Growth performance, carcass traits and gut health of broiler chickens fed diets incorporated with single cell protein

  • Objective: This study was conducted to evaluate single cell protein (SCP), produced from Methylococcus species, as a protein source on the growth performance, carcass traits and gut health of broiler chickens. Methods: Ten iso-nitrogenous and iso-caloric diets containing 0 (Control), 2.5%, 5%, 7.5%, and 10% SCP replacing either soybean meal (T1 to T5) or fish meal (T6 to T10) were formulated. Each diet prepared for starter (0 to 14 days), grower (15 to 21 days), and finisher (22 to 42 days) phases was offered to four replicates of 10 chicks each (n = 400). Growth performance at different phases and carcass characteristics and intestinal morphology on 42nd day of trial were measured. Results: Body weight gain in groups fed 2.5% and 5% SCP diets were comparable to control during different phases and cumulatively, however lower (p<0.01) in 7.5% and 10% SCP diets. Feed conversion ratio was better (p<0.01) in 2.5% and 5% SCP diets. Dressing percentage, abdominal fat percentage and meat:bone ratio were not affected (p>0.05) by SCP inclusion in the diets. However, breast percentage was higher (p<0.01) in 2.5% and 5% SCP groups and thigh percentage higher in 7.5% and 10% SCP groups. Total microbial count in duodenum, jejunum and ileum were not affected (p>0.05) by SCP inclusion up to 10% in diets. Duodenal villi length and crypt depth were highest (p<0.01) in group fed 5% SCP diets and lowest in group fed 10% SCP diets. Jejunal villi length and crypt depth as well as ileal villi length were lowest (p<0.01) in group fed 10% SCP diets. Body weight gain, feed consumption, feed conversion ratio and gut health were better (p<0.01) in broilers fed fish meal based diets compared to soybean meal based diets. Conclusion: It was concluded that inclusion of SCP up to 5% replacing soybean meal in broiler diets is beneficial in improving growth rate, breast yield and gut health status.

Dietary spray-dried plasma improves intestinal morphology of mated female mice under stress condition

  • Background: Stress causes inflammation that impairs intestinal barrier function. Dietary spray-dried plasma (SDP) has recognized anti-inflammatory effects and improvement of gut barrier function. Therefore, the purpose of this study was to investigate the effects of dietary SDP on intestinal morphology of mated female mice under stress condition. Results: Villus height, width, and area of small intestines were low on gestation day (GD) 3 or 4 under stress conditions, and higher later (Time, P < 0.05). Crypt depth of colon was low on GD 4 and higher later (Time, P < 0.05). Meanwhile, the SDP treatments improved (P < 0.05) intestinal morphology, indicated by increased villus height, villus width, villus area, and ratio between villus height and crypt depth of small intestines and crypt depth of colon, and by decreased crypt depth of small intestines, compared with the control diet. The SDP treatments also increased (P < 0.05) the number of goblet cells in intestines compared with the control diet. There were no differences between different levels of SDP. Conclusion: Dietary SDP improves intestinal morphology of mated female mice under stress condition.

  • 망상어, Ditrema temmincki 소화관의 형태 조직화학적인 특징은 다음과 같이 요약할 수 있다. 체장에 대한 식도에서 항문까지의 소화관 길이의 비 (RLG)는 0.89정도이며, 소화관에는 식도와 담관 입구사이의 팽창부인 위(胃)가 없다. 소화관은 형태 조직화학적인 특징에 의하여 식도, 식도 - 장이행부, 전장, 중장, 후장, 장 - 직장 이행부, 직장, 직장 - 항문 이행부, 항문으로 나눌 수 있다. 점막주름은 직장에서 가장 복잡한 형태를 보이며, 근육층은 식도와 항문에서 발달된 상태를 나타낸다. 점막주름의 상피층은 식도 전방부에서는 입방세포로 구성되며, 나머지 부위 에서는 원주상피로 구성된다. 소화관에는 PAS 반응에 양성을 나타내는 배상세포와 다당류 흡수세포 (Polysaccharid Absorptive Cell)가 관찰되는데, 후자는 다당류 계통의 영양물질의 흡수기능을 가진다. 망상어 소화관에서 다당류 계통의 영양물질은 주로 후장부에서 흡수된다.

