Chemical Composition and Fermentation Characteristics of Storage Sections of the Round Bale Silage of Fresh Rice Straw at Yonchon of Gyeonggi-do (연천지역에 있어서 생볏짚 원형곤포사일리지의 부위별 사료성분 및 발효품질)

  • The purpose of this study is to discuss the ways to evaluate the effectiveness of storage sections of the round bale silage of fresh rice straw (RS). This study evaluated, the changes of the fermentation characteristics and chemical composition of the different sectors of the RS after a certain period; a month later after the bailing, samples of three different RS (bale one, bale two, and bale three) were taken according to the three different sectors (top, center, and the bottom) of the bale. According to the findings of this study there was no significant difference in the RS's chemical composition content among the different sectors (top, center, or the bottom) of the bale. The possible reason for this is believed that on the basis of different sectors of the bale, the moisture (the water soluble nutrient) movement didn't occur and was locked in and couldn't escape, because the dry matter content of the rice straw was as high as $70\%$. After immediately harvesting the paddies, using the fresh rice straw to produce RS at the same time when the dry matter content is in between $26{\sim}40\%$, the chemical composition was found to be of a good quality. Accordingly, it is recommended that when the dry matter content is in between $35{\sim}45\%$, in order to produce RS with the fine chemical composition of 'Yonchon' region, bailing should be done simultaneously with the harvesting of paddies, before the first frost appears.

Characteristics and Genesis of Terrace Soils in Yeongnam Area -II. Physico-Chemical Properties (영남지역(嶺南地域)에 분포(分布)된 단구지토양(段丘地土壤)의 특성(特性)과 생성연구(生成硏究) -제(第) 2 보(報): 물리화학적(物理化學的) 특성(特性))

  • To clarify the soil characteristics and genesis of the clayey terrace soils distributed in Yeongcheon (inland region) and Yeongil (coastal region), 9 typifying pedons including a recent local deposits were studied. The physico-chemical properties found were summarized as follows; 1. The contents of available water of the terrace soils were ranged from 12.0 to 20.3%. They were increased with increaseing clay content up to 35%, and then decreased at clay content of 35 to 55%, but were maintained about 12 to 13% when the clay content exceeds 55%. 2. The amount of water stable aggregates in the surface soils were ranged from 55.0 to 81.1%, and were low in cultivated upland where water erosion were accelerated while high in the paddy where the double cropping were practiced. The index of aggregate in cultivated upland were higher in the surface soils but the paddy soils were higher in subsoils. 3. The colloidal reactions checked by the differences between pH in $0.01N-Na_2SO_4$ and $H_2O$ indicate exchangeable acidity to exchangeable neutrality, which means a sort of Red-yellow soil forming process had been prevailed during genesis of the terrace soils. 4. Cation exchange capacity of the clayey terrace soils were averaged about 15.8-20.2me/100g of fine earth and 35.6-52.6me/100g of clay. The rates of base saturation were 32.7 to 57.6% and the rates decreased with increasing the elevations of terraces. 5. The activities of free iron oxides were 0.061 to 0.739 and the values decreased with increasing elevation while the crystallinity index of iron oxides positively correlated with elevations the values ranged from 0.067 to 0.537.

