Loliolide is one of the most ubiquitous monoterpenoid compounds found in algae, and its potential therapeutic effect on various dermatological conditions via agent-induced biological functions, including anti-oxidative and anti-apoptotic properties, was demonstrated. Here, we investigated the effects of loliolide on hair growth in dermal papilla (DP) cells, the main components regulating hair growth and loss conditions. For this purpose, we used a three-dimensional (3D) DP spheroid model that mimics the in vivo hair follicle system. Biochemical assays showed that low doses of loliolide increased the viability and size of 3D DP spheroids in a dose-dependent manner. This result correlated with increases in expression levels of hair growth-related autocrine factors including VEGF, IGF-1, and KGF. Immunoblotting and luciferase-reporter assays further revealed that loliolide induced AKT phosphorylation, and this effect led to stabilization of β-catenin, which plays a crucial role in the hair-inductive properties of DP cells. Further experiments showed that loliolide increased the expression levels of the DP signature genes, ALP, BMP2, VCAN, and HEY1. Furthermore, conditioned media from loliolide-treated DP spheroids significantly enhanced proliferation and the expression of hair growth regulatory genes in keratinocytes. These results suggested that loliolide could function in the hair growth inductivity of DP cells via the AKT/β-catenin signaling pathway.
Light exerts two primary roles in plant growth and development. Plants acquire all biochemical energy required for growth and propagation solely from light energy via photosynthesis. In addition, light serves as a medium through which plants recognize environmental fluctuations, such as photoperiod and presence of neighboring animals and plants. Plants therefore constantly monitor the direction, intensity, duration, and wavelength of environmental light and integrate these light signals into the intrinsic regulatory programs to achieve an optimized growth in a given light condition. Although light regulates all aspects of plant growth and developmental aspects, the molecular mechanisms and signaling cascades involved have not been well established until recently. However, recent advances in genetic tools and plant transformation techniques greatly facilitated the elucidation of molecular events in plant photomorphogenesis. This mini-review summarizes the gist of recent findings in deetiolation and suppression of shade avoidance response as classic examples of the phytochrome-mediated photomorphogenesis.

It has been shown that dysregulation of IGF-1 signaling is associated with tumor incidence and progression, whereas blockade of the signaling can effectively inhibit carcinogenesis. Although several mechanisms of anticancer activity of quercetin were proposed, molecular targets of quercetin have not been identified yet. Hence, we assessed the effect of quercetin on IGF-1 signaling inhibition in BK5.IGF-1 transgenic (Tg) mice, which over-expresses IGF-1 in the skin epidermis. A quercetin diet (0.02% wt/wt) for 20 weeks remarkably delayed the incidence of skin tumor by 2 weeks and reduced tumor multiplicity by 35% in a 7,12-dimethylbenz(a)anthracene (DMBA)-tetradecanoyl phorbol-13-acetate (TPA) two stage mouse skin carcinogenesis protocol. Moreover, skin hyperplasia in Tg mice was significantly inhibited by a quercetin supplementation. Further analysis of the MT1/2 skin papilloma cell line showed that a quercetin treatment dose dependently suppressed IGF-1 induced phosphorylation of the IGF-1 receptor (IGF-1R), insulin receptor substrate (IRS)-1, Akt and S6K; however, had no effect on the phosphorylation of PTEN. Additionally, the quercetin treatment inhibited IGF-1 stimulated cell proliferation in a dose dependent manner. Taken together, these data suggest that quercetin has a potent anticancer activity through the inhibition of IGF-1 signaling.

Leptin is a type of adipokine mainly produced by adipocytes and reported to be overproduced in prostate cancer. However, it is not known whether it stimulates the proliferation of prostate cells. In this study, we investigated whether benign prostatic hyperplasia epithelial cells (BPH-1 cells) infected with Trichomonas vaginalis induced the proliferation of prostate cells via a leptin signaling pathway. To investigate the effect of crosstalk between adipocyte leptin and inflamed epithelial cell in proliferation of prostate cells, adipocytes 3T3-L1 cells were incubated in conditioned medium of BPH-1 cells infected with T. vaginalis (T. vaginalis-conditioned medium, TCM), and then the adipocyte-conditioned medium (ATCM) was identified to cause proliferation of prostate cells. BPH-1 cells incubated with live T. vaginalis released pro-inflammatory cytokines, and conditioned medium of these cells caused migration of adipocytes. When prostate stromal cells and BPH-1 cells were incubated with adipocyte conditioned medium containing leptin, their growth rates increased as did expression of the leptin receptor (known as OBR) and signaling molecules such as JAK2/STAT3, Notch and survivin. Moreover, blocking the OBR reduced this proliferation and the expression of leptin signaling molecules in response to ATCM. In conclusion, our findings show that inflamed BPH-1 cells infected with T. vaginalis induce the proliferation of prostate cells through leptin-OBR signaling. Therefore, it is likely that T. vaginalis contributes to prostate enlargement in BPH via adipocyte leptin released as a result of inflammation of the prostate.

