Microbial Amelioration of Acid Mine Drainage Impaired Soil using the Bacterial Consortia of Klebsiella sp. and Raoultella sp.

  • Acid mine drainage (AMD) resulting from pyrite oxidation in mining areas, subsequently leads to soil acidification accompanied by lowering pH and high concentration of metals and metalloids in its surrounding environment. Regarding to this, the microbial amelioration has been considered as a promising option for a more cost-effective and eco-friendlier countermeasure, compared to the use of alkaline chemicals. This study was aimed to evaluate influencing factors in microbially-mediated amelioration of acidic soil spiked by simulated AMD. For this, microcosm experiments were conducted by acid-neutralizing bacterial consortium (dominated by Klebsiella sp. and Raoultella sp.) under the various conditions of AMD spikes (0-2,500 mg SO42-/L), together with acidic mine soil (0-100 g) or sphagnum peat (0-5 g) in the 200 mL of nutrient medium. The employed bacterial consortium, capable of resisting to high level of sulfate concentration (up to 1,500 mg SO42-/L) in low pH, generated the ammonium while concomitantly reduced the sulfate, subsequently contributing to the effective soil stabilization with an evolution of soil pH up to neutral. Furthermore, it demonstrates that suitable condition has to be tuned for successful microbial metabolism to facilitate with neutralization during practical application.

Effects of L-tryptophan, Fructan, and Casein on Reducing Ammonia, Hydrogen Sulfide, and Skatole in Fermented Swine Manure

  • The effects of daily dietary Bacillus subtilis (Bs), and adding L-tryptophan, fructan, or casein to fecal fermentation broths were investigated as means to reduce the production of noxious gas during manure fermentation caused by ammonia, hydrogen sulfide ($H_2S$), and 3-methylindole (skatole). Eighty swine ($50.0{\pm}0.5kg$) were equally apportioned to an experimental group given Bs in daily feed, or a control group without Bs. After 6 weeks, fresh manure was collected from both groups for fermentation studies using a $3{\times}3$ orthogonal array, in which tryptophan, casein, and fructan were added at various concentrations. After fermentation, the ammonia, $H_2S$, L-tryptophan, skatole, and microflora were measured. In both groups, L-tryptophan was the principle additive increasing skatole production, with significant correlation (r = 0.9992). L-tryptophan had no effect on the production of ammonia, $H_2S$, or skatole in animals fed Bs. In both groups, fructan was the principle additive that reduced $H_2S$ production (r = 0.9981). Fructan and Bs significantly interacted in $H_2S$ production (p = 0.014). Casein was the principle additive affecting the concentration of ammonia, only in the control group. Casein and Bs significantly interacted in ammonia production (p = 0.039). The predominant bacteria were Bacillus spp. CWBI B1434 (26%) in the control group, and Streptococcus alactolyticus AF201899 (36%) in the experimental group. In summary, daily dietary Bs reduced ammonia production during fecal fermentation. Lessening L-tryptophan and increasing fructan in the fermentation broth reduced skatole and $H_2S$.

Lowered Substrate pH Reduced the Bicarbonate Injury during Vegetative Growth of 'Ssanta' Strawberry (혼합상토의 pH 저하가 영양생장 중인 '싼타' 딸기의 중탄산 피해 경감에 미치는 영향)

  • Objective of this research was to investigate the influence of lowered substrate pH on the reduction of bicarbonate injury in the vegetative growth of 'Ssanta' strawberry. The acid substrate was formulated by mixing sphagnum peat moss with pine bark (5:5, v/v) and the pH, EC, and CEC of the substrate were 4.07, $0.46dS{\cdot}m^{-1}$, and $91.3cmol^+{\cdot}kg^{-1}$, respectively. To adjust the pH of acid substrate, various amount of dolomitic lime [$CaMg(CO_3)_2$] were incorporated with the rate of 0 (untreated), 1, 2, 3, and $4g{\cdot}L^{-1}$. Then, mother plants were transplanted and grown with fertilizer solution containing $240mg{\cdot}L^{-1}$ of the $HCO_3{^-}$ and equal concentrations of essential nutrients to Hoagland solution. In growth of 'Ssanta' strawberry, fresh weight of mother plants were the highest in the treatment of $2g{\cdot}L^{-1}$ dolomitic lime such as 102.1 g followed by 94.7 g in $1g{\cdot}L^{-1}$, 91.2 g in $3g{\cdot}L^{-1}$, 75.7 g in $0g{\cdot}L^{-1}$ and 72.3 g in $4g{\cdot}L^{-1}$ treatments. The dry weight showed a similar tendency to fresh weight. At 140 days after transplanting, 5.8, 9.8, 11.8, 8.8, and 5.0 daughter plants were derived from each of the mother plants in the treatments of 0, 1, 2, 3, and $4g{\cdot}L^{-1}$ dolomitic lime, respectively. The highest occurrence of daughter plants were observed in the treatments $2g{\cdot}L^{-1}$ dolomitic lime. The substrate pH and bicarbonate concentration of 'Ssanta' strawberry seedlings in the 1 and $2g{\cdot}L^{-1}$ dolomitic lime treatments were maintained at a proper range such as 5.6 to 6.2. The micro-nutrient contents of above ground tissue in mother plants were the highest in $2g{\cdot}L^{-1}$ and the lowest in $4g{\cdot}L^{-1}$ dolomitic lime treatment. The above results indicate that incorporation rate of dolomitic lime in acid substrate with the pH of around 4 is $2g{\cdot}L^{-1}$ to raise the 'Ssanta' strawberry in propagation.

