The majority of dyes belong to the chromophoric class known as donor-acceptor systems. The essential structural feature of such systems is the presence of one or more electron donating groups conjugated to one or more electron withdrawing groups via an unsaturated bridge. The pyrazolin-5-one system is an effective electron acceptor residue, and can also act as a weak electron donor. In our experiments, the various symmetrical and unsymmetrical H-chromophores were synthesized in the indoxyl, imidazo[1,2-a]pyridin-2-one, pyrazolin-5-one, and pyridin-2,6-dione residues, resulting in cross-conjugated donor-acceptor systems. And the visible light absorption was then associated with the migration of electron density from the donor region of the molecule to the acceptor region. Also, it was useful to prepare related non-cross-conjugated donor acceptor chromophores by combining these residues with other electron donor or acceptor moieties. For convenience such chromophores are referred to as merocyanines.

Bacteriophytochromes are phytochrome-like light-sensing photoreceptors that use biliverdin as a chromophore. To study the biochemical properties of the Deinococcus radiodurans bacteriophytochrome (DrBphP) protein, two anti-DrBphP mouse monoclonal antibodies (2B8 and 3H7) were generated. Their specific epitopes were identified in our previous report. We present here fine epitope mapping of these two antibodies by using truncation and substitution of original epitope sequences in order to identify minimized epitope peptides. The previously reported original epitope sequences for 2B8 and 3H7 were truncated from both sides. Our analysis showed that the minimal peptide sequence lengths for 2B8 and 3H7 antibodies were nine amino acids (RDPLPFFPP) and six amino acids (PGEIEE), respectively. We further characterized these peptides in order to investigate their reactivity after single deletion and single substitution of the original peptides. We found that single-substituted 2B8 epitope (RDPLPAFPP) and dual-substituted 3H7 epitope (PGEIAD) showed significantly increased reactivity. These two antibodies with high reactivity for the short modified peptide sequences are valueble for developing new peptide tags for protein research.
We carried out the melt copolymerization reactions of 1,2-bis(diethylamino)tetramethyldisilane with several aryldiols such as, 4,4'-biphenol, 4,4'-isopropylidenediphenol, 9H-fluoren-9,9-dimethanol, and 4,4'-(9-fluorenylidene) bis(2-phenoxyethanol) to afford poly[oxy(arylene)oxy(tetramethyldisilylene)]s containing fluorescent aromatic chromophore groups in the polymer main chain: poly[oxy(4,4'-biphenylene)oxy(tetramethyldisilylene)], poly[oxy{(4,4'-isopropylidene) diphenylene}oxy(tetramethyldisilylene)], poly[oxy(9H-fluorene-9,9-dimethylene) oxy(tetramethyldisilylene)], and poly[oxy{4,4'-(9-fluorenylidene)bis(2-phenoxyethylene)}oxy(tetramethyldisilnylene)]. These prepared materials are soluble in common organic solvents such as $CHCl_3$ and THF. The obtained polymers were characterized by several spectroscopic methods such as $^1H$, $^{13}C$, and $^{29}Si$ NMR. Further, FTIR spectra of all the polymers exhibited characteristic Si-O stretching frequencies at 1014-1087 $cm^{-1}$. These polymeric materials in THF showed strong maximum absorption peaks at 268-281 nm, strong maximum excitation peaks at 263-291 nm, and strong maximum fluorescence emission bands at 314-362 nm due to the presence of tetramethyldisilylene and several arylene chromophores in the polymer main chain. TGA thermograms indicated that most of the polymers were stable up to $200^{\circ}C$ with a weight loss of 3-16% in nitrogen.
Antitumor antibiotic HS-1 was purified from the culture broth of a streptomyces floridae SHS-1372 which had been isolated from soil, by solvent extraction, silica gel column chromatography and gel filtration. It was confirmed that HS-1 was active against gram positive bacteria and cancer cells(K562, P388, MCF-7, HT-29). Through the analysis of UV spectrum, melting point, IR spectrum, FAB-MS, $_{1}$H-NMR, $_{13}$C-NMR, 2D-NMR spectra, HS-1 could be identified sa the actinomycin X$_{2}$ antibiotics containing actinocine chromophore and peptides consisted of threonine, proline, methylvaline, sarcosine, D-valine and 4-ketoproline.
Spatially fixed bichromophoric systems with nonidentical chromophores have been extensively employed for studies of electron transfer and excitation transfer. Excitation hopping behavior between two naphthy1 moieties on 7,14-dihydro-7,14-ethanodibenz[a,h] anthracene(DEA)has been explored by the time-resolved fluorescence anisotropy measurements. The experimentally obtained value of the hopping rate in DEA agrees at least qualitatively with that calculated on the basis of the Dexter's theory, but disagrees with that calculated on the basis of the Forster's theory, indicates that for a pair of donor and acceptor with inter-chromophore separation as short as 4.5 $\AA$, excitation transfer via electron exchange is a predominant process.