Forkhead box P3-positive (Foxp3+)-inducible Tregs (iTregs) are readily generated by TGF-β1 at low TCR signaling intensity. TGF-β1-mediated Foxp3 expression is further enhanced by retinoic acid (RA) and lactoferrin (LF). However, the intensity of TCR signaling required for induction of Foxp3 expression by TGF-β1 in combination with RA and LF is unknown. Here, we found that either RA or LF alone decreased TGF-β1-mediated Foxp3 expression at low TCR signaling intensity. In contrast, at high TCR signaling intensity, the addition of either RA or LF strongly increased TGF-β1-mediated Foxp3 expression. Moreover, decreased CD28 stimulation was more favorable for TGF-β1/LF-mediated Foxp3 expression. Lastly, we found that at high signaling intensities of both TCR and CD28, combined treatment with TGF-β1, RA, and LF induced robust expression of Foxp3, in parallel with powerful suppressive activity against responder T cell proliferation. Our findings that TGFβ/RA/LF strongly generate high affinity Ag-specific iTreg population would be useful for the control of unwanted hypersensitive immune reactions such as various autoimmune diseases.
SOCS-3와 IGF-I은 근육의 분화 과정 및 근비대 기전에 있어 매우 중요한 조절자 역할을 하는 유전자 및 성장인 자이며, 최근 골격근에서 IGF-I과 SOCS-3 유전자의 상호작용에 관한 연구의 필요성이 제기되고 있다. 본 연구에서는 C2C12 myotube에서 IGF-I이 SOCS-3 유전자 발현에 미치는 영향에 대해 알아보기 위해 4일간 분화시킨 C2C12 myotube에 IGF-I을 다양한 농도(0-200 ng/ml) 및 시간(3-72 시간)에 따라 처리하였다. 그 결과 IGF-I이 SOCS-3 유전자의 단백질 발현을 시간 의존적으로 유의하게 증가시켰으며, 3 시간에서 mRNA 발현을 증가시키고, 시간이 지남에 따라 긴 시간에서는 농도 의존적으로 발현이 감소하였음을 알 수 있었다. 또한 면역형광 염색을 통해 IGF-I이 myotube에서 SOCS-3의 단백질을 발현 시켰음을 뚜렷하게 관찰 할 수 있었다. 위 결과들을 바탕으로 본 연구에서는 IGF-I의 처리가 분화된 근육 세포인 C2C12 myotube에서 SOCS-3 유전자 발현에 유의한 영향을 미쳤음을 증명하였다. 이러한 결과는 선행연구에서 보고한 운동이 SOCS-3 유전자 발현을 증가시킴에 있어서 IGF-I이 중추적인 역할을 한 것으로 생각된다. 그러나 IGF-I에 의한 SOCS-3 유전자 발현 조절 기전에 있어 관련 신호 전달체계 및 골격근 관련 유전자 발현에 미치는 영향에 관한 연구는 보다 더 이루어져야 할 것이라 사료된다.

Objective: Discovering the mechanism of cell specification is important to manipulate cellular lineages. To obtain lineage-specific cell lines, the target lineage needs to be promoted, and counterpart lineages should be suppressed. Embryos in the early blastocyst stage possess two different cell populations, the inner cell mass (ICM) and trophectoderm. Then, cells in the ICM segregate into epiblasts (Epi) and primitive endoderm (PrE). PrE cells in embryos show specific expression of platelet-derived growth factor (PDGF) and its receptor, PDGF receptor A (PDGFRA). In this study, we suppressed PDGF signaling using two methods (CRISPR/Cas9 injection and inhibitor treatment) to provide insight into the segregation of embryonic lineages. Methods: CRISPR/Cas9 RNAs were injected into parthenogenetically activated and in vitro fertilized embryos. The PDGF receptor inhibitor AG1296 was treated at 0, 5, 10, and 20 µM concentration. The developmental competence of the embryos and the number of cells expressing marker proteins (SOX2 for ICM and SOX17 for PrE) were measured after the treatments. The expression levels of the marker genes with the inhibitor were examined during embryo development. Results: Microinjection targeting the PDGF receptor (PDGFR) A reduced the number of SOX17-positive cell populations in a subset of day 7 blastocysts (n = 9/12). However, microinjection accompanied diminution of Epi cells in the blastocyst. The PDGF receptor inhibitor AG1296 (5 µM) suppressed SOX17-positive cells without reducing SOX2-positive cells in both parthenogenetic activated and in vitro fertilized embryos. Within the transcriptional target of PDGF signaling, the inhibitor significantly upregulated the Txnip gene in embryos. Conclusion: We identified that PDGF signaling is important to sustain the PrE population in porcine blastocysts. Additionally, treatment with inhibitors was a better method to suppress PrE cells than CRISPR/Cas9 microinjection of anti-PDGF receptor α gene, because microinjection suppressed number of Epi cells. The PDGF receptor might control the number of PrE cells by repressing the proapoptotic gene Txnip. Our results can help to isolate Epi-specific cell lines from blastocysts.
