Reproductive Cycle of the Ark Shell, Scapharca subcrenata, on the West Coast of Korea

  • Monthly changes in the gonad index (GI), egg-diameter composition, gonadal development, reproductive cycle of the ark shell, Scapharca subcrenata, were investigated by histological method and morphometric data. This species is dioecious and oviparous. The gonad is located among the subregion of mid-intestinal gland, digestive diverticula and the outer fibromuscular layers compacted by the fibrous connective tissues and muscle fibers. The gonad index sharply increased in May, reached the maximum value in June, and then gradually decreased from July to December. The reproductive cycle of this species can be divided into six successive stages: early active stage (January to May), late active stage (June to July), ripe stage (June to August), partially spawned stage (July to September), degenerative stage (August to December), and resting stage (January to April). S. subcrenata spawns once a year between July and early September, and the main spawning occurred between July and August when the water temperatures were above $20^{\circ}C$. This evidence suggest that timings of maturation and spawning are closely related to water temperatures. Even though the spawning period was once a year, it is assumed that the number of spawning frequencies (broods) may occur more than twice during the spawning season.

Gonadal Maturation, RNA/DNA Ratio of Mature Eggs, and the Effect of Refrigeration on Egg Viability and Sperm Motility in Korean Walleye pollock Theragra chalcogramma (동해안 북부 자망에서 어획된 명태(Theragra chalcogramma)의 생식소 성숙과 포획 후 경과시간에 따른 성숙란의 RNA/DNA ratio 및 냉장보관 정자의 활력)

  • We conducted a study to 1) determine the indicators of gonadal maturity in male and female Korean walleye pollock Theragra chalcogramma for the purposes of artificial insemination; 2) establish the RNA/DNA ratio of mature eggs in this species; and 3) monitor the effect of refrigerated storage on egg viability and the motility of sperm collected from dead adult males. During the spawning season, the color of female gonads changed from orange to transparent, and that of male gonads changed from pale orange to milky white. The DNA content and RNA/DNA ratio of mature eggs were maintained without significant changes for approximately 6 h when eggs were preserved at $4^{\circ}C$. Sperm could be obtained from both milt and undiluted semen. Sperm obtained from milt ceased moving on the second day after isolation, while over 60% of sperm obtained from semen showed movement until the 13th day. Seven attempts were made to artificially inseminate mature eggs, of which two resulted in successful fertilization. The successful inseminations produced 94,000 and 5,000 fertilized eggs, respectively. This study shows that artificial insemination of walleye pollock is a viable strategy when natural propagation is not possible.

Isolation of cDNAs for Gonadotropin-II of Flounder (Paralichthys olivaceus) and Its Expressions in Adult Tissues

  • Gonadotropin (GTH) is a pituitary glycoprotein hormone that regulates gonadal development in vertebrates. In teleosts, two types of gonadotropins, GTH-I and GTH-II, are produced in the pituitary, and they comprised of common ${\alpha}$ and distinct ${\beta}$ subunits. In the present study, the cDNAs encoding GTH ${\alpha}\;and\;GTH-II{\beta}$ subunits were cloned and sequenced from flounder (Paralichthys olivaceus) pituitary cDNA library. The nucleotide sequence of the a subunit was 619 bp long, encoding 124 amino acids, and that of the $GTH-II{\beta}$ subunit was 538 bp long, encoding 145 amino acids. GTH subunits had well conserved cysteines, when aligned with other members of the glycoprotein family. The ${\beta}$ subunit of gonadotropin II ($GTH-II{\beta}$) had a different N-linked glycosylation site. RT-PCR analysis showed an increase of GTH II mRNA levels in association with gonadal development, and also showed that the mRNA expression of the ${\alpha}$ subunit was detected only in tissues from pituitary glands.

