Bioluminescence 반응에 의한 ATP 측정을 이용한 젖소 유방염 진단에 관한 연구 (Diagnostic test for bovine mastitis by the determination of ATP based on firefly bioluminescence)

  • This study was carried out to diagnostic test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are follow; 1. The infection rate of bovine mastitis investigated with 521 cows in 47 dairy farms were found to be 3.6% of clinical form and 44.1% of subclinical form according to the degree of infection. 2. The light yield produced in firefly bioluminescence system was proportional to the concentration of ATP giving stright line within the range of 100PM~1uM. 3. When the number of somatic cell in milk was determined by the ATP assay and compared with three conventional methods such Fossomatic, California mastatic test (CMT), and rolling ball viscometer (RBV), it was shown that r=0.92 for Fossomatic, 0.63 for CMT and 0.7 for RBV. 4. The microorganisms causing mastitis were isolated Staphylococcus sp. (53.3%), Streptococcus sp. (17.9%), Micrococcus sp. (13.5%), Gram negative bacilli (6.3%), Gram positive bacilli (5.5%) and Yeast-like fungi (5.4%). 5. The endogeneous ATP levels of bacteria in a raw milk determined by the firefly bioluminescence system and compared with the results of the conventional methods. The correlation was 0.88 for raw milk.

  • Blood is one of the most important clinical specimens for the isolation of bacteria. A rapid isolation and a high isolation rate of bacteria are very important in blood culture because bacteremic patients are mostly in grave condition. Various blood culture media which support growth of most fastidious bacteria are available commercially. However, growth of bacteria are frequently delayed because of antibacterial activity of blood. Sodium polyanethol sulfonate(Liquoid) has been reported to inactivate the antibacterial substance and disrupt phagocytic cells. The beneficial effect of SPS is well recognized in the isolation of gram-positive bacteria. However, the effect does not seem to be prominent for gram-negative bacilli isolation mainly due to the rapidity of their growth. It has been experienced with Sal. typhi that the growth is much slower than that of other gram-negative bacilli. For the rapid growth of the organism, use of bile broth has been recommended. Although Sal. typhi is the most frequently isolated organism at present, about one half of total isolates are other organisms and, in case bile broth is used, other media which support growth of these organisms should be used together. Fluid thioglycollate medium(FTM) which is always used in blood culture to isolate anaerobes is inferior to brain heart infusion(BHI) for the isolation of aerobes. This study was done to determine the effect of SPS on the isolation of Sal. typhi from blood. During the Sep. 1973 to Sep. 1974 study period, 2460 blood cultures were made from the Severance hospital patients: BHI and FTM sets 1431 specimens, BHI with SPS(0.05%) and FTM sets 396 specimens, BHI and FTM with SPS sets 359 specimens, BHI and BHI with SPS sets 274 specimens. Mean incubation time required for the macroscopic detection of growth of Sal. typhi were 3.5 days on BHI and 2.7 days on BHI with SPS. The 0.8 day difference was statistically significant. On FTM the mean incubation time was 3.8 days while it was 2.9 days on FTM with SPS. The 0.9 day difference was statistically significant. The result on BHI with and without SPS sets showed faster growth on BRI with SPS in 7 specimens and slower growth in one specimen and the remaining 12 showed growth at the same time. These specimens had mean incubation time of 3.2 days on BHI and 2.3 days on BHI with SPS. The 0.9 day difference was statistically significant. This study indicates beneficial effect of SPS for the rapid isolation of Sal. typhi from clinical blood specimens.