막내골화와 연골내골화 등의 정상적인 골격 성장은 섬유아세포 성장인자 (FGF) 와 이들 수용체들 (FGFR) 에 의한 신호 전달체계에 의해 조절된다. 또한 전사조절인자인 Runx2 ($Cbfa1/Pebp2{\alpha}A/AML3$) 는 골아세포분화 뿐만 아니라 골형성에도 필수적인 유전자로 알려져 있다. FGF signaling이 mouse의 두개관과 하악에서의 연골 형성에 어떤 영향을 미치고 있으며, 이 과정에서 Runx2의 연관성을 알아보고자 in vivo 및 in vitro 실험을 시행하여 다음과 같은 결과를 얻었다. 두개관과 하악을 Alcian blue 염색한 결과 시상두개봉합부 연골은 태생 16일부터, Meckel's 연골은 태생11일부터 형성되기 시작하였다. 이들 연골세포들의 성상을 알아보기위한 in situ hybridization 결과 시상두개봉합부 연골 및 Meckel's 연골 모두에서 type II collagen은 발현되었으나, Type X collagen은 발현되지 않았다. Runx2 mRNA는 시상두개봉합부 연골과 Meckel's 연골 모두에서 발현되지 않았지만, 이들 연골들의 가장자리를 둘러싸고 있는 독특한 발현양상을 나타내었다. 두개봉합부에서의 FGF2 protein의 국소적 적용은 두개봉합부 하방의 연골형성을 억제하였다. 또한 하악의 Meckel's 연골 발생 부위에 FGF2 protein의 국소적 적용 역시 Meckel's 연골의 형성을 억제하였다. FGF2 protein은 시상두개봉합부상의 bead 주위로 Runx2의 발현을 유도하였다. 이 결과들을 종합해볼 때, FGF signaling은 골아세포와 연골아세포가 공존하고 있는 조직에서의 연골 형성을 억제하고 있음을 시사해 주고 있으며, 이 과정에서 FGF signaling은 부분적으로 Runx2 유전자의 발현을 조절하므로써 연골세포의 증식과 분화에 관여할 것으로 사료된다.
식물의 생활사 동안 물부족 스트레스는 식물의 생장과 발달에 해로운 영향을 끼치는 대표적인 스트레스이다. 유용 작물의 생산성을 증가시키기 위하여, 물부족 스트레스를 극복하는 일은 식물학 연구 분야에서 가장 중요한 이슈로 대두되어 왔다. 식물의 호르몬 중에서 앱시스산은 물부족 스트레스에 대해 식물이 저항성을 나타내는데 가장 중요한 호르몬으로서 역할을 수행하며, 씨앗의 발아, 기공의 개폐, 유묘의 성장과 같은 다양한 발달 과정에도 관여하고 있다. 그러므로, 앱시스산에 의해 매개되는 식물의 신호전달 기작을 명확히 이해하는 것은 물부족 스트레스에 대한 내성을 갖는 유용 식물을 생산해내기 위해 가장 효과적인 방법이 될 것이다. 한편, 인산화, 유비퀴틴화와 같은 번역 후 변형 과정은 식물이 다양한 환경적 스트레스 하에서 신속하게 적응을 하기 위해 가장 효율적인 기작으로 인식되어 왔는데, 이는 전사수준에서의 조절과 달리 이미 존재하는 신호전달 물질의 활성과 안정성을 직접적으로 빠르게 조절할 수 있기 때문이다. 본 총설에서는 앱시스산의 신호전달 과정과 관련된 최근 연구 동향을 업데이트하고자 하며, 특히 이러한 신호전달 과정을 앱시스산 수송, 인식, 신호전달 및 번역 후 변형 과정에 초점을 맞추어 알아보고자 한다. 또한 그러한 조절 기작이 농업분야에서 유용 작물을 생산하는데 어떻게 적용될 수 있는지에 대한 향후 전망에 대해서도 기술하고자 한다.
본 연구에서는 고혈당으로 인해 유발되는 당뇨병성 신장병증의 대표적인 증상인 사구체 경화증을 완화시키는 nobiletin의 효능에 대해 알아보고자 하였다. 신장 세포인 HRMC를 이용하여 고혈당에서의 세포외 기질 축적 단백질의 발현과 경화에 관여하는 신호 전달 억제 효능을 확인한 결과 nobiletin은 고혈당의 자극에 의해 증가하는 섬유화 단백질인 collagen IV, fibronectin 그리고 CTGF의 발현을 억제하였으며, 여기에 관여하는 TGF-β1-Src-caveolin-1 신호 전달 경로를 통해 사구체 경화증을 억제하는 것을 확인하였다. 따라서 nobiletin은 고혈당으로 유도된 당뇨병성 신장병증에 있어 사구체 경화증을 예방하는 기능성 성분으로서의 활용 가능성을 확인하였다.
