Isolation and In Vitro Culture of Vascular Endothelial Cells from Mice

  • In cardiovascular disorders, understanding of endothelial cell (EC) function is essential to elucidate the disease mechanism. Although the mouse model has many advantages for in vivo and in vitro research, efficient procedures for the isolation and propagation of primary mouse EC have been problematic. We describe a high yield process for isolation and in vitro culture of primary EC from mouse arteries (aorta, braches of superior mesenteric artery, and cerebral arteries from the circle of Willis). Mouse arteries were carefully dissected without damage under a light microscope, and small pieces of the vessels were transferred on/in a Matrigel matrix enriched with endothelial growth supplement. Primary cells that proliferated in Matrigel were propagated in advanced DMEM with fetal calf serum or platelet-derived serum, EC growth supplement, and heparin. To improve the purity of the cell culture, we applied shearing stress and anti-fibroblast antibody. EC were characterized by a monolayer cobble stone appearance, positive staining with acetylated low density lipoprotein labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethyl-indocarbocyanine perchlorate, RT-PCR using primers for von-Willebrand factor, and determination of the protein level endothelial nitric oxide synthase. Our simple, efficient method would facilitate in vitro functional investigations of EC from mouse vessels.

  • Src-family kinases are critically involved in the control of cytoskeleton organization and in the generation of integrin-dependent signaling responses, inducing tyrosine phosphorylation of many signaling and cytoskeletal proteins. Activity of the Src family of tyrosine kinases is tightly controlled by inhibitory phosphorylation of a carboxy-terminal tyrosine residue, inducing an inactive conformation through binding with its SH2 domain. Dephosphorylation of C-ter tyrosine, as well as its deletion of substitution with phenylalanine in oncogenic Src kinases, leads to autophosphorylation at a tyrosine in the activation loop, thereby leading to enhanced Src activity. Beside this phophorylation/dephosphorylation circuitry, cysteine oxidation has been recently reported as a further mechanism of enzyme activation. Mounting evidence describes Src activation via its redox regulation as a key outcome in several circumstances, including growth factor and cytokines signaling, integrin-mediated cell adhesion and motility, membrane receptor cross-talk as well in cell transformation and tumor progression. Among the plethora of data involving Src kinase in physiological and pathophysiological processes, this review will give emphasis to the redox component of the regulation of this master kinase.

SOME EVIDENCE REGARDING REPAIRING, RECOVERY AND OVER-COMPENSATING PROCESSES DURING ONTOGENESIS, AFTERX-RAY-IRRADTATION OF BEAN SEEDS

  • Exposing plant organs to high doses of ionizing irradiation, penetrating into the plant tis-sues and cells, along the track structure of particles, lesions, and sublesions are formed on the molecules and organelles. As a result, disorders in the growth and development as well as chlorophyll-deficiency symptoms occur. The time scale of their reparation, recovery and over compensation during ontogenesis, constitutes a question of high theoretical and practical importanced, with special regard to nuclear fallout. With an aim to model the “ut supra”stated phenomena, the seeds of bean, Echo elit licensed variety, were irradiated by 300 Gy dose of X-ray-irradiation (120 kV:4.5 mA). According to the data obtained, the biosynthesis of photosynthetic pigments, will have been completed by the beginning of flowering. In consequence of the overcompensation of the repairing processes, the organs of plants developed from irradiated seeds, showed a partly differing correlative growth, compared to those of control plants. In order to characterize the vivo response of radiation-injured plants, a new method and approach were used. The changes of the electric capacitance of the plants during their ontogenesis, were continously monitored and recorede via a computer-aided and controlled measurement. In view of the data collected in such a way, the repairing plants may respond more quickly and intensively to the changes of environmental factors.

