Morphological studies on the hemal node in the Korean native cattle (한우 혈절에 관한 형태학적 연구)

  • The hemal nodes of the Korean native cattle were studied by gross anatomy and light microscopy. Hemal nodes were spherical or ovoid in shape, and red or black in colors. A hemal node consisted of a thick capsule and a hilum, and had extensive subcapsular and deep sinuses distended by a great number of erythrocytes. Although a few lymphatic nodules and tissues were seen in the parenchyma, no typical cortex and medulla was defined. Blood vessels occurred, but lymph vessel was not observed in nodes. The stroma of the hemal node was composed of reticular cells and fibers. The parenchyma consisted of many erythrocytes and lymphocytes, and a few macrophages and megakaryocytes. The capsule and trabecula was a collagenous connective tissue with smooth muscle cells. B-lymphocytes were principally located in the lymphatic nodules of the hemal node. T-lymphocytes were scattered in the diffuse lymphatic tissues of the hemal node.

Effect of Mixed Extract of Panax Notoginseng, Rehmanniae Radix and Acanthopanacis Cortex (AIF) on Experimentally Induced Osteoarthritis

  • The objective of the present study was to evaluate the effect of a mixed extract of three herbs, Panax Notoginseng, Rehmanniae Radix and Acanthopanacis cortex (AIF), for the treatment of horses with experimentally induced osteoarthritis. Twelve healthy male horses were included in this study. Horses were assigned to one of two groups: the AIF group (n=6) or the control group (n=6). Osteoarthritis was induced in all horses by intraarticular injection of sodium monoiodoacetate (0.12 mg/kg). Horses in the AIF group received 3 g of AIF with food daily, and those in the control group received food only. Treatment began on the day of intraarticular injection. Clinical and radiographic evaluations were performed every 2 weeks. At week 12, horses were euthanatized, and postmortem gross pathologic and histologic examinations of the middle carpal joint were performed. There were no significant differences in clinical values between the two groups. Radiographic evaluation revealed that the percentages of narrowness of joint space width in the control group were significantly higher than those in the AIF group (p<0.02). On gross pathologic examination, the mean total dimensions of articular cartilage erosions and fibrillations in the control group ($101.5{\pm}41.5mm^2$) were significantly wider than those in the AIF group ($29.3{\pm}39.7mm^2$; p<0.01). On histopathologic evaluation, significantly higher grades of staining intensity and lower empty lacunae (EL) ratios were found in the AIF group (p<0.03). The present study revealed that AIF had significant disease modifying effects in horses with experimentally induced osteoarthritis.

Structure of hemolymph node in WKY rats (WKY 랫드의 혈림프절 구조)

  • It has been known that the hemolymph node is one of the lymphoid organs found normally in ruminants and some rodents, and this organ shares morphological and functional characteristics of lymph node and spleen. To clanify the rigorous definition of morphological structures and functions of the hemolymph node in Wistar Kyoto(WKY)rats, we examined these organs of WKY rats gross anatomically and light microscopically. The hemolymph nodes were normally found in the abdominal cavity and in the neck of WKY rats. This organ was surrounded by a thin connective tissue capsule and there was a hilus. The parenchyma comprised a cortex of lymphatic nodules and diffuse lymphatic tissues, and a medulla of diffuse lymphatic tissues arranged in cords. Afferent and efferent lymph vessels were observed but there was no extensive subcapsular and medullary sinuses. These sinuses were filled with erythrocytes. The stroma of hemolymph nodes was composed of reticular cells and fibers, and many lymphocytes, granulocytes, erythrocytes, plasma cells, macrophages and megakaryocytes were supported by the reticular network. The above findings suggest that the hemolymph nodes of WKY rats may take part in blood formation, blood filtration and immune reaction.

