Two New Marine Sponges of the Genus Halichondria (Halichondrida: Halichondriidae) from Uljin, Korea

  • Two new marine sponges, Halichondria jangseungenesis n. sp. and H. nagokenesis n. sp., of the family Halichondriidae, were collected from Uljin-gun, Gyeongsangbuk-do, Korea by SCUBA diving during the period from Apr 2007 to Aug 2007. Based on their spicule composition and skeletal structure, H. jangseungenesis n. sp. appears to have a close similarity with H. panicea (Pallas, 1766); however, they differ in length of spicule. The spicule length of oxea of H. jangseungenesis n. sp. is shorter than that of H. panicea. Based on their spicule composition and growth form, H. nagokenesis n. sp. is quite similar to H. cylindrata Tanita and Hoshino, 1989; however, but they differ in length of spicule. The spicule length of oxea of H. nagokenesis n. sp. is longer than that of H. cylindrata.

  • In order to treat the dyeing wastewater, the UV/TiO₂/H₂O₂ system was investigated, and proper pretreatment methods were examined to reduce the load on the system considering economical and technical efficiency. The results of this study were as follows: 1.UV/TiO₂/H₂O₂ system with pretreatment process was adopted, the result of Chemical coagulation and pH control units was pH 11→ coagulation → pH 4 and the optimum dosage of FeCl₃ was 600㎎/ℓ 2. Proper dosage of TiO₂in the UV/TiO₂/H₂O₂ system with pretreatment process was 2g/ℓ and H₂O₂ was 1000㎎/ℓ, UV contact time was 20min to get below 200㎎/ℓ of $COD_{Cr}$

Analysis of opposing histone modifications H3K4me3 and H3K27me3 reveals candidate diagnostic biomarkers for TNBC and gene set prediction combination

  • Breast cancer encompasses a major portion of human cancers and must be carefully monitored for appropriate diagnoses and treatments. Among the many types of breast cancers, triple negative breast cancer (TNBC) has the worst prognosis and the least cases reported. To gain a better understanding and a more decisive precursor for TNBC, two major histone modifications, an activating modification H3K4me3 and a repressive modification H3K27me3, were analyzed using data from normal breast cell lines against TNBC cell lines. The combination of these two histone markers on the gene promoter regions showed a great correlation with gene expression. A list of signature genes was defined as active (highly enriched H3K4me3), including NOVA1, NAT8L, and MMP16, and repressive genes (highly enriched H3K27me3), IRX2 and ADRB2, according to the distribution of these histone modifications on the promoter regions. To further enhance the investigation, potential candidates were also compared with other types of breast cancer to identify signs specific to TNBC. RNA-seq data was implemented to confirm and verify gene regulation governed by the histone modifications. Combinations of the biomarkers based on H3K4me3 and H3K27me3 showed the diagnostic value AUC 93.28% with P-value of 1.16e-226. The results of this study suggest that histone modification analysis of opposing histone modifications may be valuable toward developing biomarkers and targets for TNBC.

Study on Apoptosis Effect and Mechanism by Bojungikki-tang on Human Cancer Cell Line H460 (폐암세포주(肺癌細胞株) H460에 대(對)한 보중익기탕(補中益氣湯)의 세포고사효과(細胞枯死效果) 및 기전연구(機轉硏究))

  • Objectives : This study was designed to evaluate the effect on cytotoxicity of Bojungikki-tang(BIT) in human lung cancer H460 cells. Methods : BIT-induced cell death was confirmed as apoptosis characterized by chromatin condensation and increase of the $sub-G_1$, DNA content. It was tested whether the water extract of BIT affects the cell cycle regulators such as, p2l/Cipl, p27/Kipl, cyclin $B_1$. Results : The data showed that treatment of BIT decreased the viability of H460 cells in a dose-dependent manner. p2l/Cip1 is gradually decreased by the addition of the cells with BIT extract. Interestingly, p27/Kip1 is not detected for 24 hr after the addition of BIT extract, however, after 24 hr, p27/Kipl markedly increased. In addition, cyclin $B_1$, decreased in a time dependent manner after the addition of the water extract. The activation of caspase -3 protease was further confirmed by degradation of procaspase-8 protease andpoly(ADP-ribose) polymerase(P ARP) by BIT in H460 cells. Moreover, BIT induced the increase of Bak expression. Conclusion : These results suggest that the extract of BIT exerts anticancer effects to induce the death of human lung cancer H460 cells via down regulation of cell cycle regulators such as p2l/Cip1, and cyclin B1 or up regulation of cell cycle regulators such as p27/Kip1. Moerover results suggest that BIT induces an apoptosis in H460 cells via activation of intrinsic caspase cascades.

Production of L-Lactic Acid from Soluble Starch by Enterococcus sp. JA-27. (Enterococcus sp. JA-27에 의한 가용성 전분으로부터 L형 젖산의 생산)

  • Lactic acid bacteria with amylolytic and acid producing activities can ferment starch directly to lactic acid thereby producing a monomer for the production of biodegradable poly lactic acid (PLA). In this study, the strain producing L-lactic acid from soluble starch was isolated from Nuruk. The isolated strain was identified as Enterococcus sp. through its morphological, cultural, biochemical characteristics as well as the 16S rDNA sequence analysis, and named Enterococcus sp. JA-27. Enterococcus sp. JA-27 produced exclusively L-lactic acid from soluble starch as a carbon source. The optimal conditions for the maximum production of L-lactic acid from Enterococcus sp. JA-27 were 30 C, pH 8, 1.5 % soluble starch as a substrate and 3.5 % tryptone as a nitrogen source, 0.1 % $K_2$$HPO_4$, 0.04 % $MgSO_4$. $7H_2$O, 0.014 % $MnSO_4$$.$4$H_2O$, 0.004% $FeSO_4$$.$$7H_2$O. Batch and fed batch culture were carried out and the former was more effective. L-Lactic acid production in the optimum medium was significantly increased in a 7 L jar fermenter, where the maximum L-lactic acid concentration was 3 g/L. For the purification of lactic acid in fermented broth, two stage ionexchange column chromatographies were employed and finally identified by HPLC.

  • The h-index is a new tool for measuring research outputs based on citation and many variants of h-index have been proposed to improve the weaknesses of h-index. The h-index and its variants can be applied to institutional evaluation in three different ways. We worked out 447 practical cases of professors from 10 business schools and examined h-index and its variants. And then we compared three evaluation methods of research institution based on the h-index and its variants.
