  • GTA 용접에서 용입, 용접부의 크기와 형상이 형성되는 아크 물리학적 현상을 이해함에 있어서 에너지 분포특성은 매우 중요한 인자이다. GTA 용접에서 아크 길이의 변화와 사용된 실드 가스 종류에 따라 음극인 텅스텐 전극 팁의 아크 루트 직경에 큰 변화를 주며, 양극인 모재 쪽의 에너지 분포에 영향을 미치며 된다. 기존의 연구자들은 저전류나 중전류 영역의 GTA 용접에서 텅스텐 전극의 선단각과 용입 형태와 의 관계를 plasma 기류 등에 의해 고찰하거나, 최대 아크압력에 미치는 전극형상의 영향에 대하여 연구하였다. 용접부에 작용하는 아크 압력의 분포는 결국 운동 에너지의 분포이다. 기존의 연구자들에 비하여 보다 간편한 실험을 통하여 양극 모재위의 아크 압력에 의한 에너지 분포특성을 규명하는 연구가 필요하다. 본 연구의 목적은 GTA 용접의 용접전류 100A~200A 영역에서 아크 길이 변화와 Ar 가스에 $H_2$ 혼합에 따른 측정된 아크 압력으로부터 기존연구자들의 아크 물리학적 결과들을 활용하여 양극 모재 위에 작용하는 전류밀도 분포를 유도하는 것이다. GTA 용접에서 아크 길이의 변화와 Ar 가스에 $H_2$ 혼합은 아크 압력분포에 큰 영향을 미치며, 이에 따라 에너지 분포특성에 많은 영향을 미친다. 아크 길이가 증가함에 따라 Ar가스와 $H_2$ 혼합가스의 에너지 분포는 감소하였고, Ar가스에 $H_2$ 혼합에 의해 아크 에너지가 증가하여 용입형상에 큰 영향을 미칠 것으로 판단된다. 이에 대한 연구는 향후 GTA 용접 응용분야 확대 적용될 것이며, 아크 물리학 연구에 기초적이고 아주 중요한 과학적인 자료가 될 것으로 판단된다.

Roles of Src-family kinase isoforms, Lyn, Fyn, Fgr, and c-Src on degranulation in RBL-2H3 mast cells (비만세포의 탈과립에 대한 다양한 Src-family kinase의 역할)

  • The rat RBL-2H3 mast cells contain various Src-family kinases. Previous reports with this cell line indicated that Lyn activation is an important initial signaling for the activation of the cells. However, the role and location of other Src-family kinase isoforms which are expressed in the cells are not clear. In this study, we now show that isoforms of Src-family kinases, Lyn, fyn, Fgr, c-Src, and Yes are differentially expressed and located differently in the cells as indicated by RT-PCR, immunoblotting analysis, and confocal microscopy. Lyn and Fgr were located on plasma membrane but on the other hand c-Src and Yes were located on intracellular organelle. All of Src-family kinases were cloned and overexpressed for investigating the roles of the isoforms. Overexpression of Fyn and Fgr, not Lyn and c-Src, stimulated Ag-induced degranulation in the cells. Our findings strongly suggest for the first time that each of Src-family kinase isoform can regulate differentially $Fc{\varepsilon}RI$-mediated signaling in RBL-2H3 mast cells.

The influence of different durations of aerobic exercise on fuel utilization, lactate level and antioxidant defense system in trained rats

  • This study investigated the influence of different durations of aerobic exercise on fuel utilization, lactate levels and antioxidative status in trained rats. Forty rats underwent physical training (T, n = 20) or non- training (NT, n = 20) for 6 weeks. For physical training, animals exercised on a treadmill for 30 min 5 days per week. At the end of week 6, the animals in each group were subdivided into BE, DE-0.5, DE-1 and DE-2, which were sacrificed at the end of week 6 without having performed exercise or after exercise on a treadmill for 0.5h, 1h and 2h, respectively, immediately before being sacrificed. The plasma glucose level in DE-2 of the NT group was significantly lower than in the other groups. Muscle and liver glycogen levels were significantly lower in DE-1 and DE-2, but there were no significant differences between DE-1 and DE-2 in the T group. Liver protein in DE-2 of the NT group was significantly lower. Muscle TG levels were decreased in DE-0.5 of the T group, while those of the NT group were decreased in DE-1. FFA levels were increased in DE-0.5 of the T group and in DE-1 of the NT group. Lactate levels were increased in DE-0.5 of the NT group, while they were increased in DE-1 of the T group. Catalase activity of the T group was lower in BE but higher in DE-0.5, DE-1 and DE-2. SOD activities were higher in trained rats, while the GSH/GSSG ratios were higher in BE, DE-0.5 and DE-1 in the T group, and there was no difference in that of DE-2. There were no differences in MDA levels in BE and DE-0.5, but they were significantly lower in DE-1 and DE-2 of the T group. Overall, the results of this study, suggest that training may improve exercise performance by facilitating the mobilization and oxidation of fat and conserving limited carbohydrate storage, and that it may delay the onset of fatigue and enhance the antioxidative defense system, but cannot support two hours of vigorous exercise.