Mouse Single Oral Dose Toxicity Studies of PGB-1, a Novel Polyglucosamine Polymer Produce from Enterobacter sp. BL-2

  • This study was conducted to obtain acute information of the oral dose toxicity of PGB-1, a novel polyglucosamine polymer produced from a new strain Enterobacter sp. BL-2 in male and female mice. In order to calculated 50% lethal dose ($LD_{50}$) and approximate lethal dose (LD), test material was once orally administered to male and female ICR mice at dose levels of 2000, 1000, 500, 250, 125 and 0 (vehicle control) ml/kg (body wt.). The mortality and changes on body weight, clinical signs, gross observation and organ weight and histopathology of principle organs were monitored 14 days after dosing with PGB-1. We could not find any mortalities, clinical signs, body weight changes and gross findings. In addition, significant changes in the organ weight and histopathology of principal organs were not observed except for some sporadic findings. The results obtained in this study suggest that PGB-1 may not be toxic in mice and may be therefore safe for clinical use. The $LD_{50}$ and approximate LD in mice after single oral dose of PGB-1 were considered over 2000 mg/kg in both female and male mice.

Regulation of Gastric Acid Secretion of Liriope platyphylla Extract in Gastroesophageal Reflux Disease

  • Objectives: The purpose of this study was to confirm the effects of Liriope platyphylla extract on relieving Gastroesophageal reflux disease (GERD) through regulation of acid secretion. Methods: 8-week-old ICR mice were divided into untreated control group (Ctrl), GERD elecitation group (GERDE), Omeprazole administrate group before GERD elicitation (OMA), and Liriope platyphylla extract administrate group before GERD elicitation (LPA). After inducing GERD, gross observation and histological examination were performed and ATP6V1B1 (ATPase H+ Transporting V1 Subunit B1), GRPR (Gastrin-releasing peptide receptor), COX-1 (Cyclooxygenase 1), 8-OHdG (8-hydroxy-2'-deoxyguanosine), Cathelicidin, p-JNK (phospho c-Jun N-terminal kinase) were observed to confirm the damage defense effect of the esophageal mucosa, acid secretion regulation, antioxidant, anti-inflammatory, mucosal protection, and apoptosis regulation Results: OMA and LPA showed lower levels of damage compared to GERDE in gross observation and histological examination. ATP6V1B1, GRPR, and 8-OHdG showed lower positive reactions in OMA and LPA than in GERDE. COX-1 were less positive in GERDE and OMA than in Ctrl, but showed higher secretion in LPA than in Ctrl. Cathelicidin showed a decreased positive reaction in GERDE, OMA and LPA compared to Ctrl, but the decrease in positive reaction was smaller in OMA and LPA compared to GERDE. p-JNK showed increased positive reaction in GERDE, OMA and LPA than in Ctrl, but the increase in the positive reaction was smaller in the OMA and LPA compared to GERDE. Conclusions: The effects of Liriope platyphylla extract on esophageal mucosal damage protection, acid secretion regulation, antioxidant, anti-inflammatory, mucosal protection and apoptosis regulation were confirmed.

An Anatomical and Histochemical Study of the Olfactory Organ in Rice-fish Oryzias sinensis(Pisces: Adrianichthyidae) in South Korea (한국산 대륙송사리 Oryzias sinensis(Pisces: Adrianichthyidae) 후각기관의 해부 및 조직화학적 연구)

  • The anatomy and histology of the olfactory organ in Oryzias sinensis was researched using a stereo microscope and light microscope. In the gross structure, the paired olfactory organs on the dorsal part of the head consist of two nostrils (a circular anterior nostril and a slit posterior nostril in a distance), a single olfactory chamber and a single accessory nasal sac. In the histological study, the epithelium of the olfactory chamber is classified into both sensory and non-sensory regions. The sensory epithelium consists of olfactory receptor neurons, supporting cells, basal cells and vesicles, and is islet in distributional pattern. The non-sensory epithelium is composed of stratified epithelial cells and two types of mucous cells (acidic and neutral cells). The epithelium of the accessory nasal sac has swollen stratified epithelial cells, mucous cells with a rich glycoprotein. Such an olfactory anatomy and histology of O. sinensis may reflect its habitat surrounding stagnant and polluted water.

