Effect of Supplemental Vitamin and/or Mineral Premixes in the Formulated Diets on Growth of Juvenile Abalone (Haliotis discus hannai) (참전복 배합사료의 비타민 및 미네랄 혼합물 첨가 효과)

  • A feeding experiment was conducted using juvenile abalone (Haliotis discus hannai) to investigate the optimum level of supplemental vitamin and/or mineral premixes in a formulated diet for practical feed formulation. Three replicate groups of abalone averaging 100 mg were fed the diets containing different levels of vitamin premix ($0\%,\;0.5\%,\;1.0\%$ and $2\%$) and /or mineral premix($0\%,\;2\%,\;4\%$ and $6\%$) for 4 months. Survival rates of abalone showed no distinctive trend by the levels of dietary vitamin premixes. Weight gain, shell growth and soft body weight of abalone were not significantly affected by the different dietary vitamin and/or mineral premixes (P>0,05). Moisture, protein and lipid contents of soft body were influenced by experimental diets (P<0.05) but these differences also had no definite trends by levels of vitamin and/or mineral premixes. These data indicate that supplemental vitamin or mineral premix is not needed in this formulated diet, therefore price of diet for juvenile abalone could be reduced.

  • Aronia (Aronia melanocarpa, Black chokeberry) is an important cash crop in domestic agriculture. We investigated the effects of plant growth regulators on shoot proliferation and rooting using in vitro tissue culture. The most effective shoot multiplication was observed on WPM (woody plant medium) supplemented with 1.0 mg/L zeatin ($8.3{\pm}1.0$ shoots/explant), while the highest rooting rate was obtained from half-strength WPM with 3.0 mg/L IBA (8.8 roots/explant). The rooted plantlets all survived in the artificial soil mixture (with a mixture of peat moss : perlite : vermiculite, 1:1:1, v/v/v) and grew up relatively uniform, ranging from 14 to 16 leaves, 8 to 10 cm in stem height, and 2.3 to 2.8 mm in stem diameter. While experimenting with 5 different varieties of Aronia, we found out that each variety had different characteristics of shoot proliferation and rooting. The total numbers of proliferated shoots per variety is as follows: $17.4{\pm}0.8$ for Nero, 14 to 15 for Purple and Mackenzie, and 10 for both Viking and Odamamachiko. Rooting rates were also various depending on the variety: 88% of Odamamachiko, 80% of Viking and Purple, and 76% of Nero and 60% of Mackenzie shoots rooted. The survival rate of the rooted plantlets was from 92% to 100%, varying by type. Further growth appeared to be better in auxin-treated plantlets, compared to untreated ones. Our results showed the possibility of establishing an effective in vitro micropropagation system for Aronia melanocarpa.

Effects of Materials of Drainage Layer at the Reclaimed Soil Base on Tree Growth at the Open Space of Saemangeum Sea Dike (새만금 방조제 개활지의 준설토 기반에 대한 배수층재 처리가 수목 생육에 미치는 효과)

  • This study was performed to compare the effects of different drainage layers on tree growth at the exposed sites of Saemangeum sea dike. 4 types of drainage layers including control(dredged soil), specially prepared bark, gravel, and wood chip were set in 150~165cm depth of soil. Pinus thunbergii and Celtis sinensis were planted after 9 months of soil treatment. Electrical conductivity(EC) of soil in all treated plots were decreased under $4dS{\cdot}m^{-1}$, and NaCl(%) was decreased under 0.05% after 1 year from soil treatment. Soil moisture at the 120cm depth of the bark treated plot was higher than that of the 180cm soil depth, below the drainage layer. It is considered that vertical mobility of water was inhibited. Organic matter(OM) at the 120cm soil depth increased at bark and wood chip treated plots. Survival rates after 4 years of P. thunbergii and C. sinensis were 100% in all treatments. The height of P. thunbergii was not significantly different among the treatments while the height of C. sinensis was significantly different among the treatments and it was highest at the bark treated plot.

