Neutrophilic asthma is corticosteroid-insensitive asthma related to absence or suppression of $T_H2$ process and increased $T_H1$ and/or $T_H17$ process. Macrolides are immunomodulatory drug that reduce airway inflammation, but their role in asthma is not fully known. The purpose of this study was to evaluate the role of macrolides in neutrophilic asthma and compare their effects with those of corticosteroids. Methods: C57BL/6 female mice were sensitized with ovalbumin (OVA) and lipopolysaccharides (LPS). Clarithromycin (CAM) and/or dexamethasone (DXM) were administered at days 14, 15, 21, 22, and 23. At day 24, the mice were sacrificed. Results: Airway resistance in the OVA+LPS exposed mice was elevated but was more attenuated after treatment with CAM+DXM compared with the monotherapy group (p<0.05 and p<0.01). In bronchoalveolar lavage fluid study, total cells and neutrophil counts in OVA+LPS mice were elevated but decreased after CAM+DXM treatment. In hematoxylin and eosin stain, the CAM+DXM-treated group showed less inflammation additively than the monotherapy group. There was less total protein, interleukin 17 (IL-17), interferon ${\gamma}$, and tumor necrosis factor ${\alpha}$ in the CAM+DXM group than in the monotherapy group (p<0.001, p<0.05, and p<0.001). More histone deacetylase 2 (HDAC2) activity was recovered in the DXM and CAM+DXM challenged groups than in the control group (p<0.05). Conclusion: Decreased IL-17 and recovered relative HDAC2 activity correlated with airway resistance and inflammation in a neutrophilic asthma mouse model. This result suggests macrolides as a potential corticosteroid-sparing agent in neutrophilic asthma.

  • 대표적인 histone deacetylase inhibitor 저해제의 일종일 sodium butyrate에 의한 인체백혈병 U937세포의 증식 억제에 관한 기전 연구를 세포주기 조절 측면에서 조사하였다. MTT assay 및 flow cytometry 분석을 통하여 sodium butyrate의 처리 농도 증가에 따른 U937 세포의 증식억제는 세포주기 G1 arrest 및 apoptosis 유발에 의한 것임을 확인하였다. RT-PCR및 Western blotting 결과에서 sodium butrate에 의한 G1 arrest는 세포주기 G1기에서 S기로의 진입에 중요한 역할을 하는 cyclin D1, E, A, cyclin-dependent kinase (Cdk) 4 및 Cdk6발현의 저해와 p21 및 p27과 같은 Cdk inhibitor의 발현 증가와 연관성이 있었다. Sodium butyrate는 또한 retinoblastoma protein (pRB)및 p130 단백질의 인산화를 저해시켰으나, S기 진행에 중요한 전사조절인자인 E2F-1 및 E2F-4의 의 발현에는 큰 영향이 없었다. 그러나 sodium butyrate에 의한 pRB 및 p130단백질의 인산화 저해는 pRB와 E2F-1및 p130과 E2F-4와의 결합력을 증사시켰다. 본 연구의 결과는 U937세포의 증식억제에 pRB/p130 인산화 억제 및 Cdk inhibitors의 발현 증가가 중요한 역할을 하고있음을 보여주는 것으로, sodium butyrate의 항암기전 이해에 중요한 자료가 될 것이다.

Differential expression of tescalcin by modification of promoter methylation controls cell survival in gastric cancer cells

  • The EF-hand calcium binding protein tescalcin (TESC) is highly expressed in various human and mouse cancer tissues and is therefore considered a potential oncogene. However, the underlying mechanism that governs TESC expression remains unclear. Emerging evidence suggests that TESC expression is under epigenetic regulation. In the present study, the relationship between the epigenetic modification and gene expression of TESC in gastric cancer was investigated. To evaluate the relationship between the methylation and expression of TESC in gastric cancer, the methylation status of CpG sites in the TESC promoter was analyzed using microarray with the Illumina Human Methylation27 BeadChip (HumanMethylation27_270596_v.1.2), gene profiles from the NCBI Dataset that revealed demethylated status were acquired, and real-time methylation-specific PCR (MSP) in gastric cancer cells was conducted. In the present study, it was demonstrated that the hypermethylation of TESC led to the downregulation of TESC mRNA/protein expression. In addition, 5-aza-2c-deoxycytidine (5'-aza-dC) restored TESC expression in the tested gastric cancer cells except for SNU-620 cells. ChIP assay further revealed that the methylation of the TESC promoter was associated with methyl-CpG binding domain protein (MBD)1, histone deacetylase (HDAC)2, and Oct-1 and that treatment with 5'-aza-dC facilitated the dissociation of MBD1, HDAC2, and Oct-1 from the promoter of TESC. Moreover, silencing of TESC increased MBD1 expression and decreased the H3K4me2/3 level, thereby causing transcriptional repression and suppression of cell survival in NCI-N87 cells; conversely, overexpression of TESC downregulated MBD1 expression and upregulated the H3K4me2 level associated with active transcription in SNU-638 cells. These results indicated that the differential expression of TESC via the modification status of the promoter and histone methylation controled cell survival in gastric cancer cells. Overall, the present study provided a novel therapeutic strategy for gastric cancer.

