In vitro Estimation of Cardiac Output for the TAH using an Adaptive Fuzzy Identifier

  • An estimation algorithm based on training of fuzzy logic system using back-propagation is proposed, in this paper, for determining cardiac output in the TAH. The proposed estimation utilizes only a motor current waveform generated from the moving actuator of the electromechanical TAH without using any extra transducers as an information source for estimation. In in vitro tests, the resultant estimation performance was acceptable to alppy the proposed algorithm to animal experiments and further clinical applications.

Study of n/γ discrimination using 3He proportional chamber in high gamma-ray fields

  • The $^3He$ proportional chamber is widely used for neutron measurement owing to its high neutron detection efficiency and simplicity for gamma-ray rejection. In general, the neutron and gamma-ray signals obtained from the $^3He$ proportional chamber can be easily separated by the difference in the pulse heights. However, for a high gamma-ray field, the gamma-ray signal cannot be precisely eliminated by the pulse height due to gamma-ray pulse pileup which causes the pulse height of gamma-ray pulse to increase and making the pulses due to neutrons and gamma rays indistinguishable. In this study, an improved algorithm for $n/{\gamma}$ discrimination using a parameter, which is the ratio of the rise time to the pulse height, is proposed. The $n/{\gamma}$ discrimination performance of the algorithm is evaluated by applying it to $^{252}Cf$ neutron signal separation from various gamma-ray exposure rate levels ranging 0.1-5 R/h. The performance is compared to that of the conventional pulse-height analysis method in terms of the gamma elimination ratio. The suggested algorithm shows better performance than the conventional one by 1.7% (at 0.1 R/h) to 70% (at 5 R/h) for gamma elimination.

한국산 유색미 속겨의 안토시아닌 색소에 의한 견직물염색 (Silk Dyeing Method in Natural Pigments - In Case of Korean Colored Rice Bran -)

  • This research was conducted to establish the efficient use of the colored rice bran fur dyeing textiles. To investigate the fabric dyeability of the colored rice bran extract, the anthocyanin pigments were extracted with water of different temperature ranges of 40 ~ 8$0^{\circ}C$ and were dyed on silk fabrics with different dyeing temperature $25^{\circ}C$~6$0^{\circ}C$, at acidic pH and neutral pH, respectively. Aluminum chloride was preheated with mordant K/S value and dyeing fastness of dyed silk fabrics were examined. The anthocyanins of the colored rice bran were stable and red color at acidic pH, red purple or purple blue at neutral pH, but unstable, blue color at alkaline pH. If extracting temperature and dyeing temperature of dyeing solution were higher, the dyeability was high, but the color of dyed fabric showed red tone. When extracting temperature was 8$0^{\circ}C$ and dyeing temperature of dyeing solution was 6$0^{\circ}C$, the dyeability was best. Without mordant, the dyeability of silk fabrics was higher in acidic pH than in neutral pH solution. With mordant, the dyeability was higher than without mordant, and also higher in acidic pH. Pretreatment of aluminum chloride resulted in the increase of color intensity and stability. The laundering fastness of dyed fabrics was good from grade 5 to grade 3-4. Because of the anthocyanins sensitivity on light radiation, the light fastness of dyed fabrics was poor from grade 3 to grade 1-2.

Effect of Extrinsic Factors on Differentiated Cardiomyocyte-like Cells from Human Embryonic Stem Cells

  • Cardiovascular diseases (CVDs) are one of the most cause of death around the world and fields of interest for cardiac stem cells. Also, current use of terminally differentiated adult cardiomyocytes for CVDs has limited regenerative capacity therefore any significant cell loss may result in the development of progressive heart failure. Human embryonic stem cells (hESCs) derived from blastocyst-stage embryos spontaneously have ability to differentiate via embryo-like aggregates (endoderm, ectoderm and mesoderm) in vitro into various cell types including cardiomyocyte. However, most effective molecule or optimized condition which can induce cardiac differentiation of hESCs is rarely studied. In this study, we developed both spontaneous and inductive cardiomyocyte-like cells differentiation from hESCs by treatment of induced-factors, 5-azacytidine, BMP-4 and cardiogenol C. On the one hand, spontaneous and inductive cardiomyocyte-like cells showed that cardiac markers are expressed for further analysis by RT-PCR and immunocytochemistry. Interestingly, BMP-4 greatly improved homogeneous population of the cardiomyocyte-like cells from hESCs CHA15 and H09. In conclusion, we verified that spontaneously differentiated cells showed cardiac specific markers which characterize cardiac cells, treated extrinsic factors can manage cellular signals and found that hESCs can undergo differentiation into cardiomyocytes better than spontaneous group. This finding offers an insight into the inductive factor of differentiated cardiomyocytes and provides some helpful information that may offer the potential of cardiomyocytes derived from hESCs using extrinsic factors.

