Prediction Model for the Cellular Immortalization and Transformation Potentials of Cell Substrates

  • The establishment of DNA microarray technology has enabled high-throughput analysis and molecular profiling of various types of cancers. By using the gene expression data from microarray analysis we are able to investigate diagnostic applications at the molecular level. The most important step in the application of microarray technology to cancer diagnostics is the selection of specific markers from gene expression profiles. In order to select markers of Immortalization and transformation we used c-myc and $H-ras^{V12}$ oncogene-transfected NIH3T3 cells as our model system. We have identified 8751 differentially expressed genes in the immortalization/transformation model by multivariate permutation F-test (95% confidence, FDR<0.01). Using the support vector machine algorithm, we selected 13 discriminative genes which could be used to predict immortalization and transformation with perfect accuracy. We assayed $H-ras^{V12}$-transfected 'transformed' cells to validate our immortalization/transformation dassification system. The selected molecular markers generated valuable additional information for tumor diagnosis, prognosis and therapy development.

  • An analytical diffusion model for flood routing with backwater effects and lateral flows is developed. The basic diffusion equation is linearized about an average depth of (H + h), and is solved using the boundary conditons which take into account the effects of backwater and lateral flows. Scouring phenomenon around pier which affects on the support function of pier and the stabilization if river bed is a complex problem depending on flow properties and river bed state as well as pier geometry. therefore, there is no uniting theory at present which would enable the designer to estimate, with confidence, the depth of scour at bridge piers. The various methods used in erosion control are collectively called upstream engineering, HEC-RAS Model, underwater blasting. They consist of reforestation, check-dam construction, planting of burned-over areas, contour plowing and regulation of crop and grazing practices. Also included are measures for proper treatment of high embankments and cuts and stabilization of streambanks by planting or by revetment construction. One phase of reforestation that may be applied near a reservoir is planting of vegetation screens. Such screens, planted on the flats adjacent to the normal stream channel at the head of a reservoir, reduce the velocity of silt-laden storm inflows that inundate these areas. This stilling action causes extensive deposition to occur before the silt reaches the main cavity of the reservoir.

a-C:H Films Deposited in the Plasma of Surface Spark Discharge at Atmospheric Pressure. Part I: Experimental Investigation

  • The aim of this work is the synthesis of a-C:H films from methane gas using surface spark discharge at the atmospheric pressure. Properties of these films have been investigated as functions of energy W delivered per a methane molecule in the discharge. The method enables the coatings to be deposited with high growth rates (up to $100 \mu\textrm{m}$/hour) onto large-area substrates. It is shown that the films consist of spherical granules with diameter of 20∼50 nm formed in the spark channel and then deposited onto the substrate. The best film characteristics such as minimum hydrogen-to-carbon atoms ratio H/C=0.69, maximum hardness $H_{v}$ =3 ㎬, the most dense packing of the granules and highest scratch resistance has been obtained under the condition of highest energy W of 40 eV. The deposited a-C:H coatings were found to be more soft and hydrogenated compared to the diamond-like hydrogenated (a-C:H) films which obtained by traditional plasmaenhanced chemical vapor deposition methods at low pressure (<10 Torr). Nevertheless, these coatings can be potentially used for scratch protection of soft plastic materials since they are of an order harder than plastics but still transparent (the absorption coefficient is about $10^4$$10^{5}$ $m^{-1}$ At the same time the proposed method for fast deposition of a-C:H films makes this process less expensive compared to the conventional techniques. This advantage can widen the application field of. these films substantially.y.
