외과영역에서의 감염증에 대한 Vistamycin 의 사용경험 (Clinical Studies on Ribostamycin sulfate)

  • Ribostamycin sulfate is a new aminoglycoside antibiotic which is effective against organisms resistant to other conventional antibiotics, Particularly E. coli, and Staphylococcus group. Our clinical studies were performed on 20 patients who were admitted to Thoracic Surgical Dept of National Medical Center from May '77 to September '77, and the following results were obtained; 1. Among 20 patients, 13 were excellently improved and 4 responded moderataly, but 3 patients were not improved. So, 85% of the patients were controlled with Ribostamycin sulfate administration. 2. The rate of effectiveness of Ribostamycin sulfate was 85.7% against E. coli, 92.3% against Staphylococcus, and 75% against klebsiella, revealing 83.3% in average. 3. No side effects were observed. 4. From our clinical studies, Ribostamycin sulfate is expected to possess in high therapeutic value in surgical infectins caused by both gram positive and negative microorganisms.

버섯 추출물이 인체 병원성 균에 미치는 항균활성의 특성 (Characteristics of Antimicrobial Activities for the Human Pathogenic Microorganism by Extracts from Korean Mushrooms)

  • 버섯 추출물의 병원 미생물에 대한 항균활성을 알아보기 위하여 66종의 버섯을 채집 동정하고, 이들을 석유에테르, 80% 에탄올 및 증류수로 추출하여 198종의 추출물을 얻은 후 19종의 인체 병원성 미생물에 대한 항균활성을 검정하였다. 그 결과, C. consors의 에탄올 추출물외에 13종의 추출물이 4종의 효모에, P. ostreatus의 에탄올 추출물 외 27종의 추출물이 3종의 사상균에, A. subrutilescons의 추출물외 78종의 추출물이 5종의 그람 음성세균에, 그리고 A. pseudoporphyria의 추출물외 59종의 추출물이 5종의 그람 양성세균에 항균활성을 보였다.

NADP+-Dependent Dehydrogenase SCO3486 and Cycloisomerase SCO3480: Key Enzymes for 3,6-Anhydro-ʟ-Galactose Catabolism in Streptomyces coelicolor A3(2)

  • Agarose is a linear polysaccharide composed of ᴅ-galactose and 3,6-anhydro-ʟ-galactose (AHG). It is a major component of the red algal cell wall and is gaining attention as an abundant marine biomass. However, the inability to ferment AHG is considered an obstacle in the large-scale use of agarose and could be addressed by understanding AHG catabolism in agarolytic microorganisms. Since AHG catabolism was uniquely confirmed in Vibrio sp. EJY3, a gram-negative marine bacterial species, we investigated AHG metabolism in Streptomyces coelicolor A3(2), an agarolytic gram-positive soil bacterium. Based on genomic data, the SCO3486 protein (492 amino acids) and the SCO3480 protein (361 amino acids) of S. coelicolor A3(2) showed identity with H2IFE7.1 (40% identity) encoding AHG dehydrogenase and H2IFX0.1 (42% identity) encoding 3,6-anhydro-ʟ-galactonate cycloisomerase, respectively, which are involved in the initial catabolism of AHG in Vibrio sp. EJY3. Thin layer chromatography and mass spectrometry of the bioconversion products catalyzed by recombinant SCO3486 and SCO3480 proteins, revealed that SCO3486 is an AHG dehydrogenase that oxidizes AHG to 3,6-anhydro-ʟ-galactonate, and SCO3480 is a 3,6-anhydro-ʟ-galactonate cycloisomerase that converts 3,6-anhydro-ʟ-galactonate to 2-keto-3-deoxygalactonate. SCO3486 showed maximum activity at pH 6.0 at 50℃, increased activity in the presence of iron ions, and activity against various aldehyde substrates, which is quite distinct from AHG-specific H2IFE7.1 in Vibrio sp. EJY3. Therefore, the catabolic pathway of AHG seems to be similar in most agar-degrading microorganisms, but the enzymes involved appear to be very diverse.

  • Marine pathogenic bacteria, such as Streptococcus parauberis, Edwardsiella tarda and Vibrio harveyi, can cause lethal infections in farmed fish, ozone and antibiotics, are employed to sterilize waters used for rearing fish to mitigate this threat. The most widely used method is treatment with sodium hypochlorite solution. However, the maintenance of a constant concentration of chlorine in rearing waters can be difficult. We investigated the potential of a mixed oxidant (MO) solution generated by electrolysis of sea water to improve water quality. We measured the survival rates of fish pathogenic bacteria exposed to different concentrations of MO (0.5, 1.0, 1.5 and 2.0 MO) and sodium hypochlorite (0.5, 1.0, 1.5 and 2.0 ppm) for various lengths of time (0, 5, 10, 15, 20, 25 and 30 min). We found a time-dependent decrease in the survival rates of the tested pathogenic microorganisms. The sterilization effect of the MO solution on pathogenic organisms was greater than that of sodium hypochlorite for gram-negative and gram-positive bacteria. We conclude that MO solution produced by electrolysis could be used to maintain a constant chlorine concentration in aquaculture systems.