Comparative Efficacy of Plant and Animal Protein Sources on the Growth Performance, Nutrient Digestibility, Morphology and Caecal Microbiology of Early-weaned Pigs

  • The present study was conducted to evaluate and compare the effects of various animal and plant protein sources on piglet' performance, digestibility of amino acids and gut morphology in weaned pigs until 28 days after weaning. The plant protein sources used were soybean meal (SBM), fermented soy protein (FSP), rice protein concentrate (RPC); and animal protein sources tested were, whey protein concentrate (WPC) and fishmeal (FM). Iso-proteinous (21%) diets were formulated and lysine (1.55%) content was similar in all the diets. The level of each protein source added was 6% by replacing SBM to the same extent from the control diet containing 15% SBM. The ADG was higher (p<0.05) in the groups fed animal proteins as compared with plant proteins at all the levels of measurement, except during 15-28 days. The highest ADG was noted in WPC and FM fed diets and lowest in SBM fed diet. The feed intake was higher in animal protein fed groups than plant proteins at all phases, but the feed:gain ratio was not affected by protein sources except during overall (0 to 14 day) measurement which was improved (p<0.05) in animal protein fed diets compared to plant protein sources. The digestibilities of gross energy, dry matter and crude protein were higher in animal protein fed groups than for plant protein fed sources. The apparent ileal digestibilities of essential amino acids like Leu, Thr, and Met were significantly (p<0.05) higher in animal proteins fed animals as compared with plant protein fed animals. But the apparent fecal digestibilities of essential amino acids like Arg and Ile were significantly higher (p<0.05) in plant protein diets than animal protein sources. The villous structure studied by scanning electron microscope were prominent, straight finger-like, although shortened and densely located in FM fed group as compared with others. The lactic acid bacteria and C. perfringens counts were higher in caecal contents of pigs fed plant proteins than the animal proteins. Overall, it could be concluded that animal protein sources in the present study showed better effects on growth performance, nutrient digestibility and gut morphology than plant protein sources.

Multi-carbohydrase application into energy and amino acid deficient broiler diets: A strategy to enhance performance of broiler chickens

  • The effect of Multi-Carbohydrase (MC) supplementation on growth performance, visceral organ weights, blood metabolites, jejunum morphology, nutrient digestibility, and carcass parameters of broiler chickens fed nutrient-deficient corn soybean-meal based diets containing high levels of non-starch polysaccharides from wheat and wheat by-products was investigated. A total of 378 one-day-old Ross 308 broiler chickens were randomly assigned to one of seven dietary treatments to give six replicates per treatment (nine birds per pen). Dietary treatments were as follows: (1) positive control (PC; commercial standard diet); (2) negative control 1 (NC-1; PC-120 kcal/kg metabolizable energy); (3) NC-2 (PC-3% standardized ileal digestibility [SID] amino acids). The remaining four dietary treatments were formulated with the addition of MC (MC; Superzyme-CSTM) into two negative controls along with two supplementation levels of MC (i.e., 0.025% and 0.05%, respectively). Improved body weight, average daily gain, and feed conversion ratio (p < 0.05) were observed in broiler chickens fed a reduced energy diet supplemented with MC compared to birds fed NC-1 diet from days 1-35. Additionally, birds fed a reduced energy diet with 0.05% MC showed comparable (p > 0.05) growth performance with birds fed PC for 35-day post-hatch. Furthermore, the addition of MC into reduced amino acid diets improved (p < 0.05) growth performance. Broiler chickens fed MC supplemented nutrient-deficient diets showed a greater (p < 0.05) villus height to crypt depth ratio than birds fed diets without MC on days 21 and 35. Similarly, improved (p < 0.05) nutrient digestibility was observed in birds fed reduced energy diets supplemented with MC compared to birds fed NC-1 on days 21 and 35. Our results suggest that MC supplementation into reduced energy or reduced amino acid diets containing wheat and wheat by-products has the potential to improve growth performance and nutrient digestibility while maintaining healthier gut morphology in broiler chickens from 1 to 35 days of age.

