Supplementation of Cassava Hay to Replace Concentrate Use in Lactating Holstein Friesian Crossbreds

  • Exp. I, the study was conducted to examine the supplementation levels of cassava hay (CH) in dairy cows. Six multiparous Holstein-Friesian crossbreds were paired and randomly assigned in a change-over design to receive three levels of CH supplement at 0, 0.8 and 1.7 kg DM/hd/d. Concentrate was supplemented at the same level (1:2; concentrate:milk yield) while urea-treated (5%) rice straw was offered on ad libitum basis. The results revealed that supplementation of CH could significantly reduce concentrate use resulting in similar milk yield (12.5, 12.12 and 12.6 kg/hd/d) and significantly enhanced 3.5% FCM (14.21, 15.70, 14.9 kg/d, respectively). Moreover, CH supplementation significantly increased milk fat and milk protein percentages especially at 1.70 kg/hd/d. Concentrate use could be significantly reduced by 27% at 1.7 kg/hd/d CH supplementation. Exp. II, supplementation of cassava hay to replace concentrate use was studied in lactating-Holstein Friesian crossbreds grazed on Ruzi grass. Six multiparous cows in mid-lactating periods were paired and randomly assiged according to a change-over-design to receive three dietary treatments, $T_1=0kg$ cassava hay (CH) in 1:2 concentrate supplementation (CS) to milk yield (MY), $T_2=1.0kg$ DM CH/hd/d in 1:3 CS to MY, $T_3=1.7kg$ DM CH/hd/d in 1:4 CS to MY, respectively. The results were found that milk yield were similar among treatments while protein, lactose and solids-not-fat percentages were highest (p<0.05) in cows receiving CH at 1.0 kgjhd/d. Most significant improvement from CH supplementation was the ability to reduce concentrate use by 42% which could provide a higher income for small-holder dairy farmers. In addition, milk thiocyanate was enhanced from 5.3 to 17.8 ppm (p<0.05) in the control and in the CH supplemented group (1.7 kg/hd/d), respectively. Moreover, CH supplementation could significantly reduce concentrate level for diary feeding thus resulted in more economical return. Cassava hay demonstrated as a potential and high-quality on-farm feed resource especially for dry season feeding in the tropics.

  • Purpose: Helicobacter (H) pylori infection has been considered the most important cause of gastritis, dyspepsia, and gastroduodenal ulcer. Radioiodine can be accumulated in the remaining thyroid tissue, salivary gland, and stomach. We investigated if the high radiation induced by radioiodine in the stomach after high dose radioiodine therapy (HD-RIT) is effective in the eradication of H. pylori infection. Materials and Methods: One hundred ninety nine patients (M:F=33:166, age $46.7{\pm}12.3$ years) who had HD-RIT (dose $159.1{\pm}25.9\;mCi$, range 120-250 mCi) after thyroidectomy due to well differentiated thyroid cancer were enrolled. To detect H. pylori infection, the urea breath tests (UBT) were performed at 1 hour before HD-RIT and at 4 weeks after HD-RIT. The results of UBT were classified as positive (${\geq}50\;dpm$) or negative (<50 dpm), and analyzed its values. Results: Of 199 patients, 103 (51.8%) patients had positive UBT before HD-RIT. Of these, 80 patients had follow-up UBT after HD-RIT. Among them, 76 (95.0%) patients had persistent positive UBT and only 4 (5.0%) patients were changed negative UBT. Among 76 patients with persistent positive UBT, 26 (34.2%) patients had increased the values of follow-up UBT, 49 (64.5%) had decreased them, and 1 (1.3%) had shown the same value. The different values of UBT between before and after HD-RIT were $62{\pm}66.1\;dpm$ in increased one of follow-up UBT, and $153.3{\pm}157.1\;dpm$ in decreased one of follow-up UBT. Conclusion: We conclude that the radiation induced by HD-RIT is ineffective in the eradication of H. pylori infection. However, it could be influential the degree or distribution of H. pylori infection.

  • UBV photoelectric observations of RS CVn type variable star HD 8358 were made using the 61cm reflector at Sobaeksan Astronomical Observatory. The data were obtained on 15 nights from October 1987 to December 1988. Double peaks of maximum light is seen from the light curve and continuous change of phase in notified from the times of maximum lights. The colors of October, 1987 - January, 1988 observations are bluer in $\Delta(b-\upsilon)$, but redder in $\Delta(\upsilon$-b), than those of November -December, 1988 observations.