Gametogenesis and Reproductive Cycle of the Rock Shell, Reishia (Thais) clavigera (Neogastropoda: Muricidae), on the West Coast of Korea

  • Gonadal development, gametogenesis, reproductive cycle, and first sexual maturity of Reishia clavigera were investigated monthly from July 1998 to June 1999 through cytological and histological observations. R. clavigera had separate sexes, and was an internal fertilizer. The ma1e penis was located near the two tentacles. The ovary and testis were composed of a great number of oogenic lobules and spermatogenic tubules, respectively. The size of ripe oocyte ranged from 130 to 140 ${\mu}$m in diameter. The peripheral cytoplasm of the germinal vesicle of the ripe oocyte in many cases were surrounded by smaller yolk granules, while the eccentric cytoplasm was occupied with larger ones. The reproductive cycle of R. clavigera could be classified into five successive stages: early active, late active, ripe, spawning, and recovery. Spawning of females occurred from early July to August when the seawater reached above 24.8$^{\circ}C$. Spawning of males occurred from early June to August in the water above 22.8$^{\circ}C$. Minimum size for sexual maturity of both sexes was above 10.0 mm in shell height. Each egg capsule was a cylinder or spindle in shape, 4-6 mm in length and 1-2 mm in width. Colors of newly spawned egg capsules showed yellowish white or pale yellow, while those with veliger larvae showed pale black, and released larvae or dead egg capsules showed black violet. The fecundity in an egg capsule ranged from 70 to 91 eggs (mean=80.28 eggs).

Cytologic Features of Metastatic Retroperitoneal Seminoma - A Case Report - (후복막강에 전이한 정상피종의 세포학적 소견 - 1예 보고 -)

  • A fine needle aspiration biopsy specimen of a retroperitoneal mass in a 26-year-old man who had had an orchiectomy for seminoma was submitted for cytologic evaluation. Cytologic features of the specimen included uniform neoplastic cells found singly or in groups of several cells intermingled with lymphocytes in a characteristic foamy, lacelike background. These cells varied from 10 to 20 m in diameter. The nuclei were round to ovoid with fine or reticular chromatin and one or more prominent nucleoli. The poorly defined cytoplasm stained pale-blue or blue with cytoplasmic vacuoles. The cytologic appearance was consistent with seminoma. Documented reports of the cytological appearance of seminoma are rare. The diagnosis of primary gonadal seminoma by fine needle aspiration biopsy is probably not indicated since the treatment of a primary gonadal tumor, regardless of its histogenesis, requires surgical resection. However, fine needle aspiration biopsy is extremely valuable in the diagnosis of extragonadal as well as metastic and recurrent seminoma.

Relationships among Gonad Weight, Liver Weight and Body Weight of Major, Common and Some Chinese Carps under Composite Culture System with Special Reference to Pond Fertilization

  • The relationship of gonad weight and liver weight with body weight of six fish species viz; Catla catla, Labeo rohita, Cirrhina mrigala, Hypophthlamichthys molitrix, Ctenopharyngodon idella and Cyprinus carpio under the influence of artificial feed, broiler manure, buffalo manure, N:P:K (25:25:0) and a control pond were examined after a rearing period of one year. The positive relationship between gonad weight and body weight was significant which showed the dependence of gonadal development on body weight in all the six fish species. The currelation coefficients were higher in female fishes. However, the major carps had a much smaller proportional gonadal weight as compared with Chinese carps and a common carp due to their faster growth rate. The overall comparison of six fish species under different experimental treatments revealed that highest liver weight was observed for C. idella closely followed by C. carpio. The maximum correlation value was observed with H. molitrix under the broiler manure. The maximum Gonadosomatic Index (GSI) remained as 32.63 for C. carpio followed by C. idella. The maximum value for Hepatosomatic Index (HSI). remained 1.99 for C. idella followed by C. mrigala.

Histological Analysis of Early Gonadal Development and Sex Differentiation in Chameleon Goby, Tridentiger trigonocephalus

  • This study describes the developmental process of gonads in chameleon goby, Tridentiger trigonocephalus from the stage of hatching to 100 days after hatching (DAH). Based on histological observation, the primordial germ cells were observed in mesentery between mesonephric duct and gut at 15 DAH (total length, TL: $6.8{\pm}0.2mm$). At 20 DAH (TL: $7.9{\pm}0.1mm$), the primordial gonad began to protrude into peritoneal cavity and developed between mesonephric duct and gut. Initial ovarian differentiation was identified by the presence of ovarian cavity and oogonia in the gonads at 55 DAH (TL: $21.1{\pm}1.3mm$). Testicular differentiation started at 65 DAH (TL: $23.7{\pm}0.9mm$) with appearance of spermatogonial cells in the gonads. These findings indicate that sex differentiation in T. trigonocephalus occurs earlier in females than males, suggesting that this species can be classified as an undifferentiated gonochorist.