  • Exposure to a traumatic events gives people many post traumatic conditions resulting Post Traumatic Stress Disorder (PTSD) or Post Traumatic Growth (PTG). Some of them may come up with acute Stress Disorder and some may having with grievances. But most of people overcome their traumatic condition with using their Emotional Intelligence and Resilience capacities. This article is focused on indicating basic mechanisms and resources in which can be lead to have better social rehabilitation along with the matters that can be important in trauma coping. The later part of the article appeals the concept of social work theory highlighting the psych dynamic approach which can be impact positively on psycho social rehabilitation. Traumatic experiences are really unpredictable and it can be resulted Post Traumatic Stress Disorders, Post traumatic growth in the end. But developing skills that required to overcoming trauma is facilitated by the Emotional Intelligence, Resilience, and Coping capacities that people having with. Exposure to a traumatic experience and not having a PTSD is determined by the many other factors such as social support system and government facilitation of the wellbeing afterwards the trauma. Here in this article the basic components of Emotional Intelligence, Resilience, and coping mechanisms have been considered as the major factors.

이상저온 조건이 고추의 생육 및 수량에 미치는 영향 (The Influence of Abnormally Low Temperatures on Growth and Yield of Hot Pepper(Capsicum Annum L.))

  • Growth and physiological disorders caused by abnormally low temperatures were evaluated in pepper, an important field crop in Korea. In addition, the effects of chemical treatment using glutamine was verified on minimizing the damages by low temperature. The growth of pepper plants in stem length and diameter was suppressed as the temperature decreased from $25^{\circ}C$, and the suppression level was the highest for plants grown for 90 days at $20^{\circ}C$. However, root growth was not affected by the different temperatures. The number of leaf and leaf area decreased at the temperatures below $25^{\circ}C$, an optimum temperature for growth. Fresh weight and dry weight decreased for plants grown at $20^{\circ}C$. Pepper fruit yield also decreased by 11% at $20^{\circ}C$ in comparison to $25^{\circ}C$. Falling blossom rate was different depending on the growth temperature, and the rate was 27.2% at $25^{\circ}C$, 35.2% at $22.5^{\circ}C$, and 41.0% at $20^{\circ}C$, indicating that falling blossom rate increased as temperature decreased. Different growth temperatures did not affected on the level of symptom of calcium deficiency and Phytopathora blight. Falling blossom was severe at abnormally low temperature of $20^{\circ}C$, but the treatment of glutamine reduced falling blossom rate and increased the yield by 7.0% as compared to control. The optimum concentration of glutamine treatment was 10 mg/L for yields.

교통사고 환자의 외상후 성장 영향요인 (Factors Influencing Post-Traumatic Growth in Traffic Accident Patient)

  • 본 연구는 교통사고 환자의 외상후 스트레스 장애 위험정도, 회복탄력성 및 사회적 관계망이 외상후 성장에 미치는 영향을 알아보기 위해 시도된 서술적 상관관계 연구이다. 연구대상자는 K도 C시 소재 100병상 이상 5개 병원의 교통사고 환자 158명이었으며, 2018년 7월 1일부터 8월 31일까지 구조화된 설문지를 이용하여 자료를 수집하였다. 수집된 자료는 t-test, ANOVA, Pearson's correlation coefficients, Multiple Regression으로 분석하였다. 외상후 성장에 미치는 영향요인을 파악한 결과, 연구의 설명력은 36.9%로 나타났으며, 대상자의 외상후 성장에 미치는 영향요인은 사회적 관계망, 외상후 스트레스 장애 위험정도 순으로 나타났다. 또한 사회적 관계망은 회복탄력성과 외상후 성장의 관계를 완전매개하였다. 이를 토대로, 교통사고 환자의 사회적 관계망이 크고 외상후 스트레스 장애 위험정도가 클수록 외상 후 성장이 높아짐을 확인하였고, 사회적 관계망은 회복탄력성과 외상후 성장의 관계를 완전매개한다는 것을 확인하였다. 따라서 교통사고 환자의 외상후 성장을 돕기 위해 사회적 관계망과 회복탄력성 향상 방안에 대해 모색할 필요가 있고, 교통사고 환자에 대한 반복적이고 장기적인 관찰을 통해 외상후 성장에 대한 추가연구가 필요할 것으로 사료된다.

