Transforming Growth $Factor-{\beta}$ Enhances Tyrosine Phosphorylation of Two Cellular Proteins in HEL Cells

  • Transforming growth $factor-{\beta}\;(TGF-{\beta})$ is a multifunctional polypeptide that exerts biological roles including cell proliferation, differentiation, extracellular matrix deposition and apoptosis in many different cell types. $TGF-{\beta}$, although known as a negative growth regulator, has not been tested in human embryo lung (HEll cells. This study attempts to understand the role of $TGF-{\beta}$ on growth control of HEL cells in relationship to tyrosine phosphorylation pattern of cellular proteins. In density-arrested HEL cells treated with $TGF-{\beta}$, analysis of Western immunoblot showed induction of tyrosine phosphorylation of two major cellular proteins (15 kDa and 45 kDa). In normal proliferating HEL cells with different concentrations of serum, further analysis indicated that the increase in tyrosine phosphorylation of a 45 kDa protein was regulated in serum concentration-dependent manner. However, in proliferating HEL cells treated with $TGF-{\beta}$, tyrosine phosphorylation of 45 kDa was down-regulated. Calcium involvement in the regulation of tyrosine phosphorylation of 45 kDa and 15 kDa proteins was also examined. Tyrosine phosphorylation of 15 kDa protein but not of 45 kDa protein was regulated by exogenous calcium. The level of tyrosine phosphorylation of 15 kDa protein was low at reduced caclium concentration and high at elevated caclium concentration. $TGF-{\beta}$ reversed the pattern of tyrosine phosphorylation of 15 kDa protein. These results suggest that tyrosine phosphorylation of 45 and 15 kDa proteins in HEL cells may be controlled depending on the physiological status of the cells, i.e., low in arrested cells and high in proliferating cells. And the tyrosine phosphorylation of the two proteins appears to be down- or up-regulated by $TGF-{\beta}$.

  • In Erhualian and Yorkshire reciprocal cross $F_1$ pig populations, we examined the mRNA expression characteristic of liver-derived IGF-1, IGF-1R, IGF-2, IGF-2R and IGFBP-3 during the embryonic and postnatal developmental periods (E50, E70, E90, D1, D20, D70, D120 and D180). Our results demonstrated that the IGF-system genes mRNA levels exhibited an ontogenetic expression pattern, which was potentially associated with the porcine embryonic development, postnatal growth, organogenesis and even the initiation and acceleration of puberty. The expression pattern of IGF-system genes showed variation in the reciprocal cross ($F_1$ YE and EY pigs). This study also involved the expression features of imprinted genes IGF-2 and IGF-2R. The parent-of-origin effect of imprinted genes was reflected by their differential expression between the reciprocal crosses populations. The correlation analysis also indicated that the regulatory network and mechanisms involved in the IGF system were a complex issue that needs to be more fully explored. A better understanding of IGF system components and their interactive mechanisms will enable researchers to gain insights not only into animal organogenesis but also into somatic growth development and even reproduction.

Sidewall effect in a stress induced method for Spontaneous growth of Bi nanowires

  • Single-crystalline Bi nanowires have motivated many researchers to investigate novel quasi-one-dimensional phenomena such as the wire-boundary scattering effect and quantum confinement effects due to their electron effective mass (~0.001 me). Single crystalline Bi nanowires were found to grow on as-sputtered films after thermal annealing at $270^{\circ}C$. This was facilitated by relaxation of stress between the film and the thermally oxidized Si substrate that originated from a mismatch of the thermal expansion. However, the method is known to produce relatively lower density of nanowires than that of other nanowire growth methods for device applications. In order to increase density of nanowire, we propose a method for enhancing compressive stress which is a driving force for nanowire growth. In this work, we report that the compressive stress can be controlled by modifying a substrate structure. A combination of photolithography and a reactive ion etching technique was used to fabricate patterns on a Si substrate. It was found that the nanowire density of a Bi film grown on $100{\mu}m{\times}100{\mu}m$ pattern Si substrate increased over seven times higher than that of a Bi sample grown on a normal substrate. Our results show that density of nanowire can be enhanced by sidewall effect in optimized proper pattern sizes for the Bi nanowire growth.