High Transmission Rate of Germline Chimerism Using Cultured Primordial Germ Cells in Chickens.

  • Although primordial germ cells(PGCs) have been used in the production of germline chimera, efficiency has not been satisfactory. The Present study was conducted to improve efficiency of germline chimera production using the cultured gonadal PGCs(gPGCs). Germline chimeric chickens were produced by transfer of cultured gonadal primordial germ cells from Korean Ogol Chicken (KOC) to White Leghorn (5.5-day-old) and cultured in vitro for 10 days. Approximately 200 gPGCs (2-day-old) recipient embryos from which blood had been withdrawn via the dorsal aorta prior to the injection. Recipient embryos were incubated until hatching. Germline chimerism of the chickens reaching maturity was examined by mating them with Korean Ogol Chicken. Donor-derived offspring were identified as germline chimeric chickens based on their feather color. The frequency of germline transmission of donor PGCs ranged 1.9∼60.7%. There was no difference between both sexes. Therefore, it can be concluded that efficiency of germline chimerism can be improved via using cultured gPGCs.

  • 꼬막, Tegillarca granosa은 우리나라 남해 및 서해안 일대에서 서식하는 중요한 수산자원 중의 하나이지만, 이에 대한 생물학적 기초연구는 거의 없는 실정이다. 따라서 1995년 2월부터 1996년 1월까지 전남 벌교연안에서 채집한 꼬막의 생식소발달, 생식세포형성과정 및 생식주기를 조사하였다. 1. 꼬막은 자웅이체이면서 난생이고, 생식소는 내장낭의 간중장선을 싸고 있는 결합조직으로부터 족부의 근육층까지 분포한다.2. 미분화간층직과 호산성 과립세포들이 초기 활성기의 소낭에 풍부하게 나타나기 시작하여 완숙기에는 거의 없어지는 것으로 보아, 이들은 생식소 및 생식세포형성과 발달에 영향을 공급하는 영향세포로 생각된다.3. 생식소발달, 생식세포형성과정, 조직분화과정 및 세포학적 특성에 따라, 이들의 생식주기를 초기 활성기, 완숙기, 부분 방출기, 방출 및 비활성기 등으로 구분할 수 있다. 4. 방란 및 방정은 수온 2$0^{\circ}C$정도되는 6월하순부터 시작되고, 주산란시기는 수온이 23-24$^{\circ}C$정도 되는 7-8월이며, 완숙란의 크기는 50-60$\mu\textrm{m}$이다.5. 비활성기는 9월에서 이듬해 4월까지 지속되는 비교적 긴 기간이며, 초기 활성기도 비교적 길어 1월에서 5월까지 지속되는 반면, 후기활성, 완숙 및 방출기는 비교적 짧아, 5월에서 8월까지 모두 완료된다.6. 비만도의 월별 변화는 생식소의 발달, 생식주기 및 수온과 밀접한 관계가 있다.7. 사용된 재료 433개체 중 1개체가 자웅동체현상을 나타내었다.

Effect of thymectomy on the female reproductive cycle in neonatal guinea pigs

  • Objective: The appropriate function of the hypothalamic-pituitary-gonadal axis is essential for maintaining proper reproductive function. In female mammals, the hypothalamic-pituitary-gonadal axis regulates reproductive changes that take place in the estrus cycle and are necessary for successful reproduction. This study was conducted to investigate the effect of thymectomy on the estrus cycle in neonatally thymectomized guinea pigs. Methods: In this study, 12 female guinea pigs, six thymectomized and six sham-operated, were studied. The effects of neonatal thymectomy at 5-7 days of age on parameters of the reproductive axis were examined in female guinea pigs. Gonadotropin and 17β-estradiol levels were assessed at regular intervals (days 0, 3, 6, 9, 12, and 15) of the estrus cycle, and the time of vaginal opening in the thymectomized and shamoperated guinea pigs was determined. Results: Significant reductions in gonadotropins and 17β-estradiol levels during estrus cycle were found in neonatally thymectomized female guinea pigs compared to sham-operated guinea pigs. Conclusion: The results of this study underscore the importance of the thymus in the neonatal period for normal female reproductive function.