Bioavailability and Comparative Pharmacokinetics of Two Enrofloxacin Formulations in Broiler Chickens (육계에서 경구투여시 enrofloxacin 제제에 따른 생체이용율 및 약물동태)

  • In order to compare the pharmacokinetic profiles of enrofloxacin-HCL)ENFLX-HCL) and enrofloxacin-KOH (ENFLX-KOH) after oral administration in broiler chickens, the study was performed. The chickens used in this study weighed $1.82 {\pm}0.2 kg$ and clinically healthy. The dose of intravenous and lral administration was 5 mg per kg of body weight as enrofloxacin. After intravenous injection of enrofloxacin, it showed two-compartment model with the rapid distribution phase and the slow elimination phase. The mean apparent volume of distribution (Vd) was 2.70 l/kg. The mean half-life of elimination and distribution showed 8.26 h and 0.44 h, respectively. The mean area under curve (AUC) was calculated as $19.7 {\mu} g{\cdot} h/ml$. After oral administration of ENFLX-HCL and ENFLX-KOH with a rate of dose 5 mg of enrofloxacin/kg of body weight, Both of the products were showed one-compartment model unlike that of i.v. enrofloxacin standard solution showed the mean bioavailability of 79.64% for the ENFLX-KOG and 86.24% for the ENFLX-HCL. The mean total body clearance of the former was 0.24 l/kg/h and the latter 0.42 l/kg/h. Both enorfloxacin formulations seemed to have good tissue distribution and penetration as indicated by large volume of distribution : 2.72 l/kg for the -KOH and 4.44 l/kg for the -HCL. With the results obtained in this study, ENFLX-HCL could be used in place of its salt form in chickens.

Cytotoxicity of Impregnated Dental Gingival Retraction Cord Extracts in Immortalized Human Oral Fibroblasts and Keratinocytes in vitro

  • This study evaluated cell viability and cytokine release in immortalized human oral fibroblasts (hTERT-hNOFs) and keratinocytes (IHOK) exposed to a dental-impregnated gingival retraction cord. To prepare the extracts, dental gingival retraction cords impregnated with aluminum chloride hexahydrate were immersed in a cell culture medium for 24 h at 37 ℃. hTERT-hNOFs and IHOK were cultured for 24 h. The cell culture medium was removed and extracts of the dental gingival retraction cords were added. After incubation with the extract solution, cell viability was evaluated using an MTT assay. The levels of the cytokines IL-1α and IL-8 were measured in the supernatants of each cell type. The cell viability after exposure to the extract solution for 10 min exceeded 70 % in both cell types. The ET50 values for hTERT-hNOF and IHOK were 35.75 and 28.98 min, respectively. For IHOK, the IL-1α level was (5.35 ± 5.22) pg/mL at 10 min, (3.58 ± 5.38) pg/mL at 20 min, and (2.85 ± 4.28) pg/mL at 60 min of exposure (p > 0.05). The IL-8 level in IHOK was (67.16 ± 18.70) pg/mL at 10 min, (78.36 ± 7.50) pg/mL at 20 min, and (111.9 ± 26.10) pg/mL at 60 min of exposure (p > 0.05). Cytokine release was not observed from hTERT-hNOFs. Based on these results, cell viability and cytokine release were confirmed in cells exposed to the impregnated gingival retraction cord. In addition, the application of the extracts to hTERT-hNOF and IHOK during the actual contact time and determination of ET50 may be beneficial for evaluating the biocompatibility of dental-impregnated gingival retraction cords.

Couette-Poiseuille flow based non-linear flow over a square cylinder near plane wall

  • A numerical study on the flow over a square cylinder in the vicinity of a wall is conducted for different Couette-Poiseuille-based non-uniform flow with the non-dimensional pressure gradient P varying from 0 to 5. The non-dimensional gap ratio L (=$H^{\ast}/a^{\ast}$) is changed from 0.1 to 2, where $H^{\ast}$ is gap height between the cylinder and wall, and $a^{\ast}$ is the cylinder width. The governing equations are solved numerically through finite volume method based on SIMPLE algorithm on a staggered grid system. Both P and L have a substantial influence on the flow structure, time-mean drag coefficient ${\bar{C}}_D$, fluctuating (rms) lift coefficient ($C_L{^{\prime}}$), and Strouhal number St. The changes in P and L leads to four distinct flow regimes (I, II, III and IV). Following the flow structure change, the ${\bar{C}}_D$, $C_L{^{\prime}}$, and St all vary greatly with the change in L and/or P. The ${\bar{C}}_D$ and $C_L{^{\prime}}$ both grow with increasing P and/or L. The St increases with P for a given L, being less sensitive to L for a smaller P (< 2) and more sensitive to L for a larger P (> 2). A strong relationship is observed between the flow regimes and the values of ${\bar{C}}_D$, $C_L{^{\prime}}$ and St. An increase in P affects the pressure distribution more on the top surface than on bottom surface while an increase in L does the opposite.

