Large-Scale Culture of Hepatitis A Virus in Human Diploid MRC-5 Cells and Partial Purification of the Viral Antigen for Use as a Vaccine

  • A large-scale culture of hepatitis A virus in human diploid MRC-5 cells was conducted. In a roller bottle culture, the virus was grown to a maximum titer in 3 weeks after infection. Over 95% of the cell-associated virus was excreted after culturing the infected cells in suspension media without fetal bovine serum for 3 days. The cultured virus was inactivated with formalin, concentrated by ultrafiltration, and partially purified by ultracentrifugation in a non-ionic gradient medium of Renocal. Two separate peak fractions showing high anti-HAY ELISA titer were pooled and about 40% of HAV antigen was recovered by this purification procedure. Of the partially purified vaccine, the protein pattern in SDS-PAGE and immunogenicity in mice were compared with a commercial HAV vaccine. In SDS-PAGE, the purified vaccine in this study and the commercial vaccine showed almost the same protein pattern. The seroconversion rate of the purified vaccine in mice was not different from that of the commercial vaccine. Therefore, we could prepare a good grade of HAV vaccine by a simple purification procedure although the purification itself was not completed.

  • The results of conventional operative resection of diffuse subaortic stenosis[tunnel subaortic stenosis] have been less than satisfactory. We have performed modified Konno operation to enlarge the outflow tract of both ventricle with a patch in three patients with diffuse tunnel subaortic stenosis in the department of Thoracic and Cardiovascular Surgery, Seoul National University Children`s Hospital. Preoperative left ventricular aortic pressure gradients were 135 mmHg, 50 mmHg, 80 mmHg respectively in these patients. After right ventriculotomy, the septotomy was done and extended beyond the limit of the stenosis and fibrous and/or muscular tissue was removed from each edge of septal incision. After adequate widening of the tunnel subaortic stenosis with various patches, right ventricle was closed primarily or with prosthetic patches successfully. One patient who was diagnosed as complete atrioventricular septal defect had complete AV block preoperatively and was implanted permanent pacemaker, and others who were sinus rhythm preoperatively have no serious postoperative rhythm disturbance. One patient developed mild postoperative aortic insufficiency and one who had preoperative aortic insufficiency has still same grade of aortic insufficiency after operation. All had good postoperative hemodynamics without any postoperative residual pressure gradient.

Structure and Characteristics of Diffusion Flame behind a Bluff-Body in a Divergent Flow(I) (확대유로내의 Bluff-Body 후류확산화염의 구조 및 특성 (1))

  • An experimental study is carried out on turbulent diffusion flames stabilized by a circular cylinder in a divergent duct flow. A commercial grade gaseous propane is injected from two slits on the rod as fuel. Flame stability limits, as well as size and temperrature of recirculation zone, are measured by direct and schlieren photographs to clarify the characteristics and structure of diffusion flames and to assess the effect of various divergent angle of duct. The results of the present study are as follows. Temperature in the recirculation zone decreases with increasing divergent angle. The blow-off velocity in parallel duct is higher than that in divergent duct. Critical blow-off velocity is expected to be about 8-12 degree through blow-off velocity pattern. Regardless of divergent angles, the length of recirculation zone is nearly constant, and this length becomes longer with rod diameter. Pressure gradient has an effect on the eddy structure in shear layer behind the rod. With the increase of divergent angle, large scale eddies by dissipated energy in shear layer are split into small scale eddies, and the flame becomes a typical distributedreacting flame.

  • This paper is the study for the directional solidification of the ingot through the thermal analysis simulation and structural change of casting furnace. The activation analysis of metal impurities were also detected the total number of 10 different metals, but the concentration distribution showed no significant positional deviations in the same position from the top to the bottom. With the results of thermal analysis simulation, the silicon as a whole has reached the melting temperature as the retention time 80 min. The best cooling conditions showed at the upper cooling temperature $1,400^{\circ}C$ and cooling time 60min. The fabricated wafers showed the superior etching result at the grain boundary than that of existing commercial wafers.

