Bacillus thuringiensis의 내독소(內毒素)와 플라스미드의 분리에 관한 연구 (Studies on the Isolation of δ-Endotoxin and Plasmids in Bacillus thuringiensis)

  • Bacillus thuringiensis subspecies중 HD-1, HD-9, HD-73과 finitimus의 내독소와 플라스미드분리에 대하여 얻은 실험결과는 다음과 같다. 1. 밀도구배에 의한 ${\delta}$-내독소 정제는 NaBr, CsCl, Renografin-76으로 행하여 밀도구배 재료간 독소 수율은 Renografin-76>CsCl>NaBr순으로 독소와 아포의 분리가 양호하였고 그 순도는 약 98%정도였다. 2. 정제한 내독소는 SDS-PAGE, 현미경 관찰, 생물검정으로 조사하였고, SDS-PAGE에 의한 하위단위체의 분자량은 약 66,000과 130,000달돈 이었다. 3. 내독소의 결정단백질의 외부형태는 finitimus를 제외한 3균주는 모두 이중피라마드형이었다. 4. 담배나방 유충에 대한 살충실험 결과 $0.1{\mu}g/ml$의 내독소 농도에서 HD-1은 60%, HD-73은 100% 치사율을 보였다. 5. B. thuringiensis의 플라스미드 크기는 약 0.5~120Kb 정도였고 그 수는 HD-1, HD-9, HD-73에서 각각 12, 3, 11개 였고, finitimus 균주에는 플라스미드가 없었다.

β-galactosidse 함유하는 재조합 대장균 세포를 이용한 1,2-hexanediol galactoside의 합성 (Enzymatic Synthesis of 1, 2-Hexanediol Galactoside by Whole Cells of β-Galactosidase-containing Recombinant Escherichia coli)

  • 최근 화장품에 쓰이는 방부제에 대한 피부 부작용이 보고되고 있다. 그 중에서 1,2-hexanediol (HD)는 방부기능과 보습기능을 가지고 있는 물질로 알려져 있는데, 본 연구에서는 보다 안전한 화장품용 소재를 개발하기 위하여 β-galactosidase (β-gal)을 함유하고 있는 대장균 세포를 이용한 transgalactosylation 반응을 통하여 1,2-hexanediol galactoside (HD-G)을 합성하는 연구를 수행하였다. 이 반응은 높은 농도의 lactose (300 g/l) 조건에서 24시간 동안 수행되었으며, 반응 시간이 12시간 지난 후부터 새로운 spot이 TLC 분석으로 확인되었다. 이 spot을 잠정적으로 HD-G로 추정한 후, 이 물질을 silica gel chromatography로 정제한 후, mass spectrometry 분석을 통하여, 이 물질이 HD-G의 sodium adduct ion ([M+Na]+, m/z=303.1423)임을 확인하였다. 추가적으로 정제된 HD-G를 시판되는 β-gal을 이용하여 가수분해하고, 유리된 단당류를 TLC로 분석하여 보았다. 이 때, galactose 한 분자가 HD-G로부터 유리 되는 것을 확인하였다. 결론적으로, 재조합 대장균 β-gal을 이용하여 HD로부터 HD-G이 합성되었음을 확인할 수 있었으며, 앞으로 추가연구를 통하여 항균력의 변화와 피부세포에 대한 세포독성 변화를 조사할 예정이며, 기능성에 변화가 없는 보다 안전한 화장품용 방부제가 개발되기를 기대하고 있다.