Effects of Benzoic Acid and Thymol on Growth Performance and Gut Characteristics of Weaned Piglets

  • A total of 144 weaned crossed pigs were used in a 42-d trial to explore the effects of different concentrations/combinations of benzoic acid and thymol on growth performance and gut characteristics in weaned pigs. Pigs were randomly allotted to 4 dietary treatments: i) control (C), basal diet, ii) C+1,000 mg/kg benzoic acid+100 mg/kg thymol (BT1), iii) C+1,000 mg/kg benzoic acid+200 mg/kg thymol (BT2) and, iv) C+2,000 mg/kg benzoic acid+100 mg/kg thymol (BT3). Relative to the control, pigs fed diet BT3 had lower diarrhoea score during the overall period (p<0.10) and improved feed to gain ratio between days 1 to 14 (p<0.05), which was accompanied by improved apparent total tract digestibility of ether extract, Ca and crude ash (p<0.05), and larger lipase, lactase and sucrose activities in the jejunum (p<0.05) at d 14 and d 42. Similarly, relative to the control, pigs fed diet BT3 had higher counts for Lactobacillus spp in digesta of ileum at d 14 (p<0.05), and pigs fed diets BT1, BT2, or BT3 also had higher counts of Bacillus spp in digesta of caecum at d 14 (p<0.05), and lower concentration of ammonia nitrogen in digesta of caecum at d 14 and d 42 (p<0.05). Finally, pigs fed diet BT3 had higher concentration of butyric acid in digesta of caecum at d 42 (p<0.05), and a larger villus height:crypt depth ratio in jejunum and ileum at d 14 (p<0.05) than pigs fed the control diet. In conclusion, piglets fed diet supplementation with different concentrations/combinations of benzoic acid and thymol could improve feed efficiency and diarrhoea, and improve gut microfloral composition. The combination of 2,000 mg/kg benzoic acid+100 mg/kg thymol produced better effects than other treatments in most measurements.

  • 2014년 9월부터 2015년 8월까지 국내 9개 지점에서 채집된 Anguilla japonica 454개체의 생물학적 특성을 조사한 결과, silvering index (SI) 구분과 GSI의 차이가 큰 pseudo-silver eel이 소양호에서 2개체(PS1~PS2)와 금강하구에서 1개체(PS3)가 확인되었다. Pseudo-silver eel은 외부형태 기준으로는 S2 단계였으나, GSI, 소화관지수 (Gut index) 및 안경지수 (Eye index)에서는 모두 yellow eel 단계인 Y1 및 Y2였고, 생식소발달단계와 난경에서도 Y2 단계였다.

Evaluation of glycerol encapsulated with alginate and alginate-chitosan polymers in gut environment and its resistance to rumen microbial degradation

  • Objective: To determine the effect of gut pH and rumen microbial fermentation on glycerol encapsulated in alginate and alginate-chitosan polymers. Methods: Glycerol was encapsulated at 2.5%, 5%, 7.5%, or 10% (w/w) with sodium alginate (A) and alginate-chitosan (AC) polymers. Surface morphology and chemical modifications of the beads were evaluated using scanning electron microscopy and Fourier transform infrared (FTIR) spectra. Encapsulation efficiency was determined at the 5% glycerol inclusion level in two experiments. In experiment 1, 0.5 g of alginate-glycerol (AG) and alginate-chitosan glycerol (ACG) beads were incubated for 2 h at $39^{\circ}C$ in pH 2 buffer followed by 24 h in pH 8 buffer to simulate gastric and intestinal conditions, respectively. In experiment 2, 0.5 g of AG and ACG beads were incubated in pH 6 buffer at $39^{\circ}C$ for 8 h to simulate rumen conditions. All incubations were replicated four times. Free glycerol content was determined using a spectrophotometer and used to assess loading capacity and encapsulation efficiency. An in vitro experiment with mixed cultures of rumen microbes was conducted to determine effect of encapsulation on microbial fermentation. Data were analyzed according to a complete block design using the MIXED procedure of SAS (SAS Institute, Cary, NC, USA). Results: For AG and ACG, loading capacity and efficiency were 64.7%, 74.7%, 70.3%, and 78.1%, respectively. Based on the FTIR spectra and scanning electron microscopy, ACG treatment demonstrated more intense and stronger ionic bonds. At pH 6, 36.1% and 29.7% of glycerol was released from AG and ACG, respectively. At pH 2 minimal glycerol was released but pH 8 resulted in 95.7% and 93.9% of glycerol released from AG and ACG, respectively. In vitro microbial data show reduced (p<0.05) fermentation of encapsulated glycerol after 24 h of incubation. Conclusion: The AC polymer provided greater protection in acidic pH with a gradual release of intact glycerol when exposed to an alkaline pH.