  • Background: Tumor necrosis factor(TNF)-$\alpha$ and Interleukin(lL)-$1{\beta}$ are thought to play a major role in the pathogenesis of the septic syndrome, which is frequently associated with adult respiratory distress syndrome(ARDS). In spite of many reports for the role of TNF-$\alpha$ in the pathogenesis of ARDS, including human studies, it has been reported that TNF-$\alpha$ is not sensitive and specific marker for impending ARDS. But there is a possibility that the results were affected by the diversity of pathogenetic mechanisms leading to the ARDS because of various underlying disorders of the study group in the previous reports. The purpose of the present study was to evaluate the roles of TNF-$\alpha$ and IL-$1{\beta}$ as a predictable marker for development of ARDS in the patients with septic syndrome, in which the pathogenesis is believed to be mainly cytokine-mediated. Methods: Thirty-six patients of the septic syndrome hospitalized in the intensive care units of the Asan Medical Center were studied. Sixteens suffered from ARDS, whereas the remaining 20 were at the risk of developing ARDS(acute hypoxemic respiratory failure, AHRF). In all patients venous blood samples were collected in heparin-coated tubes at the time of enrollment, at 24 and 72 h thereafter. TNF-$\alpha$ and IL-$1{\beta}$ was measured by an enzyme-linked immunosorbent assay (ELISA). All data are expressed as median with interquartile range. Results: 1) Plama TNF-$\alpha$ levels: Plasma TNF-$\beta$ levels were less than 10pg/mL, which is lowest detection value of the kit used in this study within the range of the $mean{\pm}2SD$, in all of the normal controls, 8 of 16 subjects of ARDS and in 8 in 20 subjects of AHRF. Plasma TNF-$\alpha$ levels from patients with ARDS were 10.26pg/mL(median; <10-16.99pg/mL, interquartile range) and not different from those of patients at AHRF(10.82, <10-20.38pg/mL). There was also no significant difference between pre-ARDS(<10, <10-15.32pg/mL) and ARDS(<10, <10-10.22pg/mL). TNF-$\alpha$ levels were significantly greater in the patients with shock than the patients without shock(12.53pg/mL vs. <10pg/mL) (p<0.01). There was no statistical significance between survivors(<10, <10-12.92pg/mL) and nonsurvivors(11.80, <10-20.8pg/mL) (P=0.28) in the plasma TNF-$\alpha$ levels. 2) Plasma IL-$1{\beta}$ levels: Plasma IL-$1{\beta}$ levels were less than 0.3ng/mL, which is the lowest detection value of the kit used in this study, in one of each patients group. There was no significant difference in IL-$1{\beta}$ levels of the ARDS(2.22, 1.37-8.01ng/mL) and of the AHRF(2.13, 0.83-5.29ng/mL). There was also no significant difference between pre-ARDS(2.53, <0.3-8.34ngfmL) and ARDS(5.35, 0.66-11.51ng/mL), and between patients with septic shock and patients without shock (2.51, 1.28-8.34 vs 1.46, 0.15-2.13ng/mL). Plasma IL-$1{\beta}$ levels were significantly different between survivors(1.37, 0.4-2.36ng/mL) and nonsurvivors(2.84, 1.46-8.34ng/mL). Conclusion: Plasma TNF-$\alpha$ and IL-$1{\beta}$ level are not a predictable marker for development of ARDS. But TNF-$\alpha$ is a marker for shock in septic syndrome. These result could not exclude a possibility of pathophysiologic roles of TNF-$\alpha$ and IL-$1{\beta}$ in acute lung injury because these cytokine could be locally produced and exert its effects within the lungs.