Parathyroid Hormone-Related Protein Promotes the Proliferation of Patient-Derived Glioblastoma Stem Cells via Activating cAMP/PKA Signaling Pathway

  • Background and Objectives: Glioblastoma (GBM) is an aggressive primary brain tumor characterized by its heterogeneity and high recurrence and lethality rates. Glioblastoma stem cells (GSCs) play a crucial role in therapy resistance and tumor recurrence. Therefore, targeting GSCs is a key objective in developing effective treatments for GBM. The role of Parathyroid hormone-related peptide (PTHrP) in GBM and its impact on GSCs remains unclear. This study aimed to investigate the effect of PTHrP on GSCs and its potential as a therapeutic target for GBM. Methods and Results: Using the Cancer Genome Atlas (TCGA) database, we found higher expression of PTHrP in GBM, which correlated inversely with survival. GSCs were established from three human GBM samples obtained after surgical resection. Exposure to recombinant human PTHrP protein (rPTHrP) at different concentrations significantly enhanced GSCs viability. Knockdown of PTHrP using target-specific siRNA (siPTHrP) inhibited tumorsphere formation and reduced the number of BrdU-positive cells. In an orthotopic xenograft mouse model, suppression of PTHrP expression led to significant inhibition of tumor growth. The addition of rPTHrP in the growth medium counteracted the antiproliferative effect of siPTHrP. Further investigation revealed that PTHrP increased cAMP concentration and activated the PKA signaling pathway. Treatment with forskolin, an adenylyl cyclase activator, nullified the antiproliferative effect of siPTHrP. Conclusions: Our findings demonstrate that PTHrP promotes the proliferation of patient-derived GSCs by activating the cAMP/PKA signaling pathway. These results uncover a novel role for PTHrP and suggest its potential as a therapeutic target for GBM treatment.

Effect of Omeprazole on Membrane P-Type ATPase and Peptide Transport in Helicobacter pylori

  • We investigated the growth-inhibitory mechanism of Helicobacter pylori by omeprazole (OMP) and its activated sulfenamide (OAS). Using dithiothreitol (DTT) and 5,5'-dithio-bis[2-nitrobenzoic acid] (DTNB; Ellman's reagent), we first determined the relationship between the binding capacity of these compounds to H. pylori membrane and its significance to membrane P-type ATPase activity. After incubation of the intact H. pylori cells with either OMP or OAS, the residual quantity of free SH-groups on the cell membrane was measured, and, the resulting values were plotted as a function of time. From this experiment, we found that there was a considerable difference in the membrane-binding rates between OMP and OAS. At neutral pH, the disulfide bond formation on H. pylori membrane was completed within 2 min of incubation of the intact cells with OAS. By OMP, however, it was gradually formed, exceeding 10 min of incubation for completion, whereby, the extent of P-type ATPase inhibition appeared to be proportional to the disulfide forming rate. From this data, it was suggested that the disulfide formation might directly affect enzyme activity. Since OMP per se cannot yield a disulfide bond with cysteine, it is predicted that the enzyme inactivation must be caused by the OAS form. Accordingly, we postulated that, under the neutral pH, OMP could be converted to OAS in the course of transport. By extrapolating the inhibitory slopes, we could evaluate K₁ values, relating to their minimal inhibitory concentrations (MICs) for H. pylori growth. In these MIC ranges, H. pylori uptake or vesicular export of nutrients such as peptides were totally prohibited, but their effect in Escherichia coli were negligible. From these observations, we strongly suggest that the P-type ATPase activity is essential for the survival of H. pylori cells in particular.

Effects of Temperature and Salinity on the Growth of Marine Benthic Microalgae for Phytoremediation (식물환경복원을 위한 저서미세조류의 성장에 미치는 수온과 염분의 영향)

  • To improve sediment quality in eutrophic coastal areas using benthic microalgae, we examined the effects of temperature and salinity on the growth of benthic microalgae Achnanthes sp., Amphora sp., Navicula sp. and Nitzschia sp. isolated from Sujeong Bay, Korea, using batch cultures. The maximum growth rates were obtained under the combined temperature and salinity conditions of $25^{\circ}C$ and 25 psu for Achnanthes sp. (0.60 /day), $15^{\circ}C$ and 25 psu for Amphora sp. (0.56 /day), $20^{\circ}C$ and 30 psu for Navicula sp. (0.53 /day), $20^{\circ}C$ and 25 psu for Nitzschia sp. (0.48 /day). Considering these results of temperature and salinity conditions required for optimum growth (${\geq}$ 70% of maximum specific growth rate), Amphora sp. Navicula sp. and Nitzschia sp. were characterized as eurythermal and euryhaline species, while Achnanthes sp., which exhibited extremely low survival at low temperature. In conclusion, Amphora sp., Navicula sp. and Nitzschia sp. may be useful species for phytoremediation, to control eutrophication and hypoxic water and thus improve environmental conditions of polluted coastal areas.

