Determination of the Solution Structure of Malonyl-CoA by Two-Dimensional Nuclear Magnetic Resonance Spectroscopy and Dynamical Simulated Annealing Calculations

  • In order to understand the initial interaction of the substrates malonate, ATP, and CoA with malonyl-CoA synthetase, the catalytic product malonyl-CoA was characterized by NMR spectroscopy and molecular modeling. To assign proton and carbon chemical shifts, two-dimensional $^1H-^1H$ DQF-COSY and $^1H-^{13}C$ HMBC experiments were used. The structure of malonyl-CoA in the solution phase was determined based on distance constraints from NOESY and ROESY spectra. The structures were well-converged around the pantetheine region with the pairwise RMSD value of 0.08 nm. The solution structure exhibited a compact folded conformation with intramolecular hydrogen bonds among its carbonyl and hydroxyl groups. These findings will help us to understand the initial interaction of malonate and CoA with malonyl-CoA synthetase.

  • The combination of atom transfer radical polymerization (ATRP) and click chemistry was employed for the efficient preparation of well-defined block copolymers. Bromo terminated poly(methyl acrylate) (pMA-Br) was prepared by an ATRP initiator, ethyl-2-bromoisobutyrate (EBiB). Subsequently, the bromine chain end of pMA-Br was converted to an azide group by simple nucleophilic substitution reaction. ${\alpha}-Alkyn-{\omega}-bromo-functionalized$ polystyrene was also synthesized by ATRP using the alkyn-functionalized initiator, propargyl-2-bromoisobutyrate (PgBiB). In both cases, the conversion was limited to a low level to ensure a high degree of chain end functionality. Then the coupling reaction between the azide end group in $pMA-N_3$ and alkyn-functionalized PgBiB-pSt was performed by Cu(I)catalysis. This coupling reaction was monitored by gel permeation chromatography (GPC). The synthesized block copolymer was characterized by FT-IR, $^1H-NMR$ spectroscopy and $^1H-^1H$ COSY correlation spectroscopy.

  • A total of five compounds namely, 2-methylanthracene-9, 10-dione (1), 1-hydroxy-6-methylanthracene-9,10-dione (2), $\beta$-sitosterol (3), stigmasterol (4) and sitosterone (5) were isolated from the stem extracts of Polygonum lanatum for the first time. The structures of the isolated compounds (1-5) were established by extensive spectroscopic studies, including 2D NMR such as $^{1}H-^{1}H$ COSY, HSQC and HMBC studies. The crude extracts and purified compound (1) were screened for antimicrobial activity against a wide range of Gram-positive and Gram-negative bacteria and fungi by the disc diffusion method. The n-hexane and dichloromethane soluble partitionates of the methanolic extract revealed mild to moderate inhibition of growth of the test organisms. The cytotoxic potential of the extractives and compound 1 was also determined by using brine shrimp lethality bioassay, where the extractives demonstrated significant cytotoxic activities.

Isolation and Characterization of α-Glucosidase Inhibitor Produced by Bacillus sp. SKU31-1 Strain (Bacillus sp. SKU31-1가 생산하는 α-Glucosidase 저해제 분리 및 특성 조사)

  • In the course of screening for ${\alpha}$-glucosidase inhibitor produced by microorganism, the active compound was isolated from the culture filtrate of Bacillus sp. SKU31-1 using a series of chromatography procedures. The structure of the active compound was elucidated as 5-amino-1-hydroxymethyl-1, 2, 3, 4-cyclohexanetetrol on the basis of spectroscopic evidence obtained and comparison with data from the literature. The active compound showed potent inhibitory activity against ${\alpha}$-glucosidase with an $IC_{50}$ value of $1.9{\mu}M$ for maltose and 4.9 mM for sucrose. A Lineweaver-Burk plot indicated that its inhibition of ${\alpha}$-glucosidase was competitive, with a $K_i$ value of 0.15 mM.