  • In spite the fact that H. pylori infection might be the causative organisms of acute and chronic gastritis, peptic ulcer diseases and the definition as the class I carcinogen by WHO IARC, still debates exist about the relationship between H. pylori and gastric carcinogenesis. Epidemiological and animal studies demonstrated a link between gastric cancer and chronic infection with H, pylori, but the exact mechanism responsible for the development of gastric cancer in H. pylori-infected patients still remain obscure. In order to declare the clear association, definate evidences like that decrement in the incidence of gastric cancer after the eradication of H. pylori in designated area compared to noneradicated region or the blockade of specific mechanism acting on the carcinogenesis by H. pylori infection. The other way is to identify the upregulating oncogenes or downregulating tumor suppressor genes specifically invovled in H. pylori-associated carcinogenesis. For that, we established the animal models using C57BL/6 mice strain. Already gastric carcinogenesis was developed in Mongolian gerbils infected with H. pylori, but there has been no development of gastric cancer in mice model infected with H. pylori after long-term evaluation. Significant changes such as atrophic gastritis were observed in mice model. However, we could observe the development of mucosal carcinoma in the stomach of transgenic mice featuring the loss of TGF-beta sig naling by the expressions of dominant negative forms of type II receptor specifically in the stomach. Moreover, the incidence of gastric adenocarcinoma was significantly increased in group administered with both MNU and H. pylori infection than MNU alone, signifying that H. pylori promoted the gastric carcinogenesis and there might be host susceptibility genes in H. pylori-associated gastric carcinogenesis. Based on the assumption that chronic, uncontrolled inflammation might predispose to carcinogenesis, there have been several evidences showing chronic atrophic gastritis predisposed to gastric carcinogenesis in H. pylori infection. Although definite outcome of chemoprevention was not drawn after the longterm administration of anti-inflammatory drug in H. pylori infection, the actual incidence of atrophic gastritis and molecular evidence of chemoprevention could be obtained. Selective COX-2 inhibitor was effective in decreasing the development of gastric carcinogenesis provoked by H. pylori infection and carcinogen like in chemoprevention of colon carcinogenesis.

  • This investigation has been carried out to clarify structural architecture of cerebral neuroendocrine systems in the fifth instar lanra of cabbage butterfly Pieris rapae. In order to examine the cerebral neurosecretorv cell systems the brain and retrocerebral neuroendocrine complex were histochemically stained with the paraldehvde fuchsin. The brain of the fifth instar laMa contains three kinds of neurosecretorv cells: medial, lateral and tritocerebral neurosecretorv cells. The axon bundles of medial and lateral neurosecretory cells form medial neurosecretory pathway(MNSP) and lateral neurosecretorv pathwav(LNSP) within the brain respectively. Especially, prior to exiting the brain, the axon bundles of medial neurosecretorH cells located in both left and right cefebral hemispheres decussate in cerebral medial region and project to contralateral retrocerebral neuroendocrine complexes. Outside the brain the axon bundles of medial and lateral neurosecretory cells form the nenri corporis cardiaca(NCC) I and II respectively. The NCC I and ll run together to the retrocerebral neuroendocrine complex, forming the large nenre bundles in both left md right sides. The anon bundles of tritocerebral neurosecretory cells which pass through the brain along the tritocerebral neurosecretory pathway (TNSP) form the Ncc III outids the train. some of the Ncc I and it terminate in the corpus cardiacum, while the others pass through the corpus cardiacum without termination. The nerve bundle which passes the corpus cardiacum forms the nenrus corforis allatum(NCA) I which runs between the corpus cardiacum and the corpus allatum. Theyt are finally innervated to the corpus allatum. The Ncc III Projects to the corpus cardiRcum. However, most of NCC III priss through the corpus cardiacum without branching and then run down for another organ.

Regional differences in phylogenetic group of Escherichia coli strains isolated from children with urinary tract infection in Korea

  • Purpose: We phylogenetically analyzed the Escherichia coli strains isolated from children with urinary tract infection (UTI) in 2 regions of Korea. Virulence factors (VFs) and antibiotic resistance of the strains were also determined to compare the possible differences. Methods: A total of 138 E. coli strains were collected from the 2 regions; Gyeongin (78 strains) and Gyeongnam (60 strains). The phylogenetic groups were determined using the triplex polymerase chain reaction (PCR) method and multiplex PCRs were used to detect 7 VFs genes (fimH, papC, iutA, hlyA, sfa/focDE, afa/draBC, and kpsMT II). We also tested for antibiotic resistance. Results: Phylogenetic groups, B2 (61.6%) and D (26.8%), comprised the majority of all isolated strains. Regional comparisons revealed that more B2 strains and fewer non-B2 (A+B1+D) strains were found in Gyeongnam, than in the Gyeongin region (P=0.033), and certain VFs were predominantly detected in Gyeongnam (P<0.05). Neither regional nor phylogenetic differences, in antibiotic resistance of the strains, were significant. Conclusion: We were able to confirm that the geographic location is an important determinant of the distribution of the phylogenetic groups and VFs among the E. coli strains that cause UTI in children.