Effects of Aluminium on Growth, Chlorophyll Content, ALAD Activity and Anatomy of Root rind Shoot in Azuki Bean (Vigna angularis) Seedlings (Aluminium이 팥(Vigna angularis) 유식물의 생장, 엽록소함량, ALAD활성 및 뿌리와 경엽부의 형태에 미치는 영향)

  • The toxic effects of aluminium (Al) on growth, chlorophyll content, $\delta-aminolevulinic$ acid dehydratase (ALAD) activity and anatomy of root and shoot were investigated in 7-day-old azuki bean (Vigna angularis) seedlings. Significant depressions in root elongation was observed in the low concentrations of Al (50, 100 $\muM)$ and increasing Al concentrations caused a sharp decline of root and shoot growth. The degree of inhibition was dependent upon Al supply. Exposure to 50 $\muM$ Al or more inhibited root elongation within 1 day. In the 50 $\muM$ Al treatments, a recovery of root growth was seen after 7 days exposure. In contrast, lateral root initials was little affected by Al exposure. Al toxicity symptoms and growth responses were more well developed in the roots than in the shoots. Analysis of Al localization in root cells by hematoxylin stAlning showed that Al entered root apices and accumulated in the epidermal and cortical cells immeadiately below the epidermis. There was a good positive correlation between the level of chlorophyll and ALAD activity. Increasing Al concentrations caused a decrease in total chlorophyll contents, accompanied by proportional changes in ALAD activity, suggesting a cootr-dinated reduction of a photosynthetic machinery. Al exerted specific influence on the morphology of root ann shoot. At higher concentrations of Al the roots induced drastic anatomical changes. The epidermal cells were disorganized or destructed while the cortical cells exhibited distortion of cell shape and/or disintegration. The diameter of root and transectional area of cortical cells decreased considerably with Al treatment. In the shoot Al also enhanced reduction of diameter of shoot and cell size. Gross anatomy of leaves treated with Al did not differ significantly from the controls, except for fewer and smaller chloroplast. Our results indicate that toxic effect of Al appear to be manifested primarily in roots and secondarily on shoots, and changes in root morphology are related to changes in the root growth patterns. Results are further discussed in re181ion to the findings in other plant species, and it is concluded that Al causes morphological, structural and, presumably, functional damage to the roots of the species investigated.

Functional Anatomy and Histology of the Olfactory Organ in Korean Eel Goby, Odontamblyopus lacepedii (Pisces: Gobiidae)

  • For Odontamblyopus lacepedii with small and turbid eyes, the gross structure and histology of the olfactory organ, which is important for its survival and protection of the receptor neuron in estuarial environment and its ecological habit, was investigated using a stereo, light and scanning electron microscopes. Externally, the paired olfactory organs with two nostrils are located identically on each side of the snout. These nostrils are positioned at the anterior tip of the upper lip (anterior nostril) and just below eyes covered with the epidermis (posterior nostril). Internally, this is built of an elongated olfactory chamber and two accessory nasal sacs. In histology, the olfactory chamber is elliptical in shape, and lined by the sensory epithelium and the non-sensory epithelium. The sensory epithelium of a pseudostratified layer consists of olfactory receptor neurons, supporting cells, basal cells and lymphatic cells. The non-sensory epithelium of a stratified layer has swollen stratified epithelial cells and mucous cells with acidic and neutral sulfomucin. From these results, we confirmed the olfactory organ of O. lacepedii is adapted to its ecological habit as well as its habitat with burrows at the muddy field with standing and murky waters.

Osteoarthritis Model Induced by Monosodium Iodoacetate: Good Model for Long-term Drug Efficacy Test (Monosodium Iodoacetate 유발 골관절염 모델: 장기적인 약물효능 평가에 적합한가?)

  • Objectives This study was aimed to offer basic data in long term drug efficacy test using monosodium iodoacetate-induced osteoarthrits model. Methods Sixty male rats were divided into normal and osteoarthritic group. Rats of normal group were injected with 0.1 ml physiological saline, and rats of osteoarthritic group were inected with 0.1 ml monosodium iodoacetate (3 mg/ml) into each left and right knee joint cavities. Gross examination, proteoglycan contents and histopathological examination on the knee joint were performed at 10, 20, 40, and 60 days after injection. Results Grossly, degenerative changes at 10 days, desquamation at 40 days, and ulceration of articular cartilages at 60 days were observed. Proteoglycan contents in articular cartilages were decreased rapidly to 40 days, after than decreased gradually. Osteoarthritic scores were increased rapidly to 20 days, after than increased gradually to 60 days. Conclusions From above results, osteoarthritis model induced by a single intra-articular injection of monosodium iodoacetete is useful model for long term drug efficacy test.