Two polyacetylene compounds having diyn-ene chromophore were isolated from fresh Korean ginseng roots through solvent fractionation, partition and silica gel column chromatography. The low pressure semi-preparative liquid chromatography and high performance preparative liquid chromatography were used for final separation of polyacetylenic fractions. The chemical structures of these polyacetylenes were determined to be heptadeca-1,8-dien-4,6-diyn-3,10-diol and heptadeca-1,4-dien-6,8-diyn-3,10-diol by UV, FT-IR, $^1H\;NMR,\;^{13}C\;NMR,$ mass spectra and elemental analysis.
In high temperature ceramic glazes, a stable range of pink-red colors producing $Cr_2O_3-SnO_2-CaO-SiO_2$ pigments are factored by Cassiterite and Malayaite relationship with $Cr_2O_3$. The experiment described the effect of $CrCl_3$ by adding $H_3BO_3$ as a mineralizer to increase the formation of Malayaite crystal, substituting $CrCl_3$ instead of $Cr_2O_3$ in pigment as a chromophore. Synthesized pigments were analyzed by XRD, FT-IR, Raman Spectroscop, UV and UV-vis. The result shows the differences in amount of crystal phases and oxidation state of Cr ion, which causes the color change. The melting point of $CrCl_3$ is lower than $Cr_2O_3$ which act as a mineralizer and makes the pigment synthesized in lower temperature at $1200^{\circ}C$. Holding 3 h firing at $900^{\circ}C$ where the synthesize forms shows better effect of Malayaite crystal phases and increasing engaged effect of $CrCl_3$ where the color pigmentation is more defined then in $Cr_2O_3$.
The iLOV protein belongs to a family of blue-light photoreceptor proteins containing a light-oxygen-voltage sensing domain with a noncovalently bound flavin mononucleotide (FMN) as its chromophore. Owing to advantages such as its small size, oxygen-independent nature, and pH stability, iLOV is an ideal candidate over other reporter fluorescent proteins such as GFP and DsRed. Here, for the first time, we describe the feasibility of applying LOV domain-based fluorescent iLOV as a metal sensor by measuring the fluorescence quenching of a protein with respect to the concentration of metal ions. In the present study, we demonstrated the inherent copper sensing property of the iLOV protein and identified the possible amino acids responsible for metal binding. The fluorescence quenching upon exposure to Cu2+ was highly sensitive and exhibited reversibility upon the addition of the metal chelator EDTA. The copper binding constant was found to be 4.72 ± 0.84 µM. In addition, Cu2+-bound iLOV showed high fluorescence quenching at near physiological pH. Further computational analysis yielded a better insight into understanding the possible amino acids responsible for Cu2+ binding with the iLOV protein.
A novel fullerene derivative with photoresponsive azobenzene group was designed and synthesized, and its photoresponsive properties were reported. Starting from 4-nitrophenol, compound 1, which is containing fullerene moiety connected to azobenzene group through covalent linkage was synthesized by 5 steps. All the intermediates and the final compound were characterized by $^1H$, $^{13}C$-NMR, FAB-Mass or elemental analysis. Compound 1 exhibited the expected photoresponsive behavior. Chloroform solution($10^{-5}M$) of it served to maximize the absorption at 351 nm corresponding to the trans-azobenzene chromophore. Irradiation of this solution with 365 nm light resulted in photoisomerization to cis-azobenzene, as evidenced by decrease in the absorbance at 351 nm and an increase in absorbance at 450nm. A photostationary state was reached within about 150 s. Thermal reversion to the original spectrum was observed over the course of about 6 h at room temperature in the dark. However, exposure to bright sun light for about 5 s also effect almost complete reversion to the trans-isomer. This indicates that there is no strong steric influence on the trans-cis reversible isomerization of compound 1.
In high temperature ceramic glazes, a stable range of pink-red colors that produced $Cr_2O_3-SnO_2-CaO-SiO_2$ pigments were factored by Cassiterite($SnO_2$) and Malayaite($CaSnSiO_5$) by $Cr_2O_3$. The experiment examined the influence of $CrCl_3$, a Sn-Cr substitution added with a mineralizer ($H_3BO_3$), as a chromophore in pigments. The experiment also studied the effect of $H_3BO_3$ (2 wt%) when added to malayaite($CaSnSiO_5$) to see if the crystalline reaction will increase. $Cr_2O_3$ was also substituted with $CrCl_3$ in order to prove how much influence $CrCl_3$ had on the $H_3BO_3$. Malayaite and cassiterite were the basic compound materials and the experiment was conducted both with and without mineralizers (2 wt% of $H_3BO_3$). Each compound was synthesized at 800, 1000, 1200, 1300, 1400, $1500^{\circ}C$ for 2 h. Synthesized pigments were analyzed by XRD, FT-IR and UV-Vis. The temperature variation produced two crystal phases that showed the different engaging effects of Cr oxidation. $CrCl_3$ produced a better effect on the malayaite crystal phase, resulting in a more defined pigmentation of the pink-red coloration compared to $Cr_2O_3$.
