Characterization and in vitro inhibition studies of protease inhibitor from the mushroom Pleurotus floridanus (PfTI) towards the pest Papilio demoleus is studied. The addition of 1 mM Mn2+, Na2+, Ba2+ and Ni 2+ enhanced the PfTI activity. The ICP-atomic emission spectrum showed the presence of Ca2+, Mg2+ and Zn2+ in the PfTI. Surfactants SDS and CTAB at a concentration of 1% reduced the PfTI activity whereas, the nonionic detergents Triton X and Tween 80 increased the activity. The inhibitory activity gradually decreased with increase in concentration of DMSO and H2O2. The activity was increased by dithiothreitol up to a concentration of 80 μM and inactivated at 140 μM. The activity of PMSF modified PfTI was drastically reduced to 0.234 U/mL at 4 mM concentration and similar results were obtained for modification of cysteine by N-Ethylmaleimide at slightly higher concentrations. The complex of trypsin and PfTI showed complete loss in fluorescence intensity at 343 nm compared with control. In vitro inhibition studies of PfTI with midgut proteases isolated from citrus pest P. demoleus with protease activity of 1.236 U was decreased to 0.613 U by 50 μL (0.1 mg/mL) of the inhibitor. Inhibitor was stable up to 0.04 M concentration of HCl.
Isotope-dilution inductively coupled plasma mass spectrometry was used to determine trace amounts of Cd, Cu, Pb, Ni and Zn in sediment. Sediment samples were dissolved by microwave digestion with addition of mixed acid ($HNO_3$, HF and $HClO_4$). Lead was determined after separation of alkaline and alkaline earth metals by an ammonium pyrrolidenedithiocarbarmate (APDC) solvent extraction. The other elements were determined after separation of iron, tin and titanium by hydroxide precipitation. Recovery efficiency of the analyte elements was not satisfactory, but most of matrix elements causing the isobaric interference could be effectively eliminated by the separation. Good agreement was achieved with the certified values in the analysis of the two sediment reference materials.
These studies were carried out to investigate the properties of H-Y antigen purified by immunoaffinity chromatography using monoclonal H-Y antibody. Immunoaffinity column was prepared by the coupling of monoclonal antibody to the Aminolink Coupling Gel. Murine testis supernatant was applied onto the column and eluted by O.lM glycine-HCl buffer and 31${\mu}g$ of H-Y Ag was eluted from one testis. Purified H-Y Ag strongly reacted with Con A and lentil from 6 different kinds of lectins tested, which may indicate that sugar moiety of H-Y Ag is composed of glucose, mannose and their derivatives. Con A-sepharose affinity column was used to purified H-Y Ag based on that H-Y Ag is glycoprotein. The fraction eluted by 0.2M Me-${\alpha}$-D-mannoside from the column loaded with murine testis supernatant was identified to be H-Y Ag by dot blot test. Molecular weight of the purified H-Y Ag was estimated by Sepharose G-75 gel filtration and SDS-PAGE, and showing that it was about 67,000 dalton. In fluorescence test, the ratio of XY embryos and XX embryos was 1:1.
In order to compare phosphorus (P) behavior of livestock manure compost (LMC) and chemical fertilizer (CF) applied to the sea-reclaimed land soils, incubation experiments were conducted for five weeks. Four soils differing textural classes, sandy loam and clay loam, and electric conductivity (EC) value, high and low, were applied with CF and LMC. LMC was applied at the level of 0, 1, 2, and 3% on the soil weight basis and CF was applied at the same levels of P as LMC. The results showed that increase of P application rate of CF and LMC led to linearly increase available $P_2O_5$ and $0.01M\;CaCl_2$ extractable P contents regardless of soil texture and EC. However, 0.01M $CaCl_2$ extractable P from soil applied with CF was significantly higher than with LMC. Correlation analysis between $0.01M-CaCl_2$ extractable P and fractionated P by different extraction methods showed that $0.01M-CaCl_2$ extractable P positively correlated with KCl-P (soluble and exchangeable P) and HCl-P (Ca and Mg bound P). However, NaOH-P (Fe and Al bound P and organic P) and residual P was adverse. The amount of NaOH-P significantly influenced to the amount of 0.01M $CaCl_2$ extractable P of CF and LMC in the soils. The application of LMC at sandy loam soil could be carried out in the consideration of nutrient leaching and crop uptake.