Effect of Herb Distillate on Hepatic Xanthine Oxidase Activity and Serum Lipid Profiles in Carbon Tetrachloride-Administered Rats

  • In order to evaluate the hepatoprotective effect of an herb distillate, ie., a mixture of 28 traditional Korean herbs, germanium, tormarine and Gijangsoo (Gijang water), $CCl_4$ was intraperitoneally administered to rats before or after supplementation of the diluted herb distillate (HD) for 2 weeks. Then hepatic xanthine oxidase activity and serum lipid profiles were determined. The experimental groups had higher feed intake than the normal control (NC), but had lower weight gain. Water intake and the amount of feces were not significantly different, but urine was excreted in lower amounts in all the experimental groups compared to the NC. Liver weights in the HD-supplemented groups were lower than that of the distilled water-supplemented groups (DW-groups) after $CCl_4$-administration. Serum ALT activities in all the experimental groups were higher than that of the NC-group. However, the increasing activity of serum ALT in the HD-supplemented groups (HD-groups) was lower than that of the DW-groups. Total serum and LDL-cholesterol levels were higher in all the $CCl_4$-administered groups than in the NC-groups, and serum HDL-cholesterol levels were lower in all the experimental groups compared with the NC-groups. Meanwhile, the increasing rate of total serum and LDL-cholesterol levels and the decreasing rate of HDL-cholesterol in the HD-groups were lower than that of the DW-groups. But, levels of serum TG were similar among all the experimental groups. The activities of hepatic xanthine oxidase (XOD) type O of the $CCl_4$-administered rats showed a significant increase in and an increasing rate of XOD in the HD-groups, which was lower than that of the DW-groups. On the other hand, GST activities in all the experimental groups were significantly decreased, and the decreasing rate was lower in the HD-groups than in the DW-groups. The hepatic contents of GSH and LPO in all the rats were not changed by $CCl_4$ administration. These results suggest that the decreased liver damage in the HD-supplemented groups was due to the inhibition of XOD-type O activity by constituents of HD, as well as by a prevention/inhibition of serum lipid profile changes in $CCl_4$-treated rats. However, further detailed studies are needed to support this hypothesis.

Analysis of Peroxide Curing Agents in EVA Compounds and Vulcanizates

  • Poly(ethylene-co-vinyl acetate) compounds and vulcanizates containing dicumyl peroxide (DCP) (HD) were prepared, and the curing agent and reaction products were analyzed using gas chromatography/mass spectrometry (GC/MS). Samples containing trially cyanurate (TAC) and DCP (HDT) were also prepared and analyzed. Some raw DCP were decomposed in the injector region of GC to produce acetophenone. DCP was detected in the HD compound but was not observed in the HD vulcanizate, and instead acetophenone and 2-phenyl-2-propanol were detected. Both DCP and TAC were observed in the HDT compound but not in the HDT vulcanizate, where acetophenone and 2-phenyl-2-propanol were detected. Thus, some of the DCP decomposed during the compounding process. The formation mechanism for acetophenone and 2-phenyl-2-propanol during the crosslinking reaction was identified, and differences in the crosslinking reactions of HD and HDT compounds were discussed.

Comparison of Characteristics between Insecticidal and Noninsecticidal Basillus thuringiensis Strains belonging to Serotype H8a8b

  • A noninsecticidal strain, Bacillus thuringiensis NTB-88, isolated from Korean soil, had a typical bipyramidal parasporal inclusion and its serotype is identical to B. thuringiensis subspmorrisoni (H8a8b). To elucidate differences between insecticidal and noninsecticidal strains, we compared strain NTB-88 to other toxic B. thuringiensis subsp. morrisoni strains (HD-12 and PG-14). Restriction endonucleases digested plasmid DNA patterns showed that strain NTB-88 was different from lepidopteran-toxic strain, HD-12, but it was similar to dipteran-toxic strain, PG-14. The gene type of strain NTB-88 was different from those of other insecticidal strains, Furthermore, the NH2-terminal amino acid sequence of crystal protein of strain NTB-88 had no relation to those of the previously known $\delta$-endotoxins in other toxic strains as well as HD-12 and PG-14 strains. Therefore, the noninsecticidal crystal protein in strain NTB-88 is novel and its property is different from insecticidal ones.