Effect of Salinity and Micronutrients on the Growth Rate of Bloom from Heterosigma akashiwo (Raphidophyceae) (염분과 미량원소가 Heterosigma akashiwo (Raphidophyceae) 성장률에 미치는 영향)

  • In order to understand the Heterosigma akashiwo (Raphidophyceae) bloom, we investigated the cell division pattern of its strains under various physicochemical conditions from three different origin. Among the three tested strains, HYM06HA and NFHTS-AK-1, are belong to South Korea and CCMP452 strain was from USA. Interestingly among three strains, HYM06HA was established from the cyst in Korea. Our study results showed that these three strains were indistinguishable under various salinity and iron (Fe) conditions. All strains were survived at 10 to 40 psu and the growth patterns under different iron (Fe) concentrations were observed to be similar. The growth patterns under different N:P ratios and selenium (Se) concentrations led to classification of strains into Korean and USA strain. Briefly these results indicated that the growth pattern from different strains of H. akashiwo could be dominant using their salinity tolerance. Furthermore nutritional enhancer seemed to be different based on geographical origin.

Population Structure, and Emergence and Growth Dynamics of Seedling, and Spatial Distribution of Dendropanax morbifera Lev.(Araliaceae) (황칠나무의 집단구조와 치수의 발생과 생육동태 및 공간분포)

  • A Korean endemic and evergreen small tree ' Dendropanax morbifera $L_{EV}$.(Araliaceae)' is a component of evergreen forest and mainly idstributein sourthern region and islands in Korea. A local population of D. morbifera which is located between evergreen and deciduous forest within 50m x 50m quadrate was investigated to ascertain the change of population structure, emergence and growth dynamics of seedlings and saplings, and pattern of spatial distribution by the temproal and spatial expansion of population . The result of analysis of population structure by Importnace Value(IV), evergreen forest showed a high species diversity of evergreen tree species such as Cinnamomum japonicum, Machilus japonica, Neolitsea serica, Daphniphyllum macropodum, Ligustrum japonicum, and etc, in middle and under story than in upper story where Camelia japonica and Quercus acuta were dominant. And in conterminous deciduous fores, the major component of evergreen forest in this region, Camellia japonica, Quercus acuta, evergreen tree of Lauraceae and etc. were abundant in only under story. IV of D. morbifera differed from among three story. In comparative analysis of emergence and growth dynamics of D. morbifera seedlings and saplings between evergreen and deciduous forest, emergece and density of seedlings were significantly greater in evergreen than in deciduous forest, and growth of height and basal diameter of seedlings and saplings were slightly larger in evergreen than in deciduous forest. The spatial distribution patterns by Moristia's index mapping of indivuduals using a lattice method of XY axis within this population showed that seedlings(age up to 2 years) and saplings (age>2 years and height<1m) both evergreen and deciduous forest were more or less aggregated apart from mature trees, and thougth intermediate trees(height>1m and dbh<10cm) had a aggregated distribution pattern, mature trees(dbh>10cm were uniform. In conclusion , the expansion of D. morbfera population from evergreen to deciduous forest accompanied with a mumber of evergreen woody species, and also, emergence and recruitment, and growth of seedlings were greatly influenced moisture and canopy by around community structure.

  • In order to learn the growth pattern of high yielding paddy and the effect of slaked lime and potassium on the growth pattern, a relationship between the number of panicles and tillers at different growth stages, as well as the effect of slaked lime and potassium on the increase of tillers were studied with three pot and one field experimental results. 1. The number of tillers at early stages of growth has little or negative correlation with the number of panicles. However. the correlation grows positively as the growth stage proceed and become to highly significant from the stage closing to the panicle formation. 2. Potassium is effective on increasing tillers and calcium on decreasing them. The above contradictory effect of potassium and calcium would practically be an important point for the establishment of high yielding technics of paddy, which be the one to be studied from the view point of plant physiology and soil chemistry. 3. The negative effect of calcium on tillering also seemed to be attributed to the pH rise of the media.