재사용 흡인 용기의 미생물 오염도 및 재사용 흡인 용기와 일회용 흡인 용기의 비용 분석 (Microbial Contamination of Reusable Suction Container and Cost Analysis of Reusable Suction Container and Disposable Suction Container)

  • Purpose: The purpose of this study was to check the degree of residual microbial contamination after disinfection of reusable suction containers, used in an intensive care unit (ICU) and present basic data for efficient use through cost analysis in comparison to disposable suction containers. Methods: This study was conducted on 32 reusable suction containers used in an ICU on a selected specific day. After disinfection and washing, specimens were collected from the used containers and cultured to check for microbial contamination. Additionally, a comparative narrative study analyzes the cost of using reusable suction containers and disposable suction containers. Data were analyzed with the SPSS WIN 20.0 program using real numbers and percentage ${\chi}^2$-test. Results: As a result of the study, microorganisms were found in all samples where in 30 were gram-positive (62.5%) while 13 were gram-negative (27.1%). Based on level of contamination, microorganisms were less than 10CFU/ml in 18 samples (56.3%); 11-99CFU/ml in six samples (18.8%); and more than 100CFU/ml in eight samples (25%). Cost per day for a reusable suction container was $10,655+{\alpha}$ while cost per day for a disposable suction container was 10,666 won. Conclusion: This study found that reusable suction containers, even after disinfection, accounted for factors of potential infection as well as microbial contamination. So, disposable suction containers are superior in cost-effectiveness and highly efficient for use with infected patients.

사료용 생균제 개발을 위한 마늘 내성 유산균의 분리 (Isolation of Garlic Resistant Lactic Acid Bacteria for Feed Additives)

  • 마늘의 항균력에 저항성을 갖는 유산균을 분리하기 위해서 마늘 추출물이 첨가된 배지에서 생육하는 112 균주를 김치, 젓갈, 장아찌류로부터 분리하였다. 이 중에서 14균주에 대해서 내산성 및 내담즙성 그리고, 가축 병원성 세균인 Salmonella choleraesuis, Escherichia coli, Staphylococcus aureus, Shigella flesneri에 대해 항균력을 조사한 결과, 파김치 유래의 P'GW50-2 균주가 가장 우수한 특성을 지니고 있어 생균제로 이용 가능한 균주로 선발하였다. 선발된 균주는 16S rRNA 유전자의 염기서열 분석결과 Lactobacillus plantarum으로 동정되었고 L. plantarum TJ-LP-002으로 명명하였다. L. plantarum TJ-LP-002는 Bacillus cereus, S. aureus, Clostridium perfringens와 같은 Gram 양성균과 Aeromonas hydrophila, E. coli, Pseudomonas, Salmonella, Shigella와 같은 Gram 음성균에 대해서 비교적 넓은 항균활성을 나타내었다.

각종 소독제가 구강균총에 미치는 영향에 관한 실험적 연구 (EXPERIMENTAL STUDY ON THE EFFECTS OF VARIOUS DISINFECTANTS TO ORAL MICROFLORA)

  • Though oral microgranisms were among the first to be observed by humans, the interest in oral microbiology lagged. When it became apparent that the oral microflora did influence systemic disease of the body, interest was aroused in the nature and kinds of the microgranisms. The risk of infection in dental procedures is due to the abundant flora of the mouth. This hazard can be reduced to some extent by the use of a local disinfectant. The present studies were undertaken to evaluate and compare the various disinfectants which are commonly used in clinics and hospitals. The results were as follows. 1. The bactericidal activity of the disinfectants mainly depends upon the kinds of the agents, not upon the kinds of the microorganisms. 2. In H₂O₂(3%), the bactericidal activity was greatly related to the contact time. So, at least 4 minitues of contract time was required to use it as an oral antiseptic. 3. In ethyl alcohol (70%), Pseudomonas aeruginosa and Streptococcus salivarius surived a little after 15 seconds of contact time, but, no other colony was discovered after more than 15 seconds of contact time in any kins of microorganisms. 4. Merthiolate (0.1%) showed low antibacterial activity, more in Gram-positive organisms and less in Gram-negative organisms. 5. Benzalkonium chloride (0.1%) and povidone-iodine (10%) showed the most excellent results, revealing no surviving organisms only after 14 seconds of contact time.

