Overexpresssion of the OsbZIP66 transcription factor enhances drought tolerance of rice plants

  • Drought stress is a major constraint of crop development and productivity. Plants have evolutionally developed several mechanisms at the molecular, cellular, and physiological levels to overcome drought stress. The basic Leucine zipper (bZIP) transcription factor (TF) family members are starting to be concerned about their roles in drought stress responses. In this study, we functionally characterized OsbZIP66, a rice group-E bZIP TF, to be associated with rice drought tolerance mechanisms. Expression of OsbZIP66 was significantly induced upon treatments of rice plants with drought, high salinity, and ABA. These observations and the fact that the OsbZIP66 promoter contains ten ABA-responsive elements suggest that OsbZIP66 is up-regulated by drought stress in an ABA-dependent manner. Overexpression of both OsbZIP66 in a whole plant body and specifically in roots enhanced drought tolerance of rice plants, indicating that the rice drought tolerance positively correlates with the expression levels of OsbZIP66. Thus, our results demonstrated that OsbZIP66 has a potential for use in biotechnological development of high-yielding rice plants under drought conditions.

오징어채낚기용 은백색 유인체 개발과 어획성능 (Development of the silver-white decoy for squid automatic jigging machine and fishing performance)

  • Decoys for automatic jigging machines, the body part of a squid hook, have been developed in small and existing sizes in consideration of squid food, color blindness, and retinomotor responses and in utilization of pearl pigment, PP of high transparency, and combined mixture. In comparison of the developed silver-white decoy and existing decoys, the optical characteristics were examined, and the fishing performance of small size silver-white squid hooks was assessed in application of 4 fishing boats with the squid automatic jigging machine and metal halide fishing lamp in July, 2012. The luminances of the three squid hook colors-green, dark green and silver-white-increased as the intensity of illumination increased. Among these, the increase of silver-white was particularly distinguished. As to the average contrast of squid hooks, that of silver-white was 10.33, which was the highest, and then green 1.86 and dark green -0.10 in the order. As to the fishing performance of the silver-white hook, that of the 202 Geumyeong-ho and 101Yongjin-ho which caught squids were similar to that of the existing green hook and was relatively low in the case of the Dongbu-ho. However, that of the Haengbok-ho which caught relatively small squids whose average length was 19.9cm and installed silver-white hook in all automatic jigging machines was significantly excellent. In order to enhance the fishing performance of small size silver-white hooks, therefore, it would be effective to install in every automatic jigging machines of fishing boat and to start fishing before July by which small squids are caught.

전자파를 조사한 흰쥐 간조직에서의 Mixed Function Oxidase System과 Xanthine Oxidase 활성에 미치는 녹차의 영향 (Effect of Green Tea on Mixed Functon Oxidase System and Xanthine Oxidase Activities in Rat Liver Exposed to Microwaves)

  • The purpose of this study was to investigate the changes of microsomal mixed function oxidase (MFO) and xanthine oxidase activities (XOD) in the liver of rats exposed to microwave. Sprague-Dawley male rats weighing 200$\pm$10g body weight were randomly assigned to a normal and microwave exposed(MW) groups; microwave exposed groups were divided into two groups; microwave (MW) group and green tea(GT) gropu which were fed distilled water and green tea extracts during experimental periods, respectively. Rats were irradiated with microwave at the frequency of 2.45 GHz for 15min and rats wre sacrificed at the 4th day of the microwave irradiaton. The hemoglobin level of GT group was higher than that of the normal gropu and MW group, but the hematocrit value was not significantly different among all experimental gropus. The activity of serum GOT of MW group was significantly increased but that of GT group was similar to normal group. Activities of GPT were not significantly different among all experimental groups. Liver XOD activity was significantly increased in the microwave exposed groups but green tea normalized the XOD activity. The activity of hepatic microsomal cytochrome P450 was significantly increased in MW group compared to normal group and that of GT group was similar to that of the normal group. The activity of hepatic microsomal NADPH-cytochrome P450 reductase was also significantly increased in MW group compared to normal group, but that of GT group was similar to that fo the normal group. In conclusion, the activities of MFO and XOD were elevated by microwave irradiaton, but the activation of MFO system as well as the damage of the liver by microwave were reduced by green tea supplementation.