Comparisons of Growth, Mammary Development, Serum Proteins and Serum Concentrations of Estradiol, Growth Hormone and Prolactin between Korean Native and Holstein Heifers (한우와 홀스타인 암송아지의 성장, 유선발달, 혈청 단백질과 혈청 중 Growth Hormone, Prolactin 및 Estradiol의 농도 비교)

  • Growth, mammary development, serum concentrations of growth hormone, prolactin and estradiol which affect the growth and mammary development, were investigated in Korean Native heifers and Holstein heifers from 5 months old to 15 months old. From 5 months old to 15 months old, body weights in Korean Native heifers averaged 37%, lower than those in Holstein heifers of the same age (P<.01). Teat volumes in Korean Native heifers are average 67% smaller than those in Holstein heifers. Analysis of serum protein by secondary electrophoresis showed differences in protein pattern between the breeds and between the individuals. From 5 months old to 15 months old, serum concentration of growth hormone in Korean Native heifers was averge 47% lower than that in Holstein (P<.01), yet prolactin concentrations in serum were about the same between the two breeds. However serum concentration of estradiol in Korean Native heifers was average 20% higher than that in Holstein of same age (P<.01). This result showed evidently that the decrease of the growth hormone concentration in serum, while growing up, causes to retard the growth and mammary development in Korean Native heifers compared to Holstein heifers.

A study on the correlation between airway space and facial morphology in Class III malocclusion children with nasal obstruction (비폐쇄를 보이는 III급 부정교합아동의 기도 공간 형태와 안모 골격 형태와의 상관관계 연구)

  • Objective: The aim of this study was assessment of the relationship between airway space and facial morphology in Class III children with nasal obstruction. Methods: For this study, 100 Class III children (50 boys and 50 girls) were chosen. All subjects were refered to ENT, due to nasal obstruction. Airway space measurements and facial morphology measurements were measured on lateral cephalometric radiograph. Pearson correlation analysis was used to assess the relationship between airway space and facial morphology Results: Ramal height, SNA, SNB, PFH, FHR and facial plane angle were positively related to upper PAS, and sum of saddle angle, articular angle, and genial angle, SN-GoGn, Y-axis to SN and FMA negatively related to upper PAS. Genial angle, FMA were positively related to lower PAS, and articular angle, facial depth, PFH and FHR negatively related to lower PAS. PCBL, ramal height, Mn. body length, Mn. body length to ACBL, facial depth, facial length, PFH and AFH were positively related to tonsil size. Sum of saddle angle, articular angle, genial angle, facial length, AFH, FMA and LFH were positively related to tongue gap, and IMPA and overbite was negatively related to tongue gap. Upper PAS, related to size of adenoid tissue, was mainly related to posterior facial dimension following a vertical growth pattern of face and mandibular rotation. Lower PAS and tonsil size, related to anterior-posterior tongue base position, were significantly related to each other. Lower PAS was related to growth pattern of mandible, and tonsil size was related to size of mandible and horizontal growth pattern of face. Tongue gap was related to anterior facial dimension following a vertical growth pattern of face. Conclusion: Significant relationship exists between airway space and facial morphology.

IRFP-tree: Intersection Rule Based FP-tree (IRFP-tree(Intersection Rule Based FP-tree): 메모리 효율성을 향상시키기 위해 교집합 규칙 기반의 패러다임을 적용한 FP-tree)

  • For frequency pattern analysis of large databases, the new tree-based frequency pattern analysis algorithm which can compensate for the disadvantages of the Apriori method has been variously studied. In frequency pattern tree, the number of nodes is associated with memory allocation, but also affects memory resource consumption and processing speed of the growth. Therefore, reducing the number of nodes in the tree is very important in the frequency pattern mining. However, the absolute criteria which need to order the transaction items for construction frequency pattern tree has lowered the compression ratio of the tree nodes. But most of the frequency based tree construction methods adapted the absolute criteria. FP-tree is typically frequency pattern tree structure which is an extended prefix-tree structure for storing compressed frequent crucial information about frequent patterns. For construction the tree, all the frequent items in different transactions are sorted according to the absolute criteria, frequency descending order. CanTree also need to absolute criteria, canonical order, to construct the tree. In this paper, we proposed a novel frequency pattern tree construction method that does not use the absolute criteria, IRFP-tree algorithm. IRFP-tree(Intersection Rule based FP-tree). IRFP-tree is constituted with the new paradigm of the intersection rule without the use of the absolute criteria. It increased the compression ratio of the tree nodes, and reduced the tree construction time. Our method has the additional advantage that it provides incremental mining. The reported test result demonstrate the applicability and effectiveness of the proposed approach.

  • The principle of dark field imaging is comprehensively discussed using real images of dislocations, stacking faults and gettering phenomena due to defects obtained by Cz Si wafers and LEC semi-insulating GaAs crystals. Resulution of dark field imaging is improved by Fourier transformation of Fraunhofer diffraction pattern obtained at an out-of focusing position of an objective lens.