INTENSITY AND DOPPLER VELOCITY OSCILLATIONS IN PORE ATMOSPHERE

  • Due to the simple vertical structure of magnetic field, pores can be exploited to study the transport of mechanical energy by waves along the magnetic field to the chromosphere and corona. For a better understanding of physics of pores, we have investigated chromospheric traveling features running across two merged pores from their centers at the speed about 55 km s-1, in the active region AR 11828. The pores were observed on 2013 August 24 by using high time, spatial, and spectral resolution data from the Fast Imaging Solar Spectrograph (FISS) of the 1.6 meter New Solar Telescope (NST). We infer a LOS velocity by applying the bisector method to the Ca II $8542{\AA}$ band and $H{\alpha}$ band, and investigate intensity and the line-of-sight velocity changes at different wavelengths and different positions at the pores. We find that they have 3 minutes oscillations, and the intensity oscillation from the line center is preceded by that from the core ($-0.3{\AA}$) of the bands. There is no phase difference between the intensity and the LOS velocity oscillations at a given wavelength. The amplitude of LOS velocity from near the core spectra is greater than that from the far core spectra. These results support the interpretation of the observed wave as a slow magnetoacoustic wave propagating along the magnetic field lines in the pores. The apparent horizontal motion and a sudden decrease of its speed beyond the pores can be explained by the projection effect caused by inclination of the magnetic field with a canopy.

Production of Herbicide-resistant Transgenic Plants from Embryogenic Suspension Cultures of Cucumber (오이의 배발생 현탁 배양세포로부터 제초제 저항성 형질전환 식물체 생산)

  • To develop herbicide-resistant cucumber plants (Cucumis sativus L. cv Green Angle) embryogenic suspension cultures were co-cultured with Agrobacterium tumefaciens strain LBA4404 carrying a disarmed binary vector pGA-bar. The T-DNA region of this binary vector contains the nopalin synthase/neomycin phosphotransferase Ⅱ (npt Ⅱ) chimeric gene for kanamycin resistance and the cauliflower 35S/phosphinothricin acetyltransferase (bar) chimeric gene for phosphinothricin (PPT) resistance, After co-cultivation for 48 h, embryogenic calli were placed on maturation media containing 20 mg/L PPT. Approximately 200 putatively transgenic plantlets were obtained in hormone free media containing 40 mg/L PPT. Northern blot hybridization analysis confirmed the expression of the bar gene that was integrated into the genome of five transgenic plants. Transgenic cucumber plants were grown to maturity. Mature plants in soil showed tolerance to the commercial herbicide (Basta) of PPT at the manufacturer's suggested level (3 mL/L).

LIMK1/2 are required for actin filament and cell junction assembly in porcine embryos developing in vitro