In the present study, at first, azo Schiff base ligand of (N,N'-bis(5-phenylazosalicylaldehyde)-ethylenediamine) ($H_2L$) has been synthesized by condensation reaction of 5-phenylazosalicylaldehyde and ethylenediamine in 2:1 molar ratio, respectively. Then, its cobalt complex (CoL) was synthesized by reaction of $Co(OAc)_2{\cdot}4H_2O$ with ligand ($H_2L$) in 1:1 molar ratio in ethanol solvent. This ligand and its cobalt complex containing azo functional groups were characterized using elemental analysis, $^1H$-NMR, UV-vis and IR spectroscopies. Subsequently, the interaction between native calf thymus deoxyribonucleic acid (ct-DNA) and CoL complex was investigated in 10 mM Tris/HCl buffer solution, pH = 7 using UV-vis absorption, thermal denaturation technique and viscosity measurements. From spectrophotometric titration experiments, the binding constant of CoL complex with ct-DNA was found to be $(2.4{\pm}0.2){\times}10^4M^{-1}$. The thermodynamic parameters were calculated by van't Hoff equation.The enthalpy and entropy changes were $5753.94{\pm}172.66kcal/mol$ and $43.93{\pm}1.18cal/mol{\cdot}K$ at $25^{\circ}C$, respectively. Thermal denaturation experiments represent the increasing of melting temperature of ct-DNA (about $0.93^{\circ}C$) due to binding of CoL complex. The results indicate that the process is entropy-driven and suggest that hydrophobic interactions are the main driving force for the complex formation.

As functional evaluation of mulberry fruits extracts, the protective effect on cerebral cell and antibacterial activities were carried. $1\%$ HCl-MeOH extract showed $37\%$ cytoprotective effect on hydrogen peroxide, also C3G identified mulberry fruits and cyanidin showed $52\%,\;76\%$, respectively, protective effects on oxygen-glucose deprivation (OGD). In the antibacterial activity of mulberry fruit extracts, MeOB-Cheongil extract showed the highest inhibitory activity. Salmonella typhimurium was shown inhibitory rate more than $70\%$ in all treatment groups. Also Klebsiella pneumoniae was shown inhibitory activity in all treatment groups.
In this study, Malachite Green (MG) dye removal from synthetic wastewaters by adsorption process using raw boron enrichment waste (BEW) and it's modifications (with acid and ultrasound) were aimed. 81% MG removal was obtained by BEW at optimum equilibrium conditions (time: 40 min., dosage: 500 mg/dm3, pH: 5-6, speed: 200 rpm, 298 K). MG removal from wastewaters using acid modified boron enrichment waste (HBEW) was determined as 82% at optimum conditions (time: 20 min., dosage: 200 mg/dm3, pH: 10, speed: 200 rpm, 298 K). For ultrasound modified BEW (UBEW), the highest MG removal percent was achieved as 84% at optimum conditions (time: 30 min, dosage: 375 mg/ dm3, pH: 8, speed: 200 rpm, 298 K). The equilibrium data of Malachite Green was evaluated for BEW, HBEW and UBEW adsorbents by using sorption isotherms such as Langmuir, Freundlich and Temkin models, out of which Langmuir model (R2 = 0.971, 0.987 and 0.984) gave better correlation and maximum adsorption capacity was found to be 147.05, 434.78 and 192.30 mg/g, respectively. The adsorption kinetics followed the pseudo-second-order kinetic equation for sorption of MG onto wastes. A look at thermodynamic data reveals that natural sorption is spontaneous and endothermic because of free negative energy exchange and positive change in enthalpy, respectively. The results indicated that boron enrichment waste, and HCl and ultrasound-modified boron enrichment waste served as good alternative adsorbents in dye removal from wastewater.
