  • This study carried out phylogenetic analysis of dinoflagellate Gonyaulax polygramma which was responsible for a harmful algal bloom episode in Korea in 2004. Molecular phylogenetic tree inferred from the partial LSU rDNA data showed that G. polygramma came up among the monophyletic Gonyaulax clade, but did not have apparent genetic affiliation to other Gonyaulax species. This result appears to be consistent with characteristic morphological features of G. polygramma such as epitheca sharply tapering to the apex and thecal plates ornamented with numerous longitudinal striations.

  • The red tide that occurred in Jinhae Bay in the middle of July, 1977 was caused by the microscopic dinoflagellate, Gonyaulax sp. , which is very similar to G. catenella. The cell is round, dorsoventrally flat and transdiameter is about $40\;\mu$. Many cells are chained. Dark brown, band and streak-like patches were formed in the surface water. A great deal of jelly fishes, Chrysaora quinquecirrha and Aurelia aurita, appeared in the red tide area and its surrounding waters. The number of cells was high, being $5.0\times10^5\;cells/\iota$ and among them Gonyaulax sp., predominant species, was $84.8\%$ in maximum. Transparency was less than 1 m in patches. In the surface water, temperature was $25.0^{\circ}C$, chlorinity $17.8%_{\circ}$ and dissolved oxygen $8.11m\iota/\iota\;(180\%)$ during daytime. Red tide stayed for a week. Some oysters were found damaged after the extinction of the red tide but their interrelationship was not found.

  • A study describing unrecorded and taxonomically undescribed indigenous species is in progress since 2006. Samples were collected from many sites in coastal waters and offshore of Korea as well as from Jeju Island. Since 2008, we have found ten unrecorded species of four genera belonging to family Dinophysaceae, Gonyaulacaceae, Gymnodiniaceae, and Heterocapsaceae. The species are as follows, Dinophysis elongata (2016 winter), D. nasuta (2016 winter), Gonyaulax alaskensis (2016 winter), G. diegensis (2017), G. monospina (2008), Amphidinium flagellans(2017), Heterocapsa circularisquama (2017), H. horiguchii (2017), H. lanceolata (2017), and H. pygmaea (2017) (note; The numbers in the parenthesis refer to the year in which the species was reported as unrecorded indigenous species by National Institute of Biological Resources, NIBR hereafter). Among them, seven species were described as newly recorded species in Korean waters, and three have been re-described in this study.

  • The dinoflagellates have some primitive nuclear features and are evolutionarily intermediate between prokaryotes and eukaryotes. The small subunit ribosomal RAN gene, the 5.8S ribosomal RNA gene, and the internal transcribed spacer (ITS) of Gonyaulax polyedra were cloned, and their sequences were analyzed to better understand their evolutionary position. The small subunit ribosomal RNA gene was 1,794 nt long, the large subunit ribosomal RNA gene was approximately 3,500 nt long, and the 5.8S ribosomal RNA gene was 159 nt long. The first internal transcribed spacer (ITS1) was 191 nt long, and the second internal transcribed spacer (ITS2) was 185 nt long. The intergenic spacer of the ribosomal RNA gene (IGS) was about 2,200 nt long, indicating that 5,800 nt of transcribed sequences were separated by roughly 2,200 nt of intergenic spacer. The ribosomal RNA genes were repeated many times and arranged in a head-to-tail, tandemly repeated manner. The repeating unit of ribosomal RNA gene of G. polyedra was proposed to be 8,000 nt long. Based on the lengths of ribosomal RNA, sequence alignments with representative organisms, and phylogenetic analysis on ribosomal RNA, G. polyedra appears to be one of the alveolates branched from the eukaryotic crown and, among dinoflagellates, it seems to not have emerged early.

