In vitro Callus and Somatic Embryo Induction of Six Hosta Species Native to Korea

  • Hosta is a genus of the family Asparagaceae and distributed in East Asia. There are six Hosta species (Hosta capitata (Koidz.) Nakai, H. clausa Nakai, H. jonesii M.G.Chung, H. minor (Baker) Nakai, H. venusta F.Maek., and H. yingeri S.B.Jones) native to Korea and among them, four species (H. minor, H. jonesii, H. venusta and H. yingeri) are endemic to the Korea peninsula. Hosta is generally propagated by seed, crown division or tissue culture. However, tissue culture is a more efficient method to mass proliferation, a new cultivar development and disease-free plantlet production in a limit time. Hence, we conducted this study to evaluate the influence of various plant growth regulators (PGRs) treatments on the induction of callus and somatic embryo of the six Hosta species. Leaf, petiole and root were used to select optimum tissue culture explants. Petiole explants thus only were used for callus induction and somatic embryogenesis with TDZ (0.1, 0.5 or 1.0mg/L) and NAA (0.1 or 0.5 mg/L) combinations. After 12 weeks of culture, the highest rate of somatic embryogenesis was achieved on modificated MS medium containing 1.0 mg/L TDZ and 0.1 mg/L NAA in H. capitata and H. minor (15.5%, respectively), 0.1 or 0.5 mg/L TDZ and 0.1 mg/L NAA in H. jonesii (22.2%), 1.0 mg/L TDZ and 0.5 mg/L NAA in H. yingeri (26.7%), and 0.1 mg/L TDZ and 0.5 mg/L NAA in H. venusta (53.3%). H. clausa showed very low effect on somatic embryogenesis by PGRs; 2.2%. There was interspecies difference to PGRs respond for callus and somatic embryo induction. Regenerated multiple shoots and plantlet of H. minor, H. jonesii, H. venusta and H. yingeri were obtained via somatic embryogenesis.

Chemiluminescence System with Air Pump as a Sensor for Determination of Metal Levels in Rain

  • A lab-made chemiluminescence system with air pump was developed for monitoring of some metal levels in rain. The air pump enabled injection of 17.7 $\mu$g samples into a glass cell filled with luminol-$H_2O_2$ reagent of typically 300 $\mu$L for chemiluminescence measurement. The monitored trend of total metal ions in the rain collected in our campus was compared with analytical results of each metal ion from GFAAS. The system was also demonstrated to determine $Cr^{6+}$ by reduction to $Cr^{3+}$ using $SnCl_2$. The limit of detection for $Cr^{6+}$ obtained by 4 measurements was 85.0 pg $mL^{-1}$ with a relative standard deviation of 3.4%. Although this system doesn’t have selectivity due to the characteristics of chemiluminescence, application of it to environmental monitoring as a sensor for some transition metal ions was demonstrated.

Let A be a bialgebra over $k$. Suppose $A^{{\rightarrow}{\pi}}_{{\leftarrow}i}D$ are bialgebra maps satisfying ${\pi}{\circ}i=I_D$. Set ${\Pi}=I_D*(i{\circ}s_D{\circ}{\pi}),B=\Pi(A)$ and $j:B{\rightarrow}A$ be the inclusion. Suppose that ${\Pi}$ is an algebra map. We show that (D,B) is an admissible pair and $B^{\leftarrow{\Pi}}_{\rightarrow{j}}A^{\rightarrow{\pi}}_{\leftarrow{i}}D$ is an admissible mapping system and that the generalized biproduct bialgebra $B{\times}^L_HD$ is isomorphic to A as bialgebras.

Voltammetric Determination of Bisphenol A Using a Carbon Paste Electrode Based on the Enhancement Effect of Cetyltrimethylammonium Bromide (CTAB)

  • The influence of cetyltrimethylammonium bromide (CTAB) on the electrochemical behavior of bisphenol A at the carbon paste electrode (CPE) was investigated. CTAB, with a hydrophobic C-H chain, can adsorb at the CPE surface via hydrophobic interaction and then change the electrode/solution interface, and finally affects the electrochemical response of bisphenol A, confirming from the remarkable oxidation peak current enhancement. The electrode process of bisphenol A was examined, and then all the experimental parameters which affects the electrochemical response of bisphenol A, such as pH value of the supporting electrolyte, accumulation potential and time, potential scan rate and the concentration of CTAB, were examined. Finally, a sensitive and simple voltammetric method was developed for the determination of bisphenol A. Under the optimum conditions, the oxidation peak current of bisphenol A varied linearly with its concentration over the range from $2.5\;{\times}\;10^{-8}\;to\;1\;{\times}\;10^{-6}$ mol/L, and the detection limit was found to be $7.5\;{\times}\;10^{-9}$ mol/L. This method was successfully employed to determine bisphenol A in some waste plastic samples.

  • The use of both an Alpha Hydroxy Acid, Citric Acid, and a Skin Moisturizer, Glycereth-26, formulated into a hand dishwashing detergent have been shown to be synergistic in their effects on certain skin health parameters. Each ingredient was evaluated alone and together in a hand dishwashing detergent via a 9-week use test. Panelists washed dishes using the sponge method commonly used in Asian markets and a 1:7 dilution of the concentrated dish detergent. Panelist's hands were evaluated initially and at 3-week intervals for nine weeks. After nine weeks panelist's hands showed significant improvements in Moisture Absorption and Transepidermal Water Loss (TEWL). A synergistic effect on TEWL was found between the AHA and the Moisturizer. These effects, showing an improvement in the condition of the panelist's skin are impressive, particularly since they were observed from a dishwashing product that is highly diluted and at near neutral pH during the washing process.

