The lysozymes encoded by bacteriophage T7 and K11 are both bifunctional enzymes sharing an extensive sequence homology (75%). The constructions of chimeric lysozymes were carried out by swapping the N-terminal and C-terminal domains between phage T7 and K11 lysozymes. This technique generated two chimeras, T7K11-lysozyme (N-terminal T7 domain and C-terminal K11 domain) and K11T7-lysozyme (N-terminal K11 domain and C-terminal T7 domain), which are both enzymatically active. The amidase activity of T7K11-lysozyme is comparable with the parental enzymes while K11T7-lysozyme exhibits an activity that is approximately 45% greater than the wild-type lysozymes. Moreover, these chimeric constructs have optimum pH of 7.2-7.4 similar to the parental lysozymes but exhibit greater thermal stabilities. On the other hand, the chimeras inhibit transcription comparable with the parental lysozymes depending on the source of their N-terminals. Taken together, our results indicated that domain swapping technique localizes the N-terminal region as the domain responsible for the transcription inhibition specificity of the wild type T7 and K11 lysozymes. Furthermore, we were able to develop a simple and rapid purification scheme in purifying both the wild-type and chimeric lysozymes.
This study aimed to determine dose-response (DR) curve of avian influenza (AI) virus to predict the probability of illness or adverse health effects that may result from exposure to a pathogenic microorganism in a quantitative microbial risk assessment. To determine the parametric DR relationship of several strains of AI virus, 7 feeding trial data sets challenging humans (5 sets) and chickens (2 sets) for strains of H3N2 (4 sets), H5N1 (2 sets) and H1N1 (1 set) from the published literatures. Except for one data set (study with intra-tracheal inoculation for data set no. 6), all were obtained from the studies with intranasal inoculation. The data were analyzed using three types of DR model as the basis of heterogeneity in infectivity of AI strains in humans and chickens: exponential, beta-binomial and beta-Poisson. We fitted to the data using maximum likelihood estimation to get the parameter estimates of each model. The alpha and beta values of the beta-Poisson DR model ranged 0.06-0.19 and 1.7-48.8, respectively for H3N2 strain. Corresponding values for H5N1 ranged 0.464-0.563 and 97.3-99.4, respectively. For H1N1 the parameter values were 0.103 and 12.7, respectively. Using the exponential model, r (infectivity parameter) ranged from $1.6{\times}10^{-8}$ to $1.2{\times}10^{-5}$ for H3N2 and from $7.5{\times}10^{-3}$ to $4.0{\times}10^{-2}$ for H5N1, while the value was $1.6{\times}10^{-8}$ for H1N1. The beta-Poisson DR model provided the best fit to five of 7 data sets tested, and the estimated parameter values in betabinomial model were very close to those of beta-Poisson. Our study indicated that beta-binomial or beta-Poisson model could be the choice for DR modeling of AI, even though DR relationship varied depending on the virus strains studied, as indicated in prior studies. Further DR modeling should be conducted to quantify the differences among AI virus strains.
New 7-Halo-4-hydroxy-2-allyl-N-3-(4-methoxy-2-carboxylic acid pyrrolidinyl)-2H-1,2-benzothiazine-3-carboxamide 1,1-dioxide derivatives were synthesized through the condensation of 7-halo-4-hydroxy-2-allyl-1,2-benzothiazine-3-carboxylic acid methyl ester 1,1-dioxide with 4-methoxy L-proline.
The objective of this research was to provide the characterization and method for producing anti-E. coli O157:H7 antibodies in egg-laying hens and to determine if the antibody can restrain the proliferation of E. coli O157:H7 in-vitro. Selected antigenic fractions (whole cell, outer membrane protein and lipopolysaccharide (LPS)) from E. coli O157:H7 were injected to hens in order to produce anti-E. coli O157:H7 antibodies. The immune response and the egg yolk antibodies of laying hens against the whole cell, outer membrane protein and LPS antigens were monitored by ELISA. The level of antibodies against whole cell antigen monitored through ELISA sharply increased after the initial immunization, and it was found to be maximum on day 49 however, the level was maintained up to day 70. Antibodies (5 mg/ml) directed against the whole cell inhibited E. coli proliferation 10-13 times more than outer membrane protein or LPS. The antibody response against the whole cell antigens appeared to have higher activity in restraining the proliferation of E. coli O157:H7 than antibody against outer membrane protein or LPS. Results reflected that increasing the IgY's in the egg yolk could prevent greater economic losses due to human and animal health from pathogenic bacteria i.e. E. coli O157:H7.
Ozonolysis reactions of cyclic olefins 1a-c and norbomene In in the presence of trifluoroactophenone 6 provided the corresponding cross-ozonides 7a-c and 7n. Further reactions of ozonides 7a-c and 7n with the independently prepared carbonyl oxide 11 gave diozonides of structure l0a-c and 10n. The ozonolysis of 1methylcyclopentene 12a and 1-methylcyclohexene 12b in the presence of trifluoroactophenone 6 provided exclusively ozonide 15 and 16 derived from capture of carbonyl oxide 13. All of the new ozonides have been isolated as pure substances and characterized by their 1H NMR and 13C NMR spectra.