건유기 유방염 치료제 Nophazin리 야외응용시험 (A Field Trial as Dry-cow Therapy Using Nophazin (norfloxacin mixed cephalexin) in Mastitis Control)

  • A field test using Nophazin (drying off period therapeutics) was done from May, 1989 to October, 1989. 299 quarters of 75 cows from 28 herds were tested. The results were summarized as follows. 1. At the day 7 after parturition, CMT-was 81.60%, clots were removed perfectly, removal rate of hard, hot, pain was 99.6%, only one hard 0.35% was maintained. Recovery rate of general symptoms was 100%. 2. Recovery rate in Nophazin treatment was 100% in Micro-coccus SPP, Corynebacterium SPP, and unisolated, 97.06% in Enterobacterid SPP, 95.58% in Staphylococcus SPP, 91.67% in other Bacilli, 89.19% in Streptococcus SPP, Overall recovery rate was 95.32%, And fungi did not react. 3. The ratio of isolated microorganisms was Staphylococcus SPP 49.8%, Streptococcus SPP 16.3%, Enterobacteria SPP 15.0%, Micrococcus SPP 7.9% Corynebacterium SPP 4.4%, Pseudomonas SPP 2.6%, Bacilli 2.6% and Fungi 1.3%. 4. The range of sensitivity of each microorganism to therapeutics is as follow : Staphylococcus SPP 61.1∼94.7%, Streptococcus SPP 51.4∼91.9%, Micrococcus SPP 44.4∼94.4%, Corynebacterium SPP 70∼80%, Enterobacteria SPP 11.8∼88.2%, and other Gram negative bacilli 8.3∼83.3%. Overall sensitivity was in the range of 50.4∼92.0%, Microorganisms showed the highest sensitivity (80∼94.7%) to Nophazin. 5. New intramammary infection rate was 3.96% (9 strains) at paturition, 8.37% (19 strains) at the day 7 after paturition. Persistent organism wits 6.17% .(14 strains). 6. There was no side effect of Nophazin treatment.

Multiple liver infection by Sphingomonas sp. and Bacillus sp. in a wild rodent (Apodemus agrarius): a case report

  • The first case of liver infection caused by Sphingomonas sp. and Bacillus sp. in a wild rodent is reported. A captured wild rodent, Apodemus agrarius (A. agrarius), presented with multiple liver abscess-like nodules (diameter 0.7~2.4 mm) in which Gram-positive and Gram-negative bacilli were detected simultaneously. These were grown in aerobic and anaerobic cultures, respectively, and were identified as Sphingomonas sp. and Bacillus sp., respectively, according to 16S rRNA sequencing.

  • In this study, specimens such as tongue, supragingival and subgingival biofilm were taken from total 20 scaling subjects who visited the oral prophylaxis practice lab at department of dental hygienics, J Health College in order to observe bacterial distributions and morphology using scanning electron microscopy(sem). as a result, this study came to the following conclusions: 1. According to observation of tongue, supragingival and subgingival biofilm through sem, it is found that there are round colonies of gram-positive cocci and gram-negative bacilli on blood agar medium. 2. The observation of bacterial morphology on dental biofilm through sem, cocci in chain cocci in cluster and bacillus(rod) respectively. 3. For tongue biofilm, it is found that a variety of bacterial species are detected, such as Granulicatolla adiacens(1), Gemella morbillorum(3), Streptococcus mitis(2), Streptococcus sanguinis(1), Aerococcus viridans (2), Streptococcus equinus(1), Leuconostoc spp.(1), Gemella haemolysans (1) and Lactococcus lactis spp.(1) respectively. 4. For supragingival biofilm, it is found that a variety of bacterial species detected, such as Aerococcus viridans(1), Gemella haemolysans(2), Leuconostoc spp.(2), Gemella morbillorum(1) and Pseudomonas fluoescens (1) respectively. 5. For subgingival biofilm, it is found that a variety of bacterial species detected, such as Leuconostoc spp.(1), Staphylococcus lugdunensis(1) and Streptococcus salivarius(1) respectively.