  • Effects of oral administration of electrolyte solutions were studied in experimentally dehydrated adult sheep. By the latin square method five ruminal fistulated sheep were examined and dehydrated by deprivation of feed and water for 72 hours. Tap water, physiological saline, 0.45% NaCl+120 mM/L glucose and 0.9% NaCl+1% propylene glycol solution were orally administrated after dehydration, respectively. Rehydration effect and modification of the rumen function were compared. 1. After 72 hours of deprivation of feed and water, sheep were hypertonic dehydrated and blood acid-base parameters were not significantly changed. And there was marked increase in ruminal pH and decrease in ruminal total volatile fatty acid(VFA) concentration. 2. After the fluids administration the changes in blood acid-base parameters were not significant in all groups. 3. Although glucose fermentation in the rumen was observed, 0.45% NaCl+120 mM/L glucose was more effective in rehydration than physiological saline and tap water. But it was difficult to know the rehydration effect of 0.9% NaCl+1% propylene glycol solution exactly because of excessive increase in plasma osmolality. 4. After refeeding, total concentration and proportions of ruminal volatile fatty acid(VFA) were not significantly different among groups and recovered to normal concentration but not in proportions after 2 days in all groups. 5. In vitro cultured ruminal protozoa were susceptible to the decrease of the pH and osmolality.

  • 평판형 고체산화물 연료전지 스택의 고온 밀봉 구조에 대하여 설명하고 스택 운전 후 사후 분석을 통하여 밀봉재와 금속 분리판의 계면반응에 대하여 고찰하였다. 대표적인 고온 밀봉재인 Barium-Silicate 계 결정화 유리와 Fe-Cr 계 금속 분리판은 스택의 작동온도인 $700{\sim}850^{\circ}C$ 에서 고온 반응을 통하여 계면에 반응생성물을 형성하는 것이 확인되었다. 이러한 계면반응은 장기 운전시 SOFC 스택 성능 저하의 원인이 되고, 열 싸이클(작동온도${\leftrightarrow}$상온)을 가하면 계면반응 생성물이 delamination 되어 밀봉구조가 파괴되어 수명을 단축시키게 된다. 계면반응은 Fe-Cr 계 금속 분리판의 산화물인 Cr 산화물, Fe 산화물이 밀봉유리 소재와 반응을 일으키는 것이 주요 원인으로 판명되었다. SOFC 스택에서 열 싸이클시 계면반응에 의하여 기밀도가 감소하는 현상이 확인되었으며, 밀봉 구조의 어느 부분에서 계면반응이 진행되는지 관찰하였다. 이러한 계면반응을 막기 위해서는 금속 분리판과 밀봉유리 사이에 계면반응을 억제하는 보호층을 형성하는 방법이 효과적이다. 본 연구에서는 보호층으로서 밀봉유리 및 Fe-Cr 계 금속 분리판과의 계면반응성이 낮고 열팽창 계수가 비슷한 Yttria Stabilized Zirconia 층을 APS(Atmospheric Plasma Spray) 공정을 이용하여 형성하였다. 밀봉유리/YSZ 보호층/금속분리판은 gas-tight 한 밀봉 구조를 형성하였으며, YSZ 보호층은 밀봉유리와 Fe-Cr 계 금속 분리판 소재와 계면반응을 효과적으로 억제하는 것이 확인되었다.

