연속배양에서 Microcystis aeruginosa의 질소 인 흡수와 생장 특성 (Nitrogen and Phosphorus Uptake and Growth Kinetics of Microcystis aeruginosa Cultured under Chemostats)

  • 연속배양의 평형 정상상태 조건에서 담수조류 Microcystis aeruginosa의 최대생장률 (${\mu}_m$)과 세포내 영양염의농도(Q)의 관계, 질소 결핍 상태에서 액체배지에서 영양염 농도(S)와 홉수율(V)의 관계를 각각 조사하였다. 질소제한조건에서 M. aeruginosa의 세포 당 $Q_{0N}$은 0.04 pmol $cell^{-1}$. ${\mu}_m$은 1.1 $day^{-1}$였고, 제한조건에서 최저값 $Q_{0N}$과 충분조건에서 $Q_N$을 탄소기준하여 각각 6.1 ${\mu}mol$ mg $C^{-1}$ 및 9.5${\sim}$12.4 ${\mu}mol$ mg $C^{-1}$였다. 다른 미세조류와 비교f'1면 세포의 질소 함량이 매우 높았고 질소 결핍시에도 그 함량이 높게 유지되어 질소 요구도가 높은 종이었다 질소 제한조건에서 영양염 홉수 측정 결과에서 반포화계수($K_s$)와 최대흡수율 ($V_m$)은 각각 36 ${\sim}$ 61 ${\mu}M$ 및 0.28 ${\sim}$ 0.35 ${\mu}mol\;cell\;{\cdot}\;hr^{-1}$ 범위였다. M. aeruginosa의 $K_5$값은 매우 높았고 $V_m$도 높은 편이었다. $K_s$와 흡수력 $V_m$은 미세조류의 영양상태 즉 영양염의 세포내 함량(Q)이나 제한영양염의 결핍의 강도의 영향을 받고 미세조류 생장균과도 관련이 있다. M. aeruginosa는 다른 미세조류에 비하여 세포내 질소 함량과 생장률이 높기 매문에 질소와 인에 대한 횹수율이 높을 것이다. 그러나 질소 흡수력의 반포화계수 $K_s$값이 높고 $V_m/K_s$비율은 낮아 수게에서 질소가 저농도 일 때에는 다른 미세조류와 비교하면 경쟁력이 떨어지고 질소에 대한 기질 친화력은 약한 것으로 나타났다. 낙동강 하류지역에서 M. aeruginosa가 대발생하는 시기에 수중 영양염의 농도 변동은 M. aeruginosa의 영양생리 kinetics 특성과 잘 부합하는 것으로 나타났다.

우유와 즉석섭취 메추리알에서 병원성 미생물의 생육에 미치는 지방과 저장온도의 영향 (Effect of Fat Content and Storage Temperature on the Growth and Survival Kinetics of Pathogenic Microorganisms in Milk and Ready to Eat (RTE) Quail Eggs)

  • According to the microbiological standard, Staphylococcus aureus, Clostridium perfringens and Listeria monocytogenes should not be detected in milk and egg products in Korea. Refrigerated food such as milk must be kept under $10^{\circ}C$ at retail markets. However, temperature abuse of refrigerated foods at such markets is often observed. We compared the growth and survival kinetics of S. aureus and C. perfringens at 10 and $15^{\circ}C$, and the growth kinetics of L. monocytogenes at 4 and $10^{\circ}C$ in whole and skim milk and ready-to-eat (RTE) quail eggs to evaluate their growth possibilities at retail markets. Regardless of storage temperature, the level of S. aureus reached the maximum level ($10^8-10^9CFU/ml$) in whole milk, non-fat milk and RTE quail eggs within the expiration date. Even low contamination levels of S. aureus (10 CFU/mL) grew rapidly in milk and quail eggs to reach the maximum level within the shelf life. Survival of C. perfringens in whole milk was greater than that in non-fat milk, indicating that the fat content in milk influences the survival of C. perfringens. For L. monocytogenes, the population in milk increased by 0.5-1 log CFU/mL at $4^{\circ}C$, while the populations reached the maximum level at $10^{\circ}C$ within the expiration date, regardless of initial contamination levels. In quail eggs, L. monocytogenes grew to the maximum level within the expiration date (60 days) at both temperatures. S. aureus and L. monocytogenes must be controlled to be negative, and proper temperature management should be emphasized at retail markets to protect the consumer. Since C. perfringens did not grow in milk and RTE quail eggs, there is no risk due to the growth of C. perfringens in these products at retail markets.