Characterisation of Tensile Deformation through Infrared Imaging Technique

  • It is well known that during tensile testing, a part of the mechanical work done on the specimen is transformed into heat energy. However, the ultimate temperature rise and the rate of temperature rise is related to the nature of the material, conditions of the test and also to the deformation behaviour of the material during loading. The recent advances in infrared sensors and image/data processing techniques enable observation and quantitative analysis of the heat energy dissipated during such tensile tests. In this study, infrared imaging technique has been used to characterise the tensile deformation in AISI type 316 nuclear grade stainless steel. Apart from identifying the different stages during tensile deformation, the technique provided an accurate full-field temperature image by which the point and time of strain localization could be identified. The technique makes it possible to visualise the region of deformation and failure and also predict the exact region of fracture in advance. The effect of thermal gradients on plastic flow in the case of interrupted straining revealed that the interruption of strain and restraining at a lower strain rate not only delays the growth of the temperature gradient, but the temperature rise per unit strain decreases. The technique is a potential NDE tool that can be used for on-line detection of thermal gradients developed during extrusion and metal forming process which can be used for ensuring uniform distribution of plastic strain.

  • Distribution coefficients between cation exchange resin (Dowex 50W$\times$8) and $\alpha$-hydroxyisobutric acid ($\alpha$-HIBA) are measured in order to separate traces of Sm, Eu, Gd and Dy from nuclear material. Individual separations are performed by pH gradient technique with 0.40M $\alpha$-HIBA from 3.40 to 3.60 in cation exchange resin after a group separation. Each of separated elements is determined with a fluorometric method except Gd by colorimetry. The results are applied to analyze Sm, Eu, Gd and Dy in magnesium diuranate (yellow cake).

Analysis of Oxygen Combustion Characteristics of a Low Grade Coal Using IEA-CFBC Model (IEA-CFBC 모델을 이용한 저급탄의 순산소 연소 특성 분석)

  • The application of an oxy-combustion circulating fluidized bed combustor (Oxy-CFBC) for low grade coals has recently developed in the world to meet the continuous increase of energy demand and to achieve the reduction of greenhouse gases. Since demo plants for Oxy-CFBC have been developed, the combustion properties of Oxy-CFBC in various operation conditions, such as gas flow rates, combustion temperature, fuel, and so on, should be investigated to develop design criteria for a commercial Oxy-CFBC. In this study, a computational simulation tool for Oxy-CFBC was developed on the basis of the IEA-CFBC (International Energy Agency Circulating Fluidized Bed Combustor) model. Simulation was performed under various conditions such as reaction temperature ($800^{\circ}C{\sim}900^{\circ}C$), oxygen contents (21%~41%), coal feeding rate, Ca/S mole ratio (1.5~4.0), and so on. Simulation results show that the combustion furnace temperature is higher in oxy 1 than air fired. However, the temperature gradient tended to decrease with increasing oxy mixing percent. In case of $SO_x$, the higher the Ca/S mole ratio and oxy mixing percent, the higher the desulfurization efficiency.

The Contact Metamorphism Due to the Intrusion of the Ogcheon and Boeun granites (옥천화강암과 보은화강암 관입에 의한 접촉변성작용)