전북지역 혈액투석 환자와 지속성 외래 복막 투석 환자의 영양상태 비교 (Characteristics in Nutritional Status of Patients on Hemodialysis and Continuous Ambulatory Peritoneal Patients in Chonbuk Area)

  • The present study was designed to compare the nutritional status of hemodialysis (HD) and continuous ambulatory peritoneal dialysis (CAPD) patients. Subjects were 58 HD patients (male/female = 29/29) and 33 CAPD patients (male/female = 23/10) undergoing dialysis treatment in Artificial Kidney Unit of Chonbuk National University Hospital. For nutritional assessment, %IBW (ideal body weight), %TSF (tricep skin fold), %MAC (mid arm circumference), %MAMC (mid arm muscle circumference), serum albumin, serum transferrin, TLC (total lymphocyte count), SGA (subjective global assessment) and estimated energy and protein intakes by 1-month food frequency method were used. Between HD and CAPD group, mean age (50 $\pm$ 12 vs. 52 $\pm$ 12 yr), dialysis durations (37 $\pm$ 36 vs. 30 $\pm$ 26 mon), dietary energy intakes (28.3 $\pm$ 9.0 vs. 28.8 $\pm$ 8.6 kcal/kg/day), dietary protein intakes (1.1 $\pm$ 0.4 vs. 1.2 $\pm$ 0.3 g/kg/day) and incidence of co-morbid conditions (69.0% vs. 69.7%) were not significantly different. Data by using SGA showed a higher incidence of malnutrition in CAPD patients (45.6%) than in HD patients (36.2%). %IBW (p < 0.001), %TSF (p < 0.001) and %MAC (p < 0.001) were higher in CAPD patients than were in HD patients. But serum albumin (p < 0.001) and transferrin (p < 0.001) were significantly lower in CAPD patients than were in HD patients. A higher incidence of malnutrition was shown in CAPD Patients than in HD Patients due to different dialysis type. A significant finding was that CAPD showed protein deficient malnutrition and HD did calorie deficient malnutrition. It suggests that an adequate dietary intake considering dialysis type prevents a prevalence of malnutrition.

Cloning and Expression of the Bacillus thruingiensis var. kurstaki HD-1 Crystal Protein gene in Eschelichia coli

  • Bacillus thruingiensis var. kurstaki HD-1의 내독소 단백질 유전자의 발현기작을 규명하기 위하여 이 균으로부터 내독소 단백질 유전자가 존재하는 것으로 확인된 29Md와 44Md plasmid를 분리한 후, Sau3AI 제어효소로 부분절단하고, pBR322 BamHI site에 ligation하여, E. coli HB101 strain에 transformation시켜, 3,000여개의 Ampr/Ters한 colony를 얻어 면역학적 방법과 살충적 검정으로 통해 재조합 균주 KC1을 얻었다. PKC1 plasmid DNA는 vector DNA를 포함하여 약 12kb 정도의 크기를 가지며, B. t k HD-1 내독소 단백질과 이동도가 같거나 일치하는 132kd, 117kd의 KC1 specific band 2개를 얻었다. KC1 cell extract를 첨식한 결과 약 80% 치사율을 보였다.

대장균 베타-갈락토시데이즈를 이용하여 합성된 1, 2-Hexanediol Galactoside의 NMR Spectroscopy 및 Mass spectrometry (NMR Spectroscopy and Mass Spectrometry of 1, 2-Hexanediol Galactoside synthesized using Escherichia coli β-Galactosidase)