Synchrotron SAXS Study on the Micro-Phase Separation Kinetics of Segmented Block Copolymer

  • The phase transition behavior isothermal micro-phase separation kinetics of polyester-based thermoplastic elastomer were studied using the synchrotron X-ray scattering(SAXS) method. The structural changes occurring during heating period were investigated by determining the changes of the one-dimensional correlation function, interfacial thickness and Porod constant. Based on the abrupt increases of the domain spacing and interfacial thickness, a major structural change occurring well below the melting transition temperature is suggested. Those changes are explained in terms of melting of the thermodynamically unstable hard domains or/and the interdiffusion of the hard and soft segments in the interfacial regions. SAXS profile changes during the micro-phase separation process were also clearly observed at various temperatures and the separation rate was found to be sensitively affected by the temperature. The peak position of maximum scattering intensity stayed constant during the entire course of the phase separation process. The scattering data during the isothermal phase separation process was interpreted with the Cahn-Hilliard diffusion equation. The experimental data obtained during the early stage of the phase separation seems to satisfy the Cahn-Hilliard spinodal mechanism. The transition temperature obtained from the extrapolation of the diffusion coefficient to zero value turned out to be about 147$\pm$$2^{\circ}$, which is close to the order-disorder transition temperature obtained from the Porod analysis. The transition temperature was also estimated from the inveriant growth rate. By extrapolating the inveriant growth rate to zero, a transition temperature of about 145$\pm$$\pm$$2^{\circ}$ was obtained.

The influence of sodium hypochlorite concentration on the fibrin structure of human blood clots and transforming growth factor-beta 1 release: an ex vivo study

  • Objective: This study investigated the effects of various concentrations of sodium hypochlorite (NaOCl) on human whole-blood clotting kinetics, the structure of the blood clots formed, and transforming growth factor (TGF)-β1 release. Materials and Methods: Human whole blood was collected from 5 healthy volunteers and divided into 4 groups: CG (control, 0.5 mL of blood), BN0.5 (0.5 mL of blood with 0.5 mL of 0.5% NaOCl), BN3 (0.5 mL of blood with 0.5 mL of 3% NaOCl), and BN5.25 (0.5 mL of blood with 0.5 mL of 5.25% NaOCl). The effects of NaOCl on clotting kinetics, structure of fibrin and cells, and release of TGF-β1 were assessed using thromboelastography (TEG), scanning electron microscopy (SEM), and enzyme-linked immunosobent assay, respectively. Statistical analysis was conducted using the Kruskal Wallis and Mann-Whitney U tests, followed by the post hoc Dunn test. A p value < 0.05 indicated statistical significance. Results: The blood samples in BN0.5 and BN3 did not clot, whereas the TEG of BN5.25 showed altered clot formation. Samples from the CG and BN3 groups could only be processed with SEM, which showed that the latter lacked fibrin formation and branching of fibers, as well as clumping of red blood cells with surface roughening and distortion. TGF-β1 release was significantly highest in BN3 when all groups were compared to CG (p < 0.05). Conclusions: Each concentration of NaOCl affected the release of TGF-β1 from blood clots and altered the clotting mechanism of blood by affecting clotting kinetics and cell structure.

Michelson 간섭계를 응용한 미세 상변화 현상 계측 (Probing of Microscale Phase-Change Phenomena Based on Michelson Interforometry)

  • Experimental schemes that enable characterization of phase-change phenomena in the micro scale regime is essential for understanding the phase-change kinetics. Particularly, monitoring rapid vaporization on a submicron length scale is an important yet challenging task in a variety of laser-processing applications, including steam laser cleaning and liquid-assisted material ablation. This paper introduces a novel technique based on Michelson interferometry for probing the liquid-vaporization process on a solid surface heated by a KrF excimer laser pulse (${\lambda}=248nm,\;FWHM=24\;ns$) in water. The effective thickness of a microbubble layer has been measured with nanosecond time resolution. The maximum bubble size and growth rate are estimated to be of the order of $0.1{\mu}m\;and\;1\;m/s$, respectively. The results show that the acoustic enhancement in the laser induced vaporization process is caused by bubble expansion in the initial growth stage, not by bubble collapse. This work demonstrates that the interference method is effective for detecting bubble nucleation and microscale vaporization kinetics.

Receptor Cytoplasmic 영역에 의존하는 EGF의 고친화성 결합 (Dependence of High Affinity Binding of Epidermal Growth Factor on Receptor Cytoplasmic Domain)

  • 일부의 EGF receptor 에는 EGF 가 세포표면에서 receptor 와 결합할 때 보다 높은 친화력(high affinity)을 보이고 있는데 그 이유를 설명하기 위해서 EGF receptor 의 cytoplasmic 영역을 절단하여 EGF 와의 친화력을 측정하였다. Scatchard plot 의 결과 1022 아미노산 이하로 절단된 receptor 는 high affinity 특성을 상실하였다. Triton X-100로 세포막을 제거하여 cytoskeleton 이 EGF receptor 의 구조에 미치는 영향을 조사한 결과 cytoskeleton과 결합한 receptor 보다 EGF 에 대해서 더 높은 친화력을 보였다. 따라서 cytoskeleton 이 high affinity EGF receptor 를 형성하는데 영향을 미치고 receptor 와 cytoskeleton 의 가능한 결합부위는 1022-1186 아미노산 사이인 것 같다.
