To search for the antifungal substances, various actino-mycete isolates were obtained from various soils of Korea using plate dilution method on the humic acid vitamin agar plates. In the screening procedures using a dual culture method, 32 actionomycete isolates were selected, which showed the inhibitory activity against mycelial growth of plant pathogenic fungi Altirnaria mali, Colletotrichum gloeosporides, Fusarium oxysporum f.sp. cucumerinum, Magnaporthe grisea, Phytophthora capsici, and Rhizoctonia solani. Bioassay of the crude extracts from culture filtrates and mycelial mets revealed that 12 antagonistic actionomycetes produced highly active antifungal substances. Actinomycete strain S5-55 which showed the substantial antifungal activity against the tested fungi was selected for production of the antifungal substances. Based on the cytochemical and morphological characteristics, strain S5-55 was identified as a Streptomyces species. The results of the numerical identification using the TAXON program confirmed that Streptomyces strain S5-55 was identical with Streptomyces humidus including in TAXON major cluster 19. The production of antifungal substance was most favorable when S. humidus strain S5-55 was cultivated for 10 dats on soluble starch broth supplemented with $K_2$HPO$_4$. The antifungal substances active against the plant pathogenic fungi P. capsici and M. grisea were partially purified using $\textrm{C}_{18}$ reversed-phase column chromatography.
Thirty-four strains of bacteria were isolated from Phellodendron amurense. Using Nectria haematococca as an indicator strain, the best strain, B18, was obtained by the growth rate method. The morphological, physiological and biochemical characteristics of strain B18 and its 16S DNA gene sequence were identified, and the biocontrol effect of strain B18 was assessed in pot and field tests, as well as in a field-control test. Drilling methods were used to determine the antibacterial activity of metabolites from strain B18 and their effects on the growth of pathogen mycelia and spores. The best bacteriostatic rate was 85.4%. B18 can hydrolyse starch and oxidize glucose but does not produce gas; a positive result was obtained in a gelatine liquefaction test. According to 16S DNA gene sequencing, strain B18 is Bacillus methylotrophicus (GenBank accession number: MG457759). The results of pot and field-control trials showed 98% disease control when inoculating $10^8cfu/ml$ of the strain. The disease control effect of the B18 culture liquid (concentrations of $10^8$, $2{\times}10^6$, $10^6$, $5{\times}10^5$ and $2.5{\times}10^5cfu/ml$) in the field-control test was higher than 80%, and the cure rate of the original delivery solution was 96%. Therefore, in the practical forestry production, a $2.5{\times}10^5cfu/ml$ culture liquidshould be applied in advance to achieve good control effects.
The objective of this study is to demonstrate the ability of a computer simulation of microstructural evolution in hot forging of C-Mn steels. The development of microstructure is strongly dependent on process variables and metallurgical factors that affect time history of thermodynamical variables such as temperature, strain. and strain rate during deformation. Then finite element method is applied for the prediction of microstructural evolution, and it should be coupled with heat transfer analysis to consider the change of thermodynamical properties during forming process. In this study, Yada's recrystallization model and rigid-thermoviscoplastic finite element method are employed in order to analyze microstructural evolution during hot forging process. To show the validity and effectiveness of the proposed method, experiments are accomplished and the results of experiments are compared with those of simulations.
Photobiological hydrogen production by nitrogen-fixing unicellular cyanobacteria has long been considered to be an environmentally sound and very promising method for the future supply of renewable clean energy. Using six Korean nitrogen-fixing unicellular cyanobacterial strains and the Synechococcus sp. strain Miami BG043511 we performed cultivation experiments to find out the strain-specific optimal temperature for population growth and $H_2$ production. Under $20^{\circ}C$ the population growth of all the tested strains was significantly retarded in contrasts to the faster and higher growth under 25, 30 or $35^{\circ}C$. The highest growth rates in all the 7 strains were measured under $30^{\circ}C$ while the maximal biomass yields were under $30^{\circ}C$ (strains CB-MAL 026, 054, and 055) or $35^{\circ}C$ (strains 002, 031, 058, and Miami BG043511). The difference between the maximal biomass yields at $30^{\circ}C$ and $35^{\circ}C$ was not greater than 10%. The quantity of photobiologically produced $H_2$ was only slight larger under $35^{\circ}C$ than that under $20^{\circ}C$. Our result may suggest a two-step process of $H_2$ production which includes rapid and sizable production of biomass at $30^{\circ}C$ and the following high $H_2$ production at $20^{\circ}C$ by the test strains of marine nitrogen-fixing unicellular cyanobacteria.