Evaluation of light-emitting diode colors and intensities on slaughter performance, meat quality and serum antioxidant capacity in caged broilers

  • Objective: This study was to evaluate the interaction of three different light-emitting diode (LED) light colors (white, green, and blue) and three intensities (5, 10, and 15 lx) on slaughter performance, meat quality and serum antioxidant capacity of broilers raised in three-layer cages. Methods: A total of 648 (8-days-old) male broiler chicks (Cobb-500) were randomly assigned in 3×3 factorially arranged treatments: three light colors (specifically, white, blue, and green) and three light intensities (namely, 5, 10, and 15 lx) for 35 days. Each treatment consisted of 6 replicates of 12 chicks. The test lasted for 35 days. Results: The semi-eviscerated weight percentage (SEWP) in 5 lx white was higher than that in 15 lx (p<0.01). The eviscerated weight percentage (EWP) (p<0.05) and water-loss percentage (WLP) (p<0.01) decreased in 10 lx white light than those in green light. Under blue light, the content of hypoxanthine (Hx) in muscle was lower than that under white and green light (p<0.01). The content of malondialdehyde (MDA) in 15 lx blue light was higher than that in 10 lx green light (p<0.05). Light color had an extremely significant effect on thigh muscle percentage, WLP, Hx, and crude protein content (p<0.01). Light intensity had a significant effect on SEWP (p<0.05), EWP (p<0.05), lightness (L*) value (p<0.05), WLP (p<0.01), and the contents of superoxide dismutase (p<0.05), MDA (p<0.01), glutathione peroxidase (p<0.01). Conclusion: Using white LED light with 10 lx light intensity can significantly improve the chicken quality of caged Cobb broilers, improve the content of inosine acid in chicken breast and enhance the antioxidant capacity of the body. We suggest that the broiler farm can use 10 lx white LED light source for lighting in 8 to 42 days.

망막반응으로 본 각종파장가시광선조사하에서 발육한 과두(올챙이)의 식물신경계기능변조에 관하여 (Functional Disturbances Through the Retinal Pigment Reaction of the Automatic Nervous System of Tadpoles Developed under Various Visible Rays)

  • Since Kesser first described in 1934 the functional change of the autonomic nervous system caused by certain visible rays many researchers have unanimously approved that animals flashed with a red visible ray develop parasympathicotony while those flashed with a blue visible ray develop sympathicotony. On the other hand through studies made by our colleagues it is now well known that the inner-movement of the retinal pigments of frogs is stimulated in sympathicotony and is in reverse inhibited in parasimpathicotony. It is almost evident that the mechanism by which the inner-movement of the retinal pigments is due to sympathicotony derived from the excessive secretion of adrenalin. In addition , through may recent experiments on the pharmacological action of various medicines on the retinal pigments reaction of tadpoles , ranging from the excessive secretion of adrenalin. In addition , through my recent experiments on the pharmacologtical action of various medicines on the retinal pigments reaction of tadploes, raging from every developmental stage , Ifound that the movement of the retinal pigments by adrenalin is predominant in the earlier developmental stages of taopoles around 11 mm of body length, whereas other medicines fail to give any responce to the retinal pigments in such an earlier stage. When tadpoles grown to body length of 15-16m the retinal pigments move to the complete light position while kept in adrenalin solution. Based on these facts it might be well to consider that if tadpoles were grown under the visible rays for a given period, they might show a functional change of the autonomic nervous system and thereby cause of certain change in the physciological phases of the retinal reaction. Experiments were undertaken to find this matter and also to discover the simultaneous effects of the visible radiations on the developmental process of tadpoles. The results summarized as follows ; 1. The longest wave of visible rav has an effective reaction on the growth of body length of taopoles, while the shortest wave of visible ray causes the same for the metamorphoric differentiation of tadpoles. 2. When keeping two groups of tadpoles the first group of 15 mm body length grown for the period of one week and the latter group of 20 mm body length grown for two weeks under the various visible rays. swimming in adrenalin solution, the inner-movement of the retinal pigment occurs in both groups. The movement of pigments of the first group is accelerated in a sequence of blue ray \ulcorner green ray > brown ray> red ray, and that of the latter group is also accelerated in a sequence of blue ray>green ray > brown ray and red ray. 3. When keeping tow groups of tadpoles, the first group of 20 mm body length grown for the period of two weeks, the latter group of 25 m body length grown for three wheeks, under the various visible rays in sunlight, the inner-movement of the retinal pigments occurs in both groups. The movement of pigments of the first group is accelerated in a sequence of blue ray> green ray>brown ray and red ray, and that of the latter group is also accelerated in a sequence of blue ray > brow ray>red ray. 4. In order words, there facts manifest that tadpoles grown under the various visible rays reveal functional disturbances of the autonomic nervous system, at the time of 15 mm body length by adrenalin solution, which is a unique indicator illustrating the status of sympathicotony, and at the time of 20 mm body length by sunlight. This means that the longest visible ray cause sympathicotony, while the shortest visible ray causes parasympathicotony.