  • Objective: This study was conducted to investigate the roles of LIM kinases (LIMK1 and LIMK2) during porcine early embryo development. We checked the mRNA expression patterns and localization of LIMK1/2 to evaluate their characterization. We further explored the function of LIMK1/2 in developmental competence and their relationship between actin assembly and cell junction integrity, specifically during the first cleavage and compaction. Methods: Pig ovaries were transferred from a local slaughterhouse within 1 h and cumulus oocyte complexes (COCs) were collected. COCs were matured in in vitro maturation medium in a CO2 incubator. Metaphase II oocytes were activated using an Electro Cell Manipulator 2001 and microinjected to insert LIMK1/2 dsRNA into the cytoplasm. To confirm the roles of LIMK1/2 during compaction and subsequent blastocyst formation, we employed a LIMK inhibitor (LIMKi3). Results: LIMK1/2 was localized in cytoplasm in embryos and co-localized with actin in cell-to-cell boundaries after the morula stage. LIMK1/2 knockdown using LIMK1/2 dsRNA significantly decreased the cleavage rate, compared to the control group. Protein levels of E-cadherin and β-catenin, present in adherens junctions, were reduced at the cell-to-cell boundaries in the LIMK1/2 knockdown embryos. Embryos treated with LIMKi3 at the morula stage failed to undergo compaction and could not develop into blastocysts. Actin intensity at the cortical region was considerably reduced in LIMKi3-treated embryos. LIMKi3-induced decrease in cortical actin levels was attributed to the disruption of adherens junction and tight junction assembly. Phosphorylation of cofilin was also reduced in LIMKi3-treated embryos. Conclusion: The above results suggest that LIMK1/2 is crucial for cleavage and compaction through regulation of actin organization and cell junction assembly.

Deriving the Rate Constants of Coal Char-CO2 Gasification using Pressurized Drop Tube Furnace (가압 DTF를 이용한 석탄 촤-CO2 가스화 반응상수 도출)

  • This study investigates the gasification of coal char by $CO_2$ under high pressures in a drop tube furnace(DTF). The rate constants are derived for the shrinking core model using the conventional method based on the set reactor conditions. The computational fluid dynamic(CFD) simulations adopting the rate constants revealed that the carbon conversion was much slower than the experimental results, especially under high temperature and high partial pressure of reactants. Three reasons were identified for the discrepancy: i) shorter reaction time because of the entry region for heating, ii) lower particle temperature by the endothermic reaction, and iii) lower partial pressure of $CO_2$ by its consumption. Therefore, the rate constants were corrected based on the actual reaction conditions of the char. The CFD results updated using the corrected rate constants well matched with the measured values. Such correction of reaction conditions in a DTF is essential in deriving rate constants for any char conversion models by $H_2O$ and $O_2$ as well as $CO_2$.

  • Zinc oxide is metal oxide semiconductor with the 3.37 eV bandgap energy. Zinc oxide is very attractive materials for many application fields. Zinc Oxide has many advantages such as high conductivity and good transmittance in visible region. Also it is cheaper than other semiconductor materials such as indium tin oxide (ITO). Therefore, ZnO is alternative material for ITO. ZnO is attracting attention for its application to transparent conductive oxide (TCO) films, surface acoustic wave (SAW), films bulk acoustic resonator (FBAR), piezoelectric materials, gas-sensing, solar cells and photocatalyst. In this study, we synthesized ZnO nanoparticles and defined their physical and chemical properties. Also we studied about the application of ZnO nanoparticles as a photocatalyst and try to find a enhancement photocatalytic activity of ZnO nanorticles.. We synthesized ZnO nanoparticles using spray-pyrolysis method and defined the physical and optical properties of ZnO nanoparticles in experiment I. When the ZnO are exposed to UV light, reduction and oxidation (REDOX) reaction will occur on the ZnO surface and generate O2- and OH radicals. These powerful oxidizing agents are proven to be effective in decomposition of the harmful organic materials and convert them into CO2 and H2O. Therefore, we investigated that the photocatalytic activity was increased through the surface modification of synthesized ZnO nanoparticles. In experiment II, we studied on the stability of ZnO nanoparticles in water. It is well known that ZnO is unstable in water in comparison with TiO2. Zn(OH)2 was formed at the ZnO surface and ZnO become inactive as a photocatalyst when ZnO is present in the solution. Therefore, we prepared synthesized ZnO nanoparticles that were immersed in the water and dried in the oven. After that, we measured photocatalytic activities of prepared samples and find the cause of their photocatalytic activity changes.