Development and Growth of Larvae of Four Bivalve Species (4종 조개류 유생의 발생과 성장)

  • Exposure to air and increased temperature induced successful spawning in Mytilus edulis, M. coruscus Crassostrea gigas and Pinctata fucata martensii. Developmental durations required for an egg to attain D-shaped larva and the D-shaped larva to reach pediveliger stage were estimated in these bivalves. Size of fertilized eggs was the largest (70.3 ${\mu}m$) in M. coruscus and the smallest (45.3 ${\mu}m$) in P. fucata martensii. At 17$^{\circ}C$, M. edulis and M. coruscus attained D-shaped larval stage within 48 hours after fertilization but those of C. gigas and B. fucata martensiii within 24 and 22 hours at 21 and 26$^{\circ}C$, respectively. The development duration required for a D-shaped larva to attain pediveliger stage was the longest (27 days) in M. coruscus and ranged between 20 and 22 days for the others. The shell length of the pediveliger was the longest (274.9 ${\mu}m$) in C. gigas and smallest (190.9 ${\mu}m$) in P. fucata martensii Length and height of larval shell was highly correlated with each other in all the 4 species. The shell height of C. gigas was more than the shell length beyond the size of 100 ${\mu}m$ shell length. However, shell length of the others was always longer than shell height at the larval stage.

Effects of pH and Nitrogen sources on the Pullulan Production by Aureobasidium pullulans (Aureobasidium pullulans에 의한 플루란 생산에서 pH와 질소원의 영향)

  • In this study, the effects of nitrogen sources and pH on pullulan production were investigated. As a result, the best nitrogen source in pullulan production by Aureobasidium pullulans was shown to be peptone and its product yield was 62%. Optimum concentration of carbon/nitrogen source ratio was 50/0.15 and the production of pullulan was inhibited at ratios higher than 50/0.15. Aureobasidium pullulans had produced 29.1, 27.4 and 26.5 g/L pullulan, respectively in media I, II, and III containing mixed nitrogen sources. This result showed that pullulan could be produced efficiently from mixed nitrogen source. It was found that pullulan yield with pH control was higher than that with no pH control. In fedbatch fermentation, pullulan yield obtained with a feeding rate of 0.05 N-g/L for nitrogen source was higher than that without nitrogen source feeding.

Addition of Coagulants for Phosphorus Removal from Combined Sewer Overflows (CSOs) (합류식 하수관거 월류수의 인제거를 위한 응집제 투여)

  • The coagulation of combined sewer overflows ($CSO_{s}$) was investigated by jar-testing with several commercial coagulants. $CSO_{s}$ sample showed different characteristics of coagulation from secondary wastewater with three common coagulants, aluminum sulfate, ferric chloride and polyaluminum chloride (PACl). Jar-tests showed that relatively wide range of optimal SS and T-P removal yielded with alum and ferric chloride compared with cationic polymers, though efficient SS and T-P removal can be achieved with all three coagulants. The decrease of pH was caused by the increase in dosage of aluminum sulfate, ferric chloride and PACl as coagulants. The pH was changed from 7.0 to 4.7 with the dosages of ferric chloride 25 mL/L. Aluminum sulfate revealed pH of 5.0 and PACl was highest pH of 5.4 after dosing of coagulants. The optimal pH to treat $CSO_{s}$ with aluminum sulfate were 6-6.5; with PACl 6-7, and with ferric chloride higher than 7.

Evaluation on Resistance Spot Weldability and Nugget Formation of Surface Roughness Treated Steel Sheet (표면조도 특성에 따른 저항 점 용접성 평가 및 너깃 형성 고찰)

  • With the increased use of surface textured steel sheet in body-in-white assembly, resistance spot weldability of these steels is considered to be an important subject. This study evaluated nugget formation and weldability by measuring dynamic resistance with various weld pressure, current, and weld time for steel sheet with two different surface roughnesses. The surface roughness for T-H steel ($R_{a}=1.70\;{\mu}m$) was higher than that for T-L steel ($R_{a}=1.33\;{\mu}m$), and resulted in increased contact resistance and heating for T-H steel spot welding. Therefore, at low weld current and weld cycle ranges, the T-H steel showed better weldability over the T-L steel. The evaluations of weld interface showed that the fusion zone in the T-H steel sheet was continuous in contrast to discontinuous fusion zone for T-L steel sheet at the same welding conditions. A comparison of dynamic resistance and tensile-shear strength (TSS) between T-H and T-L steel sheet suggested that high surface roughness provided larger heating at early cycle of welding and larger TSS.