Growth, Reproduction, Mortality, and Production of Laminaria japonica Areschoug on the Southeastern Coast of Korea (한국 동남해안에서 다시마(Laminaria japonica Areschoug)의 성장, 성숙, 사망률 및 생산)

  • Growth, reproduction, mortality, and production of Laminaria japonica were experimentally studied at a cultivation ground on the coast of llkwang, where the largest amount of cultivated Laminaria has been produced in Korea. For this experiment, young sporophytes (0.33 cm in mean length) grown in the laboratory were transplanted at the depth of 3 m and field surveys on them were conducted twice a month from December, 1995 to August, 1996. Plants exhibited an annual life span; they were completely dead by August. Frond width, thickness, and wet weight showed similar pattern of seasonal growth and reached their maxima in July, but frond length showed no more increment after May. Maximum mean frond length and weight were 199.8 cm and 333.0 g wet wt., respectively. Overall meristematic growth in length and weight were 384.0 cm and 393.6 g wet wt., respectively. Absolute growth rates (AGR) which were calculated from the length of tissue developed from meristem varied seasonally; AGR of length and weight reached maxima in March (3.6 $cm{\cdot}d^{-1}$) and May (3.8 g wet $wt{\cdot}d^{-1}$), respectively. Absolute attrition rates gradually increased from February to July. Seasonal differences in growth and attrition rates appeared to be related to seawater temperature and nitrogen concentration in seawater. Reproductive sporophytes bearing sprorangium sorus began to occur from April, and the ratio of sorus area to blade area reached its maximum in July (0.034). Survival rate was exponentially decreased; more than 90% of plants decayed within 56 days after outplanting. After February, mortality was size-specific; mortality of smaller plants less than 30 cm in length were relatively higher. Maximum biomass occured in July (285.6 kg wet $wt{\cdot}m^{-2}$) and annual production was 758.7 kg wet $wt{\cdot}m^{-2}$.

Expression of Several Biologic Markers as Prognostic Markers in Non-Small Cell Lung Cancers (폐암조직에서 생물학적 지표들의 예후인자로서의 비교검토)

  • Background: Despite modern diagnostic, staging, and therapeutic advances, esp. with molecular biologic techniques, the 5-year survival rate of all cases of lung cancer does not exceed 15%. Also, the incidence of lung cancer of both sex in Korea is increasing year by year and the lung cancer is one of the leading causes of cancer death. Therefore, it is strongly needed to develop the new combination of treatment modalities including neoadjuvant chemotherapy and to identify tumor specific characteristics with staging or prognostic markers. Here we present the clinical significance of several biologic tumor markers to use as a prognostic markers in patients with non-small cell lung cancers. Method: The survival has correlated with the expressibility of proliferative cell nuclear antigen (PCNA), epidermal growth factor receptor(EGFR), p53 and/or blood group antigen A(BGAA) using immunohistochemistry in 46 patients with non-small cell lung cancers. Results: 1) The expression rates of PCNA, EGFR, p53 and BGAA were 80.6%, 61.3%, 45.9% and 64.3%, respectively and those were not correlated to cell types or clinical stges. 2) The expression of BGAA was correlated with better survival in median survival and in 2-year survival rate and that of PCNA was correlated with worse survival in median survival and 2-year survival rate. 3) The expression of EGFR or p53 was not valuable to predict prognosis in non-small cell lung cancers. 4) With simultaneous applications of PCNA, EGFR and p53 immunostain, the patients with 2 or more negative expressions showed better prognosis than the patients with 2 or more positive expressions. Conclusion: It is suggested that the expression of blood group antigen may be a positive prognostic factor and that of PCNA may be a negative prognostic factor. Also, the combination of expressions of PCNA, EGFR and p53 may be used as a negative prognostic factor.