Anti-Obesity Effects of Salted and Unsalted Doenjang Supplementation in C57BL/6J Mice Fed with High Fat Diet (식이유도 비만 Mice에서 된장 및 무염된장의 항비만 효과)

  • The aim of the present study is to elucidate the anti-obesity effects of Doenjang with and without salt in C57BL/6J mice. For the analysis, a total of forty mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HD), high-fat diet supplemented with 20% Doenjang group (DJ), high-fat diet supplemented with 20% unsalted Doenjang group (NS). During the study period, food intake and body weight were measured daily and weekly, respectively. The animals were sacrificed after 12 weeks. Body weight gain, epididymal fat pad weight and serum triglyceride levels of DJ group were found to be significantly lower than those of the HD and NS groups. Serum total-cholesterol levels of DJ and NS groups were significantly lower as compared to the HD group. There were significant decreases in plasma insulin and leptin levels in DJ group compared with the HD and NS groups. We did not observe any significant changes in the expression of hepatic lipogenic-related gene $PPAR{\gamma}$ among the HD, DJ and NS groups. However, ACC expression was found to be significantly decreased in DJ group. Lipolysis-related gene ($PPAR{\alpha}$ and CPT-1) expression was significantly higher in the DJ group as compared to HD and NS groups. In conclusion, the results of this study showed that Doenjang supplementation lowers body weight gain and improves obesity-related parameters.

Identification of a Potential Gene for Elevation ω-3 Concentration and its Efficiency for Improving ω-6/ω-3 Ratio in Soybean

  • This present study was to identify a novel candidate gene that contribute to the elevated α-linolenic acid (ALA, ω-3) concentration in PE2166 from mutagenesis of Pungsannamul. Major loci qALA5_1 and qALA5_2 were detected on chromosome 5 of soybean through quantitative trait loci mapping analyses of recombinant inbred lines. With next generation sequencing of parental lines and Pungsannamul, and recombinant analyses, a potential gene, Glyma. 05g221500 (HD) controlling elevated ALA concentration was identified. HD is a homeodomain-like transcriptional regulator that may regulate the expression level of microsomal ω-3 fatty acid desaturase (FAD3) genes responsible for the conversion of linoleic acid into ALA in the fatty acid biosynthetic pathway. In addition, we hypothesized that combination of mutant alleles, HD and either of microsomal delta-12 fatty acid desaturase 2-1 (FAD2-1\ could reduce the ω-6/ω-3 ratio. In populations where HD, and FAD2-1A and FAD2-1B genes were segregated, combination of a hd allele from PE2166 and either of the variant FAD2-1 alleles were sufficient to reduce the ω-6/ω-3 ratio in seeds.

Estrogen deprivation and excess energy supply accelerate 7,12-dimethylbenz(a)anthracene-induced mammary tumor growth in C3H/HeN mice

  • BACKGROUND/OBJECTIVES: Obesity is a risk factor of breast cancer in postmenopausal women. Estrogen deprivation has been suggested to cause alteration of lipid metabolism thereby creating a cellular microenvironment favoring tumor growth. The aim of this study is to investigate the effects of estrogen depletion in combination with excess energy supply on breast tumor development. MATERIALS/METHODS: Ovariectomized (OVX) or sham-operated C3H/HeN mice at 4 wks were provided with either a normal diet or a high-fat diet (HD) for 16 weeks. Breast tumors were induced by administration of 7,12-dimethylbenz(a)anthracene once a week for six consecutive weeks. RESULTS: Study results showed higher serum concentrations of free fatty acids and insulin in the OVX+HD group compared to other groups. The average tumor volume was significantly larger in OVX+HD animals than in other groups. Expressions of mammary tumor insulin receptor and mammalian target of rapamycin proteins as well as the ratio of pAKT/AKT were significantly increased, while pAMPK/AMPK was decreased in OVX+HD animals compared to the sham-operated groups. Higher relative expression of liver fatty acid synthase mRNA was observed in OVX+HD mice compared with other groups. CONCLUSIONS: These results suggest that excess energy supply affects the accelerated mammary tumor growth in estrogen deprived mice.