PLGA-Loaded Gold-Nanoparticles Precipitated with Quercetin Downregulate HDAC-Akt Activities Controlling Proliferation and Activate p53-ROS Crosstalk to Induce Apoptosis in Hepatocarcinoma Cells

  • Controlled release of medications remains the most convenient way to deliver drugs. In this study, we precipitated gold nanoparticles with quercetin. We loaded gold-quercetin into poly(DL-lactide-co-glycolide) nanoparticles (NQ) and tested the biological activity of NQ on HepG2 hepatocarcinoma cells to acquire the sustained release property. We determined by circular dichroism spectroscopy that NQ effectively caused conformational changes in DNA and modulated different proteins related to epigenetic modifications and c ell cycle control. The mitochondrial membrane potential (MMP), reactive oxygen species (ROS), cell cycle, apoptosis, DNA damage, and caspase 3 activity were analyzed by flow cytometry, and the expression profiles of different anti- and pro-apoptotic as well as epigenetic signals were studied by immunoblotting. A cytotoxicity assay indicated that NQ preferentially killed cancer cells, compared to normal cells. NQ interacted with HepG2 cell DNA and reduced histone deacetylases to control cell proliferation and arrest the cell cycle at the sub-G stage. Activities of cell cycle-related proteins, such as $p21^{WAF}$, cdk1, and pAkt, were modulated. NQ induced apoptosis in HepG2 cells by activating p53-ROS crosstalk and induces epigenetic modifications leading to inhibited proliferation and cell cycle arrest.

Investigation of candidated genes for molecular characterization of DongGyeong dog populations (Gyeongju)

  • This study was carried out to investigate the characters of short-tailed dogs (DongGyeong dogs) with anatomical insights, molecular genetics in Gyeongju. The present study was conducted to further characterize of short-tailed dog population in Gyeongju. The short-tailed dog was analyzed in the distribution of 55 individual. The anatomical insights were by x-ray. For discovery of specific genes expressions were measured by Hot-start PCR analysis. Anatomy survey, the number of vertebral typical consists of more than 20. 88.9% of short-tailed dog populations consists of 3-8 vertebrates. The 54 individuals of the 47 observe the vestigial tailed of the sacrum. No detected sacrococcygeal vertebrae degradation individuals were malformation defects. The 3 genes were DEGs (differentially expressed genes) in Dong-Gyeong dogs. We succeeded in finding 3 novel DongGyeong dogs specific genes by using Hot-start PCR analysis, this study suggests that these novel genes may play role (s) in DongGyeong dogs.

  • Cytochrome P-450 3A4 (CYP3A4) is major enzyme in human liver, the role of this is detoxification and metabolizing more than 50% clinical drugs in use. Expression of CYP3A4 is transciptionally regulated by the Pregnenolone X receptor (PXR), of which human form is Steroid and Xenobiotics receptor (SXR). SXR is activated by wide range of endogenous and exogenous compounds, and then induces CYP3A4 gene expression. In the previous study, it has been known that proximal promoter (-864 to +64) does not response to chemical inducers such as pregnenolone 16a-carbonitrile (PCN), Rifampicin, Estrogen in terms of transcription of CYP 3A4 in cultured cells. Here, we developed luciferase reporter gene assay system to detect SXR-based CYP 3A4 transcriptional activity. We have used CYP3A4-Luc plasmid that contains proximal promoter of human CYP3A4 gene upstream of the luciferase gene. We did transient transfection of 3A4-luciferase gene and SXR. In the HepG2 cells transfected with CYP3A4-Luc, when rifampicin treatment was combined with histone deacetylase inhibitor (HDAC Inhibitor), such as Trichostatin A, Hc-toxin and IN 2001 of the luciferase activity was induced 10-20 fold over control.

봉독이 IgE가 매개하는 제1형 과민반응 동물모델에 미치는 항알레르기 효과 (Anti-allergic Effect sof Bee Venom on IgE-mediated Type I hypersensitivity Response in vivo)

  • Bee venom (BV), well known as a traditional Oriental medicine, has been widely used in the treatment of some immune-related diseases. However, the anti-allergic effect of BV have not been reported. In this study, we investigated the antiallergic effect of BV on triphasic cutaneous reaction (TpCR) and passive cutaneous anaphylaxis (PCA). Our results indicated that BV suppress ear swelling and vascular permeability on IgE mediated type I hypersensitivity response. Increase in ear thickness was significantly inhibited by BV in this model. BV also blocked the infiltration of immune cells into the ear. Moreover, BV suppressed expression of HDAC3, Tryptase, MCP-1 in ear tissue. These results demonstrated that BV has a suppressive effect on allergic reaction.