Structural and Chemical Characterization of Aquatic Humic Substances in Conventional Water Treatment Processes (재래식 정수처리 공정에서 수질계 휴믹물질의 구조 및 화학적 특성분석)

  • Humic substances(HS) from raw and process waters at a conventional water treatment plant were isolated and extracted by physicochemical fractionation methods to investigate their characteristics. They are characterized for their functionality, chemical composition, and spectroscopic characteristics using FT-IR(Fourier transform infrared) and $^1H-NMR$(proton nuclear magnetic resonance) spectroscopy. Humic fraction gradually decreased from 47.2% to 26.4%(from 0.97 to 0.54 mgC/L) through conventional water treatment processes. Concentration of phenolic groups in the HS fraction gradually decreased from 60.5% to 21.8%(from 12.2 to $6.0\;{\mu}M/L$ as phenolic-OH) through water treatment. In the case of carboxylic groups, the concentration increased from 39.5% to 46.9%(from 7.9 to $10.6\;{\mu}M/L$ as COOH) by pre-chlorination, but gradually decreased to 34.2%($9.4\;{\mu}M/L$ as COOH) through sedimentation and sand filtration. From the results of the FT-IR and $^1H-NMR$ spectra of HS, the content of carboxylic groups increased and ratio of aliphatic protons to aromatic protons($P_{Al}/P_{Ar}$) also increased through water treatment, which indicated the increase of aliphatic compounds.

Reduction of Highly Concentrated Perchlorate in Aqueous Medium by Newly Isolated Bacterial Strains Rhodococcus sp. YSPW01 and YSPW02 (신균주 Rhodococcus sp. YSPW01과 YSPW02를 이용한 수중 내 고농도 Perchlorate 환원 특성 연구)

  • The feasibility of perchlorate reducing-bacteria isolated from the sludge of an anaerobic digester was determined using ammonium perchlorate in aqueous medium. Growth kinetics of the two perchlorate reducing bacteria including Rhodococcus sp. YSPW01 and YSPW02 were investigated using acetate as the electron donor in batch experiment. The growth of YSPW01 and YSPW02 reached a steady-state at 26 and 9 h, respectively. The initial perchlorate concentration was completely reduced within 8 and 7 h by YSPW01 and YSPW02, respectively. The reduction rates were 2.1 and $15mg\;L^{-1}h^{-1}$ for YSPW01, and 3.2 and $15.5mg\;L^{-1}h^{-1}$ for YSPW02, at 1:1 and 5:1 ratios of acetate:perchlorate (w:w), respectively. In this study, the bacteria Rhodococcus sp. YSPW01 and YSPW02 demonstrated a potential for the perchlorate reduction, which could be further investigated for development of an efficient strategy to treat the perchlorate contaminated waters.

Preparation of Onion Hydrolysate for Usage of Sauce (조미액으로의 활용을 위한 양파 가수분해물 제조)

  • To develop an onion sauce, reaction conditions of celluclast 1.5L and pectinex on onion were investigated and organoleptic evaluations were carried out. Degree of hydrolysis(D.H) of hydrolysate by a mixture of celluclast 1.5L and pectinex was a higher than that by each enzyme. Hydrolysate by a mixture of celluclast 1.5L and pectinex(1:3v/v) showed 86% of D.H. and total sugar content of the hydrolysate was 54mg/ml. Hydrolysates showed 83~86% of D.H. at reaction temperature of $25^{\circ}C$ to 45$^{\circ}C$. Total sugar content of the hydrolysate was increased with increasing temperature. D.H. and total sugar content of hydrolysate was 76~86% and 51~63mg/ml, respectively, under acidic conditions. D.H. and total sugar content of hydrolysate were also increased with increasing time. Bitterness, sweetness and ordor of roasted pork prepared by adding onion and onion hydrolysate were significantly different(p<0.05), but color and preference between two groups were not significantly different(p<0.05) between two groups. There was no significant difference(p<0.05) in sweetness and bitterness of the roasted pork prepared by adding different amounts of onion hydrolysate, although ordor and preference of the roasted pork were significantly different(p<0.05).

Environmental Characteristics of Natural Habitat of Protothaca jedoensis in Gamak Bay, Korea (가막만 살조개, Protothaca jedoensis 서식지 환경특성)

  • In this study, environmental parameters of natural habitat were measured to find out suitable place for release and improve the productivity of Protothaca jedoensis in Gamak Bay. Water temperature, salinity of the habitat ranged from 5.4 to 27.6 and from 26.5 to $34.5\%_{\circ}$, respectively. Range of pH, DO, COD, Chl-a, T-N and T-P were $7.82{\sim}8.39,\;5.31{\sim}11.28mg/L,\;0.13{\sim}1.38mg/L,\;3.05{\sim}11.55mg/L,\;0.005{\sim}0.180mg/L\;and\;0.007{\sim}0.028mg/L$ respectively. Dominant grain size was fine silt and the IL of sediment was ranged from $0.75{\sim}7.26%$. The water content was $20.16{\sim}53.65%$ and highest value was observed in Baegyado. The COD and AVS in the sediments ranged from $0.53{\sim}8.67mg/g-dry$ and $0.002{\sim}0.113mg/g-dry$ respectively. The bottom condition of Baegyado was higher than other area.