  • The aim of this study is to determine the outbreaks of nontoxic Gonyaulax polygramma Stein in Yeosu waters in place of harmful Cochlodinium polykrikoides Margalef, which has occurred annually in the same region since 1995. The observation of cellular arrangement and structure by electron microscopy showed that G. polygramma isolated from Yeosu waters had a few spines connecting with membranes and prominent longitudinal ridges on the cell surface, with a cingular displacement 1.5 times their cell width. Furthermore, the location of the nucleus was posterior of large oval formation according to electron microscopy. On 6 August, 2004, the first bloom of G. polygramma occurred, the date of its disappearance was with a maximum cell density of 8,000 cells $ml^{-1}$ on 21 August, 2004. During the period of this study, the horizontal distribution of sea water temperature and salinity showed a strong coastal front, whereas the front of DIN (Dissolved Inorganic Nitrogen) was significantly different between the occurrence and disappearance of G. polygramma blooms. These results suggested that the process of the breakdown of stratification by wind and a low level of inorganic nitrogen play important roles in the rapid growth of G. polygramma, which is associated with a greater robustness in growth against DIN than that of C. polykrikoides in nature.

  • This study was carried out to determine the characteristics of the marine environment, including nutritional content, in order to clearly understand the outbreaks of Gonyaulax polygramma in the southern coastal areas in August, 2009. Samples were collected at 13 sites and water temperature and salinity were measured using a CTD. Field surveys were twice: the first between August 5-7, the second between August 22-24. The cell density of G. polygramma was 6,500-10,000 cells $ml^{-1}$ during the $1^{st}$ survey, whereas during the $2^{nd}$ survey the range of the cells was recorded from 8,000 to 12,500 cells $ml^{-1}$. Cochlodinium polykrikoides ranged from 0 to 105 cells $l^{-1}$ during the field survey. In water environments, the majority stations during the $1^{st}$ survey showed a nearly homogeneous water column below $1^{\circ}C$ in temperature, as well as similar profiles of salinity. However, the stratification between the surface and bottom was observed in the $2^{nd}$ survey. Regarding nutrients, Dissolved Inorganic Nitrogen (DIN) in the surface ranged from 0.144 to 0.236 mg $l^{-1}$ during the $1^{st}$ survey, and 0.082-0.228 mg $l^{-1}$ during the $2^{nd}$ survey. DIP (Dissolved Inorganic Phosphorus) did not show any difference in concentration between the $1^{st}$ and $2^{nd}$ survey. During August of 2009, the wind speed in the southern waters remained at around ${\leq}2\;m\;s^{-1}$ for about 60% of time, and there was very little precipitation during the month. Irradiance of ${\geq}10$ hr was shown in the late of August. It is thought that a low level of DIN and salinity play an important role as an essential factor for rapid growth, wide distribution and longer duration of red tide in G. polygramma.

  • In this study, we reviewed a group of marine biotoxins, namely yessotoxins (YTXs), focusing on their causative organisms, contaminated shellfish, domestic and foreign management status, and analytical methods. Although YTXs have not yet been reported in any cases of seafood contamination in South Korea, it is necessary to implement preemptive measures through continuous monitoring because there is a potential risk, due to the introduction of toxic microalgae associated with climate changes and the introduction of contaminated seafood from various countries. YTXs are produced by dinoflagellates, such as Protecratium reticulatum, Gonyaulax polygramma, Gonyaulax spinifera, and Lingulodinium polyedrum, all of which are species found along Korea's coastal areas. Analysis of YTXs in shellfish samples is mainly performed by use of LC-MS/MS after methanol extraction and SPE cartridge clean-up (HLB or strata-X). In the case of lipophilic marine biotoxins, including YTXs, pectenotoxins, and azaspiracids, the extraction and purification procedures are similar. Thus, it seems that the simultaneous analysis of several lipophilic marine biotoxins in shellfish samples is possible, and optimization is necessary. In addition, continuous monitoring studies on causative marine microalgae for YTXs in Korean coastal waters and contaminations in domestic and imported seafood are needed.

  • This study deals with the marine dinoflagellates observed in 50 samples collected in the vincinity of Yeosu from August 1979 to May 1980. Forty-nine species were identified and these taxa are attributed to 10 genera, of which 2 are infraspecific taxa. Two families, 3 genera, and 32 species are new records for the Korean coastal waters. Small dinoflagellates recorded in this study are ascribed to the use of volumetric samplers instead of nets. In this area, the armored forms are the most important components of the dinoflagellates (39 taxa). All species are illustrated by photomicroscopy. There are 49 pictures of taxa. An attempt has been made to provide more correct references to each species. There are comparative descriptive information, taxonomic comments, and distributional data for each species.
