제주도에 서식하는 까막전복(Haliotis discus)의 Macroalgae에 대한 섭식선호도 및 섭식유도물질 연구

  • 우리나라에 서식하는 전복류는 참전복(Haziotis discus hannai), 까막전복(H, disus), 말전복(H. gigantea), 시볼트전복(H. siebolidi)이 주로 서식하는 것으로 알려져 있으며, 원시복족류인 전복류는 잘 발달된 치설(radula)로 grazing 하는 섭식형태를 가지는 초식동물(herbivores)이다. 초식동물은 해중림 지역에서 다양한 생물군집이 직ㆍ간접적으로 관련되어 생태계를 유지하며(Lobban and harrison, 1994), 이들의 초식작용은 생태계에서 다양한 군집을 구조화하는 중요한 과정이다(John et al., 1992) (중략)

근친교배에 의한 북방전복 (Haliotis discus hannai)의 근교약세 현상 (Inbreeding depression of Pacific abalone, Haliotis discus hannai by inbreeding mating experiments)

  • 북방전복의 근친교배로 인한 근교약세현상의 유무를 명확히 하기 위하여 근친교배 2가계 (AA 및 BB)와 비근친교배 (ww1 및 ww2) 2가계를 같은날 동시에 생산하여 유생단계부터 동일한 수조에 혼합 사육관리한 후 친자확인 기술을 통하여 4개월째 및 10개월째 각각의 가계별 생존율 및 성장을 비교분석하였다. 성장의 경우, 4개월째 근친교배구 AA가계만이 비근친교배구 ww1 및 ww2가계보다 유의적으로 낮은 값으로 나타났지만, 10개월째는 근친교배구 AA가계 및 BB가계 모두 비근친교배구 ww1 및 ww2가계보다 유의적으로 낮은 값을 나타냈다. 또한 생존율에 있어서도 근친교배구가 비근친교배구에 비하여 낮은 값을 나타내었다. 이것은 한세대의 근친교배(근교계수 : 0.25) 에 있어서도 근교약세현상이 나타난다는 것을 보고하고 있다.

북방전복 종묘생산을 위한 농축 규조류 Caloneis schroederi와 Rhaphoneis sp.의 효과 (Effect of the concentrated-diatom of Caloneis schroederi and Rhaphoneis sp. for seedling production of Haliotis discus hannai)

  • Even though seedling production of Haliotis discus hannai has fully developed, the culture of benthic diatom as a live food for larvae is still a barrier to solve in commercial hatchery. The farmer depends on mixed microalgae which are non-selectively attached on the plate by flowing of natural seawater. The adequate diatom on the plate for the larvae in terms of quality and quantity is always significant bottleneck in the hatchery. In this study, two benthic diatom species, Caloneis schroederi and Rhaphoneis sp. were separately cultured in mass and inoculated to four tons' settlement tank of the larvae. And the larvae and the spats were cultured for nineteen and nine weeks, respectively. The result on seedling production of H. discus hannai with this method was compared to that of the farmer's traditional method as a control. With regard to variation of species composition of benthic diatom on the plate, C. schroederi and Rhaphoneis sp. were dominant for first three weeks after inoculation. But the diverse diatoms mainly, Navicula, Amphora, Cylindrotheca, Licmophora, Pleurosigma began to attache on the plate from the 4th week. The larvae attached 2.5 times more in C. schroederi tank than in the control tank. The final total biomass of the seeds in Rhaphoneis sp. tank was 3.2 times more than that of the control tank. The retared-spats in the seedling production also showed significantly higher growth and survival in the spat fed Rhaphoneis sp. or C. schroederi than those in the control group. We suggest that C. schroederi is proper for settlement of the larvae and Rhaphoneis sp. is appropriate for the growth of the larvae and spats in the commercial hatchery of H. discus hannai.

Effects of Amino Acids on Larval Settlement and Metamorphosis in Haliotis discus hannai

  • The compositions of amino acid in 6 monocultured benthic diatoms used in aquaculture of Haliotis discus hannai were analyzed, and effects of 15 artificial synthetic amino acids on the settlement and metamorphosis of H. discus hannai larvae. Results showed that the content of L-aspartic acid was highest in all diatoms, and that of L-glutamic acid was second high. In experiment using settlement slat without benthic diatom attached, the highest settlement rate (33.3 ${\pm}$ 8.8%) was obtained with L-glutamic acid, and a higher value (16.7 ${\pm}$ 3.3%) was found with L-aspartic acid at 24 h after experimental commencement, compared to that of control (8.6 ${\pm}$ 5.1%). 80 h later the metamorphosis rates of L-glutamic acid (86.7 ${\pm}$ 10.7%) and L-aspartic acid (80.0 ${\pm}$ 3.3%) groups were higher than control group(0) and other amino acids significantly. The response rate of L-glutamic acid was the highest (62.0%), and those of L-aspartic acid (30.0%) and L-threonine (25.3%) groups were also significantly higher than control group. In the experiment using settlement slat with benthic diatom attached, the best effect of various amino acids on induction of larval settlement was obtained with L-glutamic (82.0 ${\pm}$ 6.9%) and L-aspartic acid (78.7 ${\pm}$ 5.1%) at 24 h after experimental commencement. The settlement rates of L-histidine, L-leucine, L-lysine, L-methionine, L-phenylalanine, and L-tyrosine groups were significantly lower than control group. The same differences in the metamorphosis rate at 56 h after experimental commencement and in the response rate were found. It should be noted that after 80 h the metamorphosis rates of L-histidine (74.0 ${\pm}$ 12.0%) and L-lysine (87.0 ${\pm}$ 8.8%) declined rapidly compared to those of 56 h (8.0 ${\pm}$ 12.0%; 7.7 ${\pm}$ 12.0%).

