New Frontiers of Knowledge on Nepalese Plant Science

  • Nepal, with its unique geographical and ecological features due to its abrupt rise in altitude, plays significant role in biological evolution. Existence of numerous wild relatives of the present-day cultivated agricultural crop plants in this small Himalayan nation may serve as a potential source of several yet unidentified desirable genes that are needed for future incorporation in the improvement of cultivated crop plants. This report includes 82 different wild relatives of 41 genera under 19 families of 37 agricultural crops of Nepal(Table 1). It serves as the sample of the glossary of these wild relatives of crop plants in Nepal. Under food grain crop plants of gramineae, leguminoceae and polygonaceae families, 16 different wild species namely wild rices(7 species), wild relatives of wheat plant(3 species), wild arhar(3 species), wild fingermillets(1 species) and wild buckwheat(2 species) have been identified in different parts of the country. Similarly, under vegetable crop plants of Araceae, Amaranthaceae, Crucifereae, Cucurbitaceae, Dioscoreaceae, Labiteae, Leguminosae, Liliaceae, Malvaceae, Polygonaceae, Solanaceae and Umbellifereae, 37 different wild species-wild colocasia(1 species), wild amaranths(3 species), wild leafy vegetables(2 species), wild gourds(3 species), wild cucumber(1 species), wild yams(4 species), wild mints(3 species), wild fenugreeks(4 species), wild pea(1 species), wild beans(3 species), wild garlics(2 species), wild spinach(3 species), wild lady's finger(1 species), wild spinach(3 species), wild eggplants(2 species) and wild carrot(one species) have also been identified. In case of wild relatives of cultivated orchard plants, 11 different wild species namely wild mango(one species), wild banana(one species), wild strawberry(one species), wild pear(one species), wild cherries(2 species), wild apple(one species) and wild grapes(3 species) have been identified, Among 19 different wild species of economic crop plants, five wild species of sugarcane, one species of wild sunhemp, two wild relatives of cotton, three wild relatives of rose, two wild species of tobacco, four wild species of turmeric and two wild species of tea have also been identified. This report includes only sample of the total wild species of the present-day cultivated agricultural crop plants. Further exploration on this economic botany will help the country in cataloging the wild relatives of cultivated crop plants and their future use in crop improvement.

Anthropometric Data, Nutrient Intakes and Food Sources in Overweight and Obese Korean Adult Women

  • This study was conducted to investigate the dietary intake, anthropometric data, and association between two factors and main food sources contributing macronutrients for overweight and obese females. Subjects were 85 adult females (overweight : 28, obese 57) where mean age was 38.7y. The results are summarized as follows. Mean fat percent, BMI, obesity rate were 29.3, 23.9 and 15.1%, respectively for overweight women and 32.7, 28.3, 36.4%, respectively for obese women. There were significant differences for most of the anthropometric data between groups. Fat percent for all subjects was significantly correlated with weight, waist circumference, hip circumference, mid arm circumference or skinfold thickness for the triceps, subscapular and suprailiac (p < 0.001). The parameter which showed the highest correlation coefficient (r=0.6156) with fat Percentage was the waist to hip ratio. Any significant differences were not found in dietary intake of nutrients or in diet composition between groups. The mean energy intake was 2090.1kcal (104% of RDA) for overweight women and 2113.0kcal (106% of RDA) for obese women. PFC ratio for overweight was 17 : 24 : 59 and 18 : 23 : 58 for obese subjects, which can be regarded as higher fat and lower carbohydrate percentages compared to recommended PFC ratio (15 20 65) .4) Fat intake was positively correlated (r : 0.2301, p < 0.05) with the triceps skinfold thickness, protein intake was also positively correlated with waist circumference (r=0.2668, p < 0.05) or fat weight(r: 0.2406, p < 0.05) .5) The main food items that contributed to energy intake for overweight or obese subjects were similar (rice, pork, bread, grapes, barley) except com oil in overweight or instant noodle for obese group. The subjects in this study were taking less energy from rice and more energy from pork and bread than women from 98 National Health and Nutrition Survey. Because there were no significant differences of dietary data between overweight and obese group, further investigation considering basal metabolic rate or activity would be needed.

Phenolic compounds in domestic and imported grape cultivars in Korea (국내산 포도와 수입산 포도의 페놀계화합물 함량)

  • In this study, fruit phenolics were investigated with using LC/MS and HPLC analysis in order to compare the differences between domestic ('Campbell Early', 'Kyoho', 'Heukbosuk', and 'Hongju SDS') and imported ('Crimson SDS' and 'Thomson SDS') grapes. In the case of fruit characteristics, imported grape 'Crimson SDS' and 'Thompson SDS', had lighter skin weight (300-350 mg/berry) and hard flesh (5.2-5.6 kg·f) than domestic grape cultivar. The phenolic compound contents of 'Crimson SDS' skin was higher, but resveratrol (25-29 mg/kg), quercetin (350-380 mg/kg), and myricetin (31-32 mg/kg) contents were similar in to those of 'Hongju SDS'. The anthocyanin content was different from differed between grape cultivars. 'Hongju SDS' grape was showed higher in Delphinidin-3-glucose (D3G) levels, and 'Crimson SDS' was showed higher in Peonidin-3-glucoside (P3G) levels. The contents of phenolic compounds were investigated differently for each grape berry part. Catechin, epicatechin, procyanidin B1, and B2 were found in grape seeds.