광배양 시스템에서 NOx 및 SOx의 배지 pH와 미세조류 생장에 미치는 영향 (Effects of NOx and SOx on the Medium pH and microalgal growth in photo-culture system)

  • Carbon dioxide reduction technologies using photosynthetic microorganism were suggested to overcome environmental destruction caused by $CO_2$ in flue gases from power plant and other industries. However, there are many toxic constituents in flue gas including CO, NOx, SOx. Continuous and Excessive supply of these noxious gases to cells will leads to inhibition of microalgal growth along with partial cell death. In this study, we tested the noxious effect of SOx and NOx on the pH and microalgal growth under photoautotrophic culture in three microalgae of Neochloris oleoabundans, Chlorella vulgaris and Haematococcus pluvialis. As a result, SOx concentration more than 50 ppm led to the rapid reduction of pH, thereby inhibiting of the growth in Neochloris oleoabundans and Chlorella vulgaris. NOx concentration more the 100 ppm reduced the exponential growth of N. oleoabundans and C. vulgaris. And H. pluvialis exhibited low sensitivity to SOx and NOx. Consequently, the three microalgae of N. oleabundas, C. vulagaris and H. pluvialis showed the normal vegetative growth in 25 ppm of NOx and SOx. Above all, H. pluvialis was useful for the $CO_2$ sequestration of the flue gas including high concentrations of NOx and SOx.

Alcaligenes eutrophus의 배양액으로부터 균체 분리 효율에 미치는 철(Fe)계 응집제의 효과 (Effect of re-based Coagulants on Cell Separation Efficiency from the Culture Broth of Alcaligenes eutrophus.)

  • Alcaligenes eutrophus의 세포배양액(23-210 g dry weight/L)으로부터 균체 회수 효율에 미치는 철계 응집제 첨가 효과에 대해 연구하였다. 응집제로는 무기 응집제인 Fe$_2$(SO$_4$)$_3$과 무기 고분자 응집제인 Ferix-3를 사용하였다. 철계 응집제는 3-13의 넓은 pH 범위에서 응집효과를 나타내었으며, 배양액의 pH가 증가할수록 floc의 크기는 증가하였다. 균체 회수를 위한 배양액의 최적 pH는 10-13이었다. 최적 응집제의 첨가량은 세포농도가 증가함에 따라 증가하였는데, 무기응집제인 Fe$_2$(SO$_4$)$_3$보다는 무기고분자응집제인 Ferix-3이 적은 농도에서 응집 효율이 우수함을 알 수 있었다. 210 g/L의 세포농도 배양액에 Ferix-3을 1300 mg Fe/L를 첨가하면 45$\times$g의 낮은 원심력에 의해 95%이상의 균체를 회수 가능하였다. 응집제의 요구량은 배양액 중의 NH$_4$$^{+}$ 농도에 비례하여 증가하였고, 배양액 중의 NH$_4$$^{+}$ 농도가 1 g 증가하면 응집제가 0.066 g Fe$_3$$^{+}$정도 더 요구되었다.

사용종료 비위생매립지 주변 지하수의 수질변화 특성 (Evaluation of Groundwater Quality Characteristics around Uncontrolled Closed Valley Landfill)

  • 본 연구에서는 W시에 있는 비위생매립지 주변의 지하수질 약 2년간 관측하고 그 결과를 평가하였다. 주요관심 인자는 산화환원조건에 민감한 pH, 용존산소, 전기전도도, 산화환원전위, 용존 유기탄소, 암모니아, 질산성질소 및 황산이온 등이다. 본 매립지는 생활쓰레기 매립지로 매립종료 약 10년이 경과하였다. 매립지 침출수는 COD 136${\sim}$263 mg/L, T-N 121${\sim}$186 mg/L, NH3-N 14${\sim}$369 mg/L이며, 계절적 변동이 크게 나타났다. 매립지 직하부 지하수는 약산성을 띄나 전기전도도가 매우 높아 침출수 누출의 영향을 나타내었다. 염소이온의 경우 200 mg/L 이상으로 높으나 거리가 멀어짐에 따라 점차 감소하였는데 확산 및 희석 등의 자연저감효과가 큰 것으로 사료된다. 매립지 내부에서는 산화 환원전위가 음의 값을 보이나 하부에서는 350 mV까지 증가하여 혐기성환경에서 산화환경으로의 전이를 나타내었다. 암모니아는 가장 우려되는 지하수질 인자인데 대부분의 관측정에서 크게 부화되어 있으며, 먹는물 수질기준을 상회하였다. 본 매립지 하부의 오염지하수질은 시공간적으로 자연저감 현상이 뚜렷하나 지속적인 수질모니터링이 필요한 것으로 판단된다.