구취에 따른 구강 내 형태별 세균의 분포 및 운동성, 진균 균사 분석 (Analysis of fungal hyphae, distribution and motility of bacteria in oral cavity according to halitosis)

  • Halitosis is primarily caused by bacterial decay. The bacteria, which originate from biofilms such as dental plaque, show abnormal proliferation due to dental caries, periodontal diseases, soft tissue infections, and tongue diseases. Most studies on halitosis have exclusively focused on gram-negative bacteria in the oral cavity rather than on general oral microorganisms including oral fungi. This study analyzed oral fungal hyphae, as well as distribution and motility of oral microorganisms, and provided basic data on the control of halitosis. Our results revealed that the greater is the number of cocci bacteria, the higher is the halitosis value, or bad breath value (BBV), suggesting that cocci have a strongly positive correlation with halitosis (r=0.379, p=0.030). Moreover, there was no significant difference in the morphology or distribution of motile bacteria and motility score, with respect to BBV. Lastly, we investigated the relationship between halitosis and oral fungal hyphae. We found that a higher BBV corresponded with a greater number of fungal hyphae and that patients with fungal hyphae scored a higher BBV. However, this result was not statistically significant. In conclusion, this study provided the preliminary data on oral microorganisms and halitosis, but further studies are needed to analyze the relationship between oral microorganisms and halitosis.

흑마늘박 추출물의 항균 활성과 미생물 제어를 위한 시금치 세척에의 이용 (Antimicrobial Activity of Black Garlic Pomace Extract and Its Application to Cleansing of Fresh Spinach Leaves for Microbial Control)

  • 본 연구에서는 흑마늘 진액 가공 후 발생하는 흑마늘박의 활용 가능성을 검토하고자 흑마늘박 추출물의 항균성과 시금치에 대한 세척 적용 효과를 분석하였다. 흑마늘박 추출물은 주요 병원성 미생물인 L. monocytogenes, S. aureus, E. coli O157:H7, S. Typhimurium에 대해 모두 항균성을 나타냈으며, 특히 그람 양성균인 L. monocytogenes와 S. aureus에 더 효과적으로 작용하였다. 시금치에 0.5% 흑마늘박 추출물을 처리한 후 $4{\pm}1^{\circ}C$에서 9일 동안 저장하면서 미생물 수 변화를 측정하였다. 흑마늘박 추출물 처리는 대조구와 비교하여 저장 9일 동안 시금치의 총 호기성 세균 수를 1.23~1.35 log CFU/g, 효모 및 곰팡이 수는 0.82~1.12 log CFU/g 감소시켰다. 또한, 흑마늘박 추출물 처리는 저장 9일 동안 시금치의 Hunter 색도 값 및 총색차 값을 저장 초기와 유의적인 차이 없이 지속시켰기에 색도 품질 유지 측면에서 단순 물 세척 처리구보다 더 효과적이라고 판단된다. 따라서 본 연구 결과를 통해 흑마늘박 추출물은 병원성 미생물에 대해 높은 항균성을 가지면서 동시에 시금치와 같은 신선 농산물의 미생물 제어를 위한 효과적인 세척 처리 물질로써 활용될 수 있다고 판단된다.

생강나무(Lindera obtusiloba Blume)와 초피나무(Zanthoxylum piperitum DC) 추출물의 항균활성 (Antimicrobial activities of Lindera obtusiloba Blume and Zanthoxylum piperitum DC extracts)

  • 생강나무 줄기와 초피 과피의 60% ethanol과 열수 추출물의 식중독 균에 대한 항균 활성과 추출물의 열안정성에 대하여 알아보았다. 100 mg/mL의 농도에서 생강나무 줄기의 60% ethanol 추출물은 MRSA에 대하여 31.50 mm, 열수추출물은 S. aureus에 대해 25.5 mm의 생육 저해환을 보이며 가장 높은 항균활성을 보였으며, B. cereus에 대해서는 가장 낮은 항균활성을 보였다. 같은 농도에서 초피 과피의 60% ethanol 추출물의 항균효과는 S. aurues와 B. cereus에 대해 25 mm의 생육 저해환을 나타냈으며, 열수추출물은 S. aurues 에 대해 22 mm의 생육저해환을 나타내 항균 효과가 가장 크게 나타났으며, E. coli 에 대해서는 낮은 항균활성을 나타내었다. 생강나무 줄기와 초피 과피의 60% ethanol, 열수 추출물 모두 농도가 낮아짐에 따라 생육 저해환의 크기가 작아지는 현상을 보였으며, ethanol추출물이 열수 추출물에 비해 다소 높은 항균력을 나타냈다. 또 추출물의 열안정성 실험에서 열처리군과 비열처리군(대조구)을 비교했을 때 항균 효과가 감소하지 않는 것으로 보아 생강나무 줄기와 초피 과피 추출물은 열에 안정하다고 판단된다. 본 연구 결과 생강나무 줄기와 초피 과피 추출물에 미생물 부패 방지에 효과적인 항균 물질이 함유되어 있으며 비교적 열에 안정하므로 천연보존료로서 산업적 응용이 가능할 것이라 사료된다.