침술후 발생한 Prevotella intermedia/nigrescens에 의한 종격동염 -치험1례- (Mediastinitis Caused by Prevotella Intermedia/Nigrescens Occurred after Acupuncture -A case report-)

  • A 54-year-old male patient was admitted due to dyspnea with chest pain and reddish swelling in the right lteral neck and right upper which developed 2 or 3 days age. He was treated with acupuncture in the neck about one week ago prior to admission. CT scans of the chest showed density of air and abnormal soft tissue which suggested abscess of the anterior mediastinum and subcutaneous tissue. He underwent cervical and mediastinal drainage with closed thoracostomy and antibiotic therapy. Black-pigmente anaerobic nonspore-forming gram-negative bacilli were isolated from the mediastinal pus and were identified as Prevotella intermedia/nigrescens upon performing biochemical tests and API rapid ID 32A (bioMeriux, France) kit. He underwent decortication of the right side because of loculated empyema on 41st postoperative day. He was fully recovered and discharged on the 82nd hospital day. This was thought to be another case of descending necrotizing mediastinitis not caused by peritonsilar abscess but by cellulitis developed after acupuncture. Early deterction of mediastinits and aggressive drainage of mediastinal abscess are important.

SCC, CMT와 세균검사에 의한 유방염의 지도효과 연구 (A Study on the Effects of Bovine Mastitis Control Using SCC, CMT and Pathogens)

  • The study for a effect of monitoring on bovine mastitis was conduced for a year from Dec, 1989 to Nov, 1990, Sampling the bulk milk of 350 herds in Inchon city and out of 10 herds among them were carried out herds guidance, CMT, SCC, isolation of pathogens and antibiotic sensitivity tests. The results obtained were summarized as follows 1. Annual mean SCC of 1213 herds was 558, 000 cell /ml 2. The number of SCC below 500, 000 cell /ml to quarters for herds guidance was at 1st 77. 0%, End 84.8% and 3rd 80.4%. The is shown that milk quality was steadly improved. 3. The most number of isolated pathogens of bovine mastitis was Staphylococcus SPP - 402(47.2%) Streptococcus SPP - 80(18.7% ) 4. The highest rate of antibiotic sensitivity test was Stapylococcus SPP - cephalothin(76.7%) Streptococcus SPP - ampicillin(77.5%) Gram negative bacilli - tetracyclin(76.0%) 5. The effect of monitoring on bovine mastitis was improved showing that at 1st 49.0% to 3rd 72.0%

  • A halophilic bacterium was isolated from domestic marine. The bacterium was gram negative and motile. Transmission electron micrograph after cultivating for 6-, 20-, 72-, and 144 hrs showed that it was bacilli and contained intracellular granules, which were pleomorphic, larger in density by the time and considered to be PHB since they were positive on the sudan black B staining. The presence of sodium chloride was critical, because the isolated marine bacterium could not multiply and even produce any immunostimulant in the deficiency of sodium chloride. The strain produced an immunostimulant, which was investigated for the biological characteristics. The optimal conditions for the production of the immunostimulant were 1 % dextrose and 1 % yeast extract in artificial sea water for carbon and nitrogen sources, respectively. The initial pH and growth temperature for the production were 8.0 and $30^{\circ}C$ under the presence of oxigen, respectively.

  • Raoultella species are gram-negative, non-motile, aerobic bacilli that are primarily considered as environmental bacteria. Raoultella planticola is reportedly a rare cause of human infections. Also, the definite pathological mechanism of Raoultella planticola is currently unknown. We report a case of pneumonia caused by Raoultella planticola.

족부에 발생한 방선균증 - 1예 보고 - (Actinomycosis of the Foot - A Case report -)

  • The Actinomyces organism is inherent in human as normal flora found in the mouth. Outside of the mouth, this organism can cause actinomycosis. Involvement of the extremity is very rare and atypical. The authors have experienced a case of actinomycosis of the foot and report with the literature review. Histopathologic finding shows multiple sulfur granules. In histochemical stains, periodic acid-Schiff, Gomori methionine silver, and gram stain were positive, and acid fast bacilli stain was negative. The patient was treated with surgical excision and antibiotic administration with Ampicillin for 6 months.