Neuroprotective effects of resveratrol on 6-hydroxydopamine-induced damage of SH-SY5Y cell line (6-Hydroxydopamine 유발 SH-SY5Y 세포주 손상에 대한 resveratrol의 신경보호 효과)

  • Parkinson's disease is known to exhibit progressive degeneration of the dopaminergic neurons in the substantia nigra via inhibition of glutathione metabolism. It is well known that 6-Hydroxydopamine (6-OHDA) induces Parkinson's disease-like symptoms, while resveratrol (3,5,4'-trihydroxystilbene) has been shown to have anti-inflammatory and antioxidant effects. In the present study, we investigated the neuroprotective effects of resveratrol, a phytoalexin found in grapes and various plants, on 6-OHDA-induced cell damage to the SH-SY5Y human neuroblastoma cell line. Resveratrol (5 and 10 ${\mu}M$) inhibited 6-OHDA (60 ${\mu}M$)-induced cytotoxicity in SH-SY5Y cells and induced a reduction of the number of apoptotic nuclei caused by 6-OHDA treatment. Additionally, the total apoptotic rate of cells treated with both resveratrol (10 ${\mu}M$) and 6-OHDA (60 ${\mu}M$) was less than that of 6-OHDA treated cells. Resveratrol also dose-dependently (1, 5 and 10 ${\mu}M$) scavenged reactive oxygen species (ROS) induced by 6-OHDA in SH-SY5Y cells and prevented depletion of glutathione in response to the 6-OHDA-induced cytotoxicity in the glutathione assay. Overall, these results indicate that resveratrol exerts a neuroprotective effect against 6-OHDA-induced cytotoxicity of SH-SY5Y cells by scavenging ROS and preserving glutathione.

Induction of S Phase Arrest of the Cell Cycle by Piceatannol is Associated with Inhibition of Telomerase Activity in Human Leukemic U937 Cells (Piceatannol에 의한 인체 혈구암세포의 증식 억제 및 telomerase 활성 저하)

  • Piceatannol is a polyphenol that is found in abundant quantities in grapes and wine. Although recent experimental data revealed the anti-cancer potency of piceatannol, the molecular mechanisms underlying the antileukemic activity have not yet been studied in detail. In the present study, we investigated further possible mechanisms by which piceatannol exerts its anti-proliferative action in cultured human leukemia U937 cells. Exposure of U937 cells to piceatannol resulted in growth inhibition and induction of apoptosis as measured by MTT assay and flow cytometry analysis, which was associated with S phase arrest of the cell cycle. Piceatannol treatment markedly inhibited the activity of telomerase, and the levels of human telomerase reverse transcriptase (hTERT) and telomerase-associated protein-1 (TEP-1), main determinants of the telomerase enzymatic activity, were progressively down-regulated by piceatannol treatment in a dose-dependent fashion. However, the levels of cyclooxygenases (COXs) expression and prostaglandin E2 (PGE2) release were not changed in piceatannol-treated U937 cells. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of piceatannol.

CUMAP : A Chill Unit Calculator for Spatial Estimation of Dormancy Release Date in Complex Terrain (Chill Unit 축적과 휴면해제시기 공간변이 추정 프로그램 : CUMAP)

  • A chill unit has been used to estimate chilling requirement for dormancy release and risk of freezing damage. A system that calculates chill units was developed to obtain site-specific estimates of dormancy release date for grapes and evaluated in Baekgu myun near Kimje City, Chunbuk, Korea from September 2002 to March 2003. The system utilized daily minimum and maximum temperature maps generated from spatial interpolation with temperature correction for topography. Hourly temperature was temporally interpolated from the daily data using a sine-exponential equation (Patron and Logan, 1981). Hourly chill units were determined from sigmoid, reverse sigmoid, and negatively increasing sigmoid functions based on temperature ranges and summed for 24 h. Cumulative daily chill units obtained from measurements did not increase until 20 October 2002, which was used as a start date for accumulation to estimate the dormancy release date. As a result, a map of dormancy release date in the study area was generated, assuming 800 chill units as a threshold for the chilling requirement. The chill unit accumulation system, implemented using Microsoft Visual Basic and C++ (Microsoft, Redmond, WA, USA), runs in the Windows environment with ArcView (ESRl Inc., Redlands, CA, USA).