Comprehensive investigations of key mitochondrial metabolic changes in senescent human fibroblasts

  • There is a paucity of detailed data related to the effect of senescence on the mitochondrial antioxidant capacity and redox state of senescent human cells. Activities of TCA cycle enzymes, respiratory chain complexes, hydrogen peroxide (H2O2), superoxide anions (SA), lipid peroxides (LPO), protein carbonyl content (PCC), thioredoxin reductase 2 (TrxR2), superoxide dismutase 2 (SOD2), glutathione peroxidase 1 (GPx1), glutathione reductase (GR), reduced glutathione (GSH), and oxidized glutathione (GSSG), along with levels of nicotinamide cofactors and ATP content were measured in young and senescent human foreskin fibroblasts. Primary and senescent cultures were biochemically identified by monitoring the augmented cellular activities of key glycolytic enzymes including phosphofructokinase, lactate dehydrogenase, and glycogen phosphorylase, and accumulation of H2O2, SA, LPO, PCC, and GSSG. Citrate synthase, aconitase, α-ketoglutarate dehydrogenase, succinate dehydrogenase, malate dehydrogenase, isocitrate dehydrogenase, and complex I-III, II-III, and IV activities were significantly diminished in P25 and P35 cells compared to P5 cells. This was accompanied by significant accumulation of mitochondrial H2O2, SA, LPO, and PCC, along with increased transcriptional and enzymatic activities of TrxR2, SOD2, GPx1, and GR. Notably, the GSH/GSSG ratio was significantly reduced whereas NAD+/NADH and NADP+/NADPH ratios were significantly elevated. Metabolic exhaustion was also evident in senescent cells underscored by the severely diminished ATP/ADP ratio. Profound oxidative stress may contribute, at least in part, to senescence pointing at a potential protective role of antioxidants in aging-associated disease.

여러 pH에서 가열응고(加熱凝固)시켜 조제(調製)한 녹엽단백질(綠葉蛋白質)의 지방산(脂肪酸)과 Sterol조성(組成) (Fatty Acid and Sterol Composition of Leaf Protein Coagulated at Different pH)

  • 응고시(凝固時) pH에 따라 green LPC중(中)의 지방산(脂肪酸)관 sterol의 조성(組成)이 어떻게 달라지는지를 알아보고져 Italian ryegrass, red clover, oats 및 alfalfa의 녹즙(綠汁)을 pH 4, 6, 8로 조성하여 가열응고(加熱凝固)시켜 green LPC를 만들어 실험한 결과(結果)는 다음과 같다. 1. Italian ryegrass, oats 및 alfalfa의 green LPC는 linoleic acid, palmitic acid, linolenic acid의 순(順)으로 함량(含量)이 점점 많았고, 이 세 지방산(脂肪酸) methyl ester가 전체지방산(全體脂肪酸) methyl ester의 70.4%에서 86.2%를 점유했다. red clover의 green LPC는 palmitic acid, linoleic acid, linolenic acid의 순(順)으로 함량(含量)이 많았고 이 세 지방산(脂肪酸) methyl ester는 전체지방산(全體脂肪酸) methyl ester의 82.5%에서 86.2% 범위였다. 2. red clover, oats 및 alfalfa의 green LPC는 pH4, pH6, pH8에서 가열응고(加熱凝固)한 LPC순(順)으로 linolenic acid의 함량(含量)이 점점 많았다. Italian ryegrass의 green LPC는 pH4, pH8, pH6에서 가열응고(加熱凝固)시킨 LPC순(順)으로 linolenic acid의 함량(含量)이 점점 많았다. 3. red clover와 alfalfa의 green LPC에서 cholesterol이 검출(檢出)되었다. 모든 green LPC에서 campesterol, stigmasterol, sitosterol이 검출(檢出)되었다. 4. 일반적으로 pH6, pH4, pH8에서 가열응고(加熱凝固)시켜 조제(調製)한 green LPC순(順)으로 sterol의 함량(含量)이 점점 많았다. 다만 alfalfa의 green LPC는 pH4, pH6, pH8에서 가열응고(加熱凝固)시켜 조제(調製)한 LPC순(順)으로 sterol의 함량(含量)이 점점 많았다.

천연인디고를 이용한 셀룰로오스계 직물 염색의 표준화 연구: 일단계 환원/염색에 의한 마직물 염색 (Optimization of Cellulose Dyeing with Natural Indigo: Ramie dyeing by One-step Reduction/dyeing Process)

  • One-step reduction/dyeing method was applied for ramie dyeing with natural indigo powder. The effect of reduction/dyeing conditions including the pH of bath, dye temperature and time, and concentration of indigo powder and reduction agent on dye uptake and color properties were investigated. Regardless of addition of alkali, the dyed fabrics appeared in the PB Munsell color range(${\lambda}_{max}$: 660 nm) and the dye uptake was much higher with no addition of sodium hydroxide. Dyeing was carried out through the use of only sodium hydrosulfite in the bath. The maximum dye uptake was obtained at 60 for 30min. Saturated dye uptake was obtained at 2 g/L of sodium hydrosulfite concentration up to 2 g/ L of indigo powder. Whereas, at higher indigo powder concentration (4 g/L), more than 3 g/L of reducing agent concentration was required for obtaining the saturated dye uptake. Color reproducibility was reliable with color difference in the range of 0.03~0.16. Regardless of color strength, fastness to rubbing was acceptable with a 3/4~4/5. Fastness to washing, dry cleaning, and light of samples with low color strength were poor. Whereas, fastness to washing, dry cleaning, and light of samples with high color strength were very good.