  • 화장품용 방부제로 사용하고 있는 1, 2-hexanediol (HD)에 높은 농도의 lactose (300 g/l)를 넣고, 재조합 대장균 ${\beta}$-galactosidase (${\beta}$-gal)를 이용하여 galactose 한 분자를 결합시키는 transgalactosylation 반응을 시켜서, 1, 2-hexanediol galactoside (HD-gal)을 합성하였다. 그리고, 합성된 HD-gal 분자를 확인하기 위하여, HD-gal에 대한 NMR ($^1H$- and $^{13}C$-) 스펙트럼 분석과 mass 스펙트럼 분석을 실시하였다. HD-gal의 $^1H$ NMR 스펙트럼에서 HD에 갈락토실화가 되었음을 보여주는 다양한 피크를 확인하였다. $^1H$ NMR 스펙트럼의 다운필드인 ${\delta}_H$ 4.44 ppm과 ${\delta}_H$ 3.96~3.58 ppm에서 나타나는 다양한 피크들은 HD에 갈락토실화가 되었다는 것을 잘 암시하고 있으며, 또한 $^1H$ NMR 스펙트럼의 업필드에서 나타나는 ${\delta}_H$ 1.60~1.35 ppm과 0.92 ppm의 피크는 HD의 $CH_2$$CH_3$ 작용기로부터 나타나는 피크로써 HD가 본 물질에 존재한다는 것을 나타내고 있다. $^{13}C$ NMR 스펙트럼에서는 HD-gal의 알파-아노머와 베타-아노머의 구조에서 기인하는 총 21의 카본피크가 나타났고, 각 아노머마다 12개의 카본이 존재하는데 이중 ${\delta}_C$ 68.6, 60.9 and 13.2 ppm에 보이는 3개의 카본은 겹쳐서 나타나 총 24개의 피크 중 21개가 나타났다. 또한, 질량스펙트럼 분석에서는 protonated HD-gal인 281.1601 (m/z)의 peak를 확인할 수 있었다. 이를 종합하면, NMR ($^1H$- and $^{13}C$-) 스펙트럼 분석 결과와 질량분석 결과들은 우리가 기대했던 HD-gal의 구조와 매우 잘 일치하고 있다는 것을 알 수 있었다. 앞으로 추가적으로, 세균에 대한 minimum inhibitory concentrations (MICs) 조사와 human skin cell에 대한 독성연구를 추가적으로 진행할 예정이며, 이러한 결과를 근거로 항균력을 유지하면서 피부세포에 대한 독성이 감소된 화장품용 방부제의 연구/개발이 계속되기를 기대하고 있다.

병원에서의 가습기살균제 노출 사례 연구: 4차 가습기살균제 피해 신청자를 중심으로 (Case Studies of Exposures to Humidifier Disinfectant in Hospitals: Focusing on the Exposure Assessment of the Fourth Round of Applicants)

  • Objective: This study aimed to introduce cases of exposure to humidifier disinfectant (HD) in hospitals and to present their exposure characteristics. Methods: We used data from 4,393 subjects who participated in the fourth assessment survey of environmental exposure to HD conducted by the Korea Environmental Industry & Technology Institute. In this study, we selected 301 subjects who reported their place of use of HD as a hospital. Then, we classified cases as 'Hospital-provided'. 'Probably hospital-provided', 'Individual purchased', and 'Unknown' according to the supply sources of HD. Also, we introduced detailed exposure characteristics for the selected cases. Results: Of the 4,393 subjects, 301 (6.9%) reported the use of HD in 392 hospitals (including duplicate answers for the use in ${\geq}2$ hospitals). The 301 hospital-user subjects included 139 survivors and 162 non-survivors. When we classified the 392 cases by supply sources, 'Hospital-provided' was 12.2% (48 cases), 'Probably hospital-provided' was 25.5% (100 cases), 'Individual purchased' was 59.7% (234 cases), and 'Unknown' was 2.6% (10 cases). Among the 'Hospital-provided' cases, we selected six cases and provided a detailed description of the HD use in this study. Additionally, we reported details for six cases that had purchased HD upon a doctor or nurse's recommendation and for three cases that had purchased it at hospital stores. Conclusion: This study presents various cases of HD exposure in hospitals. Because there may be a considerable burden of HD exposure in public spaces, including hospitals, further studies are necessary to assess HD exposure in hospitals and public places.

피라미드 구조를 이용한 계층적 hausdorff distance 정합 (Hierarchical hausdorff distance matching using pyramid structures)

  • This paper proposes a hierarchical Hausdorff distance (HD) matching algorithm baased on coarse-to-fine approach. It reduces the computational complexity greatly by using the pyramidal structures consisting of distance transform (DT) and edge pyramids. Also, inthe proposed hierarchical HD matching, a thresholding method is presented to find an optimal matching position with small error, in which the threshold values are determined by using the property between adjacent level of a DT map pyramid. By computer simulation, the performance of the conventional and proposed hierarchical HD matching algorithms is compared in therms of the matching position for binary images containing uniform noise.