Pair Fed 흰쥐에 있어서 녹차의 항증체, 지질개선 및 항산화 효과 (Effects of Green Tea on Weight Gain, Plasma and Liver Lipids and Lipid Peroxidation in Pair Fed Rats)

  • 본 연구에서 pair feeding함으로서 사료 섭취량이 체중이나 지단백 대사에 미칠 요인을 제거한 상태에서 녹차분말이나 녹차추출물이 갖는 항 비만효과나 지질 강하효과를 비교 조사하였다. 녹차분말이나 추출물에서 증체 억제효과를 보였는데 이는 녹차성분이 소장에서의 지방흡수를 억제하거나 지방산화를 촉진하고 대사율을 올려 열량소비를 증가시킨다는 기존 연구결과와 이해를 같이 하는데, 예비실험에서 사료섭취량 자체가 억제됨을 감안 할 때 녹차 추출물이나 분말의 항비만 효과에 대해서 긍정적이다. 본 연구에서 녹차 추출물의 높은 콜레스테롤 흡수 억제력에도 불구하고 녹차의 콜레스테롤 강하효과는 나타나지 않았는데 이는 체 콜레스테롤 pool size에 강한 homeostasis 기질이 작용한 것이라고 생각할 수 있다. 식이 콜레스테롤과 지방의 흡수억제에 있어서 녹차분말과 녹차 추출물의 장내 유용성과 체 콜레스테롤의 합성과 담즙 생성에 있어서 EGCG를 포함한 차 카테킨 작용기전을 이해하기 위해서 보다 많은 연구가 필요하리라 생각된다. 녹차 분말과 추출물은 항산화 지표에도 정도에 다소 차이가 있으나 긍정적인 효과를 보였는데 녹차 카테킨은 수용성으로 bioavailability가 비교적 높은 polyphenol계 항산화 물질로서 녹차 잎의 건조중량 50% 이상이 카테킨이고 이중 80% 이상이 catechin gallate 형태로 강한 항산화 효력을 지닌 것으로 나타나 있다. 노화를 활성산소에 의한 세포의 손상과정이라 볼 때 식품으로서 녹차는 매우 중요한 입지에 있다. 차로 마시는 녹차의 섭취방법에서 분말이나 추출물로 섭취방법을 다양화하고 있는데 분말과 추출물의 영양적 효과를 비교해 볼 때 유리 (free)된 카테킨 형태로 추출물이 유리할 수 있지만 녹차분말이 식품으로서의 높은 선호도와 함께 HDL-콜레스테롤을 증가시키는 효과가 있다면 분말차의 섭취도 좋은 방법이다. 음료로서 따져 볼 때 일회 분량 2 g의 녹차를 3회 우려 마시는 경우 이로부터 섭취할 수 있는 항산화물질은 EGCG기준으로 $109{\sim}147\;mg$정도이고 이를 비타민 C 로 환산하는 경우 $142{\sim}168\;mg$이다. 천연의 항산화 물질을 자연스럽게 섭취하는 방법으로서 차의 음용을 생활화하는 것은 비만이나 퇴행성 또는 노화관련 질환의 예방차원에서 바람직하리라 본다.

석탄회-점토계 슬립의 분산상태에 따른 소결체의 물리적 특성 (Physical Properties of Sintered Body for Coal Fly Ash-clay Slip of Varying Dispersion State)

  • 석탄회.점토의 무게비가 7 : 3인 슬립에 대하여 그 해교정도에 따라 slip을 F(Flocculated), M(Moderate) 및 D(Dispersed)의 3종류로 제조하고 이로부터 소결체를 합성하여 시편의 물성과 슬립 특성과의 관계를 분석하였다. Slip F는 응집입자의 크기분포가 넓은 반면 slip D는 입도분포가 좁고 11 $\mu\textrm{m}$ 이상의 큰 응집입자가 거의 존재하지 않았다. 성형체의 기공은 슬립의 종류에 관계없이 1~4 $\mu\textrm{m}$ 범위 내에 분포하며 소결 후에는 1 $\mu\textrm{m}$ 이하의 미세기공은 거의 소멸된 반면, 큰 기공은 1 $\mu\textrm{m}$ 정도 더 성장하였다. Slip F의 경우에 소결 후 기공분포는 slip M과 D에 비하여 주피크의 높이가 높고 폭이 좁았으며, large pore limit (가장 큰 기공 크기)가 5.1 $\mu\textrm{m}$로 다른 슬립 성형체보다 작았다. 입자가 잘 분산된 슬립(slip D)보다는 약간 응집된 슬립(slip F)이 소결시 시편내 거대기공의 생성을 억제함으로서 높은 압축강도를 얻는데 유리하였다. 이상의 결과로부터 소결체의 기계적 특성은 기공분포에 크게 의존하며, 이는 슬립의 분산특성을 제어함으로서 가능함을 알 수 있다.