Comparative genetic diversity of wild and released populations of Pacific abalone Haliotis discus discus in Jeju, Korea, based on cross-species microsatellite markers including two novel loci

  • Pacific abalone Haliotis discus discus is an important fisheries resource in Jeju, Korea. For basic information about its current genetic status in relation to stock enhancement, the level and distribution of genetic variation between wild and released stocks of Pacific abalone in Jeju were examined at nine cross-species microsatellite markers including the use of two novel primers. High levels of polymorphism were observed between the two populations. A total of 146 different alleles were found at all loci, with some alleles being unique. The allelic variability ranged from five to 27 in the wild population and from four to 16 in the released sample. The average observed and expected heterozygosities were estimated to be 0.74 and 0.84 in the wild sample and 0.70 and 0.78 in the released sample, respectively. Although a considerable loss of rare alleles was observed in the released sample, no statistically significant reductions were found in heterozygosity or allelic diversity in the released sample compared to the wild population. Low but significant genetic differentiation was found between the wild and released populations. These results suggest that the intensive breeding practices for stock enhancement may have resulted in a further decrease in genetic diversity, and that the cross-species microsatellite markers used in this study represent a potentially efficient means for further genetic studies, providing beneficial information for the protection and management of H. discus discus.

북방전복 (Haliotis discus hannai)에서 분리한 Glutathione S-transferase 유전자의 분자생물학적 고찰 및 발현분석 (Molecular Characterization and Expression Analysis of a Glutathione S-Transferase cDNA from Abalone (Haliotis discus hannai))

  • 본 연구에서는 북방전복 (Haliotis discus hannai)의 대용량 염기서열 분석을 통해 GST유전자의 전장 cDNA를 동정하였다. 북방전복 GST 유전자의 총 길이는 1669 bp로 672 bp의 ORF는 총 223개의 아미노산을 코딩하고 있으며 등전점은 5.69, 분자량은 25.8 kDa으로 예측되었다. 북방전복 GST아미노산 서열은 둥근전복과 지중해 담치와 같은 패류의 GSTA와 가장 유사성이 높았으며 계통수 분석을 통해 GSTA와 하나의 그룹을 이루었다. 북방전복 GST에는 GSTA의 특징을 갖는 두 site (N-말단의 G-site, C-말단의 H-site)가 보존되어 있었고 효소활성과 구조 유지에 중요한 잔기가 종간에 매우 보존되어 있었다. 북방전복 GST 유전자의 mRNA는 관찰된 모든 조직에서 발현하고 있었으며, 특히 외투막, 아가미, 간췌장, 소화관에서 높은 발현이 확인되었다. 북방전복의 GST는 비브리오균을 인위감염 시킨 전복의 간췌장에서 감염 후 1시간 뒤 발현이 급격히 증가했다가 3시간까지 증가한 뒤 감소하였고, 혈구세포에서는 감염 3시간 경과 후 발현 정도가 최고로 증가했다가 감소하였다. 따라서 북방전복 GST는 alpha class GST의 특징을 가지며 병원체 감염에 따른 면역반응 조절에 관여할 것이라 생각되며 병원균 감염에 따른 바이오마커로 활용가능 할 것이라 예상된다.

Initial Culture Conditions for Primary Cell Populations Derived from Radula Tissue in Abalone Haliotis discus hannai

  • Abalone immortal cell lines can be used to study the physiological properties and disease mechanisms of abalone at the cellular and molecular level. As a first step for the final goal to establish abalone immortal cell lines, we examined various initial culture conditions for primary cell populations derived from Haliotis discus hannai radula tissue. The survival rate after cell isolation procedures using the enzymatic method was as low as $9.95{\pm}2.37%$. Based on three different experimental conditions for H. discus hannai radula-derived cell culture, we found that the salinity of the media and the presence of growth-promoting factors were important to support radula-derived primary cell populations during the initial culture. The growth factor-containing media adjusted to 35 psu salinity could induce 100% (8 out of 8 trials) initial cell attachment, and the rate of cell attachment reached 50-70%. The data obtained from this study will provide useful information for developing immortal cell lines from abalone species.

  • 주사전자현미경과 광학현미경을 이용하여 북방전복 아가미의 형태와 조직화학적 특징에 대하여 기재하였다. 북방전복의 아가미는 원새형의 bipectinate형 이었다. 아가미 새엽의 표면은 잘 발달된 섬모들로 덮여있었다. 새엽의 상피층은 공통적으로 단층으로 원주형의 상피세포와 점액세포로 구성되어 있었다. AB-PAS (pH 2.5와 pH 1.0) 반응과 AF-AB (pH 2.5) 반응 결과, 점액세포들은 주로 산성의 뮤코점액다당류를 함유하고 있었다.

  • The increasing of atmospheric $CO_2$ are changing the pH (ocean acidification) and temperature of the sea. Although the effects of ocean acidification on calcifying organisms have well-documented, only a few studies have examined the combined effects of ocean acidification and elevated temperature. This study investigated the effects of ocean acidification and elevated temperature for 2100 on survival and growth of juvenile abalone, Haliotis discus hannai. Ocean acidification was simulated by bubbling $CO_2$ into seawater at concentrations of 1,000 and 1,500 ppm, and temperature was set at room temperature $+2^{\circ}C$. Neither $CO_2$ nor temperature had a significant effect on survival of abalone, while both significantly affected growth. There was no significant interaction between the two factors. Shell length can be used as a growth index of abalone to access the impacts of ocean acidification and elevated temperature.