  • The purification of the MeOH extract from Impatiens basamina by repeated column chromatography led to the isolation of one new tetrahydronaphthalene (1), together with eleven known compounds (2 - 12). The structure of the new compound (1) was determined by spectral data analysis ($^1H$ and $^{13}C$-NMR, $^1H-^1H$ COSY, HSQC, HMBC, NOESY, and HR-ESI-MS). Isolated compounds (1 - 12) were evaluated for their inhibitory effects on NO production in LPS-activated murine microglial BV-2 cells and their effects on NGF secretion from C6 glioma cells. Compounds 3, 7, and 10 reduced NO levels in LPS-activated murine microglial cells with $IC_{50}$ values of 26.89, 25.59, and $44.21{\mu}M$, respectively. Compounds 1, 5, and 9 upregulated NGF secretion to $153.09{\pm}4.66$, $156.88{\pm}8.86$, and $157.34{\pm}3.30%$, respectively.

Chemical Structure of the Major Color Component from a Korean Pigmented Rice Variety (한국산 유색미에서 분리한 안토시아닌의 화학구조)

  • The major color component of a Korean pigmented rice (Oeyza sativa var. Suwon 415) was purified with Amberlite XAD-7 column and preparative paper chromatography. The purified pigment was determined as anthocyanin by paper chromatography, UV/Vis and NMR spectroscopy. The $\lambda_{max}$ of the Purified anthocyanin on UV/Vis spectrum were 529 nm and 281 nm. The $A_{440}/A_{529}$ value of the purified anthocyanin was 23% suggesting the presence of 3-glycosidic structure. The aglycone from acid hydrolysis showed bathochromic shift (18 nm) in the presence of $AlCl_3$ indicating that the anthocyanidin contained free adjacent hydroxyl groups such as cyanidin, delphinidin, petunidin or luteolinidin. The sugar moiety obtained from acid hydrolysis was determined as glucose by paper chromatography. The NMR spectra showed that the aglycone was cyanidin and the sugar was ${\beta}-D-glucopyranose$. Thus, the chemical structure of the purified anthocyanin was identified as cyanidin $3-O-{\beta}-D-glucopyranoside$.

A Systematic NMR Determination of α-D-Glucooligosaccharides, Effect of Linkage Type, Anomeric Configuration and Combination of Different Linkages Type on 13C Chemical Shifts for the Determination of Unknown Isomaltooligosaccharides

  • Prebiotic isomaltooligosaccharide preparations contain $\alpha$-D-glucooligosaccharides comprising isomaltooligosaccharides (IMOs) and non-prebiotic maltooligosaccharides (MOs). They are both glucose oligosaccharides characterized by their degree of polymerization (DP) value (from 2 to $\sim$10), linkages types and positions (IMOs: $\alpha$-(1$\rightarrow$2, 3, 6 and in a lower proportion internal 1$\rightarrow$4) linkages, MOs: α-(1$\rightarrow$4) linkages). Their structure is the key factor for their prebiotic potential. In order to determine and elucidate the exact structure of unknown IMOs and MOs, unambiguous assignments of $^{13}C$ and $^1H$ chemical shifts of commercial standards, representative of IMOs and MOs diversity, have been determined using optimized standard one and two-dimensional experiments such as $^1H$ NMR, $^{13}C$ NMR, APT and ${^1}H-{^1}H$ COSY, TOCSY, NOESY and <$^1H-{^{13}}C$ heteronuclear HSQC, HSQC-TOCSY, and HMBC. Here we point out the differential effect of substitution by a glucose residue at different positions on chemical shifts of anomeric as well as ring carbons together with the effect of the reducing end configuration for low DP oligosaccharides and diasteroisotopic effect for H-6 protons. From this study, structural $^{13}C$ specific spectral features can be identified as tools for structural analysis of isomaltooligosaccharides.