Green Light-emitting Diodes Light Stimuli during Incubation Enhances Posthatch Growth without Disrupting Normal Eye Development of Broiler Embryos and Hatchlings

  • Monochromatic green light-emitting diodes (LED) light stimuli influences the posthatch growth performance of chicks. This study was undertaken with the following objectives: i) to examine whether the green LED light stimuli induces an overheating effect by determining weight loss rate of fertile eggs during incubation period; ii) to look for the development of eyes and other primary organs at different ages of embryos and newly hatched chicks. Arbor Acres fertile broiler eggs (n = 480) were randomly assigned to 3 incubation groups and exposed to continuous white light, green light, or a dark environment (control) from the first day to 19 d of incubation. The light sourced from LED lamps with the intensity of 30 lx at eggshell level. The results showed that either green or white light stimuli during incubation did not significantly affect the weight loss rate of fertile eggs, hatching time, hatchability, chick embryo, or body weight (BW), the weight percentage of heart, liver, and eyes, as well as obvious systematic abnormalities in eye weight, side-to-side, back-to-front, or corneal diameter from 15 d of embryogenesis to 6 d of posthatch (p>0.05). Compared with the dark condition, green light stimuli during incubation tended to increase feed intake (p = 0.080), improved the BW gain of chicks during 0 to 6 day posthatch (p<0.05), and increased the percentage of pectoral muscle to the BW on 3- and 6-day-old chicks. In addition, embryos or chicks in green light had lower weight percentage of yolk retention on 19 d of embryogenesis and 1 d of posthatch in comparison to those in dark or white group (p<0.05). These results suggest that providing 30 lx green LED light stimuli during incubation has no detrimental effect on the development of eyes, heart and liver of embryos and hatchlings, but does have potential benefits in terms of enhancement of the chick growth during the early posthatch stages. In addition, the fertile broiler eggs stimulated with 30 lx green LED light during incubation does not cause an overheating effect.

요구르트의 프로바이오틱 활성과 물리화학적 및 기능적 특성에 대한 녹차 추출물의 영향 (Effect of green tea supplementation on probiotic potential, physico-chemical, and functional properties of yogurt)

  • 본 연구에서는 된장으로부터 분리된 Lactobacillus acidophilus D11 또는 Lactobacillus fermentum D37 균주로 발효시킨 요구르트의 프로바이오틱로서의 가능성, 물리 화학적 및 기능적 특성에 대한 녹차 추출물의 영향을 조사하였다. 인공 소화액에 대한 저항성과 상피 세포에 대한 부착력과 같은 프로바이오틱활성은 플레인 요구르트보다 녹차 추출물을 첨가한 요구르트에서 다소 높게 나타났는데, 이는 유산균의 수의 증가에 기인하는 것으로 추정되었다. L. acidophilus D11로 발효시킨 플레인 요구르트에 녹차 추출물을 첨가한 경우 유산균수, 유기산 함량 및 점도와 같은 요구르트의 물리 화학적 특성도 유의하게 (P<0.05) 증가하였다. 하지만 녹차 추출물은 L. fermentum D37 균주 발효시킨 요구르트의 물리 화학적 특성에는 유의한 영향을 미치지 않았다 (P>0.05). 한편, Escherichia coli O157 ATCC 43889, Salmonella enteritidis ATCC 13076 및 Salmonella typhimurium KCTC 2514에 대한 항균 활성 및 총 페놀 함량, 라디칼 소거능 및 철 환원력과 같은 항산화 활성은 L. acidophilus D11 보다는 L. fermentum D37로 발효시킨 플레인 요구르트에서 현저히 높았다. 게다가 요구르트의 항균 및 항산화 활성은 녹차 추출물의 농도에 비례하여 유의적으로 증가하였다(P<0.05). 결론적으로, L. acidophilus D11 또는 L. fermentum D37로 발효시킨 녹차 요구르트는 병원성 세균의 성장을 억제하고 체세포 내에 생성된 자유 라디칼을 제거 할 수 있는 유용한 기능성 식품으로 이용할 수 있는 것으로 판단되었다.

하천에서의 영양물질 관리를 위한 총질소 환경기준 설정에 관한 연구 (Deriving Water Quality Criteria of Total Nitrogen for Nutrient Management in the Stream)

  • The objective of this study is to suggest the water quality criteria of total nitrogen in order to efficiently manage the nutrient pollution in the stream. For this, correlations between water quality parameters were examined using the water quality data collected from the water quality monitoring network in the four rivers between 2003 and 2012. T-N showed positive correlations with T-P (0.636), COD (0.577), BOD (0.574), TOC (0.440), and SS (0.367). The statistical analysis including percentile analysis for the T-N and T-P concentrations was utilized to develop the water quality criteria of T-N. The feasibility of the suggested water quality criteria was evaluated by calculating the achievement rate to water quality target at the representative points in mid-watershed, then the draft water quality standard of T-N was suggested. The suggested water quality standard of T-N in the stream may be used to efficiently control the nutrient pollution in the public water body.