Y2O3 thin films were deposited by reactive sputtering of Y target in Ar and O2 gas mixture. Residual stress was measrued by sin2$\psi$ method of x-ray diffraction (XRD) and growth orientation was examined by measuring the relative intensity of (400) plane and (222) plane of Y2O3 films. In the case that Y2O3 films were deposited at 40$0^{\circ}C$ and at low working pressure below 0.05 torr the film had large compressive stress and (111) plane orientation. At working pressure of about 0.10 torr the film had small compressive stress and (100) orientation. Above working pressure of 0.20 torr, the films had nearly zero stress and random orientation. In the case that the (111) oriented film deposited at low working pressure below 0.05 torr, as substrate temperature decreased, (111) orientation increased. In the case the film, with (100) orientation, deposited at working pressure of about 0.10 torr, (100) orientation increased with decresing substrate temperature. These relationship of residual stress and growth orientation can be explained by the relationship of surface energy and strain energy.
The characteristics of fatigue crack growth subject to out-of-plane bending fatigue are studied in terms of crack opening behavior by using pre-cracked smooth specimens. Crack opening stress is measured by an elastic compliance method which may precisely and continuously provide many date using strain gages during experiment. The results of the short crack and the long crack arranged by crack closure concept show that the effective stress gange ratio of short crack is grester than that of long crack, and ano- malous growth behavior of short crack may be elucidated by the variation of crack opening stress. When the variation of fatigue crack growth rate is arranged versus effective stress intensity factor range. Iinear relation is held also for the short crack. It shows that growth behavior of short crack can be quantitatively represent- ed by the fracture mechanics parameter using effective stress intensity factor range.
Purpose: The main purpose of this study was to identify the correlation between the child rearing attitudes and the maternal role strain of mother with infants. Method: This study is designed as a descriptive research study and the data was collected from 82 mothers and infants by means of an interview and questionnaire in a period from July 2002 to December 2002, when they came to screen their infant's growth and developmental state at a public health center. Result: The results of this study were as follows: There was a significant positive correlation between the child rearing attitudes and maternal role strain(r.=.53, p=.000). There was a significant difference between the father's employment state and child rearing attitude of mothers(t.=5.22, p<.000). There was a significant difference between male infant and female infants in maternal role strain(t.=3.8, p=0.04). Conclusion: When the child rearing attitude was positive, the subject's maternal role strain was high. Also further research is needed on social support or other factors in the subjects in child-rearing attitudes and maternal role strain.
Finite element simulations were conducted to investigate the influence of grain growth in the superplastic blow forming process. A microstructure-based constitutive model considering grain growth effects is proposed and used in the simulations. Also, a grain growth rate equation accounting for both static and dynamic grain growth is implemented. The simulations were made using a 2D plane-strain model for constrained blow forming and an axisymmetric model for free bulging. These two models showed different features during the forming stages. However, the forming pressure-time curve and the thickness distribution obtained by both simulations explained well the deformation hardening induced by the grain growth during superplastic forming. This study shows that grain growth is an important factor in determining the material behavior during superplastic deformation.
The objective of this paper is to examine the detection limit, growth characteristics and notch curvature radius in short crack problem. Measurement techniques such as ultrasonic method and back-face strain compliance method were adapted. The fatigue crack growth rate of the short crack is slower than that of a long crack for a notched specimen. The short crack is detected effectively by ultrasonic method. A short surface crack occurs in the middle of specimen thickness and is transient to a through crack when maximum crack depth is larger than the notch curvature radius.
This paper deals with an evaluation of the residual stress due to shot peening induced in a carburized gear tooth and its application to the fatigue crack propagation problem. A practical method is proposed on the basis of the assumption that the residual stress is caused by the difference of volume expansion in the case and the core, and the influence of both the reduction of retained austenite and the strain due to shot peening are considered. The evaluated residual stress is close to the measured stress, though the surface stress is rather overestimated. The stress intensity factor is computed by the influence function method, and it is shown that the factor is decreased by the residual stress in shot peened gear tooth. The shot peening is fairly effective to the reduction of fatigue crack growth rate. The crack propagation is simulated and the resistance due to shot peening is quantitatively demonstrated and discussed.