Electron Microscopical Description on the Egg Stalk-like Structure of the Rockfish, Sebastes inermis (Teleostei: Scorpaenidae) (볼락 (Sebastes inermis) 난병 유사구조의 전자현미경적 기재)

  • The ovary of the rockfish, Sebastes inermis was cystovarian condition and consists of numerous ovarian lobules. Previtellogenic oocytes were embedded in the connective tissue of the ovarian lobules. As the oocytes grows, it protruded into the ovarian cavity and form grapes cluster-like structure with egg stalk-like structure. Yolk granules and zona radiata were not observed in the embedded early oocytes about 20 $\mu$m in diameter. In the oocytes about 80 $\mu$m in diameter, yolk granules with electron dense were abundant in the ooplasm near the egg stalk-like structure. However, pinocytosis was not obsened in the pore canal system of the zona radiata. In this stage, microfilaments, developed endoplasmic reticula, tubular mitochondria, granular materials with low electron density and active multivesicular bodies were observed in the egg stalk-like structure. This results suggest that the egg stalk-like structure of the rockfish are related with the oocyte attachment and exogenous vitellogenesis of the intial vitellogenic oocyte.

Identification of Volatile Compounds of 4 Grape Species by Storage Conditions (전자코와 GC/MS를 이용한 포도 품종별 저장 조건에 따른 휘발성 향기 성분 연구)

  • Volatile flavor compounds of 4 grape species (Campbell, Sheridan, Red globe, and Meoru) were identified during 3-day storage at either $4^{\circ}C$ or room temperature. Each sample was analyzed by solid-phase micro-extraction (SPME) method combined with gas chromatography-mass spectrometry. Also electronic nose composed of 12 different metal oxide sensors was used to differentiate flavors of grapes. Sensitivities (delta $R_{gas}/R_{air}$) of sensors from electronic nose were obtained by principal component analysis (PCA). Proportion of the first principal component was 99.30% at $4^{\circ}C$ and 99.36% at room temperature, respectively. In our result, flavor patterns of grape can be differentiated according to the storage period. The major volatile flavor compounds were 1-hexanol, hexanoic acid and its ethyl ester, and phenylethyl alcohol with the presence of butanoic acid and its ethyl ester, acetic acid, benzeneacetic acid and its ethyl ester.

Effect of Resveratrol on Cell Differentiation and Mineralization in Cultured Odontoblasts

  • Resveratrol (3,4',5,-trihydroxystilbene), a phytoalexin present in grapes, exerts a variety of actions to reduce superoxides, prevents diabetes mellitus, and inhibits inflammation. Resveratrol acts as a chemo-preventive agent and induces apoptotic cell death in various cancer cells. However, the role of resveratrol in odontoblastic cell differentiation is unclear. In this study, the effect of resveratrol on regulating odontoblast differentiation was examined in MDPC-23 mouse odontoblastic cells derived from mouse dental papilla cells. Resveratrol significantly accelerated mineralization as compared with the control culture in differentiation of MDPC-23 cells. Resveratrol significantly increased expression of ALP mRNA as compared with the control in differentiation of MDPC-23 cells. Resveratrol significantly accelerated expression of Col I mRNA as compared with the control in differentiation of MDPC-23 cells. Resveratrol significantly increased expressions of DSPP and DMP-1 mRNAs as compared with the control in differentiation of MDPC-23 cells. Treatment of resveratrol did not significantly affect cell proliferation in MDPC-23 cells. Results suggest resveratrol facilitates odontoblast differentiation and mineralization in differentiation of MDPC-23 cells, and may have potential properties for development and clinical application of dentin regeneration materials.

Volatile Flavor Components in Various Varieties of Grape(Vitis vinifera L.) (포도의 품종별 휘발성 향기성분 분석)

  • Volatile flavor components in three grape(Vitis vinifera L.) varieties were extracted by SDE(Simultaneous steam distillation and extraction) method using the mixture of n-pentane and diethylether(1:1, v/v) as an extract solvent. Grapes of the following varieties were studies : Blackolympia, Campbell and Delaware. The volatile extracts were analyzed by GC-FID and GC/MS. The totals of 77, 72 and 74 volatile flavor components were identified in Blackolympia, Campbell and Delaware, respectively. (E)-2-Hexenal(20.36%), diethylacetal(18.03%), hexanal and ethyl acetate were contained as the main compounds of Blackolympia. In Campbell, ethyl acetate(30.81%) was relatively more abundant than other compounds and among functional groups, C$\_$6/ aldehydes and alcohols were major constituents of the extract. On the other hand, in Delaware, alcohols was the major constituent group and (E)-2-hexenal(21.07%) and (E)-2-hexena1-ol(19.43%) were the main compounds. All of three grape varieties contained a large amount of hexanal, (E)-2-hexenal, hexanol, (E)-2-hexen-1-ol, thus C$\_$6/-compounds proved to be major volatile components of grape and small amount of terpenols were only detected from Delaware.