점농어 Lateorabrax sp. 초기 발육 단계에 있어서의 기아시 형태 변화-I. 후기 자어기 (Morphological Changes During Starvation in Early Developmental Stages of Spotted Sea Bass, Lateorabrax sp. I. Post-larval Stage)

  • 1996년 11월, 충청남도 보령군 보령화력발전소 내에 있는 실내 부화장에서 점농어의 수정란을 부화, 사육시키면서 절식 1일(부화후 6일째), 2일, 3일, 4일째와 연속 기아시 점농어 자어의 형태 변화 및 생존율에 미치는 영향을 조사하였다. 1. 점농어 자어는 부화 5일째부터 먹이를 먹기 시작하였으며, 무급이구는 부화 9일째 100%사망하였다. 부화 6일째(1일 기아)먹이를 투여하였던 실험구는 급이구와 마찬가지로 정상적인 성장을 하였으며, 7일째(2일 기아) 먹이를 투여한 실험구에서는 부화 후 9일째 5.3%의 생존율을 보였으나, 3~4일 기아구는 부화 10일에 100% 사망하였다. 2. 기아시 점농어 자어의 전장은 정상 먹이급여군보다 감소 성장하였다. 3. 정상 사육 개체와 기아 개체 사이의 체장에 대한 장높이(GH)와 근절높이(MH)의 비값이 다른 형태 형질에 비하여 가장 큰 변화 폭을 나타내었으며, 장높이/근절높이(GH/MH)도 큰 변화폭을 나타내어, 부화 9일 후 정상개체가 0.8, 기아 개체가 0.5이었다. 4. 기아 상태의 자어는 몸통 부분이 가늘어져서 휘어지며, 주둥이가 뾰족해졌고, 쇄골의 아래 끝이 튀어나왔다.

Growth and gut performance of young pigs in response to different dietary cellulose concentration and rearing condition

  • Objective: This experiment was conducted to investigate the effect of insoluble cellulose supplementation to diets on the growth performance, intestinal morphology, the incidence of diarrhea, nutrients digestibility, and inflammatory responses in altering environmental conditions of animals housing. Methods: A total of 108 male pigs (Duroc×[Yorkshire×Landrace]) were randomly allocated to one of three dietary treatments (cellulose 0%, 1%, 2%) and two environmental conditions (good sanitary condition vs. poor sanitary condition) to give 6 replicate pens per treatment with three pigs per each pen at 14 days post-weaning. Results: Pigs were in good sanitary condition had higher average daily gain (p<0.01) and improved feed efficiency (p<0.05) from day 1 to 14 after weaning compared to their counterparts. The interactions were found between environmental conditions and dietary treatments (day 7: crypt depth [p<0.01], villous height to crypt depth [p<0.001]; day 14: crypt depth [p<0.001], villous to crypt ratio [p<0.01]) in ileum morphology. Crypt depth was decreased (p<0.05), and villous to crypt ratio was increased (p<0.05) only in poor sanitary conditions. Pigs exposed to the good sanitary condition had higher (p<0.05) apparent ileal digestibility (day 7, gross energy; day 14, dry matter), apparent total tract digestibility (day 14, dry matter and crude protein) compared to pigs housed in the poor sanitary condition. Meanwhile, pigs fed a diet supplemented with 2% cellulose had decreased (p<0.05) apparent ileal digestibility (day 7, dry matter; day 14, crude protein), apparent total tract digestibility (day 7, dry matter; day 14, crude protein, gross energy) compared to pigs fed a diet supplemented with 0% or 1% cellulose. Conclusion: Our results indicated that a diet supplemented with 1% cellulose increased villous to crypt ratio, however feeding a diet containing cellulose (1% or 2%) impaired nutrient digestibility for 14 day after weaning in both good sanitary and poor sanitary conditions.