  • 반도체 소자의 크기가 45 nm 이하로 감소함에 따라 최소 선폭에 따른 다층 배선 연결 구조가 요구되고 있다. 그러나 고집적화 구조는 기생 저항과 정전 용량에 의한 신호지연증가 및 혼선 전력 소모의 문제가 발생한다. 이런 문제를 해결하기 위한 방법 중의 하나는 저저항 배선연결물질과 층간 절연막으로 저유전 상수를 갖는 물질을 사용하는 것이다. 본 연구는 DEMS $(H-Si(CH_3)(OC_2H_5)_2)$ 전구체를 이용하여 저유전막을 증착할 때 사용되는 PECVD (plasma enhanced chemical vapor deposition) 장비를 국내 기술로 개발하고 개발된 장비로 저유전박막을 평가한 것에 관한 것이다. 본 연구에서 평가 및 박막 종확 시 사용한 장비는 MAHA hp 1 type ((주)아토)로서 양산용 PECVD 장비이다. 변수는 C-He의 유랑, 300 mm Si 웨이퍼와 shower head 사이의 거리, 증착 압력, 구동 전력이고, 증착된 저유전막의 두께, 두께의 균일성, 굴절률, 굴절률의 균일성를 평가하였다. 구동 전력이 500W 일 때, C-He의 유량과 진공의 크기를 감소시키면 박막의 두께가 감소하고 박막의 균일성은 증가하였다. C-He의 유량을 증가시키고 shower head 와 Si 웨이퍼 사이의 거리 및 구동 압력을 감소시키면 굴절률과 굴절률의 균일성이 모두 저하되었다. 구동 전력이 700W 일 때, 박막 두께의 경우, 구동 전력이 500W 일 때의 결과와 유사하지만, 박막의 균일성, 굴절률, 굴절률의 균일성은 모든 조건에서 저하되었다.

  • Using a RF cylindrical magnetron operated with two electromagnets having a Helmholz configuration, RF magnetron plasma properties and characteristics of reactive ion ething of Si were investigated as a function of applied magnetic field strengths using 3mTorr $CF_4/H_2$ and $CHF_3$. Also, I-V characteristics of Schottky diodes, which were made of silicons etched under different applied magnetic field strengths and gas environments, were measured to investigate the degree of radiation damage during the reactive ion etching. As the magnetic field strent;th increased, ion densities and radical densities of the plasmas were increased linearly, however, the dc self-bias voltages induced on the powered electrode, where the specimen are located, were decreased exponentially. Maximum etch rates, which were 5 times faster than that etched without applied magnetic filed, were obtained using near lOOGauss, and, under these conditions, little or no radiation damages on the etched silicons were found.

Production of Lentinus edodes Mycelia in Submerged Culture and It's Hypoglycemic Effect in Diabetic Rats (표고버섯 균사체의 생산과 당뇨 유발 흰쥐에 대한 혈당강하 효과)

  • The optimum conditions for the production of Lentinus edodes mycelia and it's hypoglycemic effect was studied. Optimum pH and temperature for the production of mycelia in shaken flask culture were 5 and $30^{\circ}C$, respectively, for 24 days cultivation. Culture period for maximum production of mycelia (8.13 g/l) in 5-l jar fermenter cultivation was shortened as much as 6 days compared to shaken flask culture. The mycelial dose of 5 % proved almost equally effective in lowering the plasma glucose, total cholesterol, and triglyceride level as much as 23.0, 20.7, and 27.1%, respectively. The values of alanine transaminase and aspartate transaminase were also estimated and exhibited a substantial reduction (29.6 and 34.3%, respectively) in activity after the administration of dried mycelia.

Effect of Nifedipine on the Ampicillin Absorption (니페디핀이 암피실린의 흡수에 미치는 영향)

  • $Amino-{\beta}-lactam$ antibiotics are absorbed by the dipeptide transporter in the small intestine. These uptakes are coupled to a proton influx. The inward proton gradient is partly induced by the $Na^+/H^+$ exchanger and calcium ion is involved in control of this antiport. Interaction between ampicillin which is one of the $Amino-{\beta}-lactam$ antibiotics and nifedipine which is one of calcium channel blocking agents was studied in rats in vivo and with rabbit jejunum mounted on the Sweetana/Grass diffusion cells in vitro. Bioavailability of ampicillin was increased significantly when nifedipine was co-administered orally in rats. There were no differences in the distribution phase and the elimination phase when ampicillin was given either alone or with nifedipine intravenously. Conditions for in vitro experiments were determined. The lift rate of $O_2/CO_2$ gas was controlled to 3 bubbles/sec and ampicillin was stable in the Kreb's buffer at pH 6.0. Absorption of ampicillin was the greatest when the completely-stripped serosal membrane was used. Transport of ampicillin from mucosal to serosal side in the rabbit jejunum was enhanced by 32% in the presence of nifedipine (p=0.059). Above results suggest that nifedipine might increase the plasma level of ampicillin via the improved absorption in the intestine rather than the reduction in the elimination or/and alteration in the distribution.