Novel Anthracene Derivatives Isolated from Rice Hulls of Oryza sativa and Their Growth Inhibitory Activity of Radish Seed

  • Three new compounds orizaanthracenol (1-methoxyanthracen-2-ol, 1), 1-hydroxy-7-((2S,3R,4R,5S)-2'',3'',4''-trihydroxy-5''-(hydroxymethyl)tetrahydro-2H-pyran-1-yloxy)anthracen- 2-yl 3',7'-dimethyloctanoate (2) and 1-hydroxy-7-((2S,3R,4R,5S)-2'',3'',4''-trihydroxy-5''-(hydroxymethyl)tetrahydro-2H-pyran-1-yloxy)anthracen- 2-yl 3',7',11',15',19'-pentamethyltricosanoate (3) have been isolated and determined from the rice hulls of Oryza sativa. The structures of the compounds were elucidated using 1D and 2D NMR spectral methods, viz: 1H-NMR, 13C-NMR, DEPT, COSY and HSQC, and HMBC aided by IR, EIMS and FAB-MS. These isolated compounds were tested in bioassay on germination of radish to detect their inhibitory activities, in which compound 1 exhibited the most inhibition at the concentration of 40 ppm was 50.96% of total dry weight. Furthermore, germination and growth of radish were reduced by 27 to 48%, and dry weight of shoot and root were arranged from 50 to 52%, whilst compounds 2 and 3 exhibited weak inhibition than 1.

Ring-Opening Metathesis Polymerization and Hydrogenation of Ethyl-substituted Tetracyclododecene

  • Ring-opening metathesis polymerization (ROMP) of an ethyl-substituted tetracyclododecene (8-ethyltetracyclo[$4.4.0.1^{2,5}.1^{7,10}$] dodec-3-ene, Et-TCD) was carried out in the presence of a ternary catalyst system consisting of $WCl_6$, triisobutyl aluminium (iso$Bu_3Al$), and ethanol. The optimal molar ratio of Et-TCD/$WCl_3$/iso-$Bu_3Al$/ethanol was found as 500/1/3/2 at which the yield of ring-opened polymer was 100%. 1-Hexene was shown to be an effective molecular weight controlling agent for ROMP reaction of Et-TCD. The hydrogenation of the ring opened polymer (p-Et-TCD) was conducted successfully using Pd(5 wt %)/${\gamma}$-$Al_2O_3$ at $80^{\circ}C$ for 1 h. Chemical structures of p-Et-TCD and its hydrogenated product($H_2$-p-Et-TCD) were characterized using 2D NMR techniques ($^1H-^1H$ COSY and $^1H-^{13}C$ HSQC). The changes of physical properties such as thermal stability, glass transition temperature and light transmittance after the hydrogenation were also investigated using TGA, DSC, and UV.

  • The dried needles (1.5 kg) of Abies koreana and Abies holophylla were ground, extracted with acetone-$H_2O$ (7:3, v/v), concentrated, and fractionated with a series of hexane, methylene chloride, ethyl acetate and water on a separatory funnel. Each fraction was freeze dried, then a portion of ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The isolated compounds were identified by cellulose TLC, $^1H$, $^{13}C-NMR$, COSY, HETCOR, FAB and EI-MS. The needles of Abies koreana and Abies holophylla contained a large amount of aromadendrin-7-O-${\beta}$-D-glucopyranoside (compound III), polydatin (compound VI), (-)-rhododendrol-2-O-${\beta}$-D-glucopyranoside (compound VII), in addition to a small amount of (+)-catechin (compound I), kaempferol-3-O-${\beta}$-D-glucopyranoside (compound IV), myricetin-3-O-${\beta}$-D-glucopyranoside (compound V), naringenin-7-O-${\beta}$-D-glucopyranoside (compound II). DPPH analysis was also tested to investigate the antioxidative effects on the isolated compounds and (+)-catechin and polydatin were effective.