A hydrogel, poly(N-isopropylacrylamide-co-N,N'-dimethylaminopropyl methacrylamide), sensitive to both pH and temperature was synthesized and characterized for its welling behavior, lower critical solution temperature (LCST), and its appearance. The hydrogel with 5 mol% of N,N'-diemthylaminopropyl methacrylamide (DMAPMAAm) increased its volume phenomenonally in a lower pH range (ph 1~8) even at temperature ($37^{\circ}C$ and $40^{\circ}C$) higher than LCST. This behavior was unique compared to the temperature -sensitive hydrogel which did not exhibit any swelling in the same pH range. The hydrogel with 20 mol% of DMAPMAAm was swollen significantly at a higher pH of 12. With pH decrease from 12 to 2 water content in the gel increased from 38.8 wt% and 60.6 wt%, and 90.8 wt% for 5 mol% and 20 mol% gel, respectively. The transition pH that pH effect overwhelmed temperature effect occurred at a lower pH for a higher temperature ($40^{\circ}C$) and a lower mol% (5 mol%) of DMAPMAAm. Transparency and LCST of the gel increased with higher DMAPMAAm mol%.
This study was carried out to investigate the quality of ground water for hydroponics in Kyeongnam area in 1995. Water samples were collected and analyzed from 77 wells in green houses throughout Kyeongnam area. The values of several components in well water were as follows ; 7.4 in pH, 0.46dS/m in EC, 0.3mg/L in N $H_{4}$-N, 25.4mg/L in $Ca^{2+}$, 42.6mg/L in C $l^{[-10]}$ and 72.5mg/L in S $O_{4}$$^{2-}$. The pH value showed high positive significance of correlationships with $Ca^{2+}$ and EC. Also, the EC value showed high positive significance with N $a^{+}$, $Ca^{2+}$, S $O_{4}$$^{2-}$, $Mg^{2+}$ and C $l^{[-10]}$ .
The crystal structure of sodium sulfisoxazole hexahydrate, $C_{11}H_{12}N_3O_3SNa{\cdot}6H_2O$,has been determined by X-ray diffraction method. The compound crystallizes in the monoclinic space group $$P2_1}c$$ with a = 15.68(3), b = 7.70(2), c = 17.94(4)${\AA}$, ${\beta}$ = $118(2)^{\circ}$ and Z = 4. A total of 1717 observed reflections were collected by the Weissenberg method with $CuK{\alpha}$ radiation. Structure was solved by heavy atom method and refined by block-diagonal least-squares methods to the R value of 0.14. The conformational angle formed by the S-C(l) bond with that of N(2)-C(7), when the projection in taken along the S-N(2), is $73^{\circ}.$ The benzene ring is planar and makes an angle of $60^{\circ}$ with the plane of the isoxazole ring, which is also planar. The sodium atom has a distorted octahedral coordination of N(l) and five oxygen atoms from hydrate molecules. Sodium sulfisoxazole hexahydrate shows fourteen different hydrogen bondings in the crystal. These are six $O-H{\cdots}O-H bonds, three $O-H{\cdots}O$ bonds, two $O-N{\cdots}N,$ one $N-H{\cdots}O,O-H{\cdots}N,N-H{\cdots}O-H$ bond, with the distances in the range of 2.71 to $3.04{\AA}.$.
Photosynthetic bacteria (PSB) attract considerable interest as useful microorganisms; nevertheless, a generalized culture technique has not been previously reported owing to difficulty in their cultivation. Therefore, a simple culture technique suitable for public use was investigated. Among the PSB tested, the strain Rhodobacter azotoformans EBN-7 was the most suitable for scale-up production because it showed the highest specific growth rate (0.20 h-1) on basal medium. In scale-up cultivation (500 L), R. azotoformans EBN-7 showed 4.50 × 1010 colony-forming units mL-1 (number of viable cells), dry cell weight of 26.8 g/L, and a specific growth rate of 0.15 h-1. Cultivation using this final culture broth (as seed culture) in a 15 L simple reactor was successful, with maintenance of cell activity evident. For use as seed culture, the maximum allowable preservation period of R. azotoformans EBN-7 at 4℃ was 3 months. When R. azotoformans EBN-7 cultivated in a simple technique was applied to shrimp aquaculture water, NH4+-N was reduced from 0.61 mg/L to 0.24 mg/L (by 60.7%) in 4 days in comparison with the control. Thus, this simple culture technique using R. azotoformans EBN-7 has the potential for a good removal efficiency of NH4+-N, making seed culture easier and suitable for public use.