Breeding and Reproductive Studies on Korean Native Loach III. Early Embryonic Development (한국산 미꾸리에 관한 육종 번식학적 연구 III. 초기 난발생)

  • This study was carried out to investigate the appearance time of the second polar body for producing Gynogenesis or Triploid which could be obtained by arresting the second polar body by cold shock, and then blastoderm was used to measure fertility that revealed the nature of oogenesis, the effects of water temperature on fertility, hatchability, abnormality, viability and growth rate, and the water temperature and the breeding methods to prevent early death of larvae in Korean loach(Misgurnus anguillicaudatus) ; the results obtained in this study were summarized as follows. The second polar body was observed ont he surface of plasma disc close to micropyle within 10~40 min after fertilization at 29$^{\circ}C$. Artificial inseminatin had to be done immediately after the egg spawning because the spermatozoa of loach their mobility within 2 minutes when they were exposed to water. The amount of time needed to reach at blastoderm stage was 12 hours if fertilized eggs were incubated at 16$^{\circ}C$, 8 hours at 19$^{\circ}C$, 6 hours at 21$^{\circ}C$, 5 hours at 23$^{\circ}C$, 4 hours at 26$^{\circ}C$ and 3 hours 30 min at 29$^{\circ}C$ showing the shorter time for development of eggs at higher temperature. Fertilization rates in water temperatures of 19$^{\circ}C$, 21$^{\circ}C$, 23$^{\circ}C$, and 26$^{\circ}C$ were higher than those of water temperatures, 16$^{\circ}C$ and 29$^{\circ}C$. Water temperatures at 19$^{\circ}C$, 21$^{\circ}C$, and 23$^{\circ}C$ showed higher hatching rates that those of 16$^{\circ}C$, 26$^{\circ}C$, and 29$^{\circ}C$, while abnormal rates in 16$^{\circ}C$, 19$^{\circ}C$, 21$^{\circ}C$ and 23$^{\circ}C$ were lower than that of 26$^{\circ}C$ and 29$^{\circ}C$. Water temperatures at 16$^{\circ}C$, 19$^{\circ}C$, 21$^{\circ}C$, 23$^{\circ}C$ and 26$^{\circ}C$ respectively, were more different than 29$^{\circ}C$ in survival rates. The embryos were hatched at 72 hours after fertilization in 16$^{\circ}C$ water temperature, 48 hours in 19$^{\circ}C$, 40 hours in 21$^{\circ}C$, 32 hours in 23$^{\circ}C$, 25 hours in 26$^{\circ}C$, and 16 hours in 29$^{\circ}C$. Within three days after hatched out, the larvage grew 3mm in total length, the yolk granules were entirely consumed and the head and the trunk became thicker. Within 45 days after hatched out, the larva grew 25mm at 29$^{\circ}C$, 21mm at 26$^{\circ}C$, 16mm at 23$^{\circ}C$, 15mm at 21$^{\circ}C$, 12mm at 16$^{\circ}C$ in a 30 litreglass aquarium.

Seedling Plug and Cutting Method for Multi-propagation of Ornamental Miscanthus Spp. (조경용 억새의 대량번식을 위한 플러그묘와 삽목번식법)

  • Miscanthus species are known as a genus of eco-friendly and low-maintenance cost ornamental grasses. Plug and cutting methods were tested for multi-propagation of most promising ornamental Miscanthus species in greenhouse and field plot. The plug formation period with three different cell sizes with four cultivars (M. sinensis 'Andersson', 'Strictus', 'Gracillimus', 'Variegatus') were evaluated the seedling development stages with two irrigation types of the over-head and the bottom watering in greenhouse and field plot afterward during 2015-2016 season. In seedling plug test, the size of tray cell affected the plug formation. Bottom irrigation resulted positively on plant height, weight, root and tiller development compared with the over-head irrigation. Plug cell size affected the plant growth in the field after transplanting. All of the 3 Miscanthus species showed higher rates of successful propagation at the lower nodes before inflorescence formation (vegetative growth stage). To analyze the survival factors of M. xgiganteus cutting, the cutting time, node part, and culm diameter were tested as independent variables with the binary logistic model. The survival probability was influenced by node part and culm diameter significantly. The third and fifth node parts showed 0.12 (8X higher failure probability) and 0.02 (50X higher failure probability) times less survival probability. It means the survival probability will be increased by using older and lower part of cuttings during a vegetative growth stage before inflorescences of M. xgiganteus.