  • In the metapelites around the Ogcheon granite, the metamorphic grade increases from the biotite zone through the andalusite zone to the sillimanite zone towards the intrusion contact. In the metabasites around the Boeun granite, the metamorphic grade increases from transitional zone between the greenchist and amphibolite facies through the amphibolite facies to the upper amphibolite facies towards the intrusion contact. In the Doiri area locating near the intrusion contact of the Boeun granite, sillimanite- and andalusite-bearing metapelites are found with in 500 m away from the contact. The evidence described above indicates that the Ogcheon and Boeun granites caused low-P/T type contact metamorphism to the country rocks. The P-T condition of contact metamorphism due to the intrusion of the Ogcheon granite is $540{\pm}40^{circ}C, 2.8{\pm}0.9$ kb. The temperature condition of contact metamorphism due to the intrusion of the Boeun granite is $698{\pm}28^{\circ}C$. The wide compositional range of amphibole and plagioclase in the metabasites around the Boeun granite is due to the immisibility gab of amphibole and plagioclase and unstable relict composition resulted from an incomplete metamorphic reaction. The compositional range of stable amphibole and plagioclase decreases as a metamorphic grade increases due to a close of immiscibility gab. The thermal effect of contact metamorphism due to the intrusion of the Ogcheon and Boeun granites, are calculated using the CONTACT2 program based on a two dimensional finite difference method. In order to estimate the thermal effect of an introduced pluton, a circle with 10 km diameter and a triangle with 20 km side are used for the intrusion geometries of the Ogcheon granite and the Boeun granite, respectively. The results from the field and modeling studies suggest that the intrusion temperatures of the Ogcheon granite close to $800^{\circ}C$ and the intrusion temperature of the Boeun granite is higher than $1000^{\circ}C$. However, the intrusion temperatures can be lower than the suggested temperature, if the geothermal gradient prior to the intrusion of the Ogcheon and Boeun granites was higher than the normal continental grothermal gradient.

Surgical treatment of Supravalvular Aortic Stenosis (판막상부 대동맥협착증의 외과적 요법)

  • Background: Supravalvular aortic stenosis is a rare form of congenital cardiac anomaly involving ascending aorta distal to coronary orifice. Materials and methods: We operated 12 cases of supravalvular aortic stenosis between July 1986 and March 1997. Age ranged from 4 to 17(mean 10.2) years and 11 of them were male. Nine patients had clinical features of Williams syndrome. We experienced two types of supravalvular aortic stenosis, including 10 hour glass type and 2 diffuse type. Results: Preoperative transaortic pressure gradient ranged from 40 to 180(mean 92) mmHg by cardiac catheterization. Pulmonary stenosis was associated in 5 and 2 of them required angioplasty. Operative techniques included 6 standard aortoplasty with elliptical patch, 4 extended aortoplasty with inverted Y shaped patch, and 2 modified Brom's repair. There were no operative deaths. Postoperative echocardiographic evaluation was done at a mean interval of 12 months. Grade I or II aortic regurgitation was found in 3 cases. Postoperative cardiac catheterization revealed a mean transaortic pressure gradient of 26 (range 0 to 75) mmHg. A mean pressure drop was 78(range 30 to 114) mmHg. All patients were followed up for a mean of 40(range 1 to 67) months with uneventful clinical course. Conclusions: Our data proved the low mortality and excellent hemodynamic improvement after surgical relief of supravalvular aortic stenosis in children.

The Effects of Tunnel Geometrical Characteristics and Canopy Installation on the Ventilation and Fire Propagation (터널의 기하학적 형태 및 캐노피 설치가 터널 환기 및 화재 확산에 미치는 영향 분석)

  • Understanding the airflow characteristics within the canopy structure installed between closely adjacent tunnels either for light adaptation or for protection from snow hazards is required for the normal ventilation as well as safety system design. Grade, horizontal alignment, cross-sectional area and shape are known to substantially influence the fire smoke behavior and their influences raise great concern for the safety design. This paper aims at studying the effects of tunnel geometrical characteristics and canopy installation on the ventilation and fire propagation through CFD analysis. In the case of 145m long canopy, 50% opening ratio is preferred with respect to the airflow pattern and ventilation efficiency. When a 20MW fire occurs in a 1.8km-long tunnel and four 1250mm reversible jet fans are instantly turned on, smoke concentration at 40m downstream of the fire decrease 13% for the upgrade tunnel with 2% gradient and increases 20% for -2% gradient, compared to the standard horizontal tunnel. Backlayering is observed within 45m-long segment toward the entrance in 2% down-graded tunnel. In a rectangular tunnel, there is no significant difference of smoke concentration as well as velocity profile from the standard crown tunnel. Three-laned tunnel shows lower level of both profiles and backlayering is detected up to 50m upstream of the fire, while the risky situation rapidly disappears thereafter.