How to approach feeding difficulties in young children

  • Feeding is an interaction between a child and caregiver, and feeding difficulty is an umbrella term encompassing all feeding problems, regardless of etiology, severity, or consequences, while feeding disorder refers to an inability or refusal to eat sufficient quantities or variety of food to maintain adequate nutritional status, leading to substantial consequences, including malnutrition, impaired growth, and possible neurocognitive dysfunction. There are 6 representative feeding disorder subtypes in young children: infantile anorexia, sensory food aversion, reciprocity, posttraumatic type, state regulation, and feeding disorders associated with concurrent medical conditions. Most feeding difficulties are nonorganic and without any underlying medical condition, but organic causes should also be excluded from the beginning, through thorough history taking and physical examination, based on red-flag symptoms and signs. Age-appropriate feeding principles may support effective treatment of feeding difficulties in practice, and systematic approaches for feeding difficulties in young children, based on each subtype, may be beneficial.

Dual Roles of Autophagy and Their Potential Drugs for Improving Cancer Therapeutics

  • Autophagy is a major catabolic process that maintains cell metabolism by degrading damaged organelles and other dysfunctional proteins via the lysosome. Abnormal regulation of this process has been known to be involved in the progression of pathophysiological diseases, such as cancer and neurodegenerative disorders. Although the mechanisms for the regulation of autophagic pathways are relatively well known, the precise regulation of this pathway in the treatment of cancer remains largely unknown. It is still complicated whether the regulation of autophagy is beneficial in improving cancer. Many studies have demonstrated that autophagy plays a dual role in cancer by suppressing the growth of tumors or the progression of cancer development, which seems to be dependent on unknown characteristics of various cancer types. This review summarizes the key targets involved in autophagy and malignant transformation. In addition, the opposing tumor-suppressive and oncogenic roles of autophagy in cancer, as well as potential clinical therapeutics utilizing either regulators of autophagy or combinatorial therapeutics with anti-cancer drugs have been discussed.

흡수성과 비흡수성 내고정물 사용 후 관골 골절 환자의 자각증상의 차이 (A Comparative Study of the Subjective Symptoms of Bioabsorbable and Metallic Osteofixation System in Zygomatic Bone Fracture)

  • The surgical treatment of craniofacial disorders, using metallic internal osteofixation system has inspired an evolution with advanced operative technique and fixation devices over past two decades. As any other surgical procedures, this procedure also associates problems such as infection, palpability, loosening, and restrict craniofacial skeleton growth, which lead to undue secondary operations for removal. These problems are improved by using bioabsorbable osteofixation system. We compared the patient's subjective symptoms using bioaborbable system versus metallic osteofixation system in zygomatic bone fracture. we should take the individual steps (postoperative 2 weeks, and 1 year) in treating fractured zygoma. From August, 2001 to August, 2003, we used bioabsorbable osteofixation system in 28 patients in zygomatic fracture ($Biosorb^{TM}FX^{(R)}$) and compared 23 patients who were treated with metallic osteofixation system. There was no significant difference in the both groups in subjective symptoms and postoperative result.

불안장애의 분자유전학적 이해 (Molecular Genetics of Anxiety Disorder)

  • Anxiety disorder is likely caused by an interaction of multiple loci in brain, rather than a single locus. Hyperactive neurotransmitter circuits between the cortex, thalamus, amygdala, and hypothalamus are responsible for production of anxiety symptoms. Familial studies performed on anxiety disorder suggested that anxiety disorder should be caused by genetic etiology. Numerous linkage and association studies showed different genetic loci of anxiety disorder. Candidate genes have been focused on important neurotransmitters, neuropeptide, or genes affecting neuronal growth, development, protection or apoptosis. Anxiety disorder has various symptoms and comorbid diseases in family or proband. Therefore, further studies focused on symptomatic dimension of anxiety disorder or responses to drugs are required.