말기신부전증 환자에서 혈액투석액 나트륨 농도가 혈압, 투석간 체중증가 및 갈증에 미치는 영향 (The Effect of Hypertonic Dialysate on Hemodynamic Parameters (blood pressure, pulse rate, ultrafiltration rate), Interdialytic Weight Gain and the Incidence of Thirst with Hemodialysis Patients)

  • Hemodialysis(HD)-associated hypotension is a frequent complication, but it is difficult to manage. Until now, several maneuvers have been tried to prevent the HD-associated hypotension. Of these, the sodium content of dialysate was regarded as an important factor for maintaining blood pressure during HD. In this study, we evaluated the effect of hypertonic dialysate on blood pressure, interdialytic weight gain and the incidence of thirst. The study was done for 6 weeks successively with 3 different groups. Each patient was dialysed with 3 different dialysates for 2 weeks: Group I(Conventional HD: sodium concentration: 137 mEq/L), Group II(Hypertonic HD: 147 mEq/L) and Group III (Sequential HD: from 147 to 140 mEq/L). Hemodynamic parameters(blood pressure, pulse rate and ultrafiltration rate), biochemical parameters(hematocrits, blood urea nitrogen, creatinine, osmolality, sodium, potassium, chloride, fasting blood sugar) and complications (interdialytic weight gain & thirsty sensation) were compared among 3 groups. The results were as follows: 1. Decline of systolic blood pressure and diastolic blood pressure at the time of a 3 hour check during hemodialysis was lower in the Group II than Group I and III (p=0.002; p=0.012). and decline of diastolic blood pressure at the time of a 4 hour check during hemodialysis was lower in the Group II and III than Group I (p=0.04). 2. Incidence of hypotensive episodes during dialysis was significantly lower in Group II than group I (p=0.0287). 3. The ultrafiltration in Group III at the time of 1 hour, 2 hour and 3 hour check during hemodialysis was higher than that in Group I and II at the time of 1 hour, 2 hour and 3 hour check during hemodialysis respectively (p=0.0001; p=0.0001; p=0.0004). 4. Interdialytic weight gain was higher in Group I($3.1{\pm}0.8$) than Group I($2.8{\pm}0.8$) and III ($2.9{\pm}0.9$) (p=0.0422). 5. Hematologic and biochemical results were not significantly different among 3 Groups. 6. Frequency of thirst was different in Group I, II and III, $0.05{\pm}0.12$, $0.41{\pm}0.24$and $0.22{\pm}0.29$ respectively (p=0.0259). The results suggest that hypertonic HD was effective in preventing HD-associated hypotension but interdialytic weight gain and thirst sensations were increased as compared with a conventional method. In this situation, sequential HD seems to be an alternative method to minimizes the side effect of hypertonic HD.

Genistein Combined with Exercise Improves Lipid Profiles and Leptin Levels in C57BL/6J Mice Fed a High Fat Diet

  • The aim of this study is to determine the anti-obesity effects of genistein and exercise, separately and in combination, in mice. Fifty male C57BL/6J mice were divided into 5 treatment groups: normal diet (ND), high fat diet (HD), high fat diet with exercise (HD+Ex), high fat diet with 0.2% genistein (HD+G), high fat diet with 0.2% genistein, and exercise (HD+G+Ex). They were allowed free access to feed and water, and exercised mice engaged in swimming on a regular basis for 12 weeks. Genistein supplemented mice gained less weight, had lower energy intake, better lipid profiles, and lower leptin than the HD mice. Furthermore, when genistein was combined with exercise (HD+G+Ex) the effects were even greater. HD, HD+Ex, and HD+G mice exhibited increased hepatic CPT-1 mRNA expression. Therefore, genistein and exercise has anti-obesity effects, as shown by changes in body weight, fat accumulation, energy intake, and leptin levels.