A series of four experiments was conducted to compare nutritional values of decorticated sunflower meals against soybean meal, in diets for pigs from weaning (Exp. 1 and 2) to finishing (Exp. 3 and 4). All experimental diets were prepared compensating for the energy content by using vegetable oil and the lysine content was matched using synthetic L-Lysine HCl. Twenty-one day old pigs were fed either corn-soybean meal based diet (CSBM) or corn- twice-decorticated sunflower meal based diet (CDSM) for four weeks (Exp. 1). There was no difference in performances between treatment groups. In Exp. 2, corn-non-decorticated sunflower meal based diet (CNSM) was added to the existing two treatments. Twenty-one day old pigs were fed three experimental diets for four weeks. Pigs fed CNSM had a lower weight gain and feed intake than other treatments (p<0.05). There was no difference between pigs fed CSBM and CDSM (Exp. 2). Growth performance of growing pigs was also greater (p<0.05) in pigs fed corn starch-twice- decorticated sunflower meal based diet (CSDSM) than pigs fed corn starch-non-decorticated sunflower meal based diet (CSNSM) during the eight week feeding trial (Exp. 3). There was no difference between pig fed corn starch-soybean meal based diet (CSSBM) and CSDSM (Exp. 3). In Exp. 4, growing pigs were fed three experimental diets (CSBM, CDSM, and barley-twice- decorticated sunflower meal based diet; BDSM) until the slaughter. There was no difference in growth performance of pigs during growing and finishing periods among treatments. However, pigs fed CSBM had a higher carcass dressing percentage (p<0.05) than pigs fed CDSM and BDSM. Pigs fed BDSM diet had a lower fat tissue percentage than other groups (p<0.05). The twice-decorticated sunflower meal can be used as a substitute for soybean meal in pig diets. The performances of piglets and growing-finishing pigs were not affected when soybean meal was replaced by twice-decorticated sunflower meal. This substitution needs the contribution of synthetic lysine and vegetable oil as sources of complementary nutrients to match the nutrient profile.

Chlorella is rich in chlorella growth factor (CGF). A review of the literature has described that CGF improves the capability of a Th1-based immunity, anticancer, antioxidant antibacterial activity, growth promotion, wound healing and so on, but has not studied the effect for the metabolism and the proliferation of human skin keratinocyte. The aim of this study was to examine the effect of metabolism and the proliferation of human skin keratinocyte in vitro. CGF was extracted with an autoclaving method which is a modified hot-water extraction method from dried chlorella and conformed by means of absorbance 0.22 at 260 nm. We have measured the extracellular acidification rate (ECAR) of the CGF by Cytosensor$^{\circledR}$ Microphysiometer and evaluated responsiveness depending upon the dosage on the HaCaT cell. The ECAR for the concentrations of 0.15, 1.5, 15, 150 $\mu\textrm{g}$/ml of CGF increased as a 103.6, 128.2, 149.0 and 423.9%, respectively compared to control (0.0 $\mu\textrm{g}$/ml, 100% ECAR). The ECAR for ErbBl tyrosine kinase inhibited by 4-anilinoquinazolines, $C_{16}$H$_{14}$BrN$_3$O$_2$.HCl on tile HaCaT cells with the amounts of 10 $\mu\textrm{g}$/ml of the CCF compared with 100 $\mu\textrm{g}$/ml of rhEGF. The conclusion of the study is that CGF might increase human epidermal keratinocyte proliferation through the interaction between the epidermal growth factor receptor and itself.
Purified melanin pigment from Klebsiella sp. GSK was characterized by thermogravimetric, differential thermal, X-ray diffraction and elemental analysis. This melanin pigment is structurally amorphous in nature. It is thermally stable up to $300^{\circ}C$ and emits a strong exothermic peak at $700^{\circ}C$. Its carbon, hydrogen and nitrogen composition is 47.9%, 6.9% and 12.0%, respectively. It was used to scavenge metal ions and free radicals. After immobilizing the pigment and using it to adsorb copper and lead ions, the metal ion adsorption capacity was evaluated by atomic absorption spectroscopy (AAS) and the identity of melanin functional groups involved in the binding of metal ions was determined by Fourier transform infrared (FT-IR) spectroscopy. Batch adsorption studies showed that 169 mg/g of copper and 280 mg/g of lead were adsorbed onto melanin-alginate beads. The metal ion adsorption capacity of the melanin-alginate beads was relatively significant compared to alginate beads. The metal ion desorption capacity of HCl was greater (81.5% and 99% for copper and lead, respectively) than that of EDTA (80% and 71% for copper and lead, respectively). The ability of the melanin pigment to scavenge free radicals was evaluated by inhibition of the oxidation of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and was shown to be about 74% and 98%, respectively, compared with standard antioxidants.
